Abstract

Pakcoy (B. rapa L.) merupakan salah satu sayuran yang digemari karena rasanya enak, renyah, kaya akan vitamin serta memiliki nilai komersial. Produksi pakcoy mengalami peningkatan beberapa tahun terakhir. Peningkatan produksi pakcoy ini harus diimbangi dengan ketersediaan lahan. Hidroponik merupakan salah satu solusi dalam mengatasi keterbatasan lahan. Penelitian ini bertujuan untuk membandingkan pertumbuhan pakcoy yang ditanam di dalam dan di luar greenhouse. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 2 perlakuan, yaitu budidaya pakcoy di dalam greenhouse dan di luar greenhouse. Masing-masing perlakuan diulang sebanyak 2 kali, dan setiap ulangan terdiri dari 9 sampel tanaman pakcoy. Parameter yang diamati meliputi tinggi tanaman, jumlah daun, luas daun, berat segar, dan berat kering. Hasil penelitian menunjukkan bahwa terdapat perbedaan signifikan pada tinggi, luas daun, berat basah, dan berat kering antar perlakuan. Sedangkan jumlah daun tidak memiliki perbedaan signifikan. Rata-rata tertinggi pengukuran pada tanaman pakcoy di luar greenhouse.

Abstract

AbstractGlutathione S-Transferase is a superfamily enzyme that has many roles for living things, especially in the detoxification of Reactive Oxygen Species (ROS), one of which is rice plants. One gene of this family that has a role for plants is GSTL2 This gene is known to have a contribution to Arabidopsis and Oryza sativa L. against abiotic stress. To find out how this gene expression is requires a primer to specifically amplify this gene, and an optimal annealing temperature to support the success of the primer. This study aims to design primers and determine the optimal annealing temperature for gene amplification GSTL2. Primers were designed using the Primer3, which were then analyzed with Genious Prime based on good primer criteria and optimizing the annealing temperature which was carried out using gradient PCR. The results of this study obtained a primer pair is forward 5’-TTCGAAGGGCCAGCATTACT-3’ primer reverse 5’-CAATGTCCACCAAGCTGAA-3’. The primer pair has a length of 20 nt, with melting temperature (Tm) 59–59,4 °C, and GC content 50%. The primer forward there is a secondary structure in the form of a self-dimer with (Tm) 6.2 °C. The primer pair can amplify gene sequences GSTL2 by producing a PCR product 224 bp. The annealing temperature of 60 °C resulting in a single thick, bright DNA strand.AbstrakGlutathione S-Transferase merupakan enzim superfamily yang memiliki banyak peranan bagi makhluk hidup terutama dalam detoksifikasi Reactive Oxygen Species (ROS), salah satunya tumbuhan. Salah satu gen dari famili ini yang memiliki peranan bagi tanaman adalah GSTL2. Gen ini diketahui memiliki kontribusi bagi tanaman Arabidopsis dan Oryza sativa L. terhadap stres abiotik. Untuk mengetahui bagaimana ekspresi gen GSTL2 ini dibutuhkan primer untuk mengamplifikasi gen ini secara spesifik, di samping itu suhu annealing yang optimal untuk menunjang keberhasilan primer. Penelitian ini bertujuan untuk mendesain primer dan menentukan suhu annealing yang optimal untuk mengamplifikasi gen GSTL2. Primer didesain menggunakan tools Pick Primer dan Geneious Prime, yang kemudian dianalisis secara in silico berdasarkan kriteria primer yang baik. Serta optimasi suhu annealing yang dilakukan menggunakan gradient PCR. Hasil penelitian ini diperoleh sepasang primer dengan panjang masing-masing primer 20 nt, primer forward 5’-TTCGAAGGGCCAGCATTACT-3’ primer reverse 5’-CAATGTCCACCAAGCTGAA-3’. Pasangan primer dapat mengamplifikasi sekuen gen GSTL2 dengan menghasilkan produk PCR sebesar 224 bp pada suhu annealing 60 °C.

Abstract

Agriculture is a vital sector in meeting global food needs. The fulfillment of food needs has environmental challenges that cause plants to be exposed to abiotic stress, namely salinity stress. The purpose of this study was to determine the effect of auxin spraying concentration on rice given salt stress. The concentration of auxin sprayed was 25 µm, 37.5 µm, 50 µm, while the concentration of salt given was 0 µm, 15 µm, 30 µm, 45 µm. This research is an experimental research. The research was conducted at the Basic Biology Laboratory, Department of Biology, FMIPA, Universitas Negeri Padang Parameters observed were root length, crown length, relative water content, and dry weight. The results showed that the root length showed significant results after a two-way ANOVA test which means that there is an influence on root length while the crown length, relative water content, and dry weight did not show significant results.

Abstract

Anak usia sekolah memerlukan asupan energi dan zat gizi untuk mendukung pertumbuhan dan juga prestasi belajar anak di sekolah. Salah satu sumber asupan energi dan zat gizi bagi anak sekolah bersumber dari pangan jajanan. Anak usia sekolah sebagai generasi penerus bangsa merupakan investasi bagi suatu bangsa. Kualitas anak-anak pada masa sekarang akan menentukan kualitas suatu negara di masa depan. Edukasi keamanan pangan jajanan perlu dilakukan sejak dini secara sistematis dan juga berkelanjutan sebagai salah satu upaya untuk peningkatan kualitas sumber daya manusia. Pemberian nutrisi yang berkualitas, sehat dan sesuai dengan kuantitas yang seharusnya penting untuk menunjang pertumbuhan perkembangan anak usia sekolah agar optimal. Oleh karena itu, perlu adanya edukasi kepada siswa SD untuk meningkatkan pengetahuan terkait keamanan pangan jajanan dan mengetahui arti simbol-simbol pada bungkus jajanan. Kegiatan ini dilaksanakan pada bulan Juli 2022, dan diikuti oleh 206 peserta dari kelas 3 dan 6 SDN 02 Payakumbuh. Hasil evaluasi kegiatan menunjukkan adanya peningkatan pengetahuan siswa terhadap keamananan jajanan dan memiliki keterampilan dalam mengidentifikasi simbol-simbol pada bungkus jajanan. Kesimpulan untuk kegiatan ini sudah efektif dilaksanakan dan dapat mengedukasi siswa SD dalam  mengetahui keamanan pangan jajanan.

Abstract

Purpose of the research was to describe the result of VIII Grade Students' Science Literacy Skills analysis in Junior High School in Padang. It was a descriptive research. Samples were taken by using purposive sampling technique. Techniques of data collection used valid and reliable science literacy test and interview with students and teachers. Technique of data analysis was by calculating average score of students' science literacy test. To test data validity, it used triangulation technique. The finding shows that VIII grade students' science literacy skill in SMPN 1 Padang based on total score, basic competence and science competence has the highest average score with low category. Then, it is followed by VIII grade students' science literacy skill in SMPN Ar-risalah Padang and SMP Manjushri Padang with low category. In conclusion, VIII grade students' science literacy skill in Junior High School in Padang is in low category.

Abstract

– Coconuts, a major commodity in West Sumatra, often produce wasted coconut water, which causes pollution. To address this issue, training was conducted for member of the Ikhwatunnisa Women Farmers Group on how to make nata de coco from this waste. Nata de coco, a functional food, is produced by fermenting coconut water using Acetobacter xylinum bacteria. The training aimed to empower Ikhwatunnisa Women Farmers Group members with skills to start businesses, create jobs, and increase family income. The training included lectures, demonstrations, and hands-on practice, covering everything from preparation to packaging. Evaluations showed high participant enthusiasm; 85% gained new insights and planned to produce nata de coco independently. This activity is expected to encourage new businesses and transform coconut water waste into a valuable economic product.

Abstract

Abstract – SD Negeri 12 Padang Panjang is one of the schools that has made efforts to implement the Adiwiyata program. In 2024, the school represented the city of Padang Panjang in the Adiwiyata competition at the West Sumatra provincial level; however, it did not achieve optimal results. For the 2025 Adiwiyata program, SD Negeri 12 has set a target to become the provincial representative of West Sumatra. Based on the evaluation results from 2024, it was found that the assessment indicator for Participatory-Based Environmental Activities needs improvement. One activity that can support this effort is hydroponic cultivation. A hydroponic cultivation training was conducted by 26 fifth-grade students. The training included both theoretical material and hands-on practice in cultivating green vegetables using a hydroponic system. The training was guided by two experts who are competent in the field. This training successfully increased students' knowledge of hydroponic vegetable cultivation. Moreover, over 90% of the participants demonstrated teamwork and developed a greater awareness of environmental care. This outcome is a significant contribution to supporting SD Negeri 12 Padang Panjang Barat’s efforts toward achieving Adiwiyata School status.

Abstract

At the present time, pressures on local ecosystems put many endemic species at risk of extinction if conservation measures are not promptly implemented. This community service program aimed to empower tourism awareness groups (Pokdarwis) in Singgalang Tandikek Nature Park through biodiversity education and photography training to support conservation-based ecotourism. The program targeted local guides, providing knowledge on endemic and protected species, particularly the Sumatran striped rabbit (Nesolagus netscheri), combined with practical photography skills using smartphones and ethical documentation practices. Activities included biodiversity lectures, hands-on photography, species identification, and social media promotion of local biodiversity. Evaluation using pre-test and post-tests showed an increase in participants’ photography knowledge (mean score 7.57 to 8.71), indicating improved awareness and skills in documenting biodiversity. The program successfully enhanced local capacity to promote sustainable ecotourism while fostering conservation ethics.
