PENINGKATAN LAJU PELARUTAN TRIMETOPRIM MELALUI METODE KO-KRISTALISASI DENGAN NIKOTINAMIDA Zaini, Erizal; Halim, Auzal; Soewandhi, Sundani N.; Setyawan, Dwi

Abstract

Co-crystallization of trimethoprim with nicotinamide had been done using solvent (methanol as a solvent) and melted technique. Kofler’s hot contact methode was used to identify the solid state interaction between these two components. The solid phase was characterized by microscopic, powder X-ray diffraction, thermal DTA and FT-IR spectroscopy analysis. Dissolution rate profile was performed by  paddle methode (Type II USP), distilled water as a medium. Solid state interaction between trimethoprim and nicotinamide show a formation of  conglomerate (simple eutectic) at eutectical point 125 0C. Dissolution rate of co-crystallization product of trimethoprim and nicotinamide increase  significantly compare to physical mixture and intact trimethoprim. ABSTRAK Telah dilakukan ko-kristalisasi trimetoprim dengan bahan tambahan nikotinamida dengan metode pelarutan (menggunakan pelarut metanol) dan peleburan. Metode kontak panas Kofler digunakan untuk identifikasi awal pembentukan interaksi antar kedua komponen. Padatan hasil ko-kristalisasi dikarakterisasi  dengan analisis mikroskopik, difraksi sinar-X, termal DTA dan spektrofotometer FT-IR. Uji laju pelarutan dilakukan dengan metode dayung (tipe II USP) dengan medium air. Hasil interaksi menunjukkan pembentukan konglomerat (eutektikal) antara kedua fase kristalin dalam keadaan padat, dengan titik eutektik pada temperatur 125 0C. Laju pelarutan trimetoprim hasil ko-kristalisasi meningkat secara signifikan dibandingkan dengan campuran fisika dan senyawa tunggal trimetprim.

Abstract

The degradation of a plastic film containing polymer syntheticpolystyrene (PS) and biopolymer poly(3-hydroxybutyrate)[P(3HB)] and poly (3-hydroxybutyrate-co-3-hydroxy valerat)[P(3HB-co-3HV)] in various composition in aquous solution by in-vitro and in-vivo has been carried out. The testing was conducted based on immersion test in aquadest, river water, pH 4 solution, 6,8 and 10. Rate degradation measured use linear regression equality and parameter is time degradation 50% (t 50%), time degradation 95% (t 95%) and indicated by the slope (k). The result showed that, degradation of a plastic film containing PS and P(3HB) in pH 6 solution occurred faster, followed by ratio PS/P(3HB) 95:5, PS/P(3HB) 90:10, PS/P(3HB) 85:15 and PS/P (3HB) 80:20, in contrast degradation of pure PS (100:0) not occured. It is also observed that the biodegradation plastic film containing PS/P(3HB-co-3HV) in river water ratio PS/P(3HB-co-3HV) 95:5, PS/P (3HB-co-3HV) 90:10 and PS/P (3HB-co-3HV) 85:15, in contrast degradation of pure PS (100:0) not occurred.Keywords : biodegradation, polystyrene, poly(3-hydroxybutyrate), poly (3-hydroxybutyrate-co-3-hydroxyvalerat), plastic film.

Abstract

ABSTRACT : Characterization of Inclusion Complex Simvastatin - β - cyclodextrin prepared by Kneading Method" has been done . This study aims to improve the dissolution rate and characterize inclusion complexes simvastatin - β - cyclodextrin developed into 3 formula , the ratio between the simvastatin and β - cyclodextrin following formula I ( 1 : 1 ), the formula II ( 1 : 2 ), and the formula III ( 2 : 1 ). Inclusion complex prepared by adding water solvent to form a paste and then dried at room temperature. The results of the inclusion complexes were characterized by analysis Differential Thermal Analysis ( DTA ), X-ray diffraction, IR spectrophotometry, Scanning Electron Microscopy ( SEM ), and dissolution rate . The results of this analysis indicate that the inclusion complex formed, characterization and dissolution profiles better than the pure simvastatin and mixed physics simvastatin and β - cyclodextrin.   Key words : Inclusion complex, Simvastatin, β-Cyclodextrin, Kneading Methods   ABSTRAK : Telah dilakukan penelitian dengan judul "Karakterisasi Kompleks Inklusi Simvastatin - β-Siklodekstrin yang dibuat dengan Metoda Kneading". Penelitian ini bertujuan untuk meningkatkan laju disolusi serta mengkarakterisasi kompleks inklusi Simvastatin - β-Siklodekstrin yang dikembangkan menjadi 3 formula, dengan perbandingan antara simvastatin dan β - siklodekstrin sebagai berikut formula I (1 : 1), formula II (1 : 2), dan formula III (2 : 1). Komplek inklusi dibuat dengan cara menambahkan pelarut air hingga terbentuk masa seperti pasta dan kemudian dikeringkan pada suhu kamar. Hasil komplek inklusi ini dikarakterisasi dengan analisa Differensial Thermal Analysis (DTA), Difraksi Sinar X, Spektrofotometri Infra Red, Scanning Electron Microscopy (SEM), dan disolusi. Hasil analisa tersebut menunjukkan bahwa kompleks inklusi yang terbentuk memberikan karakterisasi dan profil disolusi yang lebih baik dibandingkan dengan simvastatin murni dan campuran fisika simvastatin dan β-siklodekstrin yang dibuat dengan penggerusan biasa. Abstract

Solid-state interaction between parasetamol and ibuprofen was studied using thermal and spectroscopic techniques. Parasetamol-Ibuprofen binary mixtures were prepared by rotary ball mill grinding method. Differential scanning calorimetry was used to investigate the stoichiometry and thermal properties of the complex between parasetamol and ibuprofen. Powder X-ray diffraction were used to investigate the type of interaction between the two components. Two endothermic peaks in a binary mixture of paracetamol and ibuprofen was observed in DSC thermograph. X-ray powder diffractogram do not shows shift and formation of new peaks and the position of each peak is a super-imposition of each component without dominates. It can be concluded that the solid interaction between paracetamol and ibuprofen for grinding effect is the formation of mixed crystal system.

Abstract
