Abstract

This research aimed to know the bioactivity of polysaccharide krestin (PSK) with a different timing on formation of mice antibody exposed by Mycobacterium tuberculosis. Mycobacterium tuberculosis was used as infection agent that had an abilitiy to be imunogenic and to lead immune system responses decreased. Polysaccharides krestin was used as immunopotentiator which had a role to activate macrophage and stimulate B cell in order to produce antibody. This research involved 30 adult female mice of Mus musculus strain Balb/C in the age of 8–10 weeks and in the weight between of 30–40 g. Mice were divided into six groups, as follows: group K as a control, was added only aquades; group K+ as positive control, was added only PSK; group K- as negative control, was exposed by M. tuberculosis only; group P1 was added PSK before being exposed by M. tuberculosis; group P2 was added PSK after being exposed by M. tuberculosis; group P3 was added PSK before and after being exposed by M. tuberculosis. Polysaccharides krestin was added by gavage with single dose of 500 μg and M. tuberculosis was exposed intraperitoneally with concentration of 5x108 bactery per ml. The bioactivity of PSK was observed by formation antibody using indirect ELISA test. The results of research were the average of OD value of K group was 0,590 ± 0,042, K+ group was 0.641 ± 0.025, K- group was 1.044 ± 0.054, P1 group was 1.032 ± 0.125, P2 group was 1.127 ± 0.042, P3 group was 1.230 ± 0.097. The conclusion of this research showed that the adding of PSK increased mice formation antibody. The time of adding before and after being exposed M. tuberculosis was the most potential to raise antibody production on mice.

Abstract

The aim of this research was to increase sum of total rat’s leucocytes and macrophages using Coriolus versicolor exposure 2-Methoxythanol (2-ME). Coriolus versicolor extract have been primary active compounds are polysaccharide krestin (PSK). This compound PSK contains of ß-glucan. The immune cells had b-glucan receptor. From this reason, Coriolus versicolor extract could increase of immune system after exposed 2-ME. This research used 30 of 3–4 month female rats, and they were divided into 5 groups. K (control), P1 (exposed by 2-ME), P2 (administered with Coriolus versicolor extract before exposed by 2-ME), P3 (administered with Coriolus versicolor extract after exposed by 2-ME), P4 (administered with Coriolus versicolor extract before and after exposed by 2-ME). Coriolus versicolor extract used dose of 300 mg/kg body weight was given with gavage. 2-ME dose of 11 mmol/kg body weight was given with injection at cavum peritoneum. Number on leucocytes and macrophages counted by using Haemocytometer Improved Neubauer. The data was analyzed with ANOVA, further more if any differences were analyzed with LSD. Result of this study showed that (1) P1 decreased on the total number of leucocytes if compared with K, but increased on the total number of macrophage. (2) P2, P3, and P4 increased the total number of leucocytes and macrophage. Suggestion that can be proposed from this research was Coriolus versicolor extract can repair and increase immune system.

Abstract

Saprolegniasis was a primary fungal disease that attacks fish eggs and fish. This disease was caused by Saprolegnia sp. The infections of Saprolegnia sp. in fish eggs can be minimized with dip treatment in formalin solution. This esearch was the control of saprolegniasis on red nila eggs (Oreochromis sp.) with formalin. This research used 400 red nila eggs. It was dipped suspension of Saprolegnia sp. (OD550 = 0,5) for 36 jam. The treatment was divided into four groups. Each group used 10 replication and each treatment with 10 eggs. The eggs in group 1 as control were not treated. The egg in-group 2, 3, and 4 were dipped formalin for five minute. The concentration of formalin in-group 2, 3, and 4 were 4 ml/L, 5 ml/L, and 6 ml/L. These data was taken after the treatment and the investigating parameter was the mean percentage of totally eggs that not infected by Saprolegnia sp. These data was analyzed by ANAVA and LSD at α = 5%. The results of this experiment showed that the increasing of formalin concentrations at each treatment caused significantly decreasing at the growth of Saprolegnia sp. on the red nila fish eggs. The control (1) group was not decreasing of the growth of Saprolegnia sp. The growth of Saprolegnia sp. in-group 2 decreased until 82%. The growth of Saprolegnia sp. in-group 3 decreased until 92%. The growth of Saprolegnia sp. in-group 4 decreased until 96%.

Abstract

Extract of testis contains the testis specific proteins. The testis proteins may raise immune responses. The immune responses are the antibodies against testis specific anti-protein. The reaction between the antibodies and the sperms can inhibit fertilization. This process has contraception effects. This research used 24 female and 20 male mice strain Balb/c. The experiment used the complete random design with 4 concentration treatments (0, 500, 1000, and 2000 μg extracts of testis) and 3 replications. Mice were injected 3 times with time interval 21 days. After 15 days from the rising of vaginal plug, 3 mice were dissected and counted the implantation. The others mice were waited until delivered of the children. The datas were analyzed by ANOVA and LSD (α = 5%). The results showed that extract of testis did not influence the amount of implantation at the uterus endometrial, but it decreased the amount of children. The effective concentration of extract testis is 2000 μg. So, this study showed that the intraperitoneum injection of the extract of testis had contraception effects.

Abstract

The aim of this study was to know the specific expose period of the Lymnaea sp.’s embyos and their age in order to utilization eggs of Lymnaea sp. in bioassay. Fifty individuals of Lymnaea sp. collected from Gonggan River Magetan were captured in laboratory. Egg cluster of each individual then was placed in petry dish which was filled by Baycarb 500 EC of 0.0001, 0.001 and 0.01 ppm. The result showed that 12 hours exposure in Baycarb 500 EC was the effective time since within 6 hours all of embryos were death and abnormalities of the survivals was detected in the embryos of 48 and 72 hours.

Abstract

An enzyme-linked immunosorbent assay has been developed which detect Streptococcus sp. Twenty Clarian gariepinus at the age of two weeks was soaked in Streptococcus sp. suspension with concentration of 108 bacteria per ml for two hours. Specimens such as blood, mucous, muscles, heart, kidney, liver, intestines and gill were collected, and assayed for the presence of Steptococcus sp. using ELISA. Result of present study showed that ELISA can be applied to detect an isolated Streptococcus sp. from tissues of Clarias gariepinus. Kidney is a primary target organ (predilection). Therefore, the kidney is a specimen of choice for diagnosis approach (es) of Streptococcus sp. infection.

Abstract

Staphylococcus aureus is an opportunistic pathogen and cause of nosocomial infection, which has leukocidin and can reduce immunity. Because of antibiotic resistance, immunomodulator is an alternative treatment for S. aureus infection. Polysaccharide krestin (PSK) from Coriolus versicolor extract contains active β-glucan that triggers immune responses' effectiveness including phagocytosis. This research aimed to know the activity of PSK against phagocytosis ability on mice infected by S. aureus. There were six treatment groups. 100 mg/kg BW PSK was given to the mice strain Balb/C by gavage. S. aureus was infected once every two weeks. Phagocytosis ability consisted of phagocytic activity and capacity, counted on slide smears of mice intraperitoneal fluid. The results showed that PSK increased phagocytic activity in the group giving PSK after infection. Furthermore, the most effective giving PSK on phagocytic capacity was before and after infection. Based on this research, PSK increased immunity by phagocytosis ability and can be useful as an immunomodulator.

Abstract

Squamocin is one of the annonaceous acetogenins produced by the Annonaceae family and displays potent anti-cancer activity against cancer cell lines. This study aimed to investigate the growth inhibition activity of squamocin coupled with nanodiamond on rats (Rattus norvegicus)-induced breast cancer. Twenty-five female R. norvegicus were divided into five groups (n = 5), including normal control (without any treatment), negative control, group treated with nanodiamond only (ND), group treated with squamocin only (SQ), and the group treated with squamocin coupled with nanodiamond (NDSQ). All of the animal models were induced for breast cancer, except for the normal control group. Breast cancer induction was performed using two doses of N-nitroso-N-methylurea (NMU) injection (50 and 30 mg/kg body weight) intraperitoneally and waited for 22 weeks until the tumor was detected to formed. Nanodiamond coupled with squamocin were administered by intraperitoneal injection (1.5 mg/kg body weight) for 5 weeks, one injection per 3 days. This study showed that the treatment with squamocin coupled with nanodiamond (NDSQ) significantly reduced the proliferation (Ki-67) and induced apoptosis (Caspase-3) of breast cancer cells, corresponding to the reduction of the thickness of the mammary ductal epithelium (p<0.001) and the lower level of CA-153 in serum. In addition, the treatment significantly reduced the malondioldehyde (MDA) and PI3KCA and increased the p53 level significantly. Altogether, in this study, we are the first to report the anti-cancer activity of squamocin in rat-induced breast cancer and the potency of nanodiamond as a carrier of squamocin to increase its anti-cancer activity.

Abstract

This research is to look for the influence of loyalty identity in the form of perceived value, experience quality and patriotism in museums that have historical value in Surabaya, East Java. The number of respondents was 160 respondents where respondents before visiting the museum had obtained information through social media. The results of data processing show that all the influence tests are said to be significant, both direct and indirect influence tests. The existence of significant influence test results can be stated that the predictor variable of loyalty can be a good antecedent because it is important to be input for the tourism industry which focuses on historical museums, so that managerial implications can help how to manage and provide satisfaction to visitors so that visitors besides feel the good atmosphere, also get the experience and desire to revisit.

Abstract

ABSTRAKTuberkulosis masih menjadi permasalahan yang belum bisa teratasi bagi seluruh dunia. Banyak upaya yang telah dilakukan dalam penanganan dan belum mendapatkan hasil yang diharapkan. Salah satu penyebab kesulitan dalam penanganan tuberkulosis paru adalah karena pasien tersebar di wilayah yang luas pada saat menjalani pengobatan dan belum terpetakan. Melalui bantuan kader kesehatan penulis mencoba untuk memetakannya. Metode pengabdian masyarakat yang digunakan adalah berupa pendidikan dan pelatihan dalam menggunakan aplikasi berbasis android dalam pemetaan dan identifikasi kontak pasien TB Paru di kabupaten Jombang melalui kerjasama dengan TB Care Yayasan Bhanu Yasa Sejahtera (YABHYSA) kabupaten Jombang dan diikuti oleh 40 orang kader pendeteksi dan pengendali TB dibawah naungan TB Care YABHYSA Jombang. Kegiatan diawali dengan penyamaan persepsi pada tim pengabdian masyarakat, kemudian kader sebagai sasaran dikumpulkan dalam satu ruangan untuk menerima penjelasan tentang aplikasi sekaligus mempraktekkan bagaimana cara menggunakannya. Hasil pelatihan menunjukkan nilai post test 100% responden lebih baik daripada pre test. Aplikasi diserahkan kepada kader untuk dapat digunakan dalam pemetaan pasien TB Paru. Berdasarkan hasil post test dapat disimpulkan bahwa kader telah mengerti dan memahami cara penggunaan aplikasi sehingga siap untuk memetakan pasien TB Paru di Kabupaten Jombang. Kata kunci: pencegahan TB; aplikasi, pemetaan. ABSTRACTTuberculosis is still an unresolved problem for the whole world. Many efforts have been made in handling and have not received the expected results. One of the causes of difficulties in treating pulmonary tuberculosis is because patients are spread over a large area at the time of treatment and have not been mapped. Through the help of health cadres the author tries to map it. The community service method used is in the form of education and training in using an android-based application in mapping and identifying contacts of pulmonary TB patients in Jombang district through collaboration with the Bhanu Yasa Sejahtera Foundation (YABHYSA) Jombang district TB Care and attended by 40 TB detection and control cadres under the auspices of TB Care YABHYSA Jombang. The activity began with the equalization of perceptions on the community service team, then the cadres as targets were gathered in one room to receive an explanation about the application as well as practice how to use it. The results of the training showed that the post test scores of 100% of the respondents were better than the pre test. Applications are submitted to cadres to be used in mapping pulmonary TB patients. Based on the results of the post test, it can be concluded that the cadres understand and understand how to use the application so they are ready to map pulmonary TB patients in Jombang Regency. Keywords : TB prevention; application; mapping
