Abstract

First report of Begomovirus infection on papaya in Bengkulu, Indonesia. A field survey was conducted during 2019, wefound a severe systemic yellow mosaic, striped green mosaic on leaves petiole, green spots on the fruit of papaya, leafmalformation, and stunting symptoms on three papaya cultivation area in Rejang Lebong, Kepahiang, Bengkulu Tengah, andSeluma, Bengkulu Province, Indonesia. A begomo-like virus was inferred to be the possible cause of the virus-disease-likesymptoms. The study aimed to identify the causal of those typical symptoms on papaya. PCR using universal primer fortranscriptional activator protein (TrAp) and replication-associated protein (Rep) gene of Begomovirus successfully amplifiedthe DNA fragments of 900 bp in all 10 detected samples, except for samples with leaf malformation and stunting symptoms. Itis indicating that those typical symptoms on papaya is associated with Begomovirus infection, while the causal of leafmalformation and stunting is unknown yet. This work is the first report of Begomovirus infected papaya in Indonesia. Severedisease incidence caused by this pathogen was observed on papaya plants in Bengkulu Province that was in the range of 42–100%. This finding is a precious information to be used for identification, and characterization the species of the virus,determining control strategies against the disease.

Abstract

Natural infection of Tobacco mosaic virus on butternut squash in Bali, Indonesia. Tobacco mosaic virus (TMV) was a newly emerging virus infecting cucumbers in Indonesia since 2017. The mosaic disease caused by TMV potentially caused yield loss cucumber in Java. In 2019, mosaic symptoms were observed in butternut squash plants in Bali and molecular detection using universal primer of Tobamovirus indicated the presence of TMV infection. Further research was conducted to determine molecular characteristics of TMV on butternut squash plants in Bali. Specific DNA bands of Tobamovirus were amplified using reverse transcription polymerase chain reaction method, followed by DNA sequencing. The DNA were successfully amplified from CP Tobamovirus using universal primers from several butternut squash samples, i.e. Denpasar, Gianyar, Buleleng, and Karangasem Districts. The homology analysis of nucleotide and amino acid sequences of TMV among isolates from Denpasar, Gianyar, Buleleng, and Karangasem Districts was ranged between 95.6 – 97.7% and 98.1 – 99.4%, respectively. This indicated that low genetic diversity of TMV among Bali isolates. The highest homology of corresponding sequences of TMV isolates from Denpasar, Gianyar, Buleleng, and Karangasem Districts was closely related to TMV Kediri-Indonesia isolate on cucumber plant. Correspondingly, the phylogenetic analysis showed that TMV Bali isolates were categorized into same cluster with Kediri-Indonesia isolates. This was the first report of TMV on butternut squash in Indonesia.

Abstract

Mosaic disease caused by Cucumber mosaic virus (CMV) and Chilli veinal mottle Virus (ChiVMV) has been distributed widely in chilli in Indonesia and considered as important disease. A research was conducted to investigate the spread and incidence of CMV and ChiVMV in Rejang Lebong, Bengkulu and to identify its insect vector. Symptomatic and asymptomatic leaf samples were collected systematically from several chillipepper fields for further detection by DAS-ELISA (Double antibody sandwich-enzyme linked immunosorbant assay) using specific antibody for CMV and ChiVMV. The result showed that infection of both CMV and ChiVMV was found with disease incidence reached 20-50%, whereas infection only by ChiVMV or CMV were 50-80% and 20-50%, respectively. One species of aphid, i.e. Aphis gossypii was found from the fields.Key words: Aphis gossypii, CMV, ChiVMV, disease incidence

Abstract

Cowpea mild mottle virus (CPMMV) is one of important virus which infects soybean and become an endemic disease since the first time reported in Java and Sumatra. Research was conducted to study the host range of CPMMV and to evaluate response of new type soybean varieties to CPMMV specific isolate.  Host range study of CPMMV CR16 isolate was conducted by sap transmission method to 11 species of indicator plants, whereas evaluation of soybean response involved 10 varieties.  The resistance response of soybean variety was based on incubation period, and of either  the disease incidence or severity. Virus infection on test plants were confirmed by dot immunobinding assay (DIBA) using specific CPMMV antisera. The CPMMV CR16 isolate was able to infect systemically 8 plant species belongs to Leguminoceae, and Solancaeae; while the virus caused local infection on Amaranthaceae. All plant species infected systemically proved to become sources of inoculum for CPMMV when they used in back inoculation to soybean.  Response of soybean varieties to CPMMV CR16 isolate can be categorized into susceptible (Detam 1, Detam 2, Detam 3, Anjasmoro, Wilis), moderated (Detam 4, Malika, Dena 1) and resistant (Argomulyo and Grobogan).

Abstract

Ledakan penyakit mosaik kuning yang disebabkan oleh Mungbean yellow mosaic India Begomovirus (MYMIV) terjadi pada tanaman kacang panjang (Vigna unguiculata subsp. sesquipedalis) di Jawa pada 2008. Namun penyakit ini belum dilaporkan di daerah lainnya di Indonesia. Tanaman kedelai di Cirebon, Jawa Barat, Bantul, DI Yogyakarta, dan Musi Banyuasin Sumatera Selatan menunjukkan gejala sistemik berupa mosaik kuning pada daun dengan tulang daun lebih gelap (vein banding) dan diduga terinfeksi MYMIV. Deteksi dan identifikasi virus dilakukan dengan metode polimerase chain reaction (PCR) dengan menggunakan primer spesifik MYMIV. Hasil amplifikasi menghasilkan pita DNA berukuran 1000 pb. Hasil ini menunjukkan bahwa tanaman kedelai yang bergejala mosaik kuning dari Cirebon, Bantul, Musi Banyuasin, dan positif terinfeksi MYMIV. Hasil analisis homologi sikuen nukleotida tanaman kedelai yang terinfeksi MYMIV asal Cirebon mempunyai homologi yang tinggi (98.6-99.2 %) dengan MYMIV dari Tegal, Subang, Klaten, Magelang, Brebes, dan Bogor yang menginfeksi tanaman kacang panjang. Analisis filogenetik menunjukkan bahwa isolat MYMIV Cirebon berada satu kelompok dengan semua sikuen MYMIV di Indonesia dan Oman, namun terpisah dengan kelompok MYMIV dari Bangladesh, Nepal, Pakistan dan India. Sikuen isolat MYMIV Cirebon telah didaftarkan ke GenBank dengan nomor aksesi LC481009. Sebelumnya MYMIV dilaporkan pada kedelai di Jawa tahun 2013. Temuan ini merupakan laporan pertama infeksi MYMIV pada kedelai di luar Jawa (Musi Banyuasin, Sumatera Selatan).

Abstract

[DETECTION OF TUNGRO VIRUS ON RICE IN BENGKULU]. Several types of viruses have been reported to infect rice plants in Indonesia, including tungro, dwarf grass, and empty dwarfs. The spread of tungro in Indonesia in the beginning was only limited to certain regions in South Sulawesi, South Kalimantan, West Nusa Tenggara and North Sulawesi, but later it expanded to East Java, Central Java and Yogyakarta. In this study a tungro disease survey was conducted in several areas in Bengkulu Province. Virus detection is done by Loop-medated isothermal amplification (LAMP). Observations in the field showed that there were typical symptoms of tungro infection in rice plants, namely orange leaves starting from leaves to 2 and 3, little tillers, stunted plant growth. The results of detection with LampPCR showed rice samples from the district Pondok Kelapa Kab Bengkulu Tengah is positively infected with tungro virus.

Abstract

[ELIMINATION OF Pineapple Mealybug Wilt-associated Virus (PMWaV) FROM PINEAPPLE PLANT BY HOT WATER TREATMENT]. Mealybug wilt of pineapple (MWP) is a devastating disease found in all the major pineapple growing regions. Closterovirus particles can be detected in both MWP symptomatic and asymptomatic pineapple. Rogue of symptomatic plants is the simplest method of controlling MWP disease, but this method is less effective as PMWaV might remains at asymptomatic plants. Use of pesticide to control mealybug and ants is not efficient and high cost. The research was conducted to develop elimination method for PMWaV-free plant by hot water treatment. PMWaV infected plant parts (leave, stem, crown) were exposed to two hot water treatments, consisting of 35 ºC for 24 hour as pre-treatment and immediately followed by hot water treatment either 56 ºC for 60 minute or 58 ºC for 40 minute in a water bath. The infected plant without hot water treatment served as the positive control, and healthy plant without hot water treatment as the negative control. PMWaV infection could be eliminated from propagative material through hot water treatment 58 ºC 40 minute in a water bath without decrease propagative material viability.

Abstract

Banana is a horticulture crop that has economic value and is widely cultivated in tropical countries. Banana production in Bengkulu province reached 259,748 quintals, then durian (110,387 quintals), tangerines (94,396 quintals) (BPS 2015). Banana bunchy top disease caused by Banana bunchy top virus (BBTV) infection is considered the most crucial virus disease affecting yield losses of a banana plantation in Asia, Africa, and the South Pacific. However, the incidence and molecular characters of BBTV has never been reported in Bengkulu. This research aims to characterize symptom variations, disease incidence, and disease severity of BBTV infection in Bengkulu and virus detection using molecular methods by polymerase chain reaction (PCR). Disease incidence of BBTV was measured based on field symptoms. The disease survey was conducted in Bengkulu city, Bengkulu Utara district, and Rejang Lebong district. The study showed that the incidence of BBTV in Bengkulu City, Bengkulu Utara, and Rejang Lebong ranged from 0% to 100%. The most common symptoms observed in the field involved vein clearing, upturned leaf, chlorotic, and ragged margins, reducing petiole length, distance, lamina width, and stunting. Banana crops that are infected with BBTV in the vegetative phase will not produce fruit. In contrast, viral infection in the generative phase causes the formation of stunted fruit that is not suitable for harvesting. Thus, the potential loss of yield due to stunted disease can reach 100%. This study's results are the first reports of BBTV infection in banana crops in Bengkulu. Disease diagnosis will form the basis of disease control strategies in banana crops.

Abstract

The tobacco mosaic virus (TMV) is a Tobamovirus that can infect cucumber plants. There is limited information about the response of cucumber varieties against TMV infection. The research was conducted to evaluate the response of cucumber varieties to TMV infection using TMV isolates from Bogor. The evaluation of resistance to TMV isolates was conducted by sap transmission method using TMV isolates from Bogor that propagated on tobacco as the source of inoculum. Variable observations covered the period of incubation (the day after inoculation/DAI), the symptoms of the disease, disease incidence, and disease severity. The presence of viral infection was observed through the accumulation of starch and inclusions bodies. Based on the results of the research, the TMV isolate was able to infect systematically all tested plants with symptoms of which appear in the form of mosaic light to heavy with the incidence of the disease reaching 100%. Response of cucumbers varieties to TMV isolate can be categorized into resistant (Bandana F1, Neptun, and Pandu), tolerant (Ethan F1), and susceptible (Vario F1).

Abstract

Bawang merah merupakan salah satu komoditas hortikultura strategis dan bernilai ekonomi tinggi. Ketersediaan bawang merah dipengaruhi oleh luas lahan budidaya dan kendala organisme pengganggu tanaman. Saat ini Kementerian Pertanian menetapkan perluasan daerah budidaya bawang merah di berbagai daerah di Indonesia, salah satunya adalah Kabupaten Rejang Lebong dan Kabupaten Kepahiang di Provinsi Bengkulu. Petani membutuhkan informasi teknik budidaya yang tepat. Kegiatan ini dilakukan bertujuan untuk memperkenalkan teknik budidaya bawang merah dan kemudian dilanjutkan dengan melakukan evaluasi sehingga diperoleh persepsi petani tentang budidaya bawang merah yang diperkenalkan. Petani antusias mengikuti kegiatan penyuluhan serta pengamatan di lapangan, Hasil evaluasi kegiatan menunjukkan bahwa sebagian besar petani memberikan persepsi yang baik terhadap teknologi budidaya bawang merah yang diperkenalkan. Kata kunci: Bawang Merah, Pengenalan, Budidaya, Evaluasi ABSTRACT Shallots are one of the strategic horticultural commodities and have high economic value. The availability of shallots is influenced by the area of cultivated land and the constraints of plant-disturbing organisms. At present the Ministry of Agriculture is establishing expansion of shallot cultivation areas in various regions in Indonesia, one of which is Rejang Lebong Regency and Kepahiang Regency in Bengkulu Province. Farmers need information on proper cultivation techniques. This activity was carried out aiming to introduce the technique of shallot cultivation and then proceed with an evaluation to obtain farmers' perceptions about the cultivation of shallots that were introduced. Farmers were enthusiastic in participating in the extension activities as well as observations in the field. The results of the evaluation showed that most farmers gave a good perception of the shallot cultivation technology introduced. Keywords: Shallot, Introducing, Farming, Evaluation
