Abstract

Baru-baru ini beberapa hasil penelitian menduga mikroplastik sebagai salah satu faktor risiko terjadinya gangguan ginjal. Mikroplastik merupakan fragmen hasil degradasi plastik yang berukuran lebih besar. Di dalam tubuh, mikroplastik akan terdistribusi ke seluruh tubuh sehingga memungkinkannya terakumulasi di suatu organ. Akumulasi mikroplastik dapat memicu terjadinya stres oksidatif, sitotoksitas, dan inflamasi yang berujung pada terganggunya fungsi suatu organ. Serum albumin, serum kreatinin, dan blood urea nitrogen merupakan biomarker yang masih rutin digunakan untuk menilai fungsi ginjal. Tujuan penelitian ini adalah untuk menjelaskan pengaruh partikel mikroplastik dalam darah terhadap kadar serum albumin, serum kreatinin dan blood urea nitrogen pada Rattus norvegicus Strain Wistar. Penelitian ini membagi 30 ekor Rattus norvegicus Strain Wistar ke dalam 1 kelompok kontrol dan 5 kelompok eksperimen. Kelompok eksperimen diberikan paparan mikroplastik peroral dengan dosis berbeda mulai dari 0,0375 mg/hari; 0,075 mg/hari; 0,15 mg/hari; 0,3 mg/hari; dan 0,6 mg/hari selama 90 hari. Hasil uji analisis data secara statistika menunjukkan adanya keterdugaan korelasi atau pengaruh partikel mikroplastik dalam darah terhadap kadar serum albumin (P = 0,009; R = -0,466) dan blood urea nitrogen (P = 0,010; R = 0,460), serta tidak adanya korelasi atau pengaruh partikel mikroplastik dalam darah terhadap kadar serum kreatinin (P = 0,418). Dapat disimpulkan bahwa pemberian paparan mikroplastik peroral selama 90 hari pada Rattus norvegicus Strain Wistar diduga dapat menyebabkan penurunan kadar serum albumin dan peningkatan blood urea nitrogen. Sedangkan kadar serum kreatinin didapatkan hasil yang hampir sama antara kelompok kontrol dan kelompok eksperimen.

Abstract

Mikroplastik merupakan partikel plastik berukuran kurang dari 5 mm. Mikroplastik yang tertelan oleh manusia selanjutnya akan memasuki traktus gastrointestinal dan mengalami penyerapan ke dalam aliran darah. Mikroplastik di dalam darah dan organ akan menyebabkan peningkatan produksi Reactive Oxygen Species (ROS) dan stres oksidatif. Salah satu akibat dari peningkatan produksi ROS pada hepar adalah terjadinya oksidasi dari Low Density Lipoprotein (LDL) menjadi Oxidized Low Density Lipoprotein (Ox-LDL). Penelitian ini merupakan penelitian eksperimental post-test only control group design. Hewan coba yang digunakan dalam penelitian ini adalah Rattus norvegicus Strain Wistar berjenis kelamin jantan sebanyak 30 ekor yang terbagi menjadi 6 kelompok (1 kelompok kontrol dan 5 kelompok eksperimental). Kelompok kontrol hanya diberikan aquabides, kelompok X1 diberikan dosis mikroplastik sebanyak 0,0375 mg/hari, kelompok X2 diberikan 0,075 mg/hari, kelompok X3 diberikan 0,15 mg/hari, kelompok X4 diberikan 0,3 mg/hari, kelompok X5 diberikan 0,6 mg/hari selama 90 hari. Setelah 90 hari, hewan coba diterminasi dan diambil sampel darahnya. Pada penelitian ini dilakukan uji komparasi pada kadar Ox-LDL plasma dengan menggunakan uji Kruskall Wallis dan didapatkan adanya perbedaan yang bermakna dari kadar Ox-LDL plasma antar kelompok dengan nilai P = 0,016. Selanjutnya dilakukan uji korelasi dengan uji Spearman dan didapatkan hubungan yang bermakna antara kadar partikel mikroplastik dalam darah dengan kadar Ox-LDL plasma dengan nilai P = 0,013.

Abstract

Latar Belakang: Sekitar 71% penyebab kematian di dunia berdasarkan data tahun 2016 disebabkan Penyakit Tidak Menular (PTM) yaitu 35% diantaranya karena penyakit jantung dan pembuluh darah. Menurut World Health Organization (WHO) pada tahun 2023, hanya 19% penderita hipertensi di Indonesia yang melakukan pengobatan dan hanya 4% yang mencapai target tekanan darah yang diharapkan. Berdasarkan rekapitulasi Indikator Keluarga Sehat (IKS) di puskesmas Wringinanom tahun 2022, persentase penderita hipertensi yang minum obat rutin di desa Sumberame adalah 39,9%. Tujuan: Mengetahui gambaran tingkat kepatuhan minum obat penderita hipertensi di dusun Sumberame, desa Sumberame, kecamatan Wringinanom.Metode: Penelitian ini merupakan studi deskriptif observasional. Teknik pengambilan sampel dengan consecutive sampling sesuai kriteria inklusi dan eksklusi. Penelitian ini diikuti oleh 40 responden dan dilakukan wawancara serta pengisian kuesioner untuk mendapatkan data. Hasil penelitian akan dianalisis menggunakan tabulasi silang dengan aplikasi SPSS.Hasil Penelitian: Hasil penelitian menunjukkan 57,5% dari responden tidak patuh dalam mengonsumsi obat hipertensi dengan alasan terbanyak adalah pengetahuan tentang hipertensi yang kurang. Sebanyak 55 % responden memiliki tingkat pengetahuan yang rendah terhadap hipertensi dan 68,2% di antaranya tidak patuh dalam mengonsumsi obat. Sebaliknya 55,6% responden dengan tingkat pengetahuan tentang hipertensi yang tinggi patuh dalam pengobatan. Simpulan: 57,5% dari responden mempunyai tingkat kepatuhan minum obat hipertensi yang rendah dan 42,5% dari responden mempunyai tingkat kepatuhan minum obat hiperrtensi yang tinggi.

Abstract

Microplastics, particularly Low-Density Polyethylene (LDPE), are environmental pollutants that can enter the body through oral exposure and potentially accumulate in the bloodstream. The presence of microplastics in systemic circulation is suspected to trigger physiological stress responses through activation of the hypothalamic–pituitary–adrenal axis. This study aimed to determine the effect of the number of microplastic particles in the blood on corticosterone hormone levels in Rattus norvegicus Wistar strain rats. This study employed an experimental design with a control group and treatment groups that received oral exposure to LDPE microplastics for 28 days. The number of microplastic particles in the blood was examined using light microscopy and polymer identification was confirmed using FTIR, while corticosterone levels were measured using the ELISA method. The results showed an increase in the number of microplastics in the blood following exposure, accompanied by elevated corticosterone levels as an indicator of physiological stress. This study recommends further investigation into the long-term effects of microplastics on the endocrine system and their implications for human health.

Abstract

Microplastics pose a potential health risk because they can enter the body orally and accumulate in various organs, including bone, potentially disrupting bone homeostasis through oxidative stress and inflammation. This study aimed to analyze the effect of oral polyethylene microplastic exposure on the femoral bone cell profile of Wistar strain Rattus norvegicus. A laboratory experimental study with a post-test only control group design was conducted using 28 male Wistar rats divided into four groups: control (X0) and treatment groups receiving LDPE microplastics (≤20 µm) at doses of 1.25 mg/day (X1), 2.5 mg/day (X2), and 5 mg/day (X3) for 28 days. Femoral bones were examined histologically using HE staining, and osteocytes, osteoblasts, and osteoclasts were counted. Results showed a significant increase in osteocyte and osteoclast numbers in treatment groups (p=0.002), while osteoblast numbers were not significantly different (p=0.655). Increased osteoclasts indicate enhanced bone resorption that may impair bone homeostasis.

Abstract

Low-density polyethylene (LDPE) microplastics are increasingly detected in the environment and can enter the human body through ingestion or inhalation. Microplastic exposure is associated with oxidative stress, inflammation, and cellular injury that may impair male reproductive health. Leydig and Sertoli cells play essential roles in spermatogenesis; therefore, damage to these cells can reduce sperm production and quality. However, evidence regarding the effects of LDPE microplastics on testicular morphology and spermatogenesis is still limited. This research aims to determine the effects of oral LDPE microplastic exposure on Leydig cell injury, Sertoli cell injury, and sperm count in male Wistar rats. This true experimental study used a post-test only control group design. Rats were divided into control and LDPE-treated groups. Testicular histopathology was evaluated, and data were analyzed using the Kruskal–Wallis test, Levene’s test, and Spearman correlation. LDPE exposure caused degenerative changes in Leydig and Sertoli cells and significantly reduced sperm count (p < 0.001). Oral LDPE microplastic exposure induces testicular cell damage and decreases sperm count, indicating potential reproductive toxicity.

Abstract

Nanoplastics are plastic particles smaller than 1000 nm formed from plastic degradation, including polyethylene (PE) These particles can enter the eye through environmental exposure or topical products and may induce oxidative stress, inflammation, and Meibomian gland dysfunctionEvidence regarding ocular toxicity from topical nanoplastic exposure remains limited. To evaluate the effects of topical polyethylene nanoplastic exposure on ocular surface inflammation and Meibomian gland histology. This experimental post-test only study used male Rattus norvegicus Wistar rats receiving topical polyethylene nanoplastic eye drops. Histopathological analysis with Hematoxylin–Eosin staining assessed PMN infiltration, normal Meibomian acini, acinar width, and gland obstruction. PMN infiltration in the palpebral conjunctiva increased significantly (p=0.014), while normal Meibomian acini decreased significantly (p=0.000). Other parameters showed no significant differences. Topical polyethylene nanoplastic exposure induces conjunctival inflammation and early Meibomian gland alterations.

Abstract

Orally ingested polyethylene (LDPE) microplastics are suspected to impair cognitive function through disturbances in the central nervous system, particularly in cognitive regions such as the cortex and hippocampus. This study aims to evaluate the effect of varying oral doses of LDPE microplastics on the cognitive function of Wistar Rattus norvegicus using the Morris Water Maze (MWM) test. This experimental study employed a post-test only control group design involving 21 male Wistar rats divided into four groups: a control group and three treatment groups receiving LDPE microplastics at low dose and high dose administered orally for 28 days (particle size ≤20 µm). Cognitive function was assessed using the latency time to locate the hidden platform in the MWM and analyzed statistically. The mean latency tended to increase with increasing dose. Using the control group as the baseline, there was a slight increase in the outcome in the low dose group, and a markedly higher increase observed in the high dose group. Statistical analysis showed a significant difference between the control group and the high dose group (p = 0.009). Oral administration of LDPE microplastics at  high dose for 28 days was associated with a significant impairment in cognitive function.

Abstract

Polyethylene microplastics are widely used in single-use products and can enter the body through contaminated food or beverages. After ingestion, these particles may cross the intestinal barrier and reach systemic circulation. Their presence in blood is suspected to induce oxidative stress through increased reactive oxygen species, potentially affecting erythrocyte membrane integrity. Erythrocytes are essential for oxygen transport, and structural damage may reduce their flexibility and lead to morphological abnormalities. This study aimed to evaluate the correlation between microplastic counts in blood and erythrocyte profiles in Wistar rats exposed orally to polyethylene microplastics. A quantitative experimental study with a post-test only control group design was conducted. Male Wistar rats were assigned to one control group and three exposure groups receiving different microplastic doses for four weeks. Microplastics were identified using light microscopy and Fourier Transform Infrared spectroscopy, while erythrocyte profiles were assessed through peripheral blood smears and hematological examination. Microplastics were detected in exposed groups, accompanied by several abnormal erythrocyte forms. However, no significant differences or correlations were observed between microplastic counts and erythrocyte parameters. Oral exposure to polyethylene microplastics did not significantly alter erythrocyte profiles, although potential morphological effects cannot be excluded.
