Abstract

Purple rice is a potential source of ferulic acid, which has antimicrobial properties. However, the inhibitory mechanism of ferulic acid on the growth of bacteria, particularly Salmonella and Listeria, has not been elucidated. This study aimed to determine the bioactivity of ferulic acid from purple rice as an antimicrobial agent against Salmonella typhimurium and Listeria monocytogenes using in vitro and in silico analyses. The antimicrobial activity of a purple rice ferulic acid extract was tested using the agar well diffusion method. Its effect on bacterial cells was observed using scanning electron microscopy. Ferulic acid was confirmed to have antimicrobial properties using in silico software to attenuate the binding of bacterial virulence factors (lipoproteins, lipopolysaccharides, and flagellins) to Toll-like receptors (TLRs) and to prevent interactions with peptidoglycans. The purple rice ferulic acid extract inhibited bacterial growth. The inhibitory effects included induction of a biofilm and shrinkage of S. Typhimurium, as well as osmotic lysis of L. monocytogenes. This activity was supported by the ability of ferulic acid to inhibit the binding of bacterial virulence factors with TLRs and block bacterial peptidoglycans. These findings indicate that the purple rice ferulic acid extract acts as antimicrobial, both directly to bacterial cells and indirectly through TLRs. We conclude that ferulic acid from Indonesian purple rice has a biological function as an antimicrobial agent.

Abstract

This study aims to co-microencapsulate the Ruellia tuberosa L. and Cosmos caudatus K. extracts, with chitosan–sodium tripolyphosphate (Na-TPP) as coating material. α-Amylase inhibition and antioxidant assays were conducted to determine the potential of microcapsules used as antidiabetic agents. The microcapsules were manufactured under the influences of pH, Na-TPP concentration, and stirring time. The optimum microencapsulation conditions were selected based on the highest encapsulation efficiency. The optimum microencapsulation conditions were a pH of 4, Na-TPP concentration of 0.15% (w/v), and stirring time of 60 min. The microcapsules exhibited an IC50 (inhibitory concentration) value of 223.64 ± 0.81 µg/mL and an α-amylase inhibition and antioxidant activity of 104.05 ± 0.88 µg/mL. The test for the release of bioactive compounds from microcapsules was conducted in HCl pH 1.2 and phosphate buffer pH 7.4 for 30–120 min. Results showed that 5.99% and 58.96% of bioactive compounds were released at pH 1.2 and 7.4, respectively, in 120 min. The Fourier transform infrared spectra showed the P=O functional group vibrations from Na-TPP at 1,213.71 cm−1 and C–N stretching vibrations from chitosan at 1,155.23 cm−1. Characterization with scanning electron microscopy and particle size analysis indicated that the microcapsules were spherical and had a mean diameter of 132.08 µm. The current study demonstrated that co-microencapsulation is a promising multifaceted approach for the enhancement of the pharmaceutical applications of plant extract combinations

Abstract

Caspase-3 is a biochemical marker for cell apoptosis. Several studies focused on exploring caspase inhibitor potential in natural compounds. Hence, in this study investigated the anthocyanins as anti-apoptotic potential activity through caspase-3 using molecular docking. Six types of anthocyanin were retrieved from PubChem database and caspase-3 protein was downloaded from Protein Data Bank. Anthocyanins and caspase-3 protein were docked using HEX 8.0 program and visualized using Discovery Studio 4.1 software. The interaction among cyanidin-3-O-glucoside, delphinidin-3-O-glucoside, pelargonidin-3-O-glucoside, peonidin-3-O-glucoside and petunidin-3-O-glucoside showed similar binding pattern on caspase-3 protein. All of them bind to BIR2 region and allosteric site of caspase-3, which are a crucial site for apoptosis regulation. Interestingly, malvidin-3-O-glucoside also interacted with caspase-3 in BIR1, BIR2 and BIR3 regions. In addition, anthocyanins-caspase-3 complex showed low energy and demonstrated several hydrogen bonds, hydrophobic interactions and van der Waals interactions, which indicated stable interaction. This study implies that all anthocyanins have potential as inhibitor of caspase-3 protein and might have potential as anti-apoptosis. Further in-vitro and in-vivo studies are need to confirm this experimental.

Abstract

The phytochemicals contained in rice bran, mainly flavonoid compounds, are predicted to have biological activity. Flavonoids are able to counteract the free radicals and degrade insulin resistance. The East Java Red Rice Bran samples, e.g., Mentik Wangi, Aek Sibundong, and Blambangan, were used in the study. Their phytochemical profiles, functional groups, antioxidant, and antidiabetic activities were investigated. The phytochemical analysis showed that the bran of Mentik Wangi, Aek Sibundong, and Blambangan contained flavonoid, triterpenoid, phenolic, tannin, and glycoside. Based on the FTIR, some functional groups were identified in three rice bran varieties, namely, the O-H stretching, C-H aliphatic, C-H sp3 stretching, C=C stretching aromatics, C=C stretching alkenes, CH2 and CH3 bonds rocking, C-H aromatic, CH-OH stretching alcohols, and C-O stretching ether or ester suggesting that rice brans are rich in phytochemical compounds. Through LC-HRMS analysis in positive ion mode, several types of flavonoids were confirmed. Pinocembrin was found in the three brands. The highest antioxidant and antidiabetic activity were observed in Blambangan rice bran with an IC50 value of 1.09 and 75.76 µg/mL, respectively. To conclude, the red rice bran phytochemical compounds exhibit potential biological activities as antioxidant and antidiabetic agents.

Abstract

Tanaman pletekan (Ruellia tuberosa L.) diketahui memiliki kandungan metabolit sekunder seperti flavonoid, fitosterol, sehingga diusulkan memiliki aktivitas biologis sebagai inhibitor enzim alpha-amilase. Mikroenkapsulasi merupakan metode yang dapat melindungi serta mengontrol pelepasan senyawa aktif. Penelitian ini bertujuan untuk melakukan fabrikasi terhadap ekstrak air pletekan dengan menggunakan bahan penyalut gum Arabik dan menguji aktivitasnya sebagai inhibitor untuk enzim alpha-amilase, serta mengetahui karakter mikrokapsul ekstrak air pletekan yang dihasilkan. Mikroenkapsulasi dilakukan pada variasi pH (3, 4, 5, dan 6) dan waktu pengadukan (30, 60, 90, dan 120 menit). Kondisi optimum mikrokapsul ditentukan berdasarkan penentuan efisiensi mikroenkapsulasi. Hasil penelitian menunjukkan bahwa pH 5 merupakan kondisi optimum dengan nilai efisiensi mikroenkapsulasi sebesar 51,3%. Sedangkan waktu pengadukan 90 menit merupakan kondisi optimum dengan nilai efisiensi mikroenkapsulasi sebesar 52,7%. Uji aktivitas inhibisi enzim alpha-amilase pada mikrokapsul dilakukan pada kondisi optimum menghasilkan nilai IC50 sebesar 71,61 µg/mL.. Karakterisasi menggunakan spektrofotometer FTIR menghasilkan bilangan gelombang 3401,51 cm-1 (O-H), 1608,77 cm-1 (C=C), dan 1081,07 cm-1 (C-O-C) yang menandakan terbentuknya mikrokapsul ekstrak air pletekan. Sedangkan hasil karakterisasi menggunakan SEM menunjukkan bahwa permukaan mikrokapsul yang dihasilkan masih heterogeny, dengan kecenderunagn berbentuk bulat, dan menghasilkan ukuran yang beragam berkisar antara 0,933 – 3,08 µm. Abstract

Menguatnya industri halal di Indonesia berimplikasi positif pada peningkatan tingkat perekonomian masyarakat. Sertifikasi halal selain sebagai bentuk perlindungan terhadap jaminan kehalalan produk, juga memberikan nilai tambah yang berimplikasi positif terhadap volume produksi dan penjualan. Sertifikasi halal adalah suatu proses untuk memperoleh sertifikat halal melalui beberapa tahap pemeriksaan untuk membuktikan bahwa bahan, proses produksi, dan sistem jaminan halal dapat memenuhi standar Badan Penyelenggara Jaminan Produk Halal. Tujuan dari kegiatan pengabdian kepada masyarakat ini adalah pendampingan pendaftaran Nomor Induk Berusaha (NIB) bagi Usaha Mikro dan Kecil (UMK); pendampingan tentang peraturan jaminan produk halal bagi industri makanan dan minuman UMK; serta pendampingan pengajuan sertifikasi halal bagi UMK. Metode pelaksanaan kegiatan yaitu melalui sosialisasi, pendampingan pengisian persyaratan, dan pengajuan sertifikasi produk halal. Hasil kegiatan adalah pemahaman pelaku usaha tentang jaminan produk halal bagi pelaku UMK di sektor pangan, pendaftaran sertifikasi halal bagi pelaku usaha, dan sertifikat halal bagi UMK yang telah turun sebanyak 10 sertifikat halal.

Abstract

Background: Rhodamin B and saccharin are chemicals that are harmful to the health of the body. Consumption of rhodamine B and saccharin for a long time can cause problems for the body such as impaired organ function. Purpose: The aim was to find out the benefits of giving purple rosella yogurt in preventing damage to the kidneys and ileum of Rattus norvegicus caused by rhodamine B and saccharin. Methods: Using RAL with 28 male Rattus novergicus aged 8-12 weeks. Group K(-) was not given any treatment, K(+R) was given rhodamine B, K(+S) was given saccharin, K(+K) was given rhodamine B and saccharin, P1 was given rhodamine B and yoghurt, P2 was given saccharin and yoghurt, P3 was given rhodamine B, saccharin, and yogurt. The dose of rhodamine B was 22.5 mg/kg, the dose of saccharin was 157.7 mg/kg, the volume of yoghurt was 1 ml/head. Administered orally with gastric tube for 14 days. The concentration of purple rosella extract used was 15% (v/v). Results: Group K(+K) showed the heaviest damage. Groups P1, P2, and P3 still found damage to the kidneys, in P1 they experienced thickening of Bowman's capsule in the glomerulus, in P2 they experienced a loss of more than 25% of the brush border in the tubules, and in P3 they experienced glomerular tufts in the glomerulus. In the ileum, P1 experienced thickening of the muscles, P2 experienced clusters of inflammatory cells in the submucosa, and P3 experienced 10-50% loss of goblet cells. Conclusion: Provision of purple rosella yogurt can prevent damage to the kidneys and ileum of Rattus novergicus due to rhodamine B and saccharin, although it is not optimal.

Abstract

Teachers and students need more knowledge to improve their competence. The educational quality can be improved by varying learning methods and supported by competition. This community service activity aims to strengthen the competence of teachers and students through learning innovation in the Biology subject and to provide learning competition of Biology Olympiad through simulation and training. The community-based research (CBR) method was performed by workshop and training involving 60 participants. As many as 100% of the participants attended the activities. The results indicated that the participants acquired new knowledge and realized their competence skills. Learning innovation and competition were able to improve their knowledge and competence. The participants are eager to apply the methods learned at school. These new competence and skill are expected to improve the quality of education

Abstract

This study aims to encapsulate Cosmos caudatus K. leaf extract in sodium alginate crosslinked with calcium chloride (CaCl2) as coating materials through freeze-drying. Antioxidant assays were performed to evaluate the applicability of the microcapsules as antidiabetic medicines. Their characteristics, antioxidant activity, and encapsulation efficiency (EE) were also examined. Results indicated that the concentration of sodium alginate and pH influenced the manufacturing process of the microcapsules. The highest EE was obtained at pH 6 and alginate concentration of 2% (w/v). The IC50 for the antioxidant activity of the microcapsules was 139.96 ± 1.094 μg/mL. Scanning electron microscopy confirmed the presence of irregular and spherical structures on the surface. Fourier transform infrared spectra showed new absorption bands at 1593.10 and 1427.59 cm−1, demexistening the existence of stretching vibrations of COO−. Absorption at 1024.14 cm−1 demonstrated C–C and C–O bond stretching vibrations in the sodium alginate–CaCl2 crosslinks. Fourier transform infrared spectrum analyses indicated that sodium alginate and CaCl2 formed chemical bonds, enabling microencapsulation. This study discovered that microencapsulation is a highly prospective and adaptable strategy for enhancing the medicinal use of plant extract.

Abstract

Children's understanding of science at RA Az Zahra is still very low, children are less active and feel less active bored and still having difficulty understanding science activities. This condition is caused by use the method used is not appropriate. The aim of the researcher in conducting this research is to know Children's understanding of science increases through the use of experimental methods. Researcher using a type of classroom action research (PTK). In this study took 10 girls aged 5-6 years as research subjects. Researcher using data collection techniques in the form of observation, performance and documentation. Results Action research can be seen in observing children's development in each cycle, namely pre-cycle with a score of 30%, a percentage of 50% in the first cycle stage and a percentage of 80% in cycle II stage. Percentage increase in understanding of science through experimental methods has achieved a success indicator of 80%. So that is the conclusion of this research proves that the experimental method has been proven to be able to increase understanding science in children aged 5-6 years. This can be seen from cycle II which reached an indicator of 80%.
