Abstract

This study is essential to analyze the toxicity and release of bioactive compounds in R. tuberosa L. extract microcapsules coated with gum Arabic and to provide information on the use of microcapsules as a drug delivery system with controlled release of active ingredients. Investigations focused on the effect of pH and timing on discharge kinetics, as well as toxicity evaluation. The release of active ingredients from microcapsules was carried out in medium variations, namely pH 2.2 and pH 7.4, with release durations of 30, 60-, 90-, 120-, and 150-min. Bioactive compounds were released by 59.23% at pH 2.2 and 58.21% at pH 7.4 within 150 min. The BSLT (brine shrimp lethal toxicity) assay was used using brine shrimp larvae (Artemia salina L.), resulting in a non-toxic classification as evidenced by an LC50 value of 5729 μg/mL. Microcapsules in optimum conditions were then characterized using FTIR (Fourier-Transform Infrared) and SEM (scanning electron microscopy) instruments. The FTIR analysis showed variations in functional groups in R. tuberosa L. extracts, gum Arabic, and microcapsules. Furthermore, the SEM examination highlighted the morphology of microcapsules that were mostly spherical.

Abstract

The present study evaluates the antibacterial efficacy of ethanolic extracts from Delonix regia leaves against diarrhea-inducing bacteria Escherichia coli and Salmonella typhimurium. Preliminary phytochemical screening revealed that D. regia leaves comprise flavonoids, alkaloids, saponins, tannins, phenolics, and terpenoids. Fourier Transform Infrared Spectroscopy (FTIR) analysis identified various functional groups in the D. regia leaf extract, including O-H, C-H, C=O, C=C, C-C, C-O, and C-O-C. Liquid Chromatography-Mass Spectrometry/Mass Spectrometry (LC-MS/MS) analysis further confirmed the presence of 16 distinct compounds comprising amino acids, alkaloids, phenolics, flavonoids, terpenoids, anthraquinones, n-acyl pyrrolidines, and fatty acids. The disc diffusion method (Kirby-Bauer) was employed for the antibacterial tests. The extracts of D. regia leaves at concentrations of 25%, 50%, 75%, and 100% generated inhibition zones measuring 5.26 mm, 6.37 mm, 7.27 mm, and 10.18 mm against E. coli, and 5.05 mm, 7.01 mm, 8.77 mm, and 10.04 mm against S. typhimurium, respectively. The commercial antibiotic ciprofloxacin (a positive control) produced inhibition zones of 30.02 mm for E. coli and 28 mm for S. typhimurium. The negative control, consisting of 10% ethanol, showed no inhibitory effect on bacterial growth. These findings indicate that the ethanolic extract of D. regia leaves possesses antibacterial properties against E. coli and S. typhimurium. It is likely that secondary metabolite compounds, such as flavonoids and phenolics, contribute significantly to the observed antibacterial activity

Abstract

This study aims to investigate the microencapsulation of C. caudatus K. leaves extract under optimal conditions, focusing on its potential as a therapeutic option for type 2 diabetes. In order to determine the inhibitory effect on the α-amylase enzyme, which plays a crucial role in glucose metabolism, biological activity assays were performed. The procedure of effectively encapsulating the substance was achieved at a pH level of 6 using sodium alginate at a concentration of 2% (w/v), resulting in a 77.2% percentage of encapsulation. The experiment involving the inhibition of α-amylase exhibited an IC50 value of 69.44 ± 0.924 μg/mL, demonstrating the significant biological activity of the microcapsules. The bioactive substances encapsulated in the microcapsules were released during intervals 30-120 min, at pH conditions of 2.2 and 7.4. The release of bioactive substances was only 2.32 % after 120 minutes at a pH of 2.2, whereas it increased to 64.54 % at a pH of 7.4. This discovery indicates that the microcapsules contain the capability to be utilized for the regulated delivery of drugs. The findings of this research demonstrate the potential to develop herbal medicines with therapeutic properties through microencapsulation, providing an excellent opportunity for managing type 2 diabetes.

Abstract

Advanced drug delivery systems are required due to the increasing advancement in science and technology, with a common example being microencapsulation. This system is affected by the manufacturing conditions and slow release of bioactive compounds inside microcapsule. In general, phytochemicals in the edible plant, Cosmos caudatus Kunth, specifically flavonoids, offer several health benefits but are susceptible to degradation which reduces biological activities. Therefore, this study aimed to encapsulate extract using spray drying method, which can be used as wall material in Gum Arabic. Microencapsulated product was also tested for biological activities. The effect of microcapsule production was investigated, including the concentration of wall material and the stirring speed. Furthermore, the optimal conditions of microcapsule were selected based on the highest encapsulation efficiency. The results showed that a concentration of 4% (w/v) Gum Arabic and a stirring speed of 800 rpm were the optimal conditions, leading to a percentage release of 71.87%. Microcapsule under these conditions produced an IC50 value of 55.09 ± 0.57 ug/mL as an inhibitor of alpha-amylase enzyme. The characterization by scanning electron microscope (SEM) showed spherical forms with a mean diameter of 152.00 um. The bioactive substance in microcapsule was released at pH 2.2 and pH 7.4 for 30 to 120 min, with a percentage release of 38.05% and 95.14%, respectively. In conclusion, this study confirmed that microencapsulation played a crucial role in the development of plant extract with retained biological functions.

Abstract

Ruellia tuberosa L. leaf and root extracts have been investigated for their biological activity and potential health advantages, including their antidiabetic, antioxidant, and antidiuretic qualities. This research evaluates the freeze-drying microencapsulation of R. tuberosa L. extracts using gum Arabic, maltodextrin, and their combination as coating materials. The resulting microcapsules were tested for encapsulation efficiency, biological activity, and controlled release. Characterization techniques included scanning electron microscopy (SEM), Fourier-transform infrared (FTIR) spectroscopy, and particle size analysis (PSA). The choice of encapsulant significantly influenced encapsulation efficiency, morphology, and biological activity. Microcapsules using a combination of gum Arabic and maltodextrin exhibited more spherical shapes and smaller particle sizes than those using either material alone. Alpha-amylase inhibition tests showed that microcapsules effectively inhibit the enzyme, with the coating combination performing best, followed by gum Arabic and then maltodextrin. All microcapsules exhibit moderate antioxidant activity, again in the same order. The active compound release was greater at pH 7.4 compared to pH 2.2 from 0 to 120 min. Therefore, freeze-drying microencapsulation with biodegradable polymers is a viable method for delivering the health benefits of R. tuberosa L. extracts, yielding a convenient powder form suitable for drug delivery systems.

Abstract

Rodhamine B and Saccharin are often misused by food producers as food additive, which, if their use is not monitored, can produce free radicals that cause oxidative stress conditions in the body, leading to dangerous damage to the body. The addition of purple rosella in yoghurt can increase the functional value of the yoghurt, namely increasing the antioxidant content. The purpose of this study was to determine the benefits of giving purple rosella yoghurt in preventing damage to the duodenum and colon of (Rattus norvegicus) caused by Rodhamine B and Saccharin. This study used Completely Randomized Design (CRD) with 28 male R. novergicus, Wistar strain in aged 8-12 weeks. Group K was not given any treatment; R was given Rodhamine B, S was given Saccharin, KRS was given Rodhamine B and Saccharin, YR was given Rodhamine B and yoghurt, YS was given Saccharin and yogurt, YRS was given Rodhamine B, Saccharin and yogurt. The dose of Rodhamine B was 22.5 mg.kg-1, Saccharin was 157.77 mg.kg-1, and the volume of yoghurt was 1 mL.head-1. They were administered orally with a gastric tube for 14 days. The concentration of purple rosella extract added to yoghurt was 15% (v/v). Parameters were observed in histopathology of the duodenum and colon with the scoring method. Data analysis was done with the Kruskal-Wallis test followed by the Mann-Whitney test. The KRS group showed the heaviest damage, namely necrosis, villi erosion, and inflammatory cell infiltration of >75% of the total visual field in the duodenum and colon. The YR, YS, and YRS groups showed significant changes compared to the R, S, and KRS groups, namely necrosis, villous erosion, and inflammatory cell infiltration, which appeared to be 25-50% of the total visual field. However, this did not match the condition of group K, which appeared to be <25% of the total visual field. The provision of purple rosella yoghurt can prevent 50% damage to the duodenum and colon of Rattus novergicus due to Rodhamine B and Saccharin. Keywords: colon, duodenum, Rodhamine B, Saccharin, yoghurt.
