Abstract

Diabetes mellitus is a chronic disease that requires good medical treatment to prevent long-term complications. The proportion of DM patients in Indonesia in 2013 was 3,7 million. Indonesia as one of the countries with the highest diversity has natural resources that can be used as an alternative medicine for diabetes mellitus, one of them is dayak onion. Dayak onions themselves contain compounds that can reduce blood glucose levels. The use of traditional medicine in the community is sometimes combined with chemical drugs. So the purpose of this study was to determine the comparison of the combination activity of dayak onion extract – metformin with single metformin to reduce mice blood sugar induced by alloxan. This research includes experimental research. The test animals used were male white mice which were divided into 2 groups, namely the combination of dayak onion and metformin and the single metformin group. The dose of dayak onion extract used in this study was 100 mg / KgBB and the dose of Metformin used was 45 mg / KgBB. Measurement of glucose levels is carried out every 3 days for 15 days. The decrease in fasting blood glucose from the control group and the test group was analyzed by the General Linear Model test at a 95% confidence level. The results of this study indicate that the combination of dayak onion tuber extract and metformin has better hypoglycemic activity than single metformin. This result can be seen from the average glucose level from day 3 to 15 the combination of dayak onion tuber and metformin is lower than the average glucose level of single metformin. The results of GLM analysis showed the sigvalue <0,05 so it can be concluded that there is a difference in glucose levels between the combination of dayak onion tuber and metformin with a single metformin.

Abstract

Hypertension remains a major global health concern due to its high prevalence and strong association with cardiovascular diseases and kidney failure. A key component of blood pressure regulation is the Angiotensin-Converting Enzyme (ACE), which catalyzes the conversion of Angiotensin I into the vasoconstrictor Angiotensin II, making it a primary target for antihypertensive drugs. Although synthetic ACE inhibitors such as ramiprilat are effective, their use is often associated with adverse effects, highlighting the need for safer alternatives. This study employs molecular docking and in silico analysis to evaluate the potential of phytochemicals from beluntas (Pluchea indica) as natural ACE inhibitors. A total of 110 phytoconstituents were screened for pharmacokinetic properties using ADMET analysis, leading to the selection of 20 ligands for docking simulations. Among these, 4,5-di-O-caffeoylquinic acid exhibited the highest binding affinity (-9.409 kcal/mol), followed by di-O-caffeoylquinic acid (-8.984 kcal/mol) and quercetin-3-O-β-D-galactopyranoside (-8.372 kcal/mol). These compounds demonstrated stronger binding affinities than the ACE natural substrate, Angiotensin I (-7.133 kcal/mol), and the ACE inhibitor, ramiprilat (-8.717 kcal/mol), suggesting their potential as competitive ACE inhibitors. The binding interactions of these compounds were characterized by hydrogen bonding with key catalytic residues (HIS368, GLU368), electrostatic stabilization, and hydrophobic interactions within the enzyme active site. Notably, caffeoylquinic acid derivatives closely mimicked the binding mode of ramiprilat, whereas quercetin glycosides exhibited a distinct interaction pattern, indicating a possible alternative inhibitory mechanism. These findings provide evidence supporting the potential of P. indica phytochemicals as natural ACE inhibitors and warrant further investigation into their therapeutic applications in hypertension management.

Tumbuhan yang satu family yaitu asteraceae menyebutkan bahwa kadar sari larut air tidak kurang dari 5%, yang berarti hasil memenuhi persyaratan materia medika indonesia.Kata kunci: Sari Larut Air, Sari Larut Etanol, Kumpai Mahung, AsteraceaeABSTRACT Determination of water-soluble and ethanol extract contents is a quantitative method for the amount of compound content in a simplicia that can be attracted by the solvent. Both methods are almost the same based on the solubility of the compounds contained in simplicia. For generations, the Meratus and Amandit Dayak communities use kumpai mahung (Eupathorium inulifolium H.B. & K) as a medicine for diarrhea and malaria. Research is still rare to use this plant, so researchers aim to determine the content of water-soluble extract of simplicia powder and mahung kumpai leaf extract by the method established by herbal pharmacopoeia. The results of the research showed that the concentration of water-soluble extracts and ethanol soluble extracts of this plants on the powder had values of 19.54% and 16.13%. The extracts have values of 19.53% and 14.55%. One family plant, Asteraceae, states that the water-soluble extract content is not less than 5%, which means the results meet the requirements of Indonesian medical material. Keywords:  Water Soluble Extract, Ethanol Soluble Extract
