Abstract

[TUNGRO DISEASE ON RICE PLANT (Oryza sativa) IN TABA PENANJUNG DISTRICT: DISEASE INCIDENCE AND MOLECULAR DETECTION OF VIRUSES]. Tungro is one of the important diseases of rice plants. Tungro disease caused by Rice tungro bacilliform virus (RTBV) and Rice tungro spherical virus (RTSV). This research aims to obtain information on the spread of the disease, vector population, and molecular detection by polymerase chain reaction method. This research was conducted from December 2019 to April 2020 in rice fields in Taba Penanjung sub-district, Bengkulu Tengah district at ± 540 meters above sea level and virus detection in the Laboratory of Biotechnology and Genetics Department of Biology Faculty of Mathematics and Natural Sciences (MIPA) and Laboratory of Plant Protection, Agriculture Faculty, Bengkulu University. Data analysis used descriptive analysis. The results showed that the highest disease incidence on the seven weeks after planting (wap) is 1.7% and the highest population of the vector on the 7 wap with an average of 7.7 vector/100m2 in the village of Taba Penanjung. Detection of viruses with the PCR method with the specific primer DAF/DAR succeeded in amplification of DNA band measuring 1,400 bp. RTSV was not detected by RT- PCR using a specific primer or RTSV. The result of this detection shows that the symptoms of tungro on rice plants in the cultivation of Taba Penanjung district associated with RTBV infection. RTSV unable to detect can be caused by many factors in the detection process, including total RNA extraction, cDNA synthesis, DNA amplification, and DNA visualization of PCR products.

Abstract

[MOLECULAR DETECTION AND GENETIC ANALYSIS OF BEGOMOVIRUS ON CHILLI IN PEMATANG DONOK VILLAGE]. Many symptoms of yellow curly leaf disease, but there have been no reports of viral species infected the chili plants.   The research was aimed  to determine the disease incidence of yellow leafcurl disease and detection of Begomovirus infected chilli plants in Kepahiang regency based on  polymerase chain reaction (PCR). Sampling was done by purposive sampling method based on the symptoms; yellow leafcurl, leaf malformation, and stunting. Virus detection with PCR method using general primer Begomovirus (SPG 1/2). The PCR product was sent to First Base, Malaysia for DNA sequencing. Sequencing data were analyzed using MEGA 6 (Molecular Evolutionary Genetics Analysis) software for phylogenetic tree construction. Based on field observations, the incidence of disease ranged from 96.4%-100% and the whitefly vector insect (Bemisia tabaci) was found. PCR using general primers Begomovirus obtained DNA bands measuring 912 bp according to the primary target. Based on the Blastn results, the nucleotide sequences of three Begomovirus samples from Pematang Donok Village had nucleotide similarities with the Pepper yellow leaf curl Indonesia virus (PYLCIV) isolates from Bali and Java contained in the GenBank database, with a nucleotide similarity value of 91.77%-98.99%.

Abstract

Pengetahuan petani yang rendah tentang bagaimana melakukan diagnosa kesehatan tanah dan tanaman mengakibatkan ketidakmampuan petani dalam memutuskan apa yang seharusnya mereka lakukan ketika melihat adanya kerusakan tanaman. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan petani tentang teknik diagnosa kesehatan tanah dan tanaman, serta meningkatkan keterampilan dalam pembuatan pupuk kompos. Kegiatan pengabdian ini berlokasi di Desa Tik Kuto Kecamatan Rimbo Pengadang Kabupaten Lebong, Provinsi Bengkulu. Khalayak yang menjadi sasaran kegiatan adalah petani tanaman hortikultura. Metode pendekatan kegiatan pengabdian dilakukan melalui dua cara, yaitu (1) penyuluhan tentang teknik diagnosa kesehatan tanah dan tanaman, serta (2) pelatihan keterampilan pembuatan pupuk kompos. Hasil kegiatan pengabdian menunjukkan bahwa tingkat pengetahuan petani tentang teknik diagnosa tanah meningkat dari skor rata-rata 1,50 (kurang tahu) menjadi 4,33 (tahu), sedangkan tingkat pengetahuan teknik diagnosa kesehatan tanaman meningkat dari skor rata-rata 1,42 (tidak tahu) menjadi tahu (4,17). Selanjutnya tingkat pengetahuan petani tentang pembuatan pupuk kompos juga mengalami peningkatan dari yang semula belum tahu sama sekali dan belum pernah membuat, setelah mengikuti kegiatan pengabdian menjadi tahu dan bisa membuat pupuk kompos dari bahan-bahan sampah organik, khususnya kulit kopi yang banyak ditemukan di Desa Tik Kuto, Kecamatan Rimbo Pengadang Kabupaten Lebong, Provinsi Bengkulu. Kegiatan pengabdian ini memberikan manfaat besar bagi petani, terutama untuk keberlanjutan usaha pertanian, sehingga ke depannya aktivitas pengabdian seperti ini perlu terus dilakukan.

Abstract

This research was conducted to 1) Analyze the perception of consumers on the attributes of chili, and 2) Know which chili products are selected by consumers based on shape,  size and which are most wanted to buy. Forty consumer respondents were taken by accidental sampling technique.  Consumers are asked to assess 10 types of the UNIB chili varieties, both monoculture and combination polyculture between varieties. The results showed that the majority of respondents stated that the size and shape attributes were important as the basis for the decision to buy chili, while the attributes of uniform size and shape are considered normal.  The variety of chili which consumers like because of their size, shape and desire to be purchased were UNIB C H13. While for this type of combination, the combination of UNIB C H13 and UNIB C H53 is desired to be purchased by consumers. This type of combination should be cultivated by farmers because it is resistant to pests, high productivity and preferred by consumers.

Abstract

Gourami fish has been cultivated and marketed by the Amanah Sejati cooperative located in Bengkulu city. Gurami is sold as fresh product. The price is not comparable with the cost of gurami cultivation. Therefore, it is necessary to make an effort to increase the economic value of the gourami fish. One of the solutions is to make it into presto gourami products. The gourami fish is then vacuum-packed to improve the quality and extend its shelf life. Community service aims to (1) provide knowledge about the type of packages, and (2) provide knowledge about how to vacuum packaging on presto gourami. These community services are coordination and socialization, counseling, and practice. These services were going well. The participants were enthusiastic about the counseling and practice.

Abstract

Pepper yellow leaf curl Aceh virus (PepYLCAV) and pepper yellow leaf curl Indonesia virus (PepYLCIV) are begomoviruses that dominate chili cultivation in Indonesia. Characterization of these two begomoviruses is essential as basic information for the development of virus control technologies. The symptoms on chili plants indicate that PepYLCIV is more severe than PepYLCAV, with severe mosaic, curling, and yellowing, whereas PepYLCAV causes only mild mosaic and yellowing. Sequencing results also show that the nucleotides and amino acids between PepYLCAV and PepYLCIV are significantly different. This suggests that the pathogenicity of the two viruses on chili plants differs. Protein structure predictions using AlphaFold3 also show significant differences. Based on available data, control of PepYLCAV and PepYLCIV can be achieved using different approaches, as these viruses have distinct sequences that may affect their pathogenicity.

Abstract

Melon cultivation has increased rapidly in recent years and is frequently associated with widespread virus-like symptoms in Bengkulu Province, Indonesia; however, thereis no formal documentation of the mixed viral infections has been available. Therefore, this study aimed to detect major viruses infecting melon crops in Bengkulu Province and to evaluate their occurrence based on field symptoms and serological detection. Field surveys were conducted at three melon cultivation sites: Kandang Limun Village (Bengkulu City), Air Sebakul Village (Central Bengkulu Regency), and Tawang Rejo Village (Seluma Regency). Symptomatic leaf samples were collected purposively. Virus detection was performed using the Dot Immunobinding Assay (DIBA) with antisera specific to Cucumber mosaic virus (CMV), Tobacco mosaic virus (TMV), and Papaya ringspot virus (PRSV). The results showed that the dominant symptoms of viral infection in melon plants included mosaic, vein banding, chlorosis, leaf curling, and stunted growth in both Snow & Rock and Merlin F1 varieties. Disease incidence varied among varieties and locations, with the highest incidence recorded in Snow & Rock (73.3%). Serological detection revealed that 100% of symptomatic samples tested positive for CMV, TMV, and PRSV, confirming that all infections occurred as mixed infections. This study represents the first report of mixed viral infections in melon crops in Bengkulu Province. These findings demonstrate that overlapping and variable field symptoms cannot be attributed to a single virus and cannot be reliably distinguished through visual diagnosis alone, highlighting the importance of laboratory-based diagnostic approaches for accurate virus identification.

Abstract

Permasalahan utama budidaya hortikultura di Desa Tapak Gedung, Kabupaten Kepahiang, Bengkulu adalah tingginya ketergantungan petani terhadap pupuk dan pestisida sintetis serta rendahnya kemampuan petani dalam mendiagnosis penyakit tanaman secara tepat. Kondisi tersebut berdampak pada tingginya biaya produksi, risiko kesehatan, serta potensi kerusakan lingkungan. Kegiatan pengabdian kepada masyarakat ini bertujuan meningkatkan pengetahuan dan keterampilan petani dalam diagnosis awal penyakit tanaman hortikultura serta penerapan teknologi pengendalian hama dan penyakit terpadu berbasis agens hayati. Metode pelaksanaan meliputi penyuluhan, pelatihan, pendampingan, dan pembuatan demplot budidaya bawang merah dengan aplikasi agens antagonis Trichoderma sp. dan Streptomyces sp. Evaluasi dilakukan melalui pre-test dan post-test, observasi partisipatif, serta keberhasilan demplot. Hasil kegiatan menunjukkan peningkatan pemahaman petani mengenai perbedaan penyakit biotik dan abiotik, kemampuan petani dalam memperbanyak dan mengaplikasikan agens hayati, serta keberhasilan penerapan pengendalian hama dan penyakit terpadu pada demplot bawang merah dengan hasil panen yang sehat dan baik. Kegiatan ini berkontribusi pada penerapan pertanian hortikultura berkelanjutan yang ramah lingkungan dan berbasis hasil riset perguruan tinggi.

Abstract

Background: In Tik Kuto Village, organic household and agricultural waste remains largely underutilized, contributing to environmental pollution and potentially reducing community health status and productivity. The lack of practical, low-cost, and easily applied waste management technologies at the household and farm levels highlights the need for community-based interventions that support sustainable and environmentally friendly practices.Aims: The specific objective is to enhance community capacity to process organic waste into Local Microorganisms (MOL), Liquid Organic Fertilizer (POC), and ecoenzymes within a Zero-Waste Management framework.Methods: The program was implemented through participatory outreach and counseling, demonstrations, technical training, and mentoring activities involving farmer groups and housewives. Educational support materials included posters and booklets. The resulting MOL served as an activator for POC, which was fermented for four weeks. Ecoenzymes were produced from fruit peels, brown sugar, and water in a 3:1:10 ratio and fermented for a minimum of three months. Program effectiveness was evaluated using pre-tests, post-tests, and direct observation of participant engagement.Results: A total of 18 farmers participated in the program. Although most participants were middle-aged and experienced farmers, their baseline knowledge of organic waste processing was low. Post-training evaluation showed substantial improvements, with understanding of waste sorting and processing reaching 94–100%. Knowledge related to materials, production steps, application dosage, and benefits of MOL, POC, and ecoenzymes increased from 6–28% to 83–94%.Conclusion: The program successfully improved participants’ knowledge and behavioral intentions regarding organic waste management at the household and community levels.

Abstract

Petani hortikultura di Desa Sumber Sari, Kabupaten Kepahiang, masih bergantung pada pestisida sintetis untuk mengendalikan hama dan penyakit tanaman. Penggunaan pestisida sintetis secara terus menerus dapat menyebabkan resistensi organisme pengganggu tanaman, mencemari lingkungan, serta menimbulkan gangguan kesehatan bagi petani maupun konsumen. Kegiatan ini bertujuan untuk meningkatkan pengetahuan petani dalam mengenali gejala penyakit tanaman serta keterampilan petani dalam memproduksi dan mengaplikasikan biopestisida secara mandiri dengan menggunakan agens hayati Trichoderma sp. dan Streptomyces sp.. Metode kegiatan meliputi sosialisasi, penyuluhan, pelatihan teknis, praktik lapangan, serta pendampingan dan evaluasi melalui pre-test dan post-test. Pelatihan ini berhasil meningkatkan pengetahuan dan kapasitas teknis petani secara signifikan mengenai pengendalian hayati, dengan seluruh peserta (100%) menyatakan ketertarikan untuk menerapkan teknologi ini. Sebanyak 80% peserta memahami proses pembuatan biopestisida, dan 60% mampu mengaplikasikannya di lahan. Kegiatan ini menunjukkan bahwa pendekatan partisipatif yang menggabungkan teori dan praktik lapangan efektif dalam membangun kesadaran serta kemandirian petani menuju pertanian berkelanjutan. Perlu penguatan kelembagaan lokal dan pelatihan lanjutan serta fasilitasi akses terhadap bahan dan informasi guna memperluas adopsi teknologi pengendalian hayati di tingkat petani.
