Abstract

Blind feeding is a feed dosage of feeding shrimp cultivation that is carried out without looking at the shrimp biomass conditions for the first 30 days. The goal is to ensure that shrimp get adequate feed intake in the early stages of growth, yet blind feeding often has an impact on the decline in water quality as a result of overfeeding. The study aimed to evaluate the effect of the blind feeding method on the growth of vaname shrimp (Litopenaeus vannamei) and water quality. The treatments to be tested were the quantity of feed: (a) 123.5 kg/100,000 shrimp; (b) 148.2 kg/100,000 shrimp; (c) 172.9 kg/100,000 shrimp; (d) 197.6 kg/100,000 shrimp; and (e) 222.3 kg/100,000 shrimp. The dose range of blind feeding referred to the habits applied by the cultivators in the field. The shrimp used was PL 12 with a density of 1,000 individuals/m3, which were maintained for 30 days in an aquarium container of 50 x 40 x 30 cm or a volume of 50 liters. The feed was given four times a day at 8.00, 12.00, 16.00, and 20.00. The results of the study showed that the blind feeding feed dosage had a significant effect on the absolute growth of shrimp and total ammonia nitrogen (P < 0.05). The blind feeding treatment (c), which was 172.9 kg/100,000 shrimps, produced the best growth with a lower total ammonium nitrogen so it is recommended that it be applied in shell cultivation in the first 30 days of maintenance.

Abstract

Aktinobakteri merupakan salah satu agen hayati yang mampu menghambat jamur patogen pada tanaman. Aktinobakteri mampu menghasilkan enzim kitinase untuk menghidrolisis dinding sel jamur sehingga menghambat pertumbuhan jamur patogen. Tujuan dari penelitian ini adalah untuk mengevaluasi kemampuan enzim kitinase dari aktinobakteri dari beberapa sumber tanah di Provinsi Jambi yang dapat menghambat jamur patogen tanaman Ganoderma boninense penyebab penyakit busuk pangkal batang kelapa sawit. Isolat diremajakan pada media starch nitrate agar, produksi kitinase dilakukan menggunakan koloidal kitin yang diperkaya, dan pengukuran aktivitas kitinase aktinobakteri dilakukan dengan menggunakan N-asetil glukosamin (GlcNAc) sebagai standar kitinase dan dibaca dengan spektrofotometer UV-VIS pada λ 584 nm. Isolat yang digunakan berasal dari dua jenis tanah yang berbeda, yaitu tanah perkebunan kelapa sawit (S16) dan tanah gambut (KT4). Hasil uji kualitatif menunjukkan nilai indeks kitinolitik S16 sebesar 1,90 dan KT4 sebesar 1,75. Uji aktivitas enzim kitinase dari kedua isolat diperoleh nilai 0,030 U/ml untuk isolat S16 dan 0,027 U/ml untuk isolat KT4. Uji penghambatan terhadap bakteri patogen Ganoderma boninense diperoleh daya hambat sebesar 54,33±1,53 dan 41,00±1,41 untuk isolat S16 dan KT4. Penelitian ini menyimpulkan bahwa aktinobakteri memiliki potensi untuk mengendalikan pertumbuhan Ganoderma boninense.

Abstract

Pencegahan penurunan keanekaragaman hayati di Indonesia mendasari Kebun Raya Banua melakukan kegiatan konservasi berupa eksplorasi dan penelitian setiap tahunnya. Kegiatan ini dilakukan untuk mengetahui jenis-jenis tumbuhan dan status konservasinya, sehingga dapat menjadi database sekaligus kontrol pembangunan di masa mendatang. Kegiatan ini dilaksanakan di Desa Kinarum Kabupaten Tabalong, Kalimantan Selatan pada Maret-Mei 2023. Metode yang digunakan adalah teknik eksploratif dengan analisis data secara deskriptif. Penentuan status konservasinya mengacu pada IUCN Red List, CITES, dan PermenLHK RI. Kegiatan ini berhasil menemukan 13 spesies yang terdaftar di IUCN Red List dengan kategori VU (rentan), NT (hampir terancam), LC (resiko rendah), dan DD (informasi kurang), 2 spesies terdaftar  di CITES dengan kategori Appendix 2, dan 1 spesies yang terdaftar di PermenLHK RI. Penemuan ini mengindikasikan perlu segeranya dilaksanakan kegiatan konservasi lainnya berupa penzonasian kawasan di lokasi ini untuk mencegah kepunahan flora tersebut.

Abstract

The study on Macarang hypolueca (Reichb.f. & Zoll.), which was collected from secondary forests around Samarinda City, East Kalimantan, involved phytochemical investigations that led to the isolation of a flavone type compound of flavonoid from the ethyl acetate fraction. To separate the compounds, silica gel column chromatography was utilized with a gradient solvent system of n-hexane and ethyl acetate, along with the addition of 5%. Infrared analysis (FTIR), mass spectrum (LC-ESI-MS), and nuclear magnetic resonance (1D- and 2D-NMR) were used to identify and elucidate the structure. Abstract

Sempur leaf (Dillenia suffruticosa) is a traditional medicine plant that contains flavonoids and is thought to be effective in treating gastrointestinal symptoms such as diarrhea. This research aims to evaluate the total flavonoid content of a sempur leaf infusion and assess its antibacterial effectiveness against two bacteria that cause gastroenteritis, namely Escherichia coli and Salmonella typhi. The infusion was carried out by heating the simplicial in distilled water for 15 minutes at 90 0C. The total flavonoid content was determined using UV-Vis Spectrophotometer with quercetin as the standard. Meanwhile, antibacterial activity was measured using the disc diffusion method with Ciprofloxacin as a positive control and distilled water as a negative control. The total flavonoid content of sempur leaf infusion was about 8.803 ± 0.6990 mgQE/g. In contrast, this sempur leaf infusion demonstrated no antibacterial activity against E. coli or S. typhi.

Abstract

Andaliman (Zanthoxylum acanthopodium D.C) is a plant widely used by the Batak people in North Sumatera. Several studies demonstrated that Andaliman fruit exhibits antioxidant activities and cytotoxic effects on cancer cells. Quality of Andaliman fruit could affected by post-harvest process including storage time. This research aims to analyze the compound, antioxidant activity and proliferation inhibition on MCF-7 cells of Andaliman fruit extracts (AFE) stored at varying storage time. A series of fresh 250 g Andaliman fruit sealed in a closed container and stored in the oven at 30ºC. After storage at 0, 24, 48, 72, 96, 120 and 144 hours, Andaliman fruit macerated using methanol and obtained successive extract yields of 4.12; 3.70; 12.79; 8.17; 8.15; 3.7 and 2.21. These extracts were analyzed for total phenolic, total flavonoids, free radical scavenging activity and proliferation inhibition on MCF-7 cells. The chemical compound analysis of AFE was performed by ultra performance liquid chromatography (UPLC). Results showed that the AFE stored at zero hours had higher total phenol content (47.32 mg gallic acid equivalent (GAE)/g of extract) and free radical scavenging activity (50.38%) at 500 µg/mL significantly compared to other extracts. The total flavonoid content of all extracts showed no difference level except for the extract at 120 hours of storage. The proliferation inhibition test on MCF-7 cells at 100 µg/mL showed that AFE stored at 72, 96 and 144 hours could inhibit MCF-7 cells above 50%. The storage time variation of Andaliman fruit may affect the total phenolic and flavonoid content, and also activity of free radical scavenging and proliferation inhibition on MCF-7. The UPLC analysis founded the major compound of AFE was predicted as α-sanshool. Analysis of chemicals substance in Andaliman fruit with varying of storage time need to be conducted to evaluated alteration of secondary metabolites contained in Andaliman fruit.

Abstract

Anggrek dapat tumbuh hampir di semua habitat salah satunya pohon/kayu (epifit). Pohon mangrove sebagai inang memiliki jenis dan karakter yang berbeda-beda. Pentingnya penelitian ini dilakukan agar kita dapat mengetahui jenis-jenis pohon mangrove sebagai inang dan juga adanya gambaran distribusi vertikal anggrek Dendrobium. Penelitian ini bertujuan untuk mengetahui distribusi vertikal anggrek Dendrobium pada pohon mangrove dan juga untuk mengetahui jenis-jenis mangrove apa yang disukai anggrek Dendrobium sebagai pohon inang. Pengumpulan data dilakukan dengan menggunakan metode petak plot dan selanjutnya dianalisis secara deskriptif kualitatif dan kuantitatif. Terdapat 6 petak plot dengan ukuran 20 m x 20 m ditempatkan secara purposive di hutan mangrove kampung Nubuai Distrik Urei Faisei Kabupaten Waropen. Hasil penelitian menunjukkan bahwa semakin besar diameter dan tinggi inang maka semakin banyak pula jumlah individu suatu spesies anggrek. Jumlah individu anggrek Dendrobium dipengaruhi oleh diameter batang dan ketinggian inang.

Abstract

Keanekaragaman dan status konservasi iktiofauna di aliran Sungai Mahakam, Kecamatan Long Iram, Kalimantan Timur, belum pernah diinformasikan, sementara banyak spesies ikan di perairan tersebut telah lama dimanfaatkan oleh masyarakat lokal sebagai ikan konsumsi dan diperdagangkan. Tujuan penelitian ini adalah untuk menganalisis keanekaragaman spesies dan status konservasi iktiofauna di aliran Sungai Mahakam. Penelitian dilaksanakan dari November sampai Desember 2022 pada empat stasiun yang ditentukan. Sampel ikan dikoleksi menggunakan metode purposive sampling dengan alat tangkap utama berupa jaring insang. Hasil penelitian menunjukkan bahwa sebanyak 1.263 individu ikan yang terdiri atas 28 spesies, 15 famili, dan 8 ordo didapat. Ordo Cypriniformes (42,85%) dan Siluriformes (32,14%) ditemukan paling dominan. Indeks keanekaragaman spesies berkisar antara 2,33–2,71 dan tertinggi di stasiun 2. Indeks kesamaan spesies seluruh stasiun berkisar antara 0,73–0,94. Status konservasi Anguilla bicolor dalam daftar merah IUCN diidentifikasi sebagai spesies hampir terancam (NT), 24 spesies beresiko rendah (LC), dua spesies kurang data (DD), dan satu spesies tidak ditemukan dalam daftar. Hypostomus plecostomus diidentifikasi sebagai spesies asing. Ketidakhadiran vegetasi di sepanjang pinggir sungai dan aktivitas manusia dapat memengaruhi keanekaragaman dan kelimpahan ikan di perairan ini. Secara umum, kualitas air di Sungai Mahakam dalam kondisi baik untuk mendukung kehidupan ikan.

Abstract

Actinomycetes are an important source for the discovery of bioactive secondary metabolite compounds. Over 10,000 bioactive metabolite compounds have been isolated from terrestrial actinomycetes with various biological activities, such as antibiotic, antiviral, anti-inflammatory, antitumor, anticancer, and antioxidant. One source of origin for actinomycetes is soil, including termite nests. Utilization of natural sources such as actinomycetes from termite nests as antioxidants and antibacterials can be an alternative source of production of new secondary metabolite compounds. This research aims to evaluate the antioxidant and antibacterial activities of actinomycete extracts from termite nests. Antioxidant activity using the DPPH free radical scavenging method, and antibacterial activity against S. aureus. Antioxidant and antibacterial activity were performed by using the TLC dot-blot, TLC bioautography, and microdilution methods to determine the inhibitory concentration of 50% (IC50), antioxidant activity index (AAI), and minimum inhibitory concentration (MIC). From the 33 extracts tested, 16 extracts showed antioxidant activity with IC50 values range of 76.64-126.22 µg/mL or AAI value > 0.05 (moderate), and 8 extracts had moderate antibacterial activity against S. aureus (MIC values range of 256-512 µg/mL). Future research for scaling-up of actinomycete culture, isolating of active compounds, determining of the chemical structure of active compounds, and further testing as antioxidants and antibacterials still need to be carried out.

Abstract

Goniothalamus macrophyllus (Blume) Hook.f. & Thomson merupakan jenis tumbuhan dari keluarga annonaceae yang memiliki banyak manfaat dengan populasi yang sedikit dan penyebaran yang terbatas habitat alaminya.  Tujuan penelitian ini adalah mengetahui fenologi pembungaan dan pembuahan Goniothalamus macrophyllus (Blume) Hook.f. & Thomson pada habitat alaminya di hutan dataran rendah Gunung Tilu Kabupaten Kuningan. Hasil dari penelitian ini adalah periode pembungaan dan pembuahan membutuhkan rata-rata ±180 hari atau 6 bulan dengan dimulai dari fase kuncup bunga kecil – fase buah. Fase kuncup bunga kecil (B1) - fase kuncup bunga besar (B2) membutuhkan rata-rata 10 hari, fase kuncup bunga besar (B2) – fase bunga mekar (B3) 10 hari, fase bunga mekar (F3) - fase buah (B4) 11 hari, dan dari fase buah (B4) hingga buah matang 30 hari. Moroflogi G. macrophyllus (Blume) Hook.f. & Thomson dibandingkan dengan jenis Goniothalamus lainnya memiliki morfologi bunga dan buah yang relatif lebih kecil.
