Abstract

Uncaria gambir Roxb. is an industrial crop commodity that has a high economic value and good prospect for farmers and exporter. Gambier of traditional processed products (raw gambier) generally have various catechin content, so it is required further handling to increase the purity of catechins from raw gambier. The research was aimed to get a process technology on the making of pure gambier and standardized catechins. Sources of raw materials was obtained from extraction process through steaming leaves and twigs of gambier (KA) using equipment from aluminum (RA) and from iron (RF). Purification of raw gambier through repeated extraction with water could decrease tannin content from 24% to 2.4% and increase catechin levels between 40% to 74%. Gambier with a low tannin content (catechin KA) through re-extraction using water had the highest increase of catechin content with a color of yellowish white product. The iron-containing equipment affected the color of the pure gambier produced, which was reddish-brown. The effect of solvent on further extraction using ethyl acetate solvent resulted in higher catechin content (up to 99%) compare to ethanol (95%). The catechin purity of KA samples was tested by looking at its stability at varying levels of acidity, showing catechins of gambier was stabile at pH 6.ABSTRAK Uncaria gambir Roxb. merupakan komoditas tanaman industri yang memiliki nilai ekonomi tinggi serta mempunyai prospek cukup baik bagi petani dan pemasok negara-negara asing. Gambir hasil olahan tradisional (gambir asalan), umumnya memiliki kandungan katekin yang beragam, sehingga diperlukan penanganan lebih lanjut untuk meningkatkan kemurnian katekin dari gambir asalan. Tujuan penelitian adalah untuk mendapatkan teknologi proses pembuatan gambir murni dan katekin terstandar. Sumber bahan baku diperoleh dari proses ekstraksi melalui pengukusan daun dan ranting tanaman gambir (KA) menggunakan peralatan dari aluminium (RA) dan dari besi (RF). Pemurnian gambir asalan melalui ekstraksi berulang dengan air dapat menurunkan kadar tanin, yaitu dari 24% sampai 2,4% dan meningkatkan kadar katekin, yaitu antara 40% sampai 74%. Gambir dengan kandungan tanin rendah (katekin KA) melalui ekstraksi ulang menggunakan air memiliki peningkatan kadar katekin tertinggi dengan warna produk putih kekuningan. Peralatan mengandung besi berpengaruh terhadap warna dari gambir murni yang dihasilkan, yaitu berwarna coklat kemerahan. Pengaruh pelarut terhadap ekstraksi lanjut menggunakan pelarut etil asetat menghasilkan kadar katekin lebih tinggi (sampai 98%) dibandingkan etanol (95%). Kemurnian katekin sampel KA diuji dengan melihat stabilitasnya pada berbagai tingkat keasaman, menunjukkan katekin dari gambir stabil pada pH 6.

Abstract

Extract of Uncaria gambier contains phenolic compounds that have function as an antioxidants. Antioxidant content depends on the extraction conditions. To obtain high antioxidant activity of the phenolic compounds, the optimization process was carried out using response surface methodology (RSM) with three variables namely  the ratio of solvent (ethyl acetat : ethanol), the ratio of sample : solvent and time of extraction with the antioxidant activity DPPH (IC50) as response. Optimization result showed a quadratic polynomial regression equation of Y = 137.25 + 5.18 (R.solvent) + 24.75 (R.sample) + 11.53 (Time of.extrac) + 6.17 (R.solvent x sample) + 3.14 (Rsolvent x Time of.extrac) + 13.79 (R.sample x Time of.extrac) – 5.44 (R.solvent2) – 15.33 (R.sample2) – 5.42 (Time of.extrac2). The optimum response was achieved at the solvent ratio of ethyl acetat : ethanol = 0,37 : 0,55, ratio of sample : solvent = 1:2 and time of extraction 20 hours.  Validation result revealed that the predicted value (IC50 = 82.89 mg/mL) of this model was reasonably close to the experimental result (IC50 = 82,735 ± 0,362 mg/mL). Visual characterization of the extracts indicated that the extracts have different colour in each treatment, depending on the ratio of the solvent used.ABSTRAK Ekstrak dari Uncaria gambir mengandung senyawa fenolik katekin yang berfungsi sebagai antioksidan. Kandungannya pada hasil ekstrak bergantung pada kondisi ekstraksi yang digunakan. Untuk mendapatkan kondisi ekstraksi terbaik senyawa fenolik dari gambir dengan aktivitas antioksidan yang tinggi dilakukan proses optimasi menggunakan metode permukaan respon (RSM) pada tiga variabel yaitu rasio pelarut, rasio sampel dengan pelarut dan lama ekstraksi dengan respons aktivitas antioksidan terhadap antiradikal DPPH (IC50). Hasil optimasi menunjukkan persamaan kuadrat polinomial regresi yaitu Y = 137,25 + 5,18 (Rpelarut) + 24,75 (Rsampel) + 11,53 (Lamaeks) + 6,17 (Rpelarut x sampel) + 3,14 (Rpelarut x lamaeks) + 13,79 (Rsampel x lamaeks) - 5,44 (Rpelarut2) - 15,33 (Rsampel2) - 5,42 (Lamaeks2). Kondisi optimum dicapai pada rasio etil asetat dengan etanol  0,37:0,55, rasio sampel dengan pelarut 1:2 dan lama ekstraksi 20 jam. Hasil validasi menunjukkan nilai prediksi (IC50 = 82.89 mg/ml) dari model tidak berbeda signifikan dengan nilai hasil validasi (IC50= 82,735 ± 0,362 mg/ml). Karakteristik produk hasil ekstraksi secara visual menunjukkan warna yang berbeda dari setiap perlakuan dan tergantung dari rasio pelarut yang digunakan.

Abstract

Bioactive compounds in Eucheuma cottonii include phenols or polyphenols with antioxidant, antibacterial, and antiviral properties. Seaweed quality, including phenolic content, is highly determined by several factors, including the growth location, cultivation technique, and harvest age. This study aimed to determine the effect of harvest age and solvent type on the phenolic content of E. cottonii. The effects of harvest age on water content and of simplicia on the extraction process were also determined. Phenolic active ingredients were extracted at three harvest ages (35, 40, and 45 days) and using four solvents (ethanol, ethyl acetate, hexane, and water). Analysis of variance was performed to determine the effects of both factors and their interactions. The older the seaweed, the lesser the water content. The powder produced at all ages is a good simplicia for materials extraction. Results showed that high phenolic content was obtained from the E. cottonii extract at the age of 35 days with ethyl acetate as the solvent.

Abstract

This research evaluated the effect of fungal pretreatment of betung bamboo fibers and enzymatic- and microwaveassisted hydrolysis on the reducing sugar yield. The enzymatic hydrolysis of the pretreated biomass was carried out with cellulase and 10 and 20 FPU/g of substrate in a shaking incubator at 50 °C and 150 rpm for 48 h. The sulfuric acid concentration used in the microwave-assisted acid hydrolysis was 1.0, 2.5, and 5%, either with or without the addition of activated carbon. Microwave irradiation (330 Watt) was applied for 5–12.5 min. The yield of reducing sugar was better with the microwave-assisted acid hydrolysis, and the yield tended to increase with an increase in the irradiation time. Based on the dry weight of the initial biomass (bamboo), pretreatment with 5% inoculum loading resulted in a higher reducing sugar yield (17.06%) than with 10% inoculum loading (14.54%). At a 1% acid concentration, the formation of brown compounds decreased, followed by a reduction in the reducing sugar yield. The addition of activated carbon at a 1% acid concentration seemed to be of no benefit with respect to the yield in the microwaveassisted acid hydrolysis. The pretreatment with the 5% inoculum loading for 12.5 min at 1% acid concentration resulted in the highest reducing sugar yield. Under these conditions, the yield was 6.3-fold that of the reducing sugar yield using 20 FPU/g of cellulase. The rate of bamboo hollocellulose hydrolysis reached 22.75% of the maximum theoretical reducing sugar reducing sugar of dry biomass.

Abstract

Abstrak Bakteri mampu menghasilkan siderofor umtuk mengkelat besi dalam lingkungan dan digunakan oleh tanaman sebagai kofaktor dalam pembentukan klorofil untuk pembentukan glukosa. Penelitian ini bertujuan untuk mendapatkan bakteri penghasil siderofor tinggi dari akar nanas Simadu, Kabupaten Subang Provinsi Jawa Barat. Bakteri diisolasi dari akar nanas Simadu, kemudian diseleksi dengan membedakan morfologinya dan diidentifikasi sebagai penghasil siderofor pada media Chrome Azurol S (CAS). Diperoleh 10 isolat bakteri (M1 sampai dengan M10) yang mampu menghasilkan siderofor. Isolat bakteri M7 memiliki kemampuan menghasilkan siderofor tertinggi. Isolat M7 teridentifikasi sebagai bakteri Gram negatif. Hasil analisis pohon filogenetik berdasarkan sekuensing 16S rDNA menunjukkan isolat kelompok genus Providencia. Dibandingkan dengan Providencia vermicola ,  isolat standar dari InaCC yang berasal dari akar Curcuma zedoaria, M7 menunjukkan produksi siderofor yang lebih tinggi pada media LB pada kondisi aerobik.[Kata kunci: Isolasi bakteri, Providencia, akumulasi siderofor, akar nanas Simadu Subang]Abstract Bacteria can produce siderophores for chelating iron in environments and are used by plants as an ingredient cofactor in building chlorophyll for glucose production. This study aims to obtain high siderophore-producing bacteria from the roots of pineapple Simadu, Subang District, West Java Province. Bacteria were isolated from Simadu pineapple roots, then selected by distinguishing their morphology and identified as producing siderophores with Chrome Azurol S (CAS) media. Obtained 10 bacterial isolates (M1 to M10) capable of producing siderophores. Bacterial isolate M7 had the highest siderophore production ability. M7 isolate was identified as Gram-negative bacteria. The results of the phylogenetic tree analysis based on 16S rDNA sequencing showed this isolate as the genus Providencia. Compared to the Providencia vermicola as reference isolate from InaCC derived from the roots of Curcuma zedoaria, M7 showed higher siderophore production in LB media under aerobic conditions.[Keywords:          Bacteria isolation, Providencia, siderophore accumulation, Simadu Subang pineapple root]

Abstract

Since ancient times, honey has been utilized as a medication, while Trigona sp. honey is a special kind of honey. Breast, colon, and lung cancers are very malignant types of cancer and need to be treated with caution This investigation intended to establish the in vitro toxicity activity of eight Trigona sp. honey opposed to breast cancer (MCF-7), colon cancer (WiDr), and lung cancer (A549). Cytotoxicity using the MTT test technique (3,4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide). The honey used is Trigona sp. honey with Indonesian standards and the concentration of honey tested is 5.5%. The results obtained were that honey Banten 1 dan 2, Bogor, Bangka Belitung, Batu Ampar, Sumedang, Garut, and Klaten inhibited MCF-7 cancer cells by 89.46, 84.56, 77.96, 86.39, 84.58, 85.26, 88.24, 84.40; honey Banten 1 dan 2, Bogor, Bangka Belitung, Batu Ampar, Sumedang, Garut, and Klaten inhibited WiDr cancer cells by 83.22, 76.74, 68.85, 75.51, 78.53, 81.43, 81.96, 78.33; honey Banten 1 dan 2, Bogor, Bangka Belitung, Batu Ampar, Sumedang, Garut, and Klaten inhibited cancer cells A549 by 52.16, 0, 63.21, 85.92, 82.60, 84.68, 69.63, 56.38. It was found that all types of honey could inhibit cells except honey from Banten 2. The highest percentages of inhibition against MCF-7, A549, and WiDr cells were 89.46, 85.92, and 83.22 from honey Banten 1, Bangka Belitung, and Banten 1.

Abstract

Penicillin G acylase (PAc) hydrolyses of the amide bond of benzylpenicillin (Pen-G) releasing PAA and 6-APA, key intermediate in the production of various semisynthetic penicillins. In this study, we optimised the production medium of PAc by RSM using two variables (xylose as inducer and CaCl2 as divalent cations) to obtain the optimum PAc specific activity from Bacillus megaterium btpacBD1. For this purpose, combinations of five different xylose concentrations (0.13 – 0.87 %) and five different CaCl2 concentrations (0.64 – 4.36 mM) were analysed, in a total of 22 experiments. CCD used for the analysis showed that in shake flask cultivations, xylose and CaCl2 showed significant effects on PAc volumetric activity and the quadratic model was in good agreement with the experimental results (R2= 0.86 (p-value < 0.0001)). The maximum specific activity (130.669 ± 50.241 units mg protein-1) was reached when xylose and CaCl2 concentrations were 0.49% and 2.4 mM, respectively, and medium pH was around 7. Under such conditions, the activity of PAc and protein concentration achieved were 1.318 ± 0.406 units mL-1 and  0.0101 ± 0.01 mg mL-1. The shake flask validation experiments demonstrated that with such medium composition the volumetric activity, protein concentration and specific activity achieved were 1.294 ± 0.171 units mL-1, 0.0102 ± 0.0003 mg mL-1 and 125.91 ± 13.309 units mg-1, respectively. When the optimum medium composition was applied in 10 L bioreactor, the optimum volumetric activity (2.0687 ± 0.0820 units mL-1) and protein concentration (0.0078 ± 0.0008 mg mL-1) were achieved 48 h after the start of the cultivation. However, the optimum PAc specific acivity (1260.52  ± 27.5711 units mg protein-1) was achieved 18 h after the start of the cultivation.

Abstract

The demand for halal products in the Islamic context continues to be high, requiring adherence to halal and haram laws in consuming food and beverages. However, individuals face the challenge of distinguishing between haram meat and permissible halal meat. This study aims to answer these challenges by designing an expert system application within the ERP framework to increase the usability functionality of the system that can differentiate between beef, pork, or a mixture of both based on the physical characteristics of the meat. The aim is to determine halal products permissible for consumption by Muslims. The research methodology includes a data collection process that involves taking 30 meat samples from various sources, and the criteria used to classify the meat will be determined based on an analysis of the physical characteristics of the meat. System administrators use expert systems to ensure proper treatment of meat during administration processes, including separating halal beef from pork and implementing different inventory procedures. The Certainty Factor (CF) inference engine deals with uncertainty even though the expert system's accuracy level is relatively good with several rules. However, these results must be studied further because the plan relies on expert opinion. Therefore, it is necessary to set the correct CF value for accurate height classification. The CF inference engine facilitates reasoned conclusions in meat classification. Functional testing confirms the smooth running of the system, validating its reliability and performance. In addition, the expert system accuracy assessment produces a commendable accuracy rate of 90%. In addition, the expert system works powerfully on various meat samples, accurately classifying meat types with high precision. This study explicitly highlights the expert system's design for meat classification in determining halal products by using the Expert System Certainty Factor. In conclusion, this expert system provides an efficient and reliable approach to classifying meat and supports the production and consumption of Halal products according to Islamic principles.

Abstract

Breadfruit (Artocarpus communis Forst.) is one of sources for ethanol production, which has high starch content (89%). Ethanol production from breadfruit starch was conducted by Simultaneous Saccharification and Fermentation (SSF) technology using microbes consortium. The aim of the research was to examine a method to produce ethanol by SSF technology using microbes consortium at high yield and efficiency. The main research consisted of two treatments, namely normal SSF and enginereed SSF. The results showed that normal SSF using aeration and agitation during cultivation could produce ethanol at 11.15 ± 0.18 g/L, with the yield of product (Yp/s) 0.34 g ethanol/g substrate; and yield of biomass (Yx/s) 0.29 g cell/g substrate, respectively. A better result was obtained using engineered SSF in which aeration was stopped after biomass condition has reached the end of the exponential phase. The ethanol produced was 12.75 ± 0.04 g/L, with the yields of product (Yp/s) 0.41 g ethanol/g substrate, and the yield of cell (Yx/s) 0.09 g cell/g substrate.

Abstract

The fungal pretreatment effect on chemical structural and morphological changes of Betung Bamboo was evaluated based on its biomass components after being cultivated by white rot fungi, Trametes versicolor. Betung bamboo powder (15 g) was exposed to liquid inoculum of white rot fungi and incubated at 270C for 15, 30 and 45 days. The treated samples were then characterized by FT-IR spectroscopy, X-Ray diffraction and SEM-EDS analyses. Cultivation for 30 days with 5 and 10% loadings retained greater selectivity compared to that of the other treatments. FTIR spectra demonstrated that the fungus affected the decreasing of functional group quantities without changing the functional groups. The decrease in intensity at wave number of 1246 cm-1 (guaiacyl of lignin) was greater than that at wave number of 1328 cm-1 (deformation combination of syringyl and xylan) after fungal treatment. X-ray analysis showed the pretreated samples had a higher crystallinity than the untreated ones which might be due to the cleavage of amorphous fractions of cellulose. The pretreated samples have more fragile than the untreated ones confirmed by SEM. Crystalline allomorph calculated by XRD analysis showed that fungus pretreatment for 30 days has transformed triclinic structure of cellulose to monoclinic structure.
