Abstract

This study aims to determine the prevalence and to identify gastrointestinal protozoa in goats in Sub-district of Labang, Bangkalan Regency, Madura, Indonesia. The research was conducted in April - July 2019 with 100 samples of goat’s stool. Examination was done in the laboratory of Parasitology in Faculty of Veterinary Medicine, Airlangga University using direct smear, sedimentation and flotation methods. The result showed that goats in Labang, Bangkalan Regency were infected by protozoa Eimeria sp. and Blastocystis sp. and the prevalence was 84%. There were no significant differences in the age and sex of goats (p > 0.05).

Abstract

The purpose of this research to know cross reaction of Haemonchus contortus protein with anti-Fasciola gigantica serum by using the Western Blot technique. The result can be used as Haemonchosis serologic diagnostic. This research was conducted using Haemonchus contortus worms obtained from abomasum, especially cow worms and Fasciola gigantica worms obtained from the bile duct and gall bladder. The worms were crushed and added with Phosphate Buffer Saline (PBS) to make homogenates and then centrifuged at 3500 rpm for 15 minutes. Homogenate Fasciola gigantica was immunized to mice to get anti-Fasciola gigantica serum at a dose of 200 μg/mice and booster 3 times with a span of two weeks. Homogenates were carried out using the SDS-PAGE technique to analyze proteins using a brilliant blue dye. Cross reaction of Haemonchus contortus protein with anti-Fasciola gigantica serum byusing Western blot technique and obtained 3 protein bands are 94.9, 52.7, 46.7 kDa.

Abstract

The aim of this study was to determine leukocytes and differential counting in chicken after infected with L2 Toxocra cati. In this study, it was used twenty chickens, wich were 14 weeks old. They were divided into 4 groups. Chickens were infected by orally with dose 0 eggs/ml L2 T. cati, 10 eggs/ml L2 T. cati, 100 eggs/ml L2 T. cati and 1000 eggs/ml L2 T. cati. Blood sampling were conducted on 2, 7 and 21 days after infection. Leukocyte value was determined by improve neubauer and differential counting stained with Wright’s stain then determined using microscope 1000x. The data was analyzed by Anova Factorial and then continued by BNJ Test at 5%. The result showed that increased of leukocytes value and eosinophil at 7 and 21 days after infection L2 T. cati with different dose, increased of leukocytes value and eosinophil on infection dose 10 eggs/ml L2 T. cati, 100 eggs/ml L2 T. cati and 1000 eggs/ml L2 T. cati. It shows that there was not intereraction between time of infection process and infection dose of L2 T. Cati.

Abstract

The aim of this research was to detect the presence of nematode worms that infects sacrificial cattle slaughtered duringEid al-Adha 1439 H in East Surabaya. This research used 24 sampels of sacrificial cattle and used digestive tract surgical method. Based on the result of the examination using a microscope with 100x magnification, there are positive samples infected with nematode worms. In caecum cattle consist of Oesophagostomum radiatum and Trichuris spp. In colon cattle consist of Oesophagostomum radiatum.

Abstract

This study aims to determine the pentastomide parasite that infects the digestive tract of giant water lizards examined in Laboratory of Parasitology Department, Faculty of Veterinary Medicine, Airlangga University. In this study, three sample were found to be infected with pentastomide parasite in the lung. The phase found was adult phase pentastomide; it has pentastomide spermatheca which shows that this parasite is female.

Abstract

The main obstacle to the cultivation of tilapia (Oreochromis niloticus) in the KJA Urban Farming Program in Surabaya is unstable water quality. This is because the reservoirs and bozem used are waters whose water quality depends on the water source so that if the environment experiences a decrease in water quality, the activity of disease agents such as parasites will increase and the health condition of fish will decrease. This condition allows fish to be more susceptible to ectoparasite infestations. This study aims to determine the correlation of water quality with the prevalence of ectoparasites in tilapia in the KJA Urban Farming Program in Surabaya. The method used is a survey method. The results of measurements of water quality parameters indicate that the temperature ranges between 29-30C, pH 7-8.5, brightness 18-30 cm, dissolved oxygen 4.8-5.3 mg / l, nitrite <0.043-2.213 mg / l, nitrate <0.008-7.781 mg / l and ammonia 0.063-0.35 mg / l. The results of the average prevalence of Trichodina ectoparasites were 73.85% and Dactylogyrus ectoparasites were 2.85%. There is a positive correlation between temperature, pH, nitrate, nitrite with the prevalence of Trichodina, and temperature, pH, and nitrate with the prevalence of Dactylogyrus. There is a negative correlation between brightness, dissolved oxygen and ammonia with the prevalence of Trichodina, and brightness, nitrite, dissolved oxygen, and ammonia with the prevalence of Dactylogyrus.

Abstract

Permintaan yang tinggi oleh konsumen terhadap produk perikanan terutama lobster (Cherax quadricarinatus) air tawar dari tahun ketahun memacu perkembangan industri budidaya lobster yang sangat pesat. Namun dalam proses budidaya tersebut banyak sekali kendala yang muncul. Salah satu kendala yang umum dihadapi dalam budidaya ikan adalah adanya serangan penyakit yang dise- babkan oleh bakteri. Beberapa strategi pencegahan penyakit yang telah diaplikasikan dalam budida- ya lobster, salah satunya menggunakan antibiotik. Salah satu antibiotik yang sering digunakan pembu- didaya adalah Chloramphenicol. Pada Lobster, penggunaan Chloramphenicol dengan dosis tinggi akan menyebabkan resistensi terhadap bakteri patogen. Pelaksanaan penelitian di Laboratorium Pendidikan Perikanan, Fakultas Perikanan dan Ke- lautan, Universitas Airlangga dan Balai Karantina Ikan Pengendalian Mutu dan Keamana Hasil Peri- kanan Kelas I Surabaya II. Penelitian ini berlangsung pada bulan November 2013 sampai bulan Ja- nuari 2014. Tujuan penelitian ini adalah untuk mengetahui lama waktu henti obat yang diperlukanagar residu antibiotik Chloramphenicol yang terdapat pada Lobster (Cherax quadricarinatus) air tawar me- nurun hingga batas aman untuk dikonsumsi. Lobster yang diberi pakan dengan campuran chlo- ramphenicol dengan dosis 2g/kg pakan terdeteksi residu Chloramphenicol tertinggi sebanyak 31,962 ppb pada minggu pertama dan terjadi penurunan residu menjadi 3.53 ppb selama delapan minggu.

Abstract

Penyakit merupakan masalah yang penting yang harus diperhatikan oleh pembudidaya ikan agar tidak mengalami kerugian. Penyakit dapat muncul di suatu perairan akibat ketidakseimbangan antara lingkungan, inang, dan patogen. Salah satu penyakit yang sering menyerang ikan hias air tawar adalah lernaeosis. Lernaeosis adalah penyakit pada ikan yang disebabkan oleh serangan ektoparasit copepoda dari genus Lernaea. Ektoparasit ini dapat ditemukan pada seluruh permukaan tubuh, mulut dan insang ikan. Lernaea dapat menyebabkan iritasi pada kulit serta lesi. Ikan maskoki (Carrassius auratus) merupakan salah satu jenis ikan hias air tawar tergolong dalam jenis ikan karper yang sering terserang Lernaea. Ikan maskoki (Carassius auratus) merupakan komoditas unggulan perikanan budidaya untuk jenis ikan hias air tawar di Tulungagung. Identifikasi dan derajat infestasi Lernaea yang menyerang ikan maskoki (Carassius auratus) di Tulungagung, Jawa Timur belum banyak diteliti sehingga perlu dilakukan penelitian pada tiga desa yaitu Desa Karang Rejo, Desa Ploso Kandang dan Desa Wajak dimana ketiga desa tersebut merupakan sentra budidaya di Kabupaten Tulungagung. Tujuan penelitian ini adalah untuk mengetahui spesies Lernaea yang menyerang ikan maskoki (Carassius auratus) dan derajat infestasi dari serangan Lernaea pada ikan maskoki yang dibudidayakan di tiga desa yang berada di Desa Karang Rejo, Desa Ploso Kandang dan Desa Wajak, Kabupaten Tulungagung, Jawa Timur. Identifikasi Lernaea dilakukan di Laboratorium Fakultas Perikanan dan Kelautan Universitas Airlangga Surabaya dan penggambaran spesimen dilakukan di LIPI Cibinong. Data hasil identifikasi spesies Lernaea yang menyerang ikan maskoki disajikan secara deskriptif dalam bentuk gambar untuk identifikasi Lernaea dan dalam bentuk tabel untuk derajat infestasi. Hasil penelitian yang dilakukan di Desa Karang Rejo, Desa Ploso Kandang dan Desa Wajak hanya ditemukan satu spesies saja, yaitu Lernaea cyprinacea. Derajat infestasi dari serangan Lernaea cyprinacea di tiga desa tersebut tergolong dalam infestasi ringan.

Abstract

Etawa crossbreed goat are meat and milk-producing livestock that can adapt to the climate in Indonesia. The barriers to raising goats include the problem of disease which is a direct influence on livestock life. This study aims to determine the prevalence of helminthiasis in Etawa Crossbreed Goat in Kalipuro District, Banyuwangi Regency. The study was conducted in January until February 2019 using 100 fecal samples. The samples examination in was performed the laboratory of Instrument Airlangga University PSDKU in Banyuwangi Prodi of Veterinary Medicine, using native, sedimentation and floatation methods. Data obtained from this study were presented descriptively and analyzed by Chi-Square test. Based on examination results, prevalence of helminthiasis in Etawa Crossbreed Goat was 43 %, which is dominately by Moniezia spp. (17%), Haemonchus spp. (15%), Trichuris spp. (9%), Strongyloides spp. (7%), Trichostrongylus spp. (7%). Furthermore, it was suggested that anthelmintic and inspection of helminthiasis in those area given periodically and continuously.

Abstract

Fasciolosis is one of the endemic parasitic diseases in Indonesia and causes significant economic losses. Fasciola gigantica is the main species found to infect livestock. This research aimed to describe the morphology of F. gigantica histologically using hematoxylin and eosin staining as well as ultrastructure morphology using scanning electron microscopy (SEM). Besides, the study was also carried out to determine the prevalence and estimation of economic losses due to fasciolosis in Berau Regency. The histological identification of flukes morphology found several organs including oral sucker (OS), ventral sucker (VS), cirrus (C), genital pore (GP), uterus (U), ovary (O), tegument (Te), testis (T), and caecum (C). In this study, ultrastructure morphology was divided into anterior and midbody region. In the anterior region, there are an oral sucker (OS) with a diameter of 304.5 µm and a ventral sucker (VS) with a diameter of 571 µm. It was also found the anterior spine with a width of 25.4 µm and a height of 22.8 µm. In the midbody, the spine has a width of ± 26.7 µm and a length of ± 38.9 µm leading posteriorly. Postmortem examination results showed that the prevalence of fasciolosis in Berau regency was 21.65% or 34 of 157 cows which were positive with an estimated loss of 228,656,475.00 IDR/16,450 USD (1 USD=13,900 IDR). The amount of loss due to fasciolosis must be a particular concern in carrying out control and prevention measures. Histology and ultrastructure morphology can become a reference in the development of anthelmintic testing that has been used so far in Indonesia.
