Abstract

Stroke is a deadly disease in Indonesia. Meanwhile, marine organisms can contribute to providing active compounds for anti-stroke. The purpose of this review is to provide an overview of stroke sufferers in Indonesia and to identify potential active compounds from marine organisms that can treat or prevent stroke. The writing method was carried out by tracing the literature related to stroke and organisms that produce anti-stroke active compounds through Google Scholar. After collecting data from various literature, the next step was to process the data using simple statistics. Marine organisms that potentially contain anti-stroke active compounds are sea cucumbers (DHA, Docosahexaenoic acid), sea urchins (flavonoids), starfish (flavonoids), and jellyfish (amino acids), while marine organism biomarkers are biological responses of all living things in the sea to pollution and environmental pressures. This review is expected to be the beginning of the development of bioprospecting based on omics: metabarcoding and metagenomics, transcriptomics, proteomics, glycomics, lipidomics, and metabolomics.

Abstract

Hypertension is a major global health concern and a leading risk factor for cardiovascular mortality. Conventional treatment with ACE inhibitors like captopril is effective but often causes adverse effects including cough, dizziness, and renal impairment. This study evaluated the antihypertensive effects of rice bran in DOCA-salt-induced hypertensive rats. In vitro analysis revealed 45.58% soluble fiber and rich amino acid content, including 2.121 mg L-Arginine, a nitric oxide precursor promoting vasodilation. Male Wistar rats were divided into five groups: negative control, positive control, captopril treatment (5 mg/kg BW), and rice bran supplementation at 1% and 2% of 25 g feed. The 2% rice bran group significantly (p < 0.05) reduced renal iNOS expression by 88.32% compared to hypertensive controls, surpassing captopril's 22.96% reduction. Histopathological examination revealed notable renal protection with complete absence of fatty degeneration and minimal glomerular inflammation in the 2% rice bran group. These findings demonstrate potent antihypertensive activity through ACE inhibition, enhanced nitric oxide bioavailability, and mitigation of oxidative and inflammatory stress, establishing rice bran as a promising natural therapeutic alternative for hypertension management

Abstract

Desa Sidorahayu, Kecamatan Wagir, Kabupaten Malang menghadapi permasalahan pengelolaan sampah organik rumah tangga yang belum optimal, khususnya limbah kulit buah dan sayuran yang berpotensi mencemari lingkungan. Keterbatasan pengetahuan dan keterampilan masyarakat dalam mengolah sampah menjadi produk bernilai guna menjadi permasalahan krusial yang memerlukan solusi aplikatif dan berkelanjutan. Program ini menawarkan penerapan Teknologi Tepat Guna (TTG) berupa inovasi eco enzim sebagai solusi pengelolaan sampah organik sekaligus upaya pemberdayaan masyarakat. Kegiatan dilaksanakan melalui tahapan sosialisasi, pelatihan, demonstrasi, praktik langsung, serta pendampingan produksi dan pemanfaatan eco enzim hingga tingkat dusun. Mahasiswa KKN Tematik berbobot 4 SKS dilibatkan sebagai fasilitator untuk mempercepat transfer pengetahuan dan meningkatkan partisipasi warga. Hasil evaluasi kegiatan semakin menegaskan efektivitas program, dengan 90% peserta menilai proses pembuatan eco enzim sangat mudah dan 88% menyatakan kegiatan ini sangat bermanfaat, hal ini menunjukkan tingkat penerimaan yang sangat baik sekaligus mengindikasikan peluang keberlanjutan program di tingkat komunitas. Selain itu, terbentuk kelompok warga pengelola eco enzim yang mulai memanfaatkan produk untuk kebutuhan rumah tangga dan kebersihan lingkungan. Program ini menunjukkan bahwa inovasi eco enzim efektif sebagai pendekatan pemberdayaan masyarakat dalam pengelolaan sampah organik secara berkelanjutan.

Abstract

Heparin is a linear polysaccharide composed of repeating 1–4 linked units of iduronic acid and glucosamine and is widely used as an anticoagulant drug. This paper reports characteristics of three-dimensional structural changes and stability of the FGF2–FGFR1 ternary complex in the presence of heparin 8- and 12-saccharide fragments with a 2S sulfation pattern and ^1C₄ iduronate conformation using in silico approach. Study was performed using molecular docking followed by 100 ns molecular dynamics (MD) simulations. Heparin 8- and 12-saccharide ligands were docked to the FGF2–FGFR1 ternary complex, then was evaluated by MD using GROMACS to monitor backbone root mean square deviation (RMSD), residue root means square fluctuation (RMSF), and key residue interactions over time. Docking suggested a more favorable initial binding energy for the 8-saccharide ligand, whereas MD revealed that the 12-saccharide ligand more effectively maintained stable interactions between FGF2 and FGFR1, as indicated by lower backbone RMSD at the end of the simulation and more persistent contacts at basic residues in the heparin-binding sites of both proteins. These dynamic stability features provide a mechanistic explanation for previously reported in vitro findings, showing that 12-saccharide 2S heparin fragments exert stronger FGF2-dependent antiangiogenic and anticancer effects than shorter fragments, thereby supporting their potential as environmentally friendly anticancer drug candidates.

Abstract

Lead has very toxic properties and cannot be degraded in waters that leads to health problems, such as disorders of the nervous system, heart, kidneys, digestive tract, and causing memory loss. One of the environmentally friendly methods that can reduce and even eliminate Pb is the biosorption. One of the microorganisms that can be utilized in biosorption technology is Aspergillus niger. In the current study, Pb biosorption was carried out by modifying Aspergillus niger which was immobilized in the Ca-alginate matrix. This study aims to determine the optimum biosorption conditions which include pH, contact time, initial Pb concentration, and amount of adsorbent. Results showed that the optimum conditions for Pb biosorption were pH 6.0, with contact time of 4 h, Pb concentration was 80 ppm, and at adsorbent concentration of 3 g, with the percentage of Pb adsorbed was 98.40%. FTIR spectra results show that the Pb biosorption process causes shifts in certain wavenumbers, from 1296.16 cm-1 to 1307.74 cm-1 which was a characteristic of the amine amide (CN) group, and a shift in wavenumber from 1122.57 cm-1 to 1168.86 cm-1 which was from carboxyl functional group. The interaction between Pb and the adsorbent surface was suggested through a physical adsorption, and hence the adsorption process was reversible.

Abstract

Tanaman pletekan (Ruellia tuberosa L.) diketahui memiliki kandungan metabolit sekunder seperti flavonoid, fitosterol, sehingga diusulkan memiliki aktivitas biologis sebagai inhibitor enzim alpha-amilase. Mikroenkapsulasi merupakan metode yang dapat melindungi serta mengontrol pelepasan senyawa aktif. Penelitian ini bertujuan untuk melakukan fabrikasi terhadap ekstrak air pletekan dengan menggunakan bahan penyalut gum Arabik dan menguji aktivitasnya sebagai inhibitor untuk enzim alpha-amilase, serta mengetahui karakter mikrokapsul ekstrak air pletekan yang dihasilkan. Mikroenkapsulasi dilakukan pada variasi pH (3, 4, 5, dan 6) dan waktu pengadukan (30, 60, 90, dan 120 menit). Kondisi optimum mikrokapsul ditentukan berdasarkan penentuan efisiensi mikroenkapsulasi. Hasil penelitian menunjukkan bahwa pH 5 merupakan kondisi optimum dengan nilai efisiensi mikroenkapsulasi sebesar 51,3%. Sedangkan waktu pengadukan 90 menit merupakan kondisi optimum dengan nilai efisiensi mikroenkapsulasi sebesar 52,7%. Uji aktivitas inhibisi enzim alpha-amilase pada mikrokapsul dilakukan pada kondisi optimum menghasilkan nilai IC50 sebesar 71,61 µg/mL.. Karakterisasi menggunakan spektrofotometer FTIR menghasilkan bilangan gelombang 3401,51 cm-1 (O-H), 1608,77 cm-1 (C=C), dan 1081,07 cm-1 (C-O-C) yang menandakan terbentuknya mikrokapsul ekstrak air pletekan. Sedangkan hasil karakterisasi menggunakan SEM menunjukkan bahwa permukaan mikrokapsul yang dihasilkan masih heterogeny, dengan kecenderunagn berbentuk bulat, dan menghasilkan ukuran yang beragam berkisar antara 0,933 – 3,08 µm.
