Abstract

Sungkai plants are beneficial immunomodulators because of the presence of flavanoid compounds that increase oxygen radicals and antibody production. Pharmaceutical preparations function as immunomodulators, typically in the form of syrups, tablets, capsules, suspensions, and emulsions. Until now, there has been no nanoparticle syrup on the market, even though nanoparticle preparations have the advantage of modifying particle size, controlling the release of active ingredients to reach the active side, protecting drugs from degradation, reducing toxicity, and minimizing side effects. The purpose of this study was to determine whether the syrup of nanoparticles in the ethanol extract of Sungkai leaves acts as an immunomodulator and the stability of the preparation during storage. The methods used were experimental in the laboratory with extraction using the maceration method. The extract obtained was formulated into a nanoparticle solution using the ionic gelation method, then formulated into FO syrup (Base syrup) and F1 syrup (Nanoparticles of ethanol extract of sungkai leaves) and then carried out transmittance tests, particle size analyzer tests, immunomodulatory tests and stability tests. The yield of the extract was 17.5%. The raw material examination of all the materials was in accordance with the literature. The particle size of the nanoparticle solution was 40 nm, that of F0 syrup was 670 nm, and that of F1 syrup was 964 nm. All syrup preparations fell into the nanoparticle size category of 10–1,000 nm. Abstract

Natural Deep Eutectic Solvent (NaDES) is an environmentally friendly extraction method to obtain soybean bioactive compounds, focusing on genistein compounds as drug candidates. The use of environmentally friendly extraction solvents could support green extraction to ensure the safety of natural medicinal candidates. HBA (Hydrogen Bonding Acceptor), yellow choline chloride (supplement in animal feed), and HBD (Hydrogen Bonding Donor) lactic acid. A UV-Vis spectrophotometer was used to detect genistein. MoE 2022.02 software was used in the molecular docking simulation, and the docking scoring methods affinity ΔG and GBVI/WSA (induced fit) were used. The PDB ID used was: 5nn8 (alpha glucosidase) and PDB ID: 7vsi (SGLT-2 Inhibitor). The results of genistein were obtained by 92,670 mg (0.9267%) in the 75 0C, 30 min ultrasonic NaDES extraction in HBD lactic acid. Genistein exhibited an affinity for the 5NN8 (alpha-glucosidase) and 7VSI (SGLT-2 Inhibitor) receptors of -6,230 and -8,768, respectively. These affinity values did not exceed the interaction values of the native ligands acarbose (alpha-glucosidase) and Empagliflozin (SGLT-2 Inhibitor), which were -8,988 and -12,302, respectively. Genistein compounds had the lowest RMSD value of 0.819 at 7vsi (SGLT-2 Inhibitor). These results suggested the possibility of a genistein pathway as a candicate diabetes drug. The NaDES extraction method demonstrated great potential for development into a green action that supported the green extraction process, and genistein was an isoflavone compound that could be a candidate for diabetes drugs.

Abstract

Excessive exposure to ultraviolet (UV) rays can cause degenerative skin diseases, such as skin cancer, white spots, redness, and sunburn. One way to protect the skin from UV exposure is by using sunscreen. This is especially important for children as it helps protect their skin from harmful sunlight. The Senduduk plant (Melastoma malabathricum L.) has antioxidant properties and can capture free radicals, thereby helping to prevent degenerative diseases. This plant is also easily found in the area surrounding SD Negeri 43/IV Kota Jambi. Therefore, the aim of this community service activity is to educate students at SD Negeri 43/IV Kota Jambi about the potential of senduduk leaves to be formulated into an infusion spray to protect the skin from sun exposure. The methods used were lectures and live demonstrations on how to make the sunscreen spray preparation. The results showed an increase in students' knowledge about sunscreen and how to make the senduduk leaf infusion spray. The implication of this activity is that children gain an early understanding of degenerative diseases, particularly those affecting the skin.
