Abstract

Molluscs are invertebrate organisms that produce secondary metabolites to adapt to their environment. These secondary metabolites are also found in fungi that live in symbiosis with molluscs. Research has shown that these symbiotic fungi produce various secondary metabolites with antibacterial and antifungal properties. This study aims to explore the potential of fungi associated with molluscs in developing bioprospecting strategies against pathogenic bacteria and fungi. The antibacterial and antifungal screening tests were conducted against the pathogenic bacteria Escherichia coli and Staphylococcus aureus, and the pathogenic fungi Candida albicans and Trichoderma harzianum, over a period of 36 hours. The results indicated that 12 isolates of mollusk-associated fungi exhibited antibacterial activity against the tested bacterial pathogens. Additionally, 4 of these 12 isolates also demonstrated antifungal activity. The antibacterial and antifungal activities were identified by the clear zones formed in the test medium. The screening tests revealed that fungi associated with molluscs possess antibacterial and antifungal activities against several tested pathogenic bacteria and fungi. Abstract

Molluscs are triploblastic coelomate metazoan organisms from the invertebrate phylum that have secondary metabolite profiles. Secondary metabolites of molluscs are formed due to adaptation to the surrounding environment. This study aims to analyse the secondary metabolite profiles of Cassidula nucleus species from Semarang waters. Cassidula nucleus samples were obtained from the mangrove ecosystem in Mangkang, Semarang, used the purposive sampling method. The methods used in this study include maceration extraction to extract compounds from mollusc samples, thin layer chromatography (TLC) tests for the separation and identification of metabolites, and antibacterial tests to evaluate the potential of the resulting mollusc extract. Maceration extraction was carried out using ethanol in a 1:7 ratio for 48 hours. The antibacterial tests was conducted on the pathogenic bacteria Eschericia coli and Staphylococcus aureus using the disc diffusion method. The extract concentrations tested were 1000 μg/disc, 500 μg/disc, 250 μg/disc, 125 μg/disc, and 62.5 μg/disc. Observations were made every 12 hours for a total of 36 hours. The research results indicated that the positive extract contained flavonoid and alkaloid compounds, which played an active role as antibacterial agents, as comfirmed by TLC test. Antibacterial activity against both pathogens was positive, as evidenced by the formation of a clear zone in the test medium. This study provides new insights into the potential bioactivity of Cassidula nucleus as natural ingredients for medical and health applications.

Abstract

Excessive UV radiation generates free radicals, damaging healthy skin cells. Bioactive compounds with photoprotection and antioxidant properties offer a promising treatment for this damage. Pigments, often sourced from marine microbes associated with marine organisms, are natural materials known for these capabilities. This study aimed to identify antioxidant and photoprotection activities in pigmented bacteria linked to gastropods. A source of pigments is marine microbes associated with marine organisms. This study aims to determine the antioxidant and photoprotection activities of pigmented bacteria associated with the gastropod. Gastropod samples were obtained from the mangrove ecosystem of Mangkang Semarang and bacteria were isolated by the spread plate method. This research method is explorative descriptive, includes colony characterization, bacteria mass culture and maceration extraction of bacteria using methanol, carotenoid content calculation, antioxidant and photoprotection activity test, compound preliminary analysis using Fourier Transform Infrared spectroscopy (FTIR) and molecular identification of bacteria. The results obtained 2 bacterial isolates derived from the gastropod Cassidula nucleus have colored colonies. Based on the calculation of the highest carotenoid content is in the isolate with the code MK.CN.3 amounting to 23.81 µg/g. Antioxidant activity test results from isolate MK.CN.3 showed IC50 value of 461 ppm with weak category and photoprotection value with SPF of 6.57 with extra protection category. Based on the analysis of crude extract, the pigment is suspected to contain carotenoids such as astaxanthin and β-carotene. The results of molecular identification showed that isolate MK.CN.3 is a strain of Isoptericola chiayiensis.  Peningakatan radikal bebas akibat radiasi sinar UV berlebih pada kulit menyebabkan kerusakan sel kulit sehat. Penanganan kerusakan kulit yang disebabkan oleh paparan sinar ultraviolet (UV) dapat dilakukan melalui pemanfaatan senyawa bioaktif yang berfungsi sebagai agen fotoproteksi untuk melindungi kulit dari radiasi UV serta memiliki aktivitas antioksidan yang mampu menetralisir radikal bebas. Salah satu bahan alam yang memiliki kemampuan fotoproteksi dan antioksidan adalah pigmen. Salah satu sumber pigmen adalah mikroba laut yang berasosiasi dengan organisme laut. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan fotoproteksi bakteri berpigmen asosiasi gastropoda. Sampel gastropoda diperoleh dari ekosistem mangrove Mangkang Semarang dan bakteri diisolasi dengan metode spread plate. Metode penelitian yang digunakan adalah deskriptif eksploratif, meliputi karakterisasi koloni, kultur masal bakteri dan ekstraksi bakteri secara maserasi menggunakan pelarut metanol, perhitungan kandungan karotenoid, uji aktivitas antioksidan dan fotoproteksi, analisis pendahuluan senyawa menggunakan Fourier Transform Infrared spectroscopy (FTIR) serta identifikasi bakteri asosiasi gastropoda secara molekuler. Hasil penelitian diperoleh 2 isolat bakteri yang berasal dari gastropoda Cassidula nucleus memiliki koloni berwarna. Berdasarkan perhitungan kandungan karotenoid tertinggi ada pada isolat dengan kode MK.CN.3 sebesar 23,81 µg/g. Hasil uji aktivitas antioksidan dari isolat MK.CN.3 menunjukkan nilai IC50 sebesar 461 ppm dengan kategori lemah dan nilai fotoproteksi dengan SPF sebesar 6,57 dengan kategori extra protection. Berdasarkan analisis ekstrak kasar pigmen diduga mengandung karotenoid jenis astaxanthin dan β – carotene. Hasil identifikasi molekuler menunjukkan bahwa isolat MK.CN.3 merupakan bakteri jenis Isoptericola chiayiensis.

Abstract

This research investigates the ocean's potential as a source of natural active ingredients with antioxidant, antibacterial, and sun protection properties. Marine gastropods are known to have symbiotic relationships with associated bacteria capable of producing pigments, which hold promise for the cosmetic and health sectors. The study aims to identify and analyze pigments produced by bacteria linked to marine gastropods and evaluate their effectiveness as antioxidants, antibacterials, and sun protectants. The method involved isolating and analyzing pigments from gastropod-associated bacteria, followed by tests to assess their properties. Results revealed two gastropod species: Telescopium telescopium (Linnaeus, 1758) and Cassidula nucleus (Gmelin, 1791), from which 21 bacterial isolates were obtained—10 from T. telescopium and 11 from C. nucleus. Among these, Micrococcus yunnanensis, a bacterium with high pigment production, was successfully isolated. At 1,000 µg/mL, its pigment's crude extract had low antioxidant activity and had a low SPF category as a photoprotective agent. Antibacterial tests showed efficacy against Pseudomonas aeruginosa and Staphylococcus aureus. HPLC and FTIR analysis suggested the pigment contains carotenoid compounds. These findings highlight the potential of M. yunnanensis pigment for sustainable health and cosmetic applications.

Abstract

Natural ingredients from animals and plants generally have potential in pharmaceutical applications. However, there are still relatively little exploration made on the active ingredients from marine life. Sea cucumbers inhabit seabed and are filter feeders, consuming all microorganisms within their vicinity, which opens the prospect of medicinal applications of sea cucumber intestinal symbiont microbes. This study aims to obtain sea cucumber intestinal symbiont microbes with potential pharmaceutical application as probiotic microbe candidate. The study was carried out by collecting samples, microbe isolation, antimicrobial screening, identification of potential probiotic microbe candidate. Characteristics of the microbe were studied by biochemical screening, and molecular identification. The study found 21 microbial isolates from Holothuria atra and 30 microbial isolates from Holothuria leucospilota. Antimicrobial activity screening results against pathogens of Bacillus cereus dan Pseudomonas aeruginosa found 13 microbial isolates with positive activity. Four species were identified by molecular identification, namely Bacillus aquimaris, Bacillus maritimus, Bacillus toyonensis, and Virgibacillus chiguensis, whereas the other nine were identified by biochemical screening. Four microbe genus, namely Rothia sp., Listeria sp., Micrococcus sp., and Staphylococcus sp, were found to be the most viable candidate for probiotics. It was concluded that Bacillus sp., Rothia sp., Micrococcus sp., and Staphylococcus sp. Exhibited the most potential as probiotic microbes.

Abstract

Gastropod association bacteria from magrove ecosystem have a great potency as antibacteria. The contamination of Escherichia coli and Bacillus cereus can cause damage in fish product. The aims of these research are to determine gastropod association bacteria that can inhibit E. coli and B. cereus growth, as well as the contaminantion bacteria in Belanak (Mugil subviridis). Gastropods sample Certhideopsilla alata, Cerithidea quoyii, Cassidula aurisfelis, Cassidula nucleus, Telescopium telescopium were collected at Mangrove Education Park, Tugu, Semarang. The resaerch procedure were isolation, antibacteria test, biochemistrically identification, and then application in Belanak fish (Mugil subviridis). There were 61 bacteria isolated. Thirteen isolates were able to inhibit E. coli and eight to B. cereus. Supernatant of GMT 3.2.5 and GMT 4.1.3 have the most widely inhibition zone, (8,48 mm and 7,84 mm). Biochemistry identification shown that GMT 3.2.5 and GMT 4.1.3 has identic characteristic with genus Micrococcus sp. and Bacillus sp. Total plate count (cfu/ml) of bacteria from Belanak fish flesh that soaked by Micrococcus sp. GMT 3.2.5 and Bacillus sp. GMT 4.1.3 supernatant were 115 x 104 and 265 x 103colony. The colony were less than negative control  (2312 x 103 colony). The association bacteria can inhibit contaminant bacteria in Belanak fish.

Abstract

Salt production in the form of coarse salt has a quality with a NaCl salt content of about 94%, which can still meet the requirements as raw material for table salt. Meanwhile, the minimum quality of salt containing NaCl content is required at least 97% for industrial salt needs. Hence, this research aimed to increase the quality of coarse salt from salt farmers to raise the salt selling value by producing health salt. The research method included making a salt house, making salt water with a baume content of about 25-29 Bé°, crystallization, and filtering / draining the Health Salt. The next step was the salt mineral test using the spectrophotometer method. The test results showed that the NaCl content reached 99.35%, while the SNI: 8027:2016 requires a minimum of 97% as industrial salt. Crystal salts are pollutants-free due to the levels of heavy metals (Cadmium, Cd), Mercury (Hg), and minerals that are harmful to health. Iodine, Magnesium, and Calcium are smaller, and the results of laboratory tests are 0.001 mg/kg when compared with SNI: 8027:2016 of 0.1% maximum. Bittern produces very prominent data, namely Magnesium (Mg) with the highest content of 616.67 mg/kg and Potassium (K) of 84.85 mg/kg. It was concluded that the resulting crystal salt qualified as industrial and health salt.

Abstract

Graphical Abstract   Highlight Research Lola snails originating from the Bangka Belitung Islands (Nasik Strait, Rebo Waters, and Ketawai Island) were identified as a maxima not to be R. nilotica species based on morphological molecular identification. Maxima and R. nilotica have the same morphological form, but they are genetically different. The clarification of the Lola snail species in this study serves as a reference in determining the conservation status of the Lola snail. The clarification of the Lola snail species serves as a reference in determining the catch quota from the Bangka Belitung Islands.     Abstract The use of Lola snails (Rochia maxima) in the Bangka Belitung has continued to increase. However, research focused on Lola snails originating from the Bangka Belitung has not been thoroughly conducted. In a previous study, the Lola snail sample found in the Bangka Belitung was Trochus niloticus (current name: Rochia nilotica). This study aimed to identify Lola snail species using the DNA barcoding as a way of clarifying Lola snail species that originated in the Bangka Belitung. Lola snail sampling was carried out at three locations, which were Nasik Strait, Ketawai, and Rebo Waters. This study was conducted through five stages, including Lola snail tissue sampling, DNA extraction, amplification by PCR, sequencing, and data analysis. The results showed that the molecular identification of the Cytochrome Oxidase Subunit 1 (COI) gene in Lola snail samples was identified as Rochia maxima species (Koch, 1844). The results of the phylogenetic tree analysis showed that Lola snails found in the Bangka Belitung were close to Tectus maximus (or Rochia maxima) species. R. maxima has a lighter shell than R. nilotica, a clear conical shape, and a circle on the body that does not widen at the edges. Unlike zoologists who said that R. maxima is the primitive form of R. nilotica, the present study found that they have the same morphology and are considered similar but genetically different. The clarification of Lola snail species can be used to determine the conservation status and catching quota of Lola snails from the Bangka Belitung.

Abstract

Nautilus bercangkang rapuh termasuk dalam kelas Cephalopoda, famili Argonautidae. Tujuan dari penelitan ini adalah untuk mengidentifikasi jenis Nautilus bercangkang rapuh yang ditemukan di perairan Teluk Tomini Kabupaten Parigi Moutong. Sampel paper Nautilus ditangkap pada kedalaman 1000 m. Hasil penelitian memperlihatkan bahwa sampel yang berhasil dikoleksi selama bulan Maret hingga bulan Desember hanya sebanyak 6 spesimen pada kedalaman 70-80 m, dengan 4 spesimen yang bertelur dan 2 spesimen tanpa telur. Dari 6 spesimen yang ditemukan, diketahui ada 2 jenis yaitu Argonauta argo dengan rerata panjang cangkang= 34,05 mm, lebar cangkang= 22,20 mm dan lebar aperture= 11,15 mm. A. argo memiliki 8 tentakel dengan 4 tentakel panjang dan 2 pendek. Warna cangkangnya lebih cerah dari cangkang A. hians, cangkangnya lebih pipih dengan dua baris duri kecil yang sangat berdekatan di sepanjang sisi dorsal dan halus, sepanjang sisi ventral ujung tebal dan meruncing. Serta A. hians yang memiliki rerata panjang cangkang 47,02 mm, lebar cangkang 33,07 mm dan lebar aperture 21,30 mm, memiliki duri yang lebih besar dan tumpul, di sepanjang sisi dorsal yang lebar, sepanjang sisi lateral aperture melebar dan tidak membentuk seperti sayap. Garis rusuk yang terdapat di sisi lateral lebih tebal dan renggang. Jenis Nautilus bercangkang rapuh yang ditemukan di Teluk Tomini, yaitu A. argo dan A. hians.
