Abstract

Abstract

Abstract

Bacillus sp. is well known for its functional capabilities such as solubilizing phosphorus (P) and potassium (K), and fixing nitrogen (N2). These bacteria can form endospores under stressed conditions, allowing long-term survival and application in biotechnological fields. This study aims to isolate Bacillus sp. capable of forming endospores and evaluates their viability on different carriers, specifically biochar and oil spill dispersant (OSD), to enhance biodegradation in contaminated environments. Soil samples from the bamboo rhizosphere were heat-shocked to isolate endospore-forming Bacillus strains, with the isolate identified as Bacillus pseudomycoides RAY21 through 16S rRNA sequencing. This strain exhibited Gram-positive characteristics, formed endospores, and demonstrated potential on various media such as Pikovskaya, Alexandrov, and N-Free Mannitol. The physiological characterization indicated optimal growth in a pH range of 6-8, salinity up to 3.5%, and thermophilic properties. Endospores from B. pseudomycoides RAY21 and B. subtilis CYA27 were tested on biochar and OSD as carriers. The results showed that endospores adhered better to biochar, but their viability was more stable in OSD over time. Notably, B. pseudomycoides RAY21 on OSD degraded 23.43% of total petroleum hydrocarbons (TPH), outperforming B. subtilis CYA27 (21.62%). In conclusion, the study demonstrates the potential of using Bacillus endospores on OSD as an effective carrier for bioremediation, particularly in degrading petroleum hydrocarbons. Future research should focus on optimizing carrier materials and exploring field-scale applications for enhanced environmental cleanup.

Abstract

Bitter leaf (Vernonia amygdalina Del.) hold significant potential for development in the pharmaceutical industry due to its high content of bioactive compounds, particularly flavonoids, which serve as a source of anti-inflammatory agents. A major challenge in processing bitter leaf into dried simplicia is the selection of an appropriate drying technique, as the drying process can alter the active compounds present in the plant. This study aimed to analyze the drying rate of bitter leaf and evaluate the most effective drying technique for preserving their anti-inflammatory properties. The drying process was conducted using an oven dryer at 40°C and 50°C, as well as a greenhouse dryer, and the reduction in moisture content over time was recorded. The dried simplicia was subsequently subjected to phytochemical screening and anti-inflammatory activity testing. The results indicated that the drying durations required to achieve a final moisture content of <10% were 44 hours at 40°C, 20 hours at 50°C, and 9 hours using the greenhouse drying method. Phytochemical analysis confirmed the presence of saponins, alkaloids, flavonoids, tannins, and steroids in the bitter leaf extract. The IC₅₀ values for the three drying methods were determined to be 25.33 ppm, 61.05 ppm, and 114.25 ppm, respectively. Among the drying methods tested, oven drying at 40°C yielded the highest anti-inflammatory activity, with an IC₅₀ value of 25.33 ppm. The regression equation obtained was y = 0.072x + 48.176, with an R² value of 0.9899, indicating a strong correlation between extract concentration and inflammation inhibition. This suggests the potential for reliable prediction of optimal dosages for pharmacological applications.

Abstract

The increasing number of fire incidents at oil and gas facilities in recent years, along with the results of fire risk assessments at the Onshore Receiving Facility (ORF) at PT XYZ, which is still ranked as medium, necessitates efforts to reduce the risk level to low. One such effort involves evaluating and developing the fire water reservoir at the ORF facility to ensure its adequacy according to standards. This research aims to enhance operational safety and environmental protection, considering the high fire risk at gas transmission facilities. The research methods include identifying fire-fighting facilities, calculating water requirements, and evaluating the capacity of existing reservoirs. Based on the analysis, a new reservoir was designed with estimated costs, locations, and alternatives to meet the fire-fighting water needs at the ORF. The fire-fighting facility at the ORF of PT XYZ, located in a densely populated area, has a water capacity of 16 m³, but the ideal requirement is 43 m³. It is recommended to build a 30 m³ ground tank or explore other alternatives, such as panel tanks, sea water, or PLN hydrant networks, to improve readiness for major fires and protect public safety around the ORF. The level of fire risk at the ORF of PT XYZ is categorized as medium (yellow). Therefore, the solution could involve building a new reservoir or a panel tank that is more resistant to the risk of cracks or leaks due to land subsidence or shifting.

Abstract

Granulated coconut sugar is a powdered coconut sugar manufactured by Purworejo farmers from the sap of the coconut. Farmers' usage of simple technologies is the primary cause of environmental problems due to poor efficiency levels and uncontrolled emission emissions. The objectives of this research were to identify the stages of the granulated coconut sugar production farmer level process, from coconut sap to farmers' granulated coconut sugar, and then analyze the total environmental impact of granulated coconut sugar production farmer level process using the Life Cycle Assessment (LCA) approach and the CML-IA Baseline calculation method. The study's scope included an inventory of "gate-to-gate" inputs and outputs, computation of each stage of the process from sap heating to granulated coconut sugar, and interpretation of the calculation results. The amount of impact created per 1 kg of farmer-produced coconut sugar was employed as the unit of function in the analysis of the consequent environmental impact. SimaPro 9.4.0.2 software was used to calculate environmental impact analysis. The GWP impact of manufacturing 1 kilogram of farmer granulated coconut sugar was 4.76E-02 kg CO2 eq, the AP impact was 9.45E-04 kg SO2 eq, and the EP impact was 2.77E-04 kg PO4 eq. The biggest GWP effect contribution indicator was the use of LPG, which was 2.89E-02 kg CO2 eq, while the largest AP and EP impact contribution indicator was the use of firewood, which was 8.16E-04 kg SO2 eq and 2.12E-04 kg PO4 eq. The improvement scenario of the granulated coconut sugar production farmer level process, namely substituting the use of firewood with rice husks, reduces 85% of the total AP impact and reduces 93% of the total EP impact.
