Abstract

Background: Collagen plays a crucial role in post-extraction wound healing. Thunnus albacares skin, a collagen source synthesized through the papain soluble collagen (PaSC) method with varying NaCl concentrations, is assessed for non-toxicity using the MTT assay on BHK-21 fibroblast cell cultures. This research is to determine cell viability resulting from synthesizing collagen powder from the skin of yellowfin tuna (Thunnus albacares) with varying NaCl concentrations using the papain soluble collagen (PaSC) method. Method: Collagen powder was synthesized by cleaning Thunnus albacares skin measuring 1x1 cm, soaking it in 0.1 M NaOH, hydrolyzing it with CH3COOH, extracting it with papain enzyme, and then dividing the samples into four groups: without NaCl, NaCl 0.9 M, NaCl 1.3 M, and NaCl 1.7 M. Samples were centrifuged (6000 rpm) and followed by freeze-drying. Cell viability was obtained by conducting a cytotoxicity test using the MTT Assay method on BHK-21 fibroblast cells. Result: The percentage of cell viability in groups K, P1, P2, P3, and P4 were 100%, 10.708%, 113.750%, 107.833%, and 105.958%, respectively. The Kruskall-Wallis test yielded a significance value 0.000, indicating a significant difference (p<0,005). The Mann-Whitney test confirmed significant differences between groups. Conclusion: Collagen powder from Thunnus albacares skin with NaCl concentrations of 0.9 M, 1.3 M, and 1.7 M showed no toxic effects, while the group without NaCl showed toxic effects. Collagen powder with a NaCl concentration of 1.7 M yielded ideal results and showed no toxic effects.

Abstract

ABSTRACTIntroduction: The skin of yellowfin tuna (Thunnus albacares) contains high protein, which could potentially be a halal collagen product. Collagen extraction using the papain-soluble collagen method has the advantage of producing a higher collagen yield compared to the acid method. Particle size, one of the physical properties of collagen, plays a crucial role in its efficacy in dentistry. This study aims to analyze the particle size of collagen powder synthesized   from Thunnus albacares skin using the papain-soluble collagen method, with varying concentrations of NaCl. Methods: Type of research is an experimental laboratory. Thunnus albacares skin was synthesized by chopping, cleaning, and soaking in a 0.1 M NaOH solution. The extraction process used the enzyme papain and 0.5 M of acetic acid. Samples were divided into four groups, each with different concentrations of NaCL: 0, no NaCL;, 0.9 M NaCl, 1.3 M NaCl, and 1.7 M NaCl. After centrifugation, the samples were freeze-dried. The particle size of collagen powder was measured using a Particle Size Analyzer test tool. The data collected was then analyzed using the Mann-Whitney test. Results: Particle size distributions are as follows: K group (3.36 nm), P1 (1.842 nm), P2 (3.36 nm), and P3 (10.12 nm). There is a significant difference in groups K-P1 and P3, P1-P2 and P3, and P2-P3 (p<0.05). However, there is no significant difference in groups K-P2 (p>0.05). Conclusion: Particle size of this research produced nano-sized collagen powder, with the lowest particle size observed in the 0.9 M NaCl group, measuring at 1.842 nm. The particle size increased in the group without NaCl and in the 1.3 M NaCl group at 3.36 nm, and reached the highest value in the 1.7 M NaCl group at 10.12 nm.

Abstract

Abstract

Background: Chitosan synthesized from Penaeus monodon shells was developed into a chitosan-gelatin suspension as an injectable bone substitute for socket preservation. Purpose: To investigate the characteristics of chitosan from P. monodon shells and their influence on the viscosity of a chitosan-gelatin suspension. Methods: P. monodon shells from Tarakan Waters were prepared using three methods: Group 1) deproteinization-depigmentation-deacetylation, Group 2) demineralization-depigmentation-deacetylation, and Group 3) deproteinization-demineralization-depigmentation-deacetylation. The chitosan was characterized by morphology, moisture and ash content, molecular weight (MW), deacetylation degree (DD), and viscosity. This chitosan was made into a chitosan-gelatin suspension with a ratio of 45:55 (w/w%) (95 ml:110 ml). The differences in viscosity of the chitosan-gelatin suspension were determined using Kruskal–Wallis and Mann–Whitney tests. The effects of the chitosan’s MW and DD on the viscosity of the chitosan-gelatin suspension were analyzed using Spearman’s correlation. Results: Group 2 had the highest moisture content (10.63%), MW (159.68 kDa), viscosity of the chitosan powder (5.53 dPa.s), and viscosity of the chitosan-gelatin suspension (40.20 cps). Group 1 had the highest ash content (27.83%) and DD (93.72%). Group 3 showed the lowest ash content (1.06%), MW (37.12 kDa), and DD (86.22%), but it had good viscosity of the chitosan-gelatin suspension (37.25 cps). A significant difference in viscosity was found between the chitosan-gelatin suspension groups. Spearman’s correlation coefficients between the viscosity of the chitosan-gelatin suspension and MW and between the viscosity of the chitosan-gelatin suspension and DD were 0.389 and −0.195, respectively. Conclusion: The viscosity of a chitosan-gelatin suspension is influenced by the MW and DD of the chitosan powder. Chitosan from P. monodon can potentially be an injectable bone substitute in socket preservation.
