Abstract

Ikan fillet tuna merupakan pengolahan inovasi ikan yang memiliki daya simpan yang singkat jika disimpan dalam suhu ruang. Penelitian ini bertujuan untuk memperpanjang daya simpan dan mempertahankan mutu fisik ikan fillet tuna dalam suhu ruangan dengan menggunakan lapisan kitosan. Penelitian ini menggunakan metode edible coating dengan merendam ikan fillet tuna pada larutan kitosan dengan berbagai variasi yakni tanpa perlakukan sebagai kontrol negatif, berbagai konsentrasi kitosan dari 1%, 1,5%, 2%, 2,5%, 3%, serta penggunaan formalin dengan konsentrasi 1% dan 10% sebagai kontrol positif selama 5 menit. Hasil menunjukkan bahwa ikan fillet tuna dengan konsentrasi 3% dapat memperpanjang daya simpan selama 4 hari sedangkan tanpa perlakuan sebagai kontrol negatif bertahan selama 2 hari, sampel dengan konsentrasi 1%, 1,5%, 2%, dan 2,5% memperpanjang daya simpan selama 3 hari. Maka dapat disimpulkan penggunaan larutan kitosan dapat digunakan sebagai pengawet ikan fillet tuna serta mempertahankan parameter fisik (tekstur, warna, aroma) dan pH ikan fillet tuna lebih baik setelah perlakukan dibandingkan sebelum perlakuan dengan larutan kitosan.

Abstract

Hand is the main transmission pathogen entry into the body which is the part of the body that is most often in direct contact with other limbs and surrounding objects. The importance of maintaining hand hygiene by always washing hands with soap is the first step in preventing transmission of COVID-19. However, during a pandemic like this, it will be difficult to find soap available everywhere, and tends to be hard to carry all the time. Therefore, this service aims to invite the people of Pematang Gubernur, Bengkulu City, to use paper soap because it is easier to use its use and flexible to carry anywhere besides being an effort to prevent the COVID-19 outbreak, it can also add to new information and skills for residents in making attractive soaps. The feedback response given by the community service implementation team to the community service participants resulted in a positive response to this activity, who initially did not know about making and use of paper soap and became aware of this.

Abstract

The increasing intensification of dragon fruit (Hylocereus sp.) cultivation is followed by an increase in disease attacks that have the potential to reduce plant productivity. This study aims to isolate, identify, and test the pathogenicity of bacteria that cause dragon fruit stem rot in Pematang Donok Village, Kepahiang, Bengkulu. Bacterial isolation was carried out from disease-infected stem tissue, followed by isolation purification. Bacterial identification was carried out based on colony morphology and Gram staining, while pathogenicity and hemolysis tests were carried out on potatoes and blood agar media, respectively. The data were analyzed descriptively qualitatively through observation of colony morphological characteristics and Gram staining, as well as pathogenicity tests on potatoes and hemolysis tests on blood agar. The isolation results obtained two bacterial isolates, namely isolates BBNK1 and BBNK2 with different colony morphology characteristics. Both isolates are Gram-positive bacteria with bacillus and coccus cell shapes. Pathogenicity tests on potatoes showed that both isolates were able to cause soft rot symptoms such as tissue softening, discoloration, and mucus formation compared to the control. Hemolysis tests showed that only isolate BBNK1 showed hemolytic activity with the formation of a clear zone around the isolate. The findings of this study offer foundational data to support future research on molecular identification and the development of more environmentally friendly and sustainable disease management strategies.

Abstract

Microbial infections pose serious threats to human health, emphasizing the need to develop effective antimicrobial materials. This study aims to synthesize, characterize, and evaluate the antimicrobial properties of ester vanillin-benzoate (1) and its chitosan-based composite material (2). Furthermore, the synthesized compound was tested for antibacterial and antifungal activities against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Candida albicans using the Kirby-Bauer diffusion method. The synthesized compounds were effective against S. aureus and B. subtilis but had minimal action against P. aeruginosa, E. coli, and C. albicans. The formation of the Schiff-base composite could increase its antibacterial activity, indicating a synergistic effect arising from the combination of compound 1 and chitosan.

Abstract

Impatiens balsamina L. has traditionally been used in Indonesia for treating skin infections and is considered a potential source of antibacterial compounds. However, the interaction between crude plant extracts and antibiotics may not always result in synergistic effects and may instead reduce antibacterial efficacy. This study evaluated the antibacterial activity and potential interaction of methanol leaf extract of I. balsamina with five antibiotics against Staphylococcus aureus ATCC 29213. The antibacterial activity was assessed using well diffusion and paper strip methods. The treatments consisted of 15% I. balsamina leaf methanol extract, 0.025% of each antibiotic, including clindamycin, chloramphenicol, cotrimoxazole, ciprofloxacin, and tetracycline, and their 1:1 combination. The extract alone produced a mean inhibition zone of 10.27±0.216 mm. Clindamycin, chloramphenicol, ciprofloxacin, and tetracycline showed inhibition zones ranging from 19.46±1.732 mm to 22.34±3.021 mm, while cotrimoxazole showed no inhibition zone. The combinations of the extract with clindamycin, tetracycline, chloramphenicol, ciprofloxacin, and cotrimoxazole produced inhibition zones of 19.54±0.155 mm, 18.57±0.422 mm, 16.37±0.879 mm, 13.28±0.257 mm, and 9.76±0.103 mm, respectively. The diffusion-based assays indicated a potential antagonistic interaction between the extract and the tested antibiotics, characterized by combined inhibitory diameters smaller than the additive sum of the individual agents. These findings suggest that the concurrent use of I. balsamina extract and antibiotics should be further evaluated using MIC-based checkerboard assays and Fractional Inhibitory Concentration Index analysis. Potensi Interaksi Antagonis Ekstrak Daun Metanol Impatiens balsamina L. dengan Lima Antibiotik terhadap Staphylococcus aureus ATCC 29213 ABSTRAK: Impatiens balsamina L. secara tradisional digunakan di Indonesia untuk mengobati infeksi kulit dan diketahui memiliki potensi sebagai sumber senyawa antibakteri. Namun, kombinasi antara ekstrak tanaman dan antibiotik tidak selalu menghasilkan efek sinergis, tetapi juga dapat menurunkan efektivitas antibakteri. Penelitian ini bertujuan untuk mengevaluasi aktivitas antibakteri dan potensi interaksi antara ekstrak metanol daun I. balsamina dengan lima antibiotik terhadap Staphylococcus aureus ATCC 29213. Aktivitas antibakteri diuji menggunakan metode difusi sumuran dan paper strip. Perlakuan terdiri atas ekstrak metanol daun I. balsamina 15%, masing-masing antibiotik 0,025%, yaitu klindamisin, kloramfenikol, kotrimoksazol, siprofloksasin, dan tetrasiklin, serta kombinasi ekstrak dan antibiotik dengan rasio 1:1. Ekstrak tunggal menghasilkan rata-rata zona hambat sebesar 10,27±0,216 mm. Klindamisin, kloramfenikol, siprofloksasin, dan tetrasiklin menghasilkan zona hambat sebesar 19,46±1,732 mm hingga 22,34±3,021 mm, sedangkan kotrimoksazol tidak menunjukkan zona hambat. Kombinasi ekstrak dengan klindamisin, tetrasiklin, kloramfenikol, siprofloksasin, dan kotrimoksazol menghasilkan zona hambat masing-masing sebesar 19,54±0,155 mm, 18,57±0,422 mm, 16,37±0,879 mm, 13,28±0,257 mm, dan 9,76±0,103 mm. Hasil uji berbasis difusi menunjukkan adanya indikasi interaksi antagonis antara ekstrak dan antibiotik yang diuji, ditandai dengan diameter hambat kombinasi lebih kecil daripada jumlah aditif dari masing-masing agen secara individu. Temuan ini menunjukkan bahwa penggunaan ekstrak I. balsamina bersamaan dengan antibiotik perlu dikaji lebih lanjut melalui uji MIC, checkerboard assay, dan Fractional Inhibitory Concentration Index

Abstract

Background: Kebiul seeds are an Indonesian medicinal plant with potential antioxidant and chemopreventive properties against cancer. The development, growth, and invasion of malignant cancer closely link to angiogenesis, which antioxidant effects can influence.Objective: This study aims to evaluate the antioxidant activity of kebiul seeds based on the IC50 and their anti-angiogenesis effects.Method: Extraction was performed using the maceration method using ethyl acetate, n-hexane, and ethanol. The anti-angiogenesis test was conducted using the chorioallantoic membrane (CAM) method, while the antioxidant activity was assessed using the diphenylpyrrolhydrazyl (DPPH) method.Results: The finding revealed the antioxidant activity values, as indicated by the IC50, were 45.225 ppm for the ethanol extract (strong antioxidant), 86.190 ppm for the n-hexane extract (strong antioxidant), and 146 ppm for the ethyl acetate extract (medium antioxidant). Anti-angiogenesis tests were conducted across various solvents and concentrations. Significant anti-angiogenesis effects were observed at concentrations of 10, 50, and 1000 ppm. The n-hexane extract yielded average scores of 0.33, 0.66, and 1.3, indicating a good effect. The ethyl acetate extract had average scores of 0.33, 0.5, and 0.66, suggesting a weak effect, while the ethanol extract showed mean scores of 1.33, 1.66, and 2, indicating a strong effect. Conclusion: Ethanol extract demonstrates the most potential as a chemopreventive agent.

Abstract

Abstract: Mangroves Rhizophora stylosa Griff. and Sonneratia alba Sm. produce secondary metabolite compounds that are useful as potential sources of new antimicrobes because they inhibit the growth of pathogenic microbes. Endophytic bacteria are bacteria that live within plant tissues and are capable of producing metabolite compounds with antimicrobial properties. This study aimed to determine the antimicrobial activity and molecular profile of the 16S rRNA gene from endophytic bacterial isolates of R. stylosa Griff. and S. alba Sm. from Enggano Island. The isolates were subcultured on Nutrient Agar (NA) medium. The results showed that five isolates (DRSE 16, DRSE 17, ASAE 3, DSAE 17, and DSAE 19) out of 40 endophytic bacterial isolates exhibited potential activity against the testing microbes (Salmonella typhi, Enterococcus faecalis, and Candida albicans). The results of 16S rRNA identification showed that the bacterial isolate DRSE 16 had similarities with the species Stenotrophomonas nitritireducens strain BR-49, isolate DRSE 17 with the species Pseudomonas putida strain XT214, isolate ASAE 3 with the species Pseudomonas aeruginosa strain D3, isolate DSAE 17 with the species Bacillus subtilis strain C-2-3-2, and isolate DSAE 19 with the species Pseudomonas otitidis strain RR017. Abstrak: Bakau Rhizophora stylosa Griff. dan Sonneratia alba Sm. menghasilkan senyawa metabolit sekunder yang berguna untuk dijadikan sumber antibiotik baru karena dapat menghambat pertumbuhan mikroba patogen. Bakteri endofit merupakan bakteri yang hidup pada jaringan tumbuhan dan dapat menghasilkan senyawa metabolit yang dapat berfungsi sebagai antimikroba. Penelitian ini bertujuan untuk mengetahui aktivitas antimikroba dan profil molekuler gen 16S rRNA dari isolat bakteri endofit bakau R. stylosa Griff. dan S. alba Sm. asal Pulau Enggano. Isolat diremajakan pada media Nutrient Agar (NA), kemudian digunakan untuk menguji aktivitas senyawa antimikroba hasil kultur sel bakteri, supernatan, dan pelet. Hasil penelitian didapatkan bahwa lima isolat potensial (DRSE 16, DRSE 17, ASAE 3, DSAE 17 dan DSAE 19) dari 41 isolat bakteri endofit terhadap mikroba uji (Salmonella typhi, Enterococcus faecalis dan Candida albicans). Hasil identifikasi 16S rRNA menunjukkan bahwa isolat bakteri DRSE 16 memiliki kemiripan dengan spesies Stenotrophomonas nitritireducens galur BR-49, isolat DRSE 17 dengan spesies Pseudomonas putida galur XT214, isolat ASAE 3 dengan spesies Pseudomonas aeruginosa galur D3, isolat DSAE 17 dengan spesies Bacillus subtilis galur C-2-3-2, dan isolat DSAE 19 dengan spesies Pseudomonas otitidis strain RR017.
