Abstract

Hyperuricemia and hypercholesterolemia’s cases are increasing. Noninvasive early detection method should be develope. Development of smart sanitary napkins based on paper chip biosensors using “Whatman” filter paper through screen printing and immobilized with pH 6 citrate buffer and iron (III)-actress- (1,10-phenanthroline) complex reagent for detection of uric acid and cholesterol oxidase enzymes and reagent 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS)) for cholesterol detection. The analysis of the paper chip biosensor has a response time of 10 minutes for uric acid detection and 25 minutes for cholesterol detection. Uric acid linearity 2 - 20 mg / dL and cholesterol 50 - 500 mg / dL. The LOD and LOQ detection areas for uric acid were 0.322 mg / dL and 0.965 mg / dL, respectively, while the cholesterol detection areas were 8.740 mg / dL and 26.221 mg / dL, respectively. NaCl interference interfering with the detection of uric acid and cholesterol was 5.746% and 6.388%, respectively and creatinine interference interfering with the detection of uric acid and cholesterol was 5.868% and 5.583%, respectively. The precision for uric acid detection was 1.252% (RSD <3.7%) and cholesterol detection was 0.059% (RSD <2.7%). The accuracy (% recovery) for uric acid detection was 100.333% and for cholesterol, the detection was 100.016%. Smart bandages can be used to analyze the initial conditions of hyperuricemia and hypercholesterolemia.

Abstract

Leukaemia is a type of heterogeneous malignancy disease in which the number of immature white blood cells in the bone marrow is increased or poorly differentiated. Inhibitors of HL-60 cell line are important drugs in the treatment of leukaemia. This study aims to identify potential inhibitors for HL-60 leukaemia cells using virtual screening with QSAR and molecular docking approaches. The best QSAR model obtained was the model with R2 testing value of 0.72. The best model was applied to the Enamine database, obtained 3,841,278 compounds with pIC50 above 9 were clustered to end up with 50 inhibitor candidate compounds. The compounds were further studied through molecular docking against CDK- 2 and BCL-2 target proteins with binding free energy analysis. Screened compounds have high inhibitory potential against CDK-2 and BCL-2 target proteins in HL-60 cells which can be proposed as potential inhibitors of HL-60 cells. These compounds can be further tested for the development of more effective antileukaemia drugs.

Abstract

The emergence of antibiotic resistance becomes a barrier in controlling infectious diseases, so it is necessary to find new alternative antibiotics. This study aimed to explore the potential of estuarine soil fungals secondary metabolites isolated from Katialada Village Gorontalo Location One and test their antibacterial activity against Staphylococcus aureus. The antibacterial parameter observed was the percent inhibition of ethyl acetate extracts against Staphylococcus aureus. The estuarine soil samples were cultured in PDA media producing five single fungal isolates differentiated based on color morphology, texture, and shape coded IS2-BTG-4.1.1, IS2-BTG-4.1.2, IS1-BTG-4.2, IS2-BTG4.3.1, and IS2-BTG-4.3.2. Those five samples were fermented using PDB media and the fermented products were extracted with ethyl acetate. The extracts were screened for its compound using a TLC plate and antibacterial testing with a single concentration of 100 g/mL in the microdilution method. The screening results showed that the five ethyl acetate extracts contained terpenoid compounds and code IS2-BTG-4.3.2 also had alkaloids. Based on the antibacterial test results, the five ethyl acetate extracts had growth inhibitory activity on the Staphylococcus aureus, with the percentage showed by IS2-BTG-4.1.1 50,2% ± 14,8%, IS2-BTG-4.1.2 35,6% ± 12,9%, IS1-BTG-4.2 13,0% ± 7,3%, IS2-BTG-4.3.1 13,6% ± 6,0%, and IS2-BTG-4.3.2 by 12,4% ± 9,2%.

Abstract

 Kepundung (Baccaurea racemosa Muell.Arg.) merupakan salah satu tanaman berkhasiat di Indonesia yang mengandung flavonoid, polifenol, tanin, dan terpenoid yang dapat berfungsi sebagai antioksidan. Kandungan senyawa dengan aktivitas antioksidan dapat dikaitkan dengan potensi aktivitas antidiabetes dari tanaman ini. Penelitian dilakukan untuk mengetahui aktivitas antioksidan dan antidiabetes ekstrak metanol, etanol, dan etil asetat dari daun kepundung. Ekstraksi dilakukan dengan metode ultrasonik. Pengujian aktivitas antioksidan dilakukan secara kuantitatif dengan metode DPPH, sedangkan pengujian aktivitas antidiabetes menggunakan metode penghambatan enzim α-amilase. Nilai IC50 pada penentuan aktivitas antioksidan dan antidiabetes ditentukan dengan menggunakan spektrofotometer UV-Vis pada panjang gelombang maksimum. Hasil penentuan antioksidan menunjukkan bahwa nilai IC50 ekstrak metanol, etanol, dan etil asetat masing-masing adalah 9,38 ± 0,15; 10,55 ± 0,09; dan 946,70 ± 2,31 ppm. Ini menunjukkan ekstrak metanol dan etanol memberikan hasil terbaik dalam aktivitas antioksidan. Nilai IC50 aktivitas antidiabetes in vitro ekstrak metanol, etanol, dan etil asetat masing-masing adalah 67,63 ± 0,36; 67,46 ± 0,23; dan 841,04 ± 1,52 ppm. Hasil ini juga menunjukkan bahwa ekstrak metanol dan etanol daun kepundung memiliki aktivitas antidiabetes yang hampir sama

Abstract

Penelitian ini bertujuan untuk mengkaji budaya kewirausahaan yang berkembang di kalangan pelaku Usaha Mikro, Kecil, dan Menengah (UMKM) di kawasan wisata Pantai Sayang Heulang, Kabupaten Garut. Fokus utama kajian ini adalah memahami nilai-nilai, sikap, dan praktik kewirausahaan yang diterapkan oleh masyarakat lokal dalam menjalankan usahanya di sektor pariwisata. Metode yang digunakan adalah pendekatan kualitatif dengan teknik observasi, wawancara mendalam, dan dokumentasi terhadap sejumlah pelaku UMKM. Hasil penelitian menunjukkan bahwa masyarakat memiliki semangat kewirausahaan yang tinggi, tercermin dari kemandirian usaha, kemampuan beradaptasi terhadap kebutuhan wisatawan, serta budaya gotong royong yang masih kuat. Namun demikian, pelaku usaha masih menghadapi berbagai kendala, seperti keterbatasan akses teknologi, pemasaran digital, dan ketergantungan pada musim wisata. Oleh karena itu, diperlukan dukungan berkelanjutan dari pemerintah dan lembaga terkait untuk memperkuat kapasitas wirausaha masyarakat lokal, melalui pelatihan, pendampingan, serta penguatan ekosistem usaha berbasis potensi lokal.

Abstract

ARTICLE HIGLIGHTS- The study identified 36 lichen species from 14 different families.- The lichen composition differed between stations, indicating disparities in air quality.- Station II (1600-1700 masl) had better air quality than Station I (1600-1700 masl), which had a higher diversity index value and more lichen coverage. - Nitrogen emissions from farming may reduce the non-nitrophilic lichen diversity and abundance.- The diversity and abundance of lichen is affected by air temperature, humidity, light intensity, and the type of bark.ABSTRACTThe Mount Merapi ecosystem is distinguished by its considerable biodiversity potential, which has led to its designation as a national park and the protection of its unique ecological characteristics. A notable example of the Mount Merapi ecosystem is the Gunung Bibi forest, which has been designated as a Sanctuary Zone within the Mount Merapi National Park, with the primary objectives of biodiversity conservation, habitat preservation, and ecosystem protection. However, the Gunung Bibi forest is susceptible to the repercussions of volcanic eruptions and the pressure of agricultural activities from the surrounding area, which may potentially impact its air quality. Given the geographical area of the forest, the most efficient method for air quality monitoring is the use of lichens diversity as bio-indicators. This study aims to analyze the relationship between lichen species diversity and air quality in the Gunung Bibi forest area of Mount Merapi National Park, and the factors that influence it. Data collection was carried out by dividing the area into two research stations based on altitude. Data collection was carried out using a purposive sampling method with analysis including lichen abundance, lichen thallus cover area, and Shannon-Wiener Diversity Index measurement. Based on the research conducted, the results of lichen species diversity found at the research site include 36 species from 13 different families. The lichen composition at the two stations differs, indicative of disparities in air quality. Station II (1600-1700 masl) exhibited indications of better air quality in comparison to Station I (1500-1600 masl), which was distinguished by a higher diversity index value, as well as a greater lichen thallus covering area. The difference in air quality between the two research stations is probably due to nitrogen emissions from agricultural activities, which limit the diversity and abundance of non-nitrophilic lichen species Environmental factors affecting lichen species diversity and abundance are air temperature, humidity, light intensity, and bark type of lichen substrate.

Abstract

Antioxidants can protect cells from free radical damage by stabilizing them. One of the compounds that has antioxidant activity is cinnamic acid. Cinnamic acid and its derivatives have several activities: antibacterial, anticancer, and antioxidant. However, the ability of cinnamic acid to capture free radicals is still relatively low. One of the efforts that can be made to increase the antioxidant activity of cinnamic acid is to modify its structure. Structure modification is an effort to improve the pharmacological activity of a compound through chemical synthesis reactions. The cinnamic acid structure can be modified by changing the carboxylic -OH group into an amine group through an N-atom acylation reaction. This study was conducted by reacting cinnamoyl chloride (1a), which is a cinnamic acid derivative with 1,2,3,4-tetrahydroisoquinoline (2b) which is a compound of isoquinoline group to produce (E)-1-(3,4-dihydroisoquinoline-2(1H)-yl)-3-phenylprop-2-en-1-one (3b) and then tested for antioxidant activity using DPPH method. The resulting product compound was yellow crystals with a yield of 81.56%. The antioxidant activity produced by the product is more significant than that of cinnamic acid compounds at the same concentration.

Abstract

Flavonoid has potential bioactivity as anticancer agents. The flavonoid of cultivated tobacco (Nicotiana tabacum), locally known as “Kasturi”, leaves was screened for its cytotoxicity against MCF-7 human breast cancer cells and non-transformed Vero cells (African normal cell kidney line) in different concentrations. This study aimed to examine the cytotoxic potential of the flavonoid of Kasturi tobacco leaves against MCF-7 human breast cancer cells. Flavonoid obtained from methanolic extracts of Kasturi tobacco leaves, which have been purified from nicotine. The flavonoid of Kasturi tobacco leaves with concentrations of 20 to 640 μg/mL were exposed to MCF-7 and Vero cells for 24 h. Cell viability was evaluated by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Flavonoid of Kasturi tobacco leaves with concentrations of 160 μg/mL decreased the MCF-7 cell viability more than 50%, with an inhibitory concentration 50 (IC50) value of 148.41 μg/mL. Meanwhile, it inhibited 50% of Vero cell viability at 255.35 μg/mL. The flavonoid of Kasturi tobacco leaves has cytotoxic activity on MCF-7 cells, and might be a potential alternative agent for human breast cancer therapy.Keywords: flavonoid, tobacco leaves, human breast cancer cells, anticancer activity

Abstract

Exposure to free radicals and oxidative stress are major factors in various degenerative diseases, such as cardiovascular disease, diabetes, cancer, and neurodegenerative diseases. Therefore, the use of natural antioxidant sources is important to reduce the negative effects of free radicals. This study compares the natural antioxidant potential of black rice, bajakah root, and kebiul seeds, whereas previous studies have only identified the antioxidant potential of a single natural ingredient. This study aims to measure the antioxidant potential, such as total polyphenols, total flavonoids, and antioxidant activity, of these three natural materials using distilled water, 70% ethanol, and 96% ethanol as solvents. The experimental research method involved testing antioxidant activity and measuring total phenolics and flavonoids. The results showed that the 70% ethanol extract of black rice had the highest total polyphenol content at 6.83±0.28 mgGAE/g (0.007±0.00%), the 70% ethanol extract of bajakah wood root had the highest flavonoid content at 8.77±0.02 mgQE/g (0.009±0.00%), and the distilled water extract of kebiul seeds showed the strongest antioxidant activity based on an IC50 value of 5.96 ± 0.01 ppm. This study concluded that black rice has potential as a primary source of phenolics and anthocyanins, while bajakah wood root and kebiul seeds are sources of flavonoids with strong antioxidant activity.

Abstract

Oxidative stress and inflammation are closely linked pathological processes. Although non-steroidal anti-inflammatory drugs (NSAIDs) are effective, their use is associated with gastrointestinal, cardiovascular, and renal side effects, highlighting the need for safer alternatives. Jamu, an Indonesian traditional medicine, is widely used, yet scientific evidence supporting its efficacy and safety remains limited. The objective of this study was to investigate the anti-inflammatory and antioxidant activities of four different Indonesian jamu brands available for purchase on e-commerce platforms in Indonesia, and examine their correlation with the total phenolics, flavonoids, and alkaloids content in those jamu. The anti-inflammatory activity of jamu was assessed in vitro by evaluating its ability to stabilize red blood cell membranes and inhibit protein denaturation, using diclofenac sodium as a positive control. The antioxidant activity was evaluated using the 1-diphenyl-2-picrylhydrazyl (DPPH) assay, with ascorbic acid as the standard antioxidant. Total phenolics, flavonoids, and alkaloids contents were determined spectrophotometrically and expressed as gallic acid equivalents (GAE), rutin equivalents (RE), and berberine equivalents (BE) per gram of jamu extract, respectively. The correlations were evaluated based on multiple regression and Pearson correlation analysis using 99% confidence level. The aqueous extracts of Jamu A, B, C, and D were able to stabilize red blood cell membranes, inhibit protein denaturation, and exhibit free radical scavenging activity. Among the extracts, Jamu B demonstrated the lowest IC50 value, indicating the most effective anti-inflammatory and antioxidant properties followed by Jamu A, Jamu D, and Jamu C. These activities were significantly correlated with their total phenolics, flavonoids, and alkaloids content (p < 0.01).
