Abstract

      The rpoB gene has been known to be a surrogate marker for detection of Multidrug Resistant Tuberculosis (MDR-TB). Mutations of rpoB gene were associated with rifampicin (RMP) resistance occur in 81 bp region called Rifampin Resistance-Determining Region (RRDR). This study was aimed to determine the types of mutations and amino acid differences in isolates of M.tuberculosis MDR compared to M.tuberculosis H37Rv. Amplification of rpoB gene fragments was carried out using nested Polymerase Chain Reaction (nested PCR). Analysis of mutation using MEGA4 showed two amino acid changes namely, E418D and H526L. Abstract

Kelompok ibu PKK Banjar Tohpati merupakan organisasi wanita yang menjadi bagian Desa Kesiman Kertalangu. Terjadinya pandemi COVID-19 telah memberikan dampak yang signifikan bagi masyarakat Bali termasuk bagi kelompok ibu PKK Banjar Tohpati khususnya dari segi kesehatan dan ekonomi. Berdasarkan hasil diskusi, mitra ingin memiliki pengetahuan tentang cara pembuatan makanan sehat yang murah dan mudah serta memiliki kandungan gizi yang tinggi untuk menjaga imunitas selama masa pandemi COVID-19. Mitra juga mengharapkan makanan yang dibuat dapat memberikan manfaat ekonomi bagi mitra. Berdasarkan permasalahan mitra ini dan hasil diskusi disepakati untuk melakukan pemberdayaan kelompok ibu PKK Banjar Tohpati untuk membuat yoghurt sebagai panganan sehat karena telah terbukti secara ilmiah memberikan manfaat kesehatan dan cukup mudah dibuat. Selain itu, yoghurt dapat menjadi produk yang dapat dipasarkan dan memberikan manfaat ekonomi bagi mitra. Metode yang digunakan dalam PKM ini adalah focus group discussion melalui pemaparan tentang konsep gizi sehat disertai dengan pelatihan pembuatan yoghurt. Mitra mendapatkan penyuluhan dari sisi ekonomi tentang cara membuat memasarkan produk dengan metode promosi online. Pengusul telah memfasilitasi mitra dengan alat dan bahan pembuatan yoghurt sebagai modal awal untuk memulai usaha dan bantuan hidup dasar untuk meringankan dampak pandemi COVID-19. Hasil evaluasi pre dan post test menunjukkan peningkatan nilai sebesar 38 point atau 44.2%. Hal ini mengindikasikan bahwa mitra paham akan konsep gizi sehat dan pemasaran online. Hasil monitoring dan evaluasi menunjukkan mitra sudah mampu secara mandiri membuat prodiuk yoghurtnya sendiri dan sudah memasarkan dilingkungan tempat tinggal mereka. Mitra juga mampu memasarkan yoghurt menggunakan media sosial seperti WhatsApp dan Facebook.

Abstract

Sentul (Sandoricum koetjape) is a tropical plant that has been used as traditional medicine in some Asian countries for decades. Research on phytochemicals and pharmacological activities of this plant extracts has been conducted and shows promising medicinal properties. This review aims to integrate knowledge about S. koetjape focusing on three main aspects namely ethnomedicinal, phytochemicals, and pharmacological, in order to encourage further research on this plant for future drug development. Traditionally, all plant parts of S. koetjape have been used for treating various health problems and diseases such as diarrhea, fever, colic, and leucorrhoea. More than 30 chemicals have been identified from S. koetjape, which the most important compounds are ring-A secotriterpene, oleane-type triterpene, secomultiflorane-type triterpene, hydroxymultiflorane triterpene, and limonoids. In vitro studies showed pharmacological potential of the extracts and phytochemicals constituents of S. koetjape including antibacterial, antifungal, antitumoral, anticancer, insecticide, and antioxidant.

Abstract

In this study we reported cultivation of bacteria associated with mangrove soil from the Ngurah Rai Mangrove Forest, Bali. Mangrove soil samples were serially diluted using sterile artificial seawater, spread onto Starch Casein M agar and incubated at 28oC for 28 days. Cultivation of mangrove soil samples yielded 165 bacterial colonies with 68 isolates were selected and purified based on different morphology. Of these 68 isolates, 22 isolates displayed antibacterial activities ranging from weak to strong inhibition against at least one of four bacterial indicators namely Staphyloccocus aureus, Streptococus mutans, Escherichia coli and Klebsiella pneumoniae using perpendicular streak method. Overall, 19 out of 22 bacteria isolates displayed weak antibacterial potential and two isolates exhibited moderate antibacterial activity. The isolate SA4 was the only bacterium with strong antibacterial potential with measured clear distance ≥ 10 mm against the four bacterial isolates. Sequence analysis based on 16S rRNA gene fragment assigned the isolate SA4 as Bacillus subtilis strain BIL/BS-168. Overall, this study confirmed the untapped potential of antibacterial activities from bacteria associated with mangrove soil.

Abstract

The increasing rate of antimicrobial resistance in the past decades has motivated the search for novel antibacterial compounds to overcome infectious diseases. Among diverse natural sources, mangrove ecosystems offer untapped sources of biological active compounds for future antibacterial medicine. This research was aimed to evaluate antibacterial activities of crude extracts of four dominant mangrove plants from the Ngurah Rai Mangrove Forest namely Rhizophora mucronata, Avicennia marina, Rhizophora apiculata, and Sonneratia alba. Roots of these four plants were extracted using methanol, chloroform, and n-hexane. These crude extracts were tested against two Gram positive bacteria (Staphylococcus aureus and Streptococcus mutans) and two Gram negative bacteria (Escherichia coli dan Klebsiella pneumoniae) by disc difussion assay. We found that 3 mg/mL of N-hexane crude extracts from R. apiculata yielded the highest zone of inhibition of 8.64 mm against S. aureus. While, 3 mg/mL of chloroform crude extract of R. apiculata yielded the highest inhibition of 19.83 mm against S. mutans. Unfortunately, no zone of inhibition was observed when crude extracts were tested against Gram negative indicator strains. Our results indicate that the root crude extracts of R. apiculata yielded the highest zone of inhibition against Gram positive indicator strains compared to root crude extracts of R. mucronata, S. alba, and A. marina. Further research is required to determine the antibacterial activities of the mangrove crude extracts against other bacterial indicator strains to determine their spectrum of activities.

Abstract

Research has been conducted to amplify and identify mutations in the rpoB gene fragment, 0,5 kb, from the isolate 134 Mycobacterium tuberculosis multidrug resistance (MDR). Amplification was performed using the method nested polymerase chain reaction (nested PCR) while sequencing was conducted in one direction using forward inner primer. Nucleotide sequence obtained was translated into amino acids using MEGA4 program. Amplification of M.tuberculosis rpoB gene fragment, 0,5 kb, was successfully carried out and sequenced. Alignment result by using MEGA4 program showed that there had been missense mutations in the rpoB gene. It had two amino acids changes to rpoB protein, they were glutamic acid to aspartic acid at codon 418 and glutamine to arginine at codon 510.

Abstract

Good and sufficient nutritional intakes are prerequisite to build a strong immune system against infectious diseases especially during the pandemic of COVID-19 that spread out widely since the early year of 2020. Currently socialization about COVID-19 have been emphasized on the preventive aspects such as washing hand with soap properly, using face mask and applying social distancing. However, these preventive aspects have not much focused on nutritional aspects. In order to build awareness of society on good nutritional intakes, this community service focused to build a group of cadres in collaboration with the women group (PKK) of Banjar Lebah Desa Sumerta Denpasar. The method of socialization was conducted via face to face meeting by still applying health protocols such as using face mask, washing hand with soap and physical distancing. Evaluation of the community service activity showed that cadres have displayed significant understanding about good nutritional intakes as indicated by increasing of post-test grades to 35% compared to the initial pre-test grades. These cadres were also successfully made yoghurt as the example of nutritional foods that can be consumed daily. In the future, these cadres were expected to actively socializing the importance of good nutritional intakes for their neighbourhood especially during the pandemic of COVID-19. Abstract

Kanker payudara dan kanker serviks merupakan jenis kanker dengan prevalensi dan angka kematian tertinggi pada wanita. Namun, deteksi dini ternyata dapat menurunkan angka kematian akibat kanker payudara secara signifikan dan skrining yang efektif serta pengobatan lesi pra-kanker dapat menurunkan angka kejadian kanker serviks. Oleh karena itu, pemberdayaan kelompok-kelompok masyarakat dalam meningkatkan kesadaran akan pentingnya pengetahuan mengenai deteksi dini gejala dan tanda, serta langkah pencegahan kanker payudara dan kanker serviks sangat dibutuhkan. Anggota kelompok masyarakat yang diberdayakan diharapkan dapat menjadi kader yang dapat membantu program pemerintah dalam memberikan edukasi mengenai deteksi dini kanker payudara dan kanker serviks kepada anggota kelompoknya, kelompok masyarakat lain, serta keluarga kader. PKMS ini memberikan pelatihan kepada kelompok penjual canang di Pasar Kreneng Denpasar mengenai penyakit kanker payudara dan kanker serviks beserta metode deteksi dini kanker tersebut. Abstract

Swamedikasi atau pengobatan sendiri tanpa resep dokter telah dilakukan secara luas oleh masyarakat untuk mengobati berbagai penyakit ringan seperti sakit kepala, demam, batuk pilek, dan lainnya. Namun dalam pelaksanaannya, sering terjadi kesalahan pengobatan karena diagnosis sendiri yang kurang tepat, kesalahan dosis/cara pakai, informasi yang bias dari iklan obat, dan sebagainya. Tata laksana swamedikasi yang tidak rasional dapat menimbulkan dampak negatif seperti efek terapi yang tidak tercapai maupun munculnya efek obat yang tidak diinginkan. Oleh karena itu, dilakukan kegiatan PKM kepada mitra yang merupakan kelompok PKK Banjar Tohpati, Desa Kesiman Kertalangu, Denpasar Timur mengenai Gerakan Masyarakat Cerdas Menggunakan Obat (Gema Cermat) yang meliputi cara memilih dan mendapatkan obat dengan benar, menggunakan obat secara rasional, serta cara menyimpan dan membuang obat dengan baik dan benar. Metode yang digunakan adalah focus group discussion, penyuluhan dan pemutaran video terkait Gema Cermat, serta pelatihan penggunaan obat sediaan khusus seperti tetes mata, salep mata, injeksi insulin, dan suppositoria. Hasil pre-test dan post-test menunjukkan peningkatan pengetahuan kader mengenai Gema Cermat sebesar 20%, dari nilai rata-rata 64 saat pre-test menjadi 80 saat post-test. Kegiatan pemberdayaan masyarakat dalam program Gema Cermat ini diharapkan dapat membantu pemerintah dan masyarakat dalam meningkatkan keterampilan dan pengetahuan masyarakat dalam penggunaan obat secara benar.

Abstract

Introduction: Surgical site infection (SSI) is the most common nosocomial infection, affecting one out of three patients undergoing surgical procedures. The incidence of SSI in clean surgical wounds is still higher, especially in inguinal hernia repair. Today, tension-free mesh hernia repair is the primary method used in inguinal hernia management to reduce the risk of recurrence. This study aims to analyze preoperative factors influencing the incidence of SSIs in inguinal hernia patients that underwent tension-free mesh hernia repair at several regional public hospitals in Bali.Methods: This is a case-control study with samples taken from seven regional hospitals in Bali. Data used in this study were obtained from patients’ medical records from 2017-February 2021. Samples included in this study are inguinal hernia patients that underwent tension-free mesh hernia repair and postoperative check-up. Samples are selected through the purposive sampling method. Statistical analysis was done using SPSS 18.0 software. Univariate and bivariate analysis was performed, and Pearson chi-square and Fisher’s exact test were applied.Results: From the 61 samples in the case group and 61 samples in the control group, the results showed a significant relationship between the incidence of SSIs and elderly patients (OR = 2.262, 95% CI 1.086-4.711), between SSIs and obesity (OR = 9.057, 95% CI 1.096-74.808), and between SSIs and diabetes mellitus (OR = 9.620, 95% CI 2.094-44.201).Conclusion: It is concluded that there is a significant relationship between these preoperative factors and the incidence of SSIs in inguinal hernia patients that underwent tension-free mesh hernia repair at seven regional public hospitals in Bali.
