Abstract

Papaya is one type of fruit that is widely consumed and cultivated by the farmer. However, genetic analysis has not been carried out on various types of papaya available on the market. This aims to determine the genetic diversity of papaya plants that can become genetic resources to fullfil food needs and genetic resources for breeders. Genetic analysis was conducted by Random Amplified Polymorphic DNA (RAPD) method using 11 papaya varieties consisting of Calina, Bangkok, Red Pomegranate, Sunrise, Orange Lady, Red Lady, Taiwan, Arum, Miba, Golden, and Local and using 15 RAPD primers consisting of OPA-1, OPA-2, OPA-8, OPA-16, OPC-4, OPC-11, OPC-13, OPC-20, OPD-20, OPE-2, OPE-6, OPE-11, OPE-14, OPM-6, and OPY-15. PCR-RAPD results were translated into binary data and then cluster analysis was conducted using the Unweighted Pair-Group Method Arithmetic (UPGMA) method using the Numerical Taxonomy and Multivariate System (NTSYS) program. The PCR-RAPD results of 11 papaya varieties successfully amplified 8 out of 15 primers that formed 112 DNA bands with 85 polymorphic bands. Genetic diversity analysis showed the results at 90% similarity coefficient formed 9 groups. Group 1 consists of Bangkok and Sunrise varieties. Group 2 consists of Red Pomegranate and Arum varieties. Groups 3 to 9 consisted of Calina, Miba, Local, Golden, Orange Lady, Red Lady, and Taiwan varieties, respectively.

Abstract

[TUNGRO DISEASE ON RICE PLANT (Oryza sativa) IN TABA PENANJUNG DISTRICT: DISEASE INCIDENCE AND MOLECULAR DETECTION OF VIRUSES]. Tungro is one of the important diseases of rice plants. Tungro disease caused by Rice tungro bacilliform virus (RTBV) and Rice tungro spherical virus (RTSV). This research aims to obtain information on the spread of the disease, vector population, and molecular detection by polymerase chain reaction method. This research was conducted from December 2019 to April 2020 in rice fields in Taba Penanjung sub-district, Bengkulu Tengah district at ± 540 meters above sea level and virus detection in the Laboratory of Biotechnology and Genetics Department of Biology Faculty of Mathematics and Natural Sciences (MIPA) and Laboratory of Plant Protection, Agriculture Faculty, Bengkulu University. Data analysis used descriptive analysis. The results showed that the highest disease incidence on the seven weeks after planting (wap) is 1.7% and the highest population of the vector on the 7 wap with an average of 7.7 vector/100m2 in the village of Taba Penanjung. Detection of viruses with the PCR method with the specific primer DAF/DAR succeeded in amplification of DNA band measuring 1,400 bp. RTSV was not detected by RT- PCR using a specific primer or RTSV. The result of this detection shows that the symptoms of tungro on rice plants in the cultivation of Taba Penanjung district associated with RTBV infection. RTSV unable to detect can be caused by many factors in the detection process, including total RNA extraction, cDNA synthesis, DNA amplification, and DNA visualization of PCR products.

Abstract

[MOLECULAR DETECTION AND GENETIC ANALYSIS OF BEGOMOVIRUS ON CHILLI IN PEMATANG DONOK VILLAGE]. Many symptoms of yellow curly leaf disease, but there have been no reports of viral species infected the chili plants.   The research was aimed  to determine the disease incidence of yellow leafcurl disease and detection of Begomovirus infected chilli plants in Kepahiang regency based on  polymerase chain reaction (PCR). Sampling was done by purposive sampling method based on the symptoms; yellow leafcurl, leaf malformation, and stunting. Virus detection with PCR method using general primer Begomovirus (SPG 1/2). The PCR product was sent to First Base, Malaysia for DNA sequencing. Sequencing data were analyzed using MEGA 6 (Molecular Evolutionary Genetics Analysis) software for phylogenetic tree construction. Based on field observations, the incidence of disease ranged from 96.4%-100% and the whitefly vector insect (Bemisia tabaci) was found. PCR using general primers Begomovirus obtained DNA bands measuring 912 bp according to the primary target. Based on the Blastn results, the nucleotide sequences of three Begomovirus samples from Pematang Donok Village had nucleotide similarities with the Pepper yellow leaf curl Indonesia virus (PYLCIV) isolates from Bali and Java contained in the GenBank database, with a nucleotide similarity value of 91.77%-98.99%.

Abstract

Pengetahuan petani yang rendah tentang bagaimana melakukan diagnosa kesehatan tanah dan tanaman mengakibatkan ketidakmampuan petani dalam memutuskan apa yang seharusnya mereka lakukan ketika melihat adanya kerusakan tanaman. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan petani tentang teknik diagnosa kesehatan tanah dan tanaman, serta meningkatkan keterampilan dalam pembuatan pupuk kompos. Kegiatan pengabdian ini berlokasi di Desa Tik Kuto Kecamatan Rimbo Pengadang Kabupaten Lebong, Provinsi Bengkulu. Khalayak yang menjadi sasaran kegiatan adalah petani tanaman hortikultura. Metode pendekatan kegiatan pengabdian dilakukan melalui dua cara, yaitu (1) penyuluhan tentang teknik diagnosa kesehatan tanah dan tanaman, serta (2) pelatihan keterampilan pembuatan pupuk kompos. Hasil kegiatan pengabdian menunjukkan bahwa tingkat pengetahuan petani tentang teknik diagnosa tanah meningkat dari skor rata-rata 1,50 (kurang tahu) menjadi 4,33 (tahu), sedangkan tingkat pengetahuan teknik diagnosa kesehatan tanaman meningkat dari skor rata-rata 1,42 (tidak tahu) menjadi tahu (4,17). Selanjutnya tingkat pengetahuan petani tentang pembuatan pupuk kompos juga mengalami peningkatan dari yang semula belum tahu sama sekali dan belum pernah membuat, setelah mengikuti kegiatan pengabdian menjadi tahu dan bisa membuat pupuk kompos dari bahan-bahan sampah organik, khususnya kulit kopi yang banyak ditemukan di Desa Tik Kuto, Kecamatan Rimbo Pengadang Kabupaten Lebong, Provinsi Bengkulu. Kegiatan pengabdian ini memberikan manfaat besar bagi petani, terutama untuk keberlanjutan usaha pertanian, sehingga ke depannya aktivitas pengabdian seperti ini perlu terus dilakukan.
