Abstract

The current work is focused on the characterization of the chromium(III)-containing low-molecular-weight (LMW) fractions of blood serum by mass spectrometry. The LMW fractions of serum binding-Cr(III) were collected by organic solvent precipitation using acetonitrile. The chromium(III) compounds used were [Cr(pic)3], where pic = 2-pyridinecarboxylato(-) or A, and the Cr(III) propionate complex, [Cr3O(OCOEt)6(OH2)3])+ or B. The new signals appeared in the mass spectra in the presence of added Cr(III) compounds, either A or B. This indicated that common LMW Cr containing serum fractions were formed on addition of Cr(III) to serum. The new signals contained three distinct peaks with a shift of 16, corresponding to ions of m/z +1083, +1099, and +1115 Da, which consisted of 4 Cr atoms with bridging ligands.

Abstract

Nanoparticles have been widely used in various fields which depends on its size and shape. Nanoparticles can be synthesized physically and chemically. However, these methods need large amount of energy, not environmental friendly and quite expensive, because requires several additional materials besides precursors. In this study, we performed the analysis of some parameters to determine the best conditions for biosynthesis using Rhizopus oligosporus to obtain the nanoparticles with specific size. This fungi was used because easy to find, cultivate, and handle. We found that grain size of Cu2O/CuO-NP and AgNP obtained are 23 nm, 55 nm respectively. In conclusion, this study confirms that some parameters investigated and Rhizopus oligosporus can be used to obtain the nanoparticles with specific size.

Abstract

The aim of this research was to study the antidiabetic activity of root extract of Ruellia tuberosa L. on rats (Rattus novergicus) induced by multiple low doses of streptozotocin. The parameters investigated were blood glucose levels, protease activities, and the histopathologic profiles of the kidneys. Three groups of rats, i.e. control, diabetic, and treatment, were used. R. tuberosa L. extracts were given orally for treatment at a dose of 250 mg/kg body weight per day for 21 consecutive days. The induced diabetes group treated with root extract of R. tuberosa L. (group III) had 60.3% lower glucose levels (119 mg/dL) compared to the group with induced diabetes only. Moreover, antidiabetic activity of R. tuberosa L extract was evidenced by the 55% lower protease activity in the treatment group compared to the untreated diabetic group. Finally, a positive effect from root extract of R. tuberosa L. was shown from the less damaged renal glomerulus and smaller gap between extracellular matrices and epithelium cells in the histopathologic profiles of the kidneys in the extract treatment group. These results show the potential of root extract of R. tuberosa L. as a natural product anti-diabetic agent.

Abstract

Purple rice is one of the main sources of ferulic acid (FA). Some studies reported anti-inflammatory properties of FA, but the interaction of FA with TNF-α signaling has not been elucidated. TNF-α is a target for anti-inflammatory drug research due to its major role in the inflammatory process. This study aims to investigate the interaction of FA with TNF-α and TNF-α receptor (TNFR) through in silico study and evaluate the drug-like properties and biological activity of FA. The interactions among FA (CID 445858), TNF-α (2AZ5), and TNFR (1NCF) were docked by Hex 8.0.0 Cuda, then visualized by Discovery Studio 2020 and LigPlot V.1.4.5. Apigenin-7-glucuronide (AG, CID 5319484) was used as the positive control. The drug-like properties were predicted by Lipinski’s rule of five and the biological activity was analyzed by PASS online. FA showed good properties as a drug-like molecule and biological activity as an anti-inflammatory. FA also showed good interaction with TNF-α and TNFR. FA bound to TNF-α at Asn92(B), Val91(B), Leu93(B), Phe124(B), Phe124(D), and Leu93(D) residues with docking energy of -214.6 kJ/mol, and bound to TNFR at Pro16(A), Glu56(B), Cys55(B), Glu54(B) residues with docking energy of -191.1 kJ/mol. FA could inhibit TNF-α – TNFR interaction by binding to TNFR at Glu54 residue, the same inhibition mechanism to AG which bind to TNFR at Glu54 and Val90. The current study shows that FA has the potential as an anti-inflammatory of TNF-α signaling and can be developed as an oral anti-inflammatory drug candidate.

Abstract

Makanan yang beredar di masyarakat saat ini, khususnya di sekolah dasar, lebih banyak berupa makanan instan yang sarat dengan bahan aditif seperti bahan pengawet, perasa, pewarna, dan pemanis, serta kadar nutrisi yang kurang untuk memenuhi kebutuhan harian anak. Tujuan kegiatan pengabdian kepada masyarakat ini adalah untuk memperkenalkan informasi dan pengetahuan tentang bahaya mengkonsumsi makanan jajanan di sekitar sekolah yang mengandung zat berbahaya seperti formalin, boraks, dan zat warna sintetis. Kegiatan ini juga bertujuan memperkenalkan metode uji makanan jajanan di sekitar sekolah yang mengandung zat berbahaya seperti formalin, boraks, dan zat warna sintetis. Peserta kegiatan adalah guru-guru SDI Surya Buana, Malang. Kegiatan dilakukan melalui tiga tahapan yaitu: 1. Pembuatan modul; 2. Penyuluhan; 3. Pelatihan serta uji bersama kandungan bahan pengawet dan pewarna berbahaya. Evaluasi dilakukan dalam 2 tahap, yaitu evaluasi, yaitu evaluasi pada saat pelaksanaan kegiatan dan evaluasi pasca pelaksanaan Kegiatan pengabdian masyarakat dilaksanakan pada tanggal 12 Agustus 2020, di SDI Surya Buana. Kegiatan dihadiri oleh Kepala SDI Surya Buana dan oleh 40 orang guru-guru. Pada kegiatan tersebut dilakukan presentasi tentang bahaya makanan jajanan yang mengandung boraks, formalin, dan zat warna tekstil. Kegiatan lainnya adalah peragaan tes uji cepat makanan jajanan menggunakan tes kit untuk uji formalin, boraks, dan zat warna. Pada awal dan di akhir acara, dibagikan kuesioner untuk evaluasi kegiatan. Hasil evaluasi menunjukkan bahwa 100% responden puas setelah mengikuti kegiatan ini. Dari kegiatan ini dapat disimpulkan bahwa pengetahuan tentang bahaya zat pengawet dan pewarna buatan sangat dibutuhkan bagi warga sekolah. Selain itu, warga sekolah perlu mengetahui tentang uji cepat zat pengawet dan pewarna buatan pada makanan jajanan di sekolah.

Abstract

Talok is a partner district with economic conditions that have developing status according District Development Index values. The developing status related with susceptible of economic shocks, natural disasters and social conflicts will be effect to underdeveloped district. This Community Empowerment Program is purpose to develop Micro, Small and Medium Enterprises (MSME) product businesses can expect to income increasing of enterprise through the Business Model Canvas (BMC) approach. Mapping survey of local potentials from partner district was observed to achieve the success of community service activity. In this survey, we collaborate with MSME partners are (1) Samiler Crackers MSME group, (2) Cassava Chips MSME group, (3) Lempeng Crackers MSME group. The data was analyses using descriptive and qualitative method by using the in-depth interview in collecting primary data needed during the research process. The BMC analysis on each element of business model canvas could improve the service quality, marketing activity, infrastructure and financial ability. Business model canvas focuses on business ideas to create value in the business. In addition, the cost of production calculation is very important for selling price determination of the product to avoid the losses condition.

Abstract

Hemodialisa merupakan modalitas terapi penyelamat hidup bagi pasien gagal ginjal kronis (GGK) dan dapat menimbulkan berbagai komplikasi. Salah satunya yang paling sering terjadi adalah hipotensi intradialisis mencapai 20-30%. Efek hemodialisis kronik yaitu fatigue memiliki prevalensi tinggi mencapai 60-97%. Fatigue akan dirasakan bila pasien mengalami hipotensi intradialisis. Penelitian ini bertujuan mengetahui hubungan hipotensi intradialisis dengan tingkat fatigue pada pasien GGK yang menjalani hemodialisis di Ruang Hemodialisa RSUD Ratu Zalecha Martapura. Metode penelitian menggunakan cross sectional dengan populasi 71 pasien GGK yang menjalani hemodialisis. Sampel penelitian diambil dengan cara Purposive Sampling sebanyak 45 orang, dianalisis dengan uji Spearman Rank menggunakan program software SPSS versi 23 tahun 2015. Instrumen penelitian menggunakan lembar observasi tekanan darah per jam selama intradialisis dan skala pengukuran kelelahan (FAS). Hasil analisis univariat dari 45 responden, sebagian besar mengalami penurunan TD sistolik yang normal (< 20 mmHg) yaitu sebanyak 25 orang (56%) dan mengalami tingkat fatigue lelah (skor ³ 22) yaitu sebanyak 29 orang (64%). Hasil analisis bivariat menunjukkan adanya hubungan antara hipotensi intradialisis dengan tingkat fatigue pada pasien GGK dengan nilai p-value < 0,05 yaitu sebesar 0,044 dan nilai correlations coefficient sebesar r: 0,257 yang menunjukkan hubungan lemah. Kesimpulannya pasien dengan penurunan TD sistolik akan mengalami tingkat fatigue lelah. Saran untuk perawat agar lebih meningkatkan monitoring selama HD berlangsung dan mengevaluasi keluhan pasien.

Abstract

The existence of phytochemicals in Cosmos caudatus Kunth, predominantly phenolic compounds, offers several health benefits. Nevertheless, the bioactive compounds are usually susceptible to degradation, and therefore may reduce their biological activity. This work aims to carry out microencapsulation of C. caudatus K. extracts by spray drying technique. The in vitro alpha-amylase inhibitory activity of the microencapsulated product is also investigated. The effect of manufacturing conditions, including pH, the concentration of wall materials, and stirring time, was evaluated. The optimal conditions for microcapsules formation were selected based on the activity of microcapsules as inhibitors for the alpha-amylase enzyme, pointing out by the lowest number of IC50. Results showed that microcapsules prepared in pH 4, 0.05% of chitosan, and 90 min stirring time had optimum efficiency, with the IC50 value of 92.85 ± 1.21 μg/mL. The FTIR (Fourier-Transform infrared) analysis showed that the –C–N stretching amine functional group appeared at wavenumber 1285 cm–1, and the –P=O phosphate bending appeared at 1206 cm–1. Characterization with PSA (particle size analyzer) and SEM (scanning electron microscope) indicated that microcapsules had predominantly spherical forms with a mean diameter of 38.92 μm. This work confirms the important role of microencapsulation in developing plant extracts with retained biological functionalities.

Abstract

Metal ion speciation in various biological systems has been extensively studied to elucidate its biological role and the toxicity of the element of interest. In the present study, chromium speciation was performed by reacting Cr(III) nutritional supplements [Cr(pic)3], where pic = 2-pyridinecarboxylato(-), and a Cr(III) propionate complex, [Cr3O(OCOEt)6(OH2)3])+, in calf serum. Cr(III) complexes in serum were fractionated using size-exclusion chromatography, and the Cr(III) concentrations in each fraction were determined using graphite furnace atomic absorption spectroscopy. The results showed that Cr(III) bound to both high-and low-molecular weight serum fractions. While Cr(III) was mainly bound toalbumin or transferrin, unknown low-molecular-weight serum fractions were also important in Cr binding. The Cr(III) distribution in serum fractions was found to be time-dependent.

Abstract

This study aims to analyze the inhibitory activities of aqueous root extract compounds of Ruellia tuberosa L. toward alpha-glucosidase protein by computational docking analysis. Three major compounds contained in the extracts (i.e., betaine, daidzein, and hispidulin) were selected as ligands; quercetin and acarbose were used as the reference. Computational docking analysis was performed using the HEX 8.0.0 program and visualized using the Discovery Studio Visualizer v19.1.0.18287 (2019 version) on the basis of the scoring functions. The interactions between ligands and alpha-glucosidase protein showed different binding patterns. The types of bonds involved in the interaction between the enzyme and these ligands were hydrogen and hydrophobic bonds. Energy generated from docking of betaine, daidzein, hispidulin, quercetin, and acarbose to alpha-glucosidase protein were −167.6, −249.5, −251.2, −241.5, and −322.1 cal/mol, respectively. Acarbose had the lowest energy, indicating that it has the strongest interaction with alpha-glucosidase, followed by hispidulin, daidzein, quercetin, and betaine. Amino acid residues that interacted with the ligands were His717, Met363, Arg608, Pro361, Phe362, Leu865, Glu869, Arg594, andAsp356. The current research shows that R. tuberosa L. aqueous root extracts have the potential to be used as an inhibitor for the alpha-glucosidase protein and as an antidiabetic agent. Nonetheless, further studies are needed to support this modeling study.
