Abstract

Gecko is one of the reptiles are used as traditional Chinese medicine which has been reported as antitumor capabilities. The purpose of this study was to determine the cytotoxic effect of rat serum that treated with gecko powder and its ability to inhibit proliferation and stimulate apoptosis of WiDr cells. Gecko powder  the treated in vivo in the rat (Rattus novergicus) SD female. Twelve mice treated with powder gecko at a dose of 9 g/kg body weight per day. Then the rat blood serum on the in vitro treated in WiDr cells. Cytotoxicity test using the MTT method. Observation of the proliferation kinetics using doubling time test. Then apoptosis test performed by the doublestaining method. Cytotoxicity assay results showed that the powder is toxic to the gecko cell WiDr. In the cytotoxicity test showed IC50rat serum with gecko powder body parts obtained at a concentration of 7,914%. The observation of cell proliferation kinetics demonstrated  rat serum with gecko powder body parts most effective in inhibiting the proliferation of WiDr cells Data apoptotic WiDr cells  showed the rat serum with gecko powder effective agency in promoting apoptosis. Rat serum with treat gecko powders bodies part are toxic and can inhibit proliferation and stimulate apoptosis of WiDr cells.

Abstract

Iridovirus was known as agents that caused serious systemic disease in freshwater and marine fishes. The mortality up to 100% of orange-spotted grouper (Epinephelus coioides) due to iridovirus infection has been reported in Indonesia. The gene encoding capsid protein of iridovirus is supposed to be conserved and has the potency for the development of control methods. The objectives of this study are to clone the gene encoding capsid protein iridovirus and to analyze their sequences. The   spleen tissues of orange-spotted grouper were collected and extracted their DNA. The DNA fragment of capsid protein of iridovirus genes were amplified by PCR using designed primers with the extraction DNA as templates. The amplified DNA fragments were cloned in pBSKSII and sequenced.  The genes encoding capsid protein of iridovirus from Jepara and Bali were successfully amplified and cloned. The Jepara clone (IJP03) contained complete open reading frame (ORF) of the gene composed by 1362 bp nucleotides which encoded 453 amino acids. Those Jepara and Bali (IGD01) clones shared 99.8% similarity in nucleotide level and 99.4% at amino acid level. Based on those sequences, Indonesian iridovirus was belonged to genus Megalocystivirus and shared 99,6-99,9% similarity on nucleotide level with DGIV, ISKNV, MCIV, and ALIV

Abstract

The availability of specific and rapid detection methods is essential for monitoring the health status of farmed species, particularly in viral disease as in this case early diagnosis is a critical factor in containing disease outbreaks. Jembrana Disease Virus (JDV) is a lentivirus that causes an acute, severe disease syndrome in infected Bali cattle in Indonesia, resulting in heavy economic losses because of the high mortalities. The virus-host interaction and the modes of transmission are still unknown. The goal of the research was to designa probe candidate of Jembrana Disease Virus based on envelope-transmembrane (env-tm) gene to optimize Jembrana disease detection method. The DNA fragment derived from env-tm of JDV was used, cloned in pGEX-TM and expressed in E.coli DH 5α. Sequence analysis was conducted with BLAST programs from NCBI. Sequence analyses of the fragments of env-tm clone, indicated that it has a very closed genetic relation with 97,68% homology identity. Probe was designed based on the conserved region of env-tm using Geneious resulted in JT2 252 bp long. BLAST analyses showed that probes had high specifity to other strains of JDV in Indonesia.Key words : probe, env-tm, JDV, specifity, sensitivity.

Abstract

Koi herpesvirus (KHV) which also known as Cyprinid herpesvirus 3 (CyHV-3), Koi herpes-like virus,and carp interstitial nephritis gill necrosis virus (CNGV), caused signifi cant morbidity and mortality in koiand common carp (Cyprinus carpio). The case fatality rate of this disease is 80–100%. Glycoprotein has beenused for vaccine development as sub unit vaccine against viruses. The aim of this research was to clone andexpress membrane glycoprotein ORF124 KHV as a candidate of recombinant vaccine. ORF124 KHV gene wassuccessfully cloned into pBSKS and sequenced. Result showed that ORF124 KHV (isolate from Indonesia) had100 % similarity with Cyprinid herpesvirus 3 strain TUMST1 (from Japan), 99% similarity with Koi herpesvirus strain KHV-U (from USA) and Koi herpesvirus strain KHV-I (from Israel). Prediction analysis of T and B cellepitopes showed that ORF124 KHV protein had 14 and 11 T cell epitopes (IAd, Rothbard/Taylor pattern),and had 10 B cell epitopes, suggested that the protein can be used as a vaccine candidate. ORF124 gene hasbeen expressed in Escherichia coli under pET32-a(+)vector.

Abstract

Sweet potatoes (Ipomoea batatas) are tuber that have high antioxidant activity. The determination of antioxidant activity based on as percentage of inhibition of free radical DPPH. The purpose of this research was to observe the activity of three local cultivars of sweet potatoes by using DPPH method and by using vitamine C as compparison. The measurement of antioxidant activity which performed on sweet potatoes juice and vitamine C with concentration 625 ppm, 1250 ppm, 2500 ppm, 5000 ppm and 10000 ppm by using free radical of DPPH 0,004% that was soluble in ethanol pa and then was measure by using spectrofotometer. Data were analyzed by using ANOVA and LSD (α = 0.05). The result of this research showed that the three cultivar of sweet potatoes juice have antioxidant activity and the purple sweet potatoes cultivar Biru Mangsi had the highest antioxidant activity, although lower than vitamine C. The highest of antioxidant activity were vitamine C, followed by purple sweet potatoe, yellow sweet potatoes (cultivar Kuning Madu) and white sweet potatoes (cultivar Kapasan).

Abstract

Pseudodiaptomus species are one of the copepods species as a superior live feed to date due to their nutrition and digestive enzyme contents. Some of them have been used as natural for rearing marine fish larvae. The purposes of this study were to compare the nutritional and protease activity between two species of Pseudodiaptomus originated from Indonesian waters, and to determine more superior species to cultivate. Two different feeds i.e. Thalassiosira sp. and milk powder were used to grow the Pseudodiaptomus species. Analysis of amino acids (AAs) and fatty acids (FAs) profiles were carried out for both the Pseudodiaptomus species samples and the feeds, while the protease activity assay was carried out only for the Pseudodiaptomus species samples. Results indicated that the nutritional and protease activity profiles of Pseudodiaptomus were affected by the types of feed. Pseudodiaptomus code P61 was more superior to Pseudodiaptomus code P71. This code P61 species contained a wide variety of essential fatty acids and exhibited stabile protease activity under the different feeding treatments. However, P61 contained a lower total AA content than P71. Both of them could be cultivated because they were complementary in nutrients to each other.

Abstract

The Community Service Activities for the Independent Campus Learning Program (PkM MBKM), which involves lecturers and students has been done. The program aims to empower the people of Padukuhan Malangrejo, Wedomartani, Ngemplak Sleman, regarding the use of natural dyes for healthy food products. The targets during this program were 14 Women Farmers Group (KWT) participants, 10 elementary school students and 10 teenagers. In this course, we did the socialization of the activities, increasing the community literacy about natural food coloring and its benefits for health, by using the learning modules for children, adolescents and members of Women Farmer Groups (KWT). The training was carried out on the production of natural dyes that can be stored, as well as the creation of colorful food and drink recipes. Evaluation of literacy activities is assessed from understanding the material at the beginning (pre-test) and end (post-test) of the activity. Food products with natural dyes were organoleptically tested in communities outside Malangrejo. Other activities include assistance in the production of BiTel pudding (beetroot and butterfly pea coloring) with limited marketing, obtaining a Hygiene and Sanitation Certificate (SLHS) from the Health Service, Sleman. Activities to plant natural food dyes are carried out in yards and less productive land. Planting methods were adjusted to the type of plant, both horizontally and vertically. The results of the activity showed an increase in community participation and understanding of the benefits of natural food coloring (16.4%), increased literacy in the youth group (4%), KWT participants (2.9%), while for elementary school students there were 40% of participants who got a score of 100 %. This activity also gave rise to Micro, Small and Medium Enterprises (MSMEs or UMKM) which produce food with natural dyes, as well as the enthusiasm of KWT Malangrejo members in using their home gardens to produce natural food dyes. The MBKM PkM program for village development, which was implemented in Padukuhan Malangrejo, was well appreciated by the community and the Wedomartani Village leader, and hopefully the programs will continue in the following year.

Abstract

Obesity increases the risk of various diseases. Black rice, renowned for its high anthocyanin content, is considered a potential functional food for preventing metabolic disorders. The current study investigated the effects of black rice crunch (BRC) on body weight and haematological profiles in obese rats. Rats were fed with high-fat diet to induce obesity and supplemented with different concentrations of BRC for 4 and 8 weeks. The results showed that high-fat diet effectively induced obesity, as evidenced by significant increase in body weight. Importantly, 75% BRC supplementation resulted in significant weight reduction in obese rats. Further analysis revealed an increase in erythrocyte numbers in obese groups supplemented with 75% BRC, but no significant changes in haemoglobin concentration or haematocrit percentage. Further investigation showed that 75% BRC led to a decrease in mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), and mean corpuscular volume (MCV), potentially affecting the size and concentration of haemoglobin within erythrocytes. The total leucocytes count increased with the high-fat diet, while BRC supplementation alone did not have significant impact. Lymphocyte percentage remained stable across the groups, indicating minimal influence of the dietary interventions. Neutrophil percentage varied initially but was not specific to BRC or the high-fat diet. Platelet count and distribution width were not significantly influenced, but mean platelet volume (MPV) increased after 8 weeks of BRC treatment, suggesting larger platelet sizes associated with obesity. Overall, the study provides important insights into the effects of BRC supplementation on body weight and haematological parameters related to obesity.

Abstract

Coronavirus Disease-19 (COVID-19) is a disease caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), this disease began to emerge at the end of 2019 and has become a pandemic to this day. Vaccination can play an important role to prevent the spread of the disease, so there are many vaccine developments, including DNA vaccines. Delivery systems such as chitosan can increase the effectiveness of vaccines. The purpose of this study was to optimize and evaluate the pEGFP-N1-S1 formula with a chitosan delivery system as a DNA vaccine candidate. Plasmids were cloned in E.coli DH5α and extracted for further evaluation with restriction enzymes and sequencing. The EGFP-N1-S1 plasmid was then formulated with chitosan nanoparticles with a mass ratio of Plasmid DNA:chitosan of 1:0.1; 1:0.2; 1:0.3; 1:0.4; 1:0.5; 1:0.6 and 1:0.7. The isolated plasmid has a concentration of 2182.32 ng/L and a purity of 1.911. Restriction results showed that the S1 gene (684 bp) was successfully inserted into the pEGFP-N1 plasmid (4813 bp). DNA:chitosan mass ratio of 1:0.6 is the optimal formulation for DNA to bind with chitosan perfectly.

Abstract

Penelitian ini bertujuan untuk menguji sitotoksisitas fraksi ekstrak kloroform gabungan organ vegetatif Vanda hasil persilangan terhadap kanker payudara cell line T47D dan kanker serviks cell line HeLa. Organ vegetatif dari tanaman tersebut diekstrak dan difraksinasi. Hasil fraksinasi dengan profil yang sama digabungkan. Fraksi gabungan diuji sitotoksisitasnya terhadap cell line T47D dan cell line HeLa menggunakan metode MTT Assay. Analisis data untuk penentuan IC50 dihitung dengan analisis probit dalam SPSS 17. Hasil penelitian menunjukan fraksi C yang paling baik dalam menghambat pertumbuhan sel kanker payudara cell line T47D dan sel kanker serviks cell line HeLa.
