Abstract

Marina and Baruna Beach are widely recognized as popular beach tourism destinations in Semarang. Its proximity to the main beach estuary causes them to significantly impact marine tourism quality. In addition, their exposure to waste may be a potential threat to biological pollution. Coliform and Escherichia coli are used as bioindicators to monitor the quality of marine tourism due to their high correlation to faecal contamination and pathogenic microbes. Samples collected were beach sand and seawater from Marina Beach, Baruna Beach and the estuary of Semarang Banjir Kanal Barat. Data were collected following the presumptive and confirmed test, followed by the conversion to MPN Table. Data obtained is combined with Government Regulations No. 22 of 2021 in Appendix VIII, specifically seawater quality standards for marine tourism. The results of this study reveal high contamination of coliform and E. coli in both beaches. Coliform result in the swimming zone at Marina Beach was 2800 MPN/100ml and at Baruna Beach was 16000 MPN/100ml. Meanwhile, Escherichia coli in the swimming zone area at Marina Beach was 400 MPN/100ml and at Baruna Beach was 1700 MPN/100ml. Overall, these results show that the contamination rate from both swimming zones is higher than the recommended seawater quality standard for marine tourism  Wisata Pantai Marina dan Pantai Baruna menjadi kawasan wisata pantai yang populer di Kota Semarang. Lokasinya yang berdekatan dengan muara sungai utama di Kota ini berdampak terhadap kualitas wisata pantai. Paparan limbah berpotensi menyebabkanpencemaran biologis. Monitoring terhadap bakteri coliform dan bakteri Escherichia coli dapat digunakan sebagai bioindikator kualitas wisata pantai, karena keberadaannya berkorelasi dengan cemaran feses dan mikroba patogen. Sampel berupa pasir pantai dan air laut diambil dari  lokasi Pantai Marina, Pantai Baruna, dan muara Sungai Banjir Kanal Barat Kota Semarang. Pengujian sampel dilaksanakan dengan presumtif test dan confirmed test, selanjutnya dikonversi dengan Tabel MPN. Data yang didapatkan akan dipadukan dengan PP Nomor 22 Tahun 2021 pada Lampiran VIII, yaitu baku mutu air laut untuk wisata bahari. Hasil penelitian menunjukkan tingginya kontaminasi bakteri coliform dan bakteri Escherichia coli dikedua pantai. Hasil coliform pada kawasan swimming zone pada Pantai Marina 2800 MPN/100ml dan Pantai Baruna 16000 MPN/100ml, serta hasil Escherichia coli pada kawasan swimming zone pada Pantai Marina 400 MPN/100ml dan Pantai Baruna 1700 MPN/100ml. Kawasan swimming zone pada wisata Pantai Marina dan Pantai Baruna mempunyai hasil yang melebihi baku mutu air laut untuk wisata bahari.

Abstract

Masyarakat nelayan perempuan pesisir di Kecamatan Tugu Semarang memiliki potensi untuk meningkatkan pendapatan desanya melalui wira usaha wisata. Potensi yang dimiliki di wilayah tersebut adalah pariwisata mngrove, sector budidaya, hasil2 produk perikanan. Semua potensi tersebut berpeluang untuk dapat meningkatkan aktifitas pembangunan sosial ekonomi, membuka lapangan pekerjaan sekaligus meningkatkan pendapatan masyarakat. Masyarakat di Kecamatan Tugu Semarang yang sangat antusias untuk meningkakan penghasilan dalam rumah tangganya namun belum ada peluang karena keterbatasan pengetahuan, fasilitas dan kesempatan. Maka salah satu tantangan dalam solusi permasalah tersebut diatas  adalah dengan terciptanya industri wisata. Membentuk inkubasi wirausaha baru (INWUB) pada masyarakat melalui tercapainya alih/transfer teknologi bahan alam khususnya tentang produk yang spesifik batik pewarna alam  mangrove. Wilayah tersebut merupakan wilayah pesisir pantai utara Semarang yang di wilayahnya banyak ditanami tanaman mangrove. Budidaya mangrove mereka lakukan dari turun temurun, kini wilayah tanaman mangrovenya sudah meluas di sepanjang pantai Mangunhardjo. Limbah tanaman mangrove biasanya terdapat dalam bentuk ranting, batang, daun atau tanaman yang tidak tumbuh. Limbah tersebut  biasannya dibakar atau dikubur dalam tanah. Team UNDIP telah memulai dalam memberikan solusi dalam pemanfaatan limbah mangrove, yaitu dengan memanfaatkan limbah mangrove sebagai bahan warna alam. Masyarakat nelayan perempuan pesisir di Kecamatan Tugu Semarang memiliki kini berpeluang untuk dapat meningkatkan aktifitas pembangunan sosial ekonomi, membuka lapangan pekerjaan sekaligus meningkatkan pendapatan masyarakat. banyak ditanami tanaman mangrove. Kini sudah berjalannya program pemberdayaan wanita nelayan pesisir  di desa Mangunhardjo Kecamatan Tugu Semarang dan telah menjadikan masyarakat lebih meningkat produksi batik mangrovenya dengan diversifikasi warna yang bervariasi. Telah dibentuk inkubasi wirausaha baru (INWUB) yang khusus produksi asesoris dari mangrove. Wira usaha ini masih dibawah koordinasi kelompok batik ”Wijaya Kusuma”. Hasil penelitian memperlihatkan bahwa batik pewarna alam dari ekstrak daun dan kulit limbah mangrove yang difiksasi dengan tawas, kapur dan tunjung menghasilkan warna batik yang berbeda-beda. Hasil penelitian batik pewarna alam dari daun limbah mangrove cenderung lebih muda warnanya dibandingkan dengan kulit limbah mangrove dan variasi warna juga terganting dari fixaxernya. Dari aplikasi teknologi pewarn limbah mangrove jadi batik, maka kini telah dibangun galeri batik limbah mangrove “Wijaya Kusuma” yang berlokasi di desa Mangunhardjo.

Abstract

This study is the further study on isolation and identification of bacterial symbionts and Enhalus acoroides and Thalassia hemprichii. The results showed that the activity of bacterial symbionts seagrass Enhalus acoroides and Thalassia hemprichii acquired seven bacterial isolates which were able to inhibit Vibrio harveyimasing - each isolate consisted of 5 Thalassia hemprichii isolates and 2 Enhalus isolates. The results of the study showed that the activity of bacterial symbionts seagrass Enhalus acoroides and Thalassia hemprichii acquired 7 bacteria isolates to inhibit Vibrio. There were 4 isolates that had bactericidal activities. Two bacterial symbionts that had the highest inhibitory zones were identified, namely Th 12 isolate, type of bacteria Bacillus macerans, meanwhile Eh 10 isolate was Micrococcus varians bacteria. The results showed that the inhibitory zone Eh 9 isolate (3.87 ± 0.25 mm) was higher than Th 20 isolate (3.14 ± 0.18 mm). Based on the results of biochemical identification of bacteria known that Eh 9 was kind of Brevibacterium sp ., while the Th 20 isolate was a type of bacteria Bacillus pumilus. The results concluded that the bacterial symbionts that comes from Enhalus acoroides and Thalassia hemprichii can serve as an antibacterial of Vibrio harveyi

Abstract

Seagrass is a community that consists a large number of organisms which associated within. Organisms associated with seagrass are gastropods. The purpose of this research is to determine the structure of gastropods community at seagrass on Pulau Panjang, Jepara. Exploration methods was applied and data was analyzed descriptively. The location of research are divided into 3 locations based on 3 different densities of seagrass : sparse, medium and dense. Every location consists of 4 stations with 3 repetitions. The research was held during Desember 2014. Sampling using quadratic a frame size of 1x1 m. The result, there are were 15 genera gastropods of 8 families as followed Trochus (Ciridae), Strombus (Strombidae), Turbo, Astralium, Dentarene, Liotina (Turbinidae), Cantharus, Nassaria, Engina  (Buccinidae), Cerithium, Rhinoclavis, Clypeomorus (Cerithiidae), Morulla (Muricidae), Columbella (Columbellidae), and Neritodryas (Neritidae). The abundance in location I 108.75 ind/????2 with dense of seagrass density, location II 91 ind/m2 with medium of seagrass density, and location III 47,25 ind/m2 with rare of seagrass density. Index value of Gastropod diversity classified medium category, index value of gastropod similarity classified high category, it means abudance from each spesies can be said to be similar, and evenly distributed and not on domination by a specific species.  Community similarity index classified in the category of medium to very large

Abstract

Microorganisms that live in association with marine invertebrates are able to produce a compound similar  to those produced by marine invertebrates and are thought to be potential as bioactive materials. Supplement  extracts from the Photobacterium phosphoreum bacterial culture have been shown to have antibacterial activity. The purpose of this study was to determine the anti-bacterial activity of the chloroform extract of Photobacterium  phosphoreum biomass against pathogenic bacteria Bacillus substilis, Staphylococcus aureus, Salmonella sp. and  Escherichia coli. The extraction process was carried out by the liquid-liquid extraction method. Fractionation was  performed using Open Column Chromatography (KKT). The antibacterial activity test was conducted by the agar diffusion method according to Kirby-Bauer. The results showed that for the antibacterial activity test of crude  extract, the largest inhibition zone diameter occurred in Escherichia coli with a concentration of 50 µg / disk at 24 h incubation, namely (9.80 ± 0.75 mm), while the smallest inhibition zone diameter occurred in Salmonella sp. by  giving a concentration of 10 µg / disk at an incubation time of 48 hours, namely (6.03 ± 0.05) mm. Of the 6  fractions resulting from crude extract fractionation, it was known that fraction 5 was the most active fraction and  inhibited the growth of the tested bacteria. The largest inhibition zone diameter occurred in Escherichia coli (9.83  ± 0.28) mm, while the smallest inhibition zone diameter occurred in Salmonella sp. (8.63 ± 0.20 mm)

Abstract

Several marine gastropods are known to have high economic value, one of which is the Spiral babylonia snail. They commonly exploited for meat, shell, or operculum. Tiger snail meat is commonly exported and consumed by fishermen. Its shell is used as a material for industrial lime and for omamen. Operculum is commonly used as a medicinal and perfume ingredient). However, the recent production has decreased due to the limited information of seasonal catch. One way to overcome these obstacles is through culture activities. The provision of seeds is an important link in the culture system, therefore the study of the spawning process and larval development is an important requirement for the development of larval rearing techniques. This study aims to determine the development process of juvenil eggs and the survival rate of tiger snails reared in the laboratory.. The study was conducted at the Laboratory of Marine Farm Subdivision in Kudus, Pt Pura Bahari, Jepara Regency. This sudy used experimental method with descriptive data analysis. Adult tiger snails were caught from Bunga waters, Demak. The snails were reared in a rectangular tube filled with 15 cm filtered sea water and 5 cm of sand. Egg capsules were reared in a 4 liters circular tube with continuous aeration. The hatched larvae were reared in a 300 liters circular tube filled with filtered seawater, continuously aerated and fed with Chlorella plankton . Our result showed that tiger snails spawn naturally and produce egg capsules. The incubation period for the capsule is 6 days, then develop into veliger larva for 12 days. The veliger larvae metamorphosed,and become juvenil. The lowest survival rate observed in the veliger stage. The survival rate of the egg and the intracapsular trochopore stages were quite high (tube I = 98.413%; tube II = 98.238%; tube III = 90.476%), then the lowest was in the veliger stage (tube I = 3,468%; tube II = 2.603%; tube III = 2,598%) and the juvenil stage is quite high compared to the previous phase (tube I = 3,413%; tube II = 2,479%; tube III = 2,351%).

Abstract

Green Mussel included to permanent animal so the interaction with its environment will affect its needed or its un-needed nutrient. Iron and zinc represent a very required micro mineral of human body. Iron plays an important role in forming red corpuscles and preventing anemia. The function and role of zinc in body for example to assist the forming of cell genetic items, forming the red corpuscle (haemoglobin), and also to assist pancreas function in digestion process. Territorial water of Jepara and Semarang are the representation areas producing Green Mussel (Perna Viridis). The aim of this research is to understand the iron and zinc content in Green Mussel. This research was conducted in December 2006 until February 2007. Materials used in this research is Green Mussel derived from Territorial water of Bandengan, Jepara and Territorial Water of Tanjung Mas, Semarang. The intake of Green Mussel sample used Purpose Random Sampling Method, which is intake of sample at random according to specific purpose. Method used in this research is Descriptive Method, while for iron and zinc measurement method using AAS. The result showed that the iron rate in Green Mussel at Territorial Water of Bandengan, Jepara degraded 459,70, 209,23 and 147,87 mg/kg from December 2006 until February 2007. While the Iron content in Green Mussel at Territorial Water of Tanjung Mas, Semarang continuously increased 227,18, 317,66 and 336,52 mg/kg from December 2006 until February 2007. The analysis result of zinc content on December until February of Jepara degraded 60,15, 8,90 and 7,73 mg/kg. While the Green Mussel Zinc of Tanjung Mas, Semarang on December 2006 until January 20.07 was degrading, from 154,10 become 9,63 and on January until February was increasing, from 9,63 mg/kg become 10,11 mg/kg.

Abstract

Natural ingredients from animals and plants generally have potential in pharmaceutical applications. However, there are still relatively little exploration made on the active ingredients from marine life. Sea cucumbers inhabit seabed and are filter feeders, consuming all microorganisms within their vicinity, which opens the prospect of medicinal applications of sea cucumber intestinal symbiont microbes. This study aims to obtain sea cucumber intestinal symbiont microbes with potential pharmaceutical application as probiotic microbe candidate. The study was carried out by collecting samples, microbe isolation, antimicrobial screening, identification of potential probiotic microbe candidate. Characteristics of the microbe were studied by biochemical screening, and molecular identification. The study found 21 microbial isolates from Holothuria atra and 30 microbial isolates from Holothuria leucospilota. Antimicrobial activity screening results against pathogens of Bacillus cereus dan Pseudomonas aeruginosa found 13 microbial isolates with positive activity. Four species were identified by molecular identification, namely Bacillus aquimaris, Bacillus maritimus, Bacillus toyonensis, and Virgibacillus chiguensis, whereas the other nine were identified by biochemical screening. Four microbe genus, namely Rothia sp., Listeria sp., Micrococcus sp., and Staphylococcus sp, were found to be the most viable candidate for probiotics. It was concluded that Bacillus sp., Rothia sp., Micrococcus sp., and Staphylococcus sp. Exhibited the most potential as probiotic microbes.

Abstract

Gracilaria verrucosa adalah salah satu jenis rumput laut merah yang memiliki kandungan bioaktif untuk antioksidan alami. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan yang terkandung di dalam Gracilaria verrucosa dengan menggunakan dua metode berbeda, yaitu penangkapan radikal bebas DPPH dan daya reduksi dengan menggunakan tiga pelarut berbeda (n – heksan, etil asetat dan metanol), mengetahui nilai total fenolat dan pigmen (klorofil a, klorofil b dan karotenoid). Penentuan total fenolat menggunakan asam galat sebagai standar dengan Folin – Ciocalteu sebagai pelarutnya. Penentuan pigmen klorofil a, klorofil b dan karotenoid dilakukan dengan spektrofotometri UV – VIS dengan Panjang gelombang 646 nm, 663 nm dan 470 nm. Hasil penelitian menunjukkan bahwa ekstrak Gracilaria verrucosa memiliki aktivitas antioksidan sangat lemah dengan nilai IC50 ekstrak etil asetat 1.274 ppm, ekstrak n – heksan 1.304 ppm dan ekstrak metanol 1.381 ppm sedangkan aktivitas antioksidan dengan daya reduksi menunjukkan nilai antioksidan ekstrak etil asetat 15,13 mgAAE/gr sampel; ekstrak n – heksan 8,50 mgAAE/gr sampel dan ekstrak metanol 5,85 mgAAE/gr sampel.

Abstract

The gonads maturity level (TKG) is a particular stage gonadal development before and after the spawning biota. This research aims to determine the gonads index of the Diadema setosum and the gonads maturity level of Diadema setosum found on Panjang Island, Jepara. The research method used was the observation method. The gonads histology was made by using paraffin method and hematoxylin-eosin staining. The measured parameters were the diameter of the shell, the Gonads Maturity Index (GI) and the Gonads Maturity Level, and environmental parameters such as temperature, pH, DO, and salinity of the water. The Gonads Maturity Level was determined on the histology of gonads. The result shows average Diadema setosum GI value of 7,334 % with an optimum maturity in December. Based on the height of individual the sea urchin (Diadema setosum) found in the category TKG III, which is 11 out of 30 Individuals and two individuals in the TKG IV category are observed histology of the gonadal showing in that phase.
