Abstract

Purple rice is rich in ferulic acid, a phenolic acid abundant in rice. Ferulic acid (FA) has long been recognized for its anti-oxidant properties, which are linked to its skin anti-aging activity owing to its ability to scavenge free radicals and pro-tect cells from damage. Skin anti-aging activity could be assessed by inhibiting skin aging enzymes. However, there was limited information on the mechanism of FA inhibition of these enzymes. This study focused on the molecular interac-tion between purple rice FA and collagenase or tyrosinase, both with and without their substrates. The interaction be-tween FA (CID 445858), collagenase (2Y6I), tyrosinase (5M8M), collagenase substrate (CID 5486160), and tyrosinase substrate (CID 6057) was docked using Hex 8.0.0 Cuda with the default settings. Discovery Studio 2020 and LigPlot software were used to visualize the docking results. Ascorbic acid (CID 54670067) and kojic acid (CID 3840) were used as positive controls for collagenase and tyrosinase, respectively. Molecular docking revealed that FA formed electro-static, hydrogen, and hydrophobic bonds with all aging enzymes, substrates, and enzyme–substrate complexes. FA could bind to both free enzymes and free substrates, consequently attenuating enzyme–substrate bonding, and preventing the substrate from binding appropriately in its normal position. Molecular dynamics simulations demonstrated that FA formed a stable bond with tyrosinase, similar to kojic acid, although the bond with collagenase was less stable. However, the pattern of each parameter during the simulation was comparable with that of ascorbic acid. The current study indicated that the FA from purple rice had the potential to act as an inhibitor of skin aging en-zymes and could be developed as a skin anti-aging candidate

Abstract

The Indonesian purple rice (IPR), a crossbreeding of black and white rice, is a potential source of ferulic acid. Up to this point, the genomic similarity between the crossbreeding and its parentals has not been determined, particularly in genes involved in ferulic acid biosynthesis. In this present work, we analysed the profile of Oryza sativa caffeic acid-O-methyltransferase (OsCOMT) gene in IPR. The genomic DNA was extracted by CTAB method, then amplified with a specific primer of OsCOMT gene. The amplicon was sequenced by Sanger method and confirmed by geneID NC_029259.1. The IPR showed an amplicon of 708 bp sequence of the OsCOMT gene, indicating the gene involved in ferulic acid biosynthesis. The IPR gene profile is similar to the parentals, but there are variations in point mutations that distinguish it from the parentals. Aspartic acid was changed to Glutamine by mutations at positions 52, 69, and 79. We suggest that IPR has a novel OsCOMT gene variant that is unique as it is the crossbreed.

Abstract

Stroke is a deadly disease in Indonesia. Meanwhile, marine organisms can contribute to providing active compounds for anti-stroke. The purpose of this review is to provide an overview of stroke sufferers in Indonesia and to identify potential active compounds from marine organisms that can treat or prevent stroke. The writing method was carried out by tracing the literature related to stroke and organisms that produce anti-stroke active compounds through Google Scholar. After collecting data from various literature, the next step was to process the data using simple statistics. Marine organisms that potentially contain anti-stroke active compounds are sea cucumbers (DHA, Docosahexaenoic acid), sea urchins (flavonoids), starfish (flavonoids), and jellyfish (amino acids), while marine organism biomarkers are biological responses of all living things in the sea to pollution and environmental pressures. This review is expected to be the beginning of the development of bioprospecting based on omics: metabarcoding and metagenomics, transcriptomics, proteomics, glycomics, lipidomics, and metabolomics.

Abstract

Hypertension is a major global health concern and a leading risk factor for cardiovascular mortality. Conventional treatment with ACE inhibitors like captopril is effective but often causes adverse effects including cough, dizziness, and renal impairment. This study evaluated the antihypertensive effects of rice bran in DOCA-salt-induced hypertensive rats. In vitro analysis revealed 45.58% soluble fiber and rich amino acid content, including 2.121 mg L-Arginine, a nitric oxide precursor promoting vasodilation. Male Wistar rats were divided into five groups: negative control, positive control, captopril treatment (5 mg/kg BW), and rice bran supplementation at 1% and 2% of 25 g feed. The 2% rice bran group significantly (p < 0.05) reduced renal iNOS expression by 88.32% compared to hypertensive controls, surpassing captopril's 22.96% reduction. Histopathological examination revealed notable renal protection with complete absence of fatty degeneration and minimal glomerular inflammation in the 2% rice bran group. These findings demonstrate potent antihypertensive activity through ACE inhibition, enhanced nitric oxide bioavailability, and mitigation of oxidative and inflammatory stress, establishing rice bran as a promising natural therapeutic alternative for hypertension management

Abstract

Desa Sidorahayu, Kecamatan Wagir, Kabupaten Malang menghadapi permasalahan pengelolaan sampah organik rumah tangga yang belum optimal, khususnya limbah kulit buah dan sayuran yang berpotensi mencemari lingkungan. Keterbatasan pengetahuan dan keterampilan masyarakat dalam mengolah sampah menjadi produk bernilai guna menjadi permasalahan krusial yang memerlukan solusi aplikatif dan berkelanjutan. Program ini menawarkan penerapan Teknologi Tepat Guna (TTG) berupa inovasi eco enzim sebagai solusi pengelolaan sampah organik sekaligus upaya pemberdayaan masyarakat. Kegiatan dilaksanakan melalui tahapan sosialisasi, pelatihan, demonstrasi, praktik langsung, serta pendampingan produksi dan pemanfaatan eco enzim hingga tingkat dusun. Mahasiswa KKN Tematik berbobot 4 SKS dilibatkan sebagai fasilitator untuk mempercepat transfer pengetahuan dan meningkatkan partisipasi warga. Hasil evaluasi kegiatan semakin menegaskan efektivitas program, dengan 90% peserta menilai proses pembuatan eco enzim sangat mudah dan 88% menyatakan kegiatan ini sangat bermanfaat, hal ini menunjukkan tingkat penerimaan yang sangat baik sekaligus mengindikasikan peluang keberlanjutan program di tingkat komunitas. Selain itu, terbentuk kelompok warga pengelola eco enzim yang mulai memanfaatkan produk untuk kebutuhan rumah tangga dan kebersihan lingkungan. Program ini menunjukkan bahwa inovasi eco enzim efektif sebagai pendekatan pemberdayaan masyarakat dalam pengelolaan sampah organik secara berkelanjutan.
