Abstract

Chitosan shares characteristic with dietary fiber which can interact with lipids and/or  cholesterol in the intestinal lumen, consequently, reduce their absorption.  The effect of chitosan concentration on the serum cholesterol levels (total cholesterol, HDL, LDL cholesterol and trigliceride) and digesta profile of Sprague Dawley rats were investigated. A two month old Sprague Dawley rats consisted of six groups with seven animals of each group were adapted for one week by giving standard diet (AIN-93). Then they were treated with hypercholesterol diet containing approximately 200 mg/kg cholesterol for one week.  At the end of this week, cholesterol level was measured. The parameter for hypercholesterolemic rats was the raising level of serum cholesterol as much as 130 mg/dl. The first three groups of rats were treated with standard diet (AIN-93) with 0%, 2.5%, and 5%  of chitosan.  The second three groups were treated with hypercholesterol diet with the same treatments as the first one. Blood was drawn from eyes (reorbital plexus) every week for analyzing cholesterol levels (total cholesterol, HDL, LDL cholesterol and triglicerides). At the end of the treatment, seven rats were killed after one day fasting then liver and caecum were collected for analyzing.Research showed that chitosan of 2.5% and 5% in the diet did not alter body and liver weight of the group with standard diet (AIN-93) treatment. Conversely, body wieght decreased and liver weight increased in the hypercholesterol diet treatments. Chitosan also increased digesta weight and cholesterol level either in standard or hypercholesterol diet treatments. Serum profiles showed that chitosan treatment of 2.5% or 5% decreased total cholesterol, LDL cholesterol and triglyceride but increased HDL cholesterol.Keyword: lipid profile, blood serum, chitosan, hypercholesterol

Abstract

Background : As one of important markers of cardiovascular complication in patients with type 2 diabetes mellitus, low density lipoprotein (LDL) shall be maintained to be in normal range. Based on previous research, consumption of probiotic yogurt contains Lactobacillus acidophilus and Bifidobacterium animalis subsp. Lactis is presumed can improve LDL in patients with type 2 diabetes mellitus but the scientific evidence is still few.Objectives: To investigate the effect of probiotic yogurt on LDL in patients with type 2 diabetes.Methods: This study was double blinded randomized controlled trial which was conducted on 32 patientss with type 2 diabetes mellitus. Subjects were divided randomly into 2 groups and given 4 weeks intervention. Subjects in control group were given conventional yogurt while subjects in intervention group were given probiotic yogurt contains L. acidophilus LA5 and B. animalis subsp. Lactis Bb12. Food intake data was collected 4 times using 24-hour recall method during intervention period. LDL of subjects was measured before and after intervention.Results: There was no significant difference between LDL before and after intervention in control group (128 – 148.5 mg/dL) and intervention group (130 – 120.5 mg/dL). Meanwhile, LDL after intervention in control group and intervention group were significantly different (p<0.05). LDL between energy and macronutrients intake categories were not significantly different.Conclusion: Probiotic yogurt has a better LDL reduction effect in patients with type 2 diabetes mellitus than conventional yogurt, although the reduction is not significant.

Abstract

Background: Generally, patients with type 2 diabetes mellitus (T2D) have dysbiosis condition. Dysbiosis can increase oxidative stress that leads to hyperglycemia. Previous researches showed that yogurt consumption can reduce blood glucose in T2D, so it can be used as an alternative healthy snack for T2D patients.Objectives: To investigate the effects of probiotic and conventional yogurt with dosage 100ml/day on fasting blood glucose (FBG) in T2D patientsMethods: Randomized controlled trial, double blind with pre-post group design. The 30 T2D patients from 3 publics health centre in Yogyakarta, were assigned to two groups. Each group, either control or intervention group, received 100 ml/d of yogurt for 4 weeks. FBG samples was assessed before and after intervention periodResults: FBG significantly decreased in both group, which was -27 mg/dL in control group (p<0.05) and -19 mg/dL in intervention group (p<0.05). No significant difference in FBG change between intervention and control group, but control group has greater reduction in FBG compared to intervention group. Conclusion: Conventional yogurt has no significant difference effect in FBG change compared to probiotic yogurt.

Abstract

The objectives of this study were to evaluate the effect of purple sweet potato (PSP) yogurt on thelipid profile of Sprague Dawley rats. The PSP yogurt was made from fresh PSP (F-PSP) and its powder (P-PSP).The viscosity, pH, total titratable acid, proximate, antioxidant activity, anthocyanin content, and total lacticacid bacteria (LAB) between those yogurts were compared. Selected yogurt was given to Sprague Dawley ratsfor 4 weeks. Serum lipid profiles were determined after 2 and 4 weeks of treatment. F-PSP yogurt has betterphysical, chemical and microbiological properties compared to P-PSP yogurt. The serum cholesterol andtriglyceride levels in F-PSP yogurt group was not significantly different compared to control group (P>0.05).However, the reduction of serum LDL cholesterol in F-PSP yogurt group was significantly higher than controlgroup. The serum HDL cholesterol in the yogurt group decreased after 2 weeks of treatment, but it increasedafter 4 weeks of treatment. F-PSP yogurt could be used as an alternative functional food that could lower theblood cholesterol level because of the ability to reduce the LDL cholesterol level.

Abstract

Bovine and porcine gelatin are usually used as ice cream stabilizer. However, Muslims areforbidden to consume foods that contain porcine or its derivatives product. Hence, in our previous study, wedeveloped a technique to produce gelatin from local sources such as fish, buffalo, and bovine. This studyaimed to determine the effect of various gelatin on the physical, chemical, and sensory properties of ice cream.Bovine, fish, and buffalo gelatin with the same concentration of 0.3% are added as ice cream stabilizer. Weevaluated the overrun, melting rate, viscosity, as well as the nutrient content. The sensory properties weretested using a hedonic test using a 7-point scale. The substitution of carboxymethyl cellulose (CMC) withvarious gelatin did not affect the overrun, the melting rate, ash and protein content as well as sensoryattributes (p>0.05). On the other hand, ice cream with various gelatin significantly affected the viscosity,moisture, fat, and carbohydrate content (p<0.05).

Abstract

Several studies found that the probiotic bacterias such as Lactobacillus acidophilus La5 and Bifidobacterium lactis Bb12 could prevent glycemia and control the blood lipid profiles. Currently, a probiotic product such as yogurt is not preferable by some consumers since the taste is sour. Therefore, this study was conducted to develop milkshakes supplemented by probiotics Lactobacillus acidophilus La5 and Bifidobacterium lactis Bb12 which are expected to be more acceptable and have a beneficial effect with the addition of prebiotics and soluble fiber glucomannan. This study aims to determine the effect of synbiotic shakes on fasting blood glucose levels and lipid profiles in hyperglycemia Wistar rat models. The study design was a pre-posttest controlled group with 48 male Wistar rats divided into 8 groups, namely the healthy control group (I); hyperglycemia control group (II); group III that received metformin; group IV that received synbiotic shake with La5 + FOS; group V that received synbiotic shake La5 + inulin; group VI that received synbiotic shake Bb12 + FOS; group VII that received synbiotic shake Bb12 + inulin; and group VIII that received synbiotic shake La5 + Bb12 + FOS. The dose of the intervention was 3.6 ml/day with an intervention duration of 28 days. The results showed that the differences in blood glucose levels were not significant (p> 0.05) except for groups IV and VI that experienced a significant increase (p <0.05). The differences in lipid profiles showed insignificant changes in LDL levels except in groups IV and V, a significant increase in HDL levels (p <0.05) in group V and VI, and insignificant change in triglycerides except in group VI. It can be concluded that the administration of synbiotic shakes with a variety of prebiotics Lactobacillus acidophilus La5 and Bifidobacterium lactis Bb12 for 4 weeks were able to maintain fasting blood glucose levels and lipid profiles in hyperglycemic rats.

Abstract

ABSTRAK Latar Belakang: Faktor risiko anemia pada remaja obesitas meningkat dengan adanya gangguan homeostasis besi yang terjadi, ditandai dengan kadar feritin dan kadar hepcidin tinggi namun kadar hemoglobin rendah yang disebabkan oleh adanya inflamasi kronik derajat ringan terkait obesitas.Tujuan: Penelitian ini dilakukan untuk mengetahui hubungan antara obesitas, faktor perantara yang memengaruhi, dan kejadian anemia pada remaja usia 12-15 tahun di SMP di Kota Yogyakarta.Metode: Penelitian yang dilakukan adalah penelitian observasional dengan rancangan penelitian potong lintang pada 68 siswa SMP di Kota Yogyakarta yang mempunyai status gizi normal dan obesitas. Pengukuran tinggi badan, berat badan, indeks massa tubuh, kadar hepcidin, kadar feritin, kadar hemoglobin, dan penilaian asupan zat gizi responden dilakukan dalam satu kurun waktu yang hampir bersamaan.Hasil: Persentase anemia pada kelompok normal sebesar 15,15% sedangkan pada kelompok obesitas sebesar 2,85%. Pada penelitian ini, tidak terdapat perbedaan secara signifikan pada prevalensi anemia antara remaja status gizi normal dan obesitas (p=0,074). Median data kadar hemoglobin pada kelompok normal 14,2 g/dl dan kelompok obesitas 14,5 g/dl. Kadar feritin pada kelompok obesitas lebih tinggi secara signifikan dibandingkan dengan kelompok normal, masing-masing 9,7 ng/ml pada kelompok normal, 11,59 ng/ml pada kelompok obesitas sedang dan 15,81 ng/ml kelompok obesitas berat. Terdapat perbedaan signifikan pada kadar leukosit pada kedua kelompok responden (p=0,0443), namun tidak terdapat perbedaan signifikan pada kadar hepcidin (p=0,511). Tidak terdapat korelasi antara kadar hepcidin dan feritin pada seluruh responden (p=0,396), serta terdapat korelasi positif tingkat rendah antara kadar feritin dan hemoglobin pada seluruh responden (p=0,0008).Kesimpulan: Terjadi proses inflamasi kronis derajat ringan pada kelompok obesitas, namun tidak terjadi gangguan metabolisme besi akibat inflamasi. Penelitian lebih lanjut dibutuhkan untuk menggali hubungan anemia dengan obesitas di masa dewasa.KATA KUNCI: anemia; gizi remaja; Hemoglobin; obesitas    ABSTRACT Background: The risk factor for anemia in obese adolescents is increased by the presence of iron homeostatic disorder that occurs, characterized by high levels of ferritin and hepcidin levels but low hemoglobin levels which is caused by mild chronic inflammation associated with obesity.Objectives: This study was conducted to determine the relationship between obesity and intermediary factors that affect the incidence of anemia in adolescents aged 12-15 years in Junior High School, Yogyakarta.Methods: This study was observational research with a cross-sectional study design on 68 junior high school students in Yogyakarta City who had normal and obese nutritional status. Measurement of height, weight, body mass index, hepcidin levels, ferritin levels, hemoglobin levels, and assessment of nutrient intake of respondents performed in a period of time.Results: The percentage of anemia in the Normal Group (NG) was 15.15% meanwhile in the Obese Group (OG) was 2.85%. In this study, there was no significant difference in the prevalence of anemia between NG and OG (p=0.074). Median data hemoglobin levels in the NG were 14.2 g/dl and in the OG was 14.5 g/dl. Surprisingly, there was a significantly higher level of ferritin found in the OG than NG, respectively 9,7 ng/ml in NG, 11.59 ng/ml in the moderately obese group, and 15.81 ng/ml severe obese. There was a significant difference in leukocyte levels between groups (p = 0.0443), however, there was no significant difference in hepcidin levels (p=0.511). There was no correlation between hepcidin and ferritin levels in all respondents (p=0.396), and there was a low positive correlation between ferritin and hemoglobin levels in all respondents (p=0.0008).Conclusions: Low-grade chronic systemic inflammation occurs in the OG, but no iron metabolism disorder occurs due to inflammation. Further study is needed to explore the correlation between anemia and obesity in adulthood.KEYWORDS: adolescents; anaemia; Haemoglobin; obesity

Abstract

Background: Protein energy deficiency is a major public health problem in Indonesia. WHO has recommended F100 made from skim milk for undernourished patient. Alternative formulas have been developed using other nutritious foods that are cheaper, easily accessible and can be used for children with lactose intolerance such as tempe flour.Objective: To investigate the effect of F100 and tempe flour formula supplementation on serum Fe and hemoglobin (Hb) levels of undernourished child.Method: This was an experimental study with randomized controlled clinical trial design and purposive sampling method. Subjects of the study were undernourished patients aged 1-10 years hospitalized at Dr. Saiful Anwar Hospital Malang. Sample consisted of 30 patients divided into 2 groups; group 1 supplemented with F100 and group 2 with tempe flour formula. Hb level was assessed by cyanmethemoglobin and serum Fe level by colorimetric method. Dietary intake data was collected by visual comstock and food recall. Data were analyzed by chi square, unpaired t-test and double linear regression.Results: Statistical analysis showed that there were no significant differences in Hb (p=0.139) and serum Fe levels (p=0.313) between both groups after treatment. Intake data indicated that there was a significant disparity in protein (p=0.019) and Fe intake (p=0.006) between the two groups, whereas energy, fat and carbohydrate intake showed no significant differences. Lastly, the association between energy and nutrient intake with serum Fe and Hb levels was not significant and the correlation was weak (r<1).Conclusion: There were no significant differences in hemoglobin and serum Fe levels between F100 group and tempe flour formula group, but there was a significant difference in protein and Fe intake.

Abstract

Background: Macrophages play an important role as part of the innate immune response in the gut and they represent one of the first lines of nonspecific defense against bacterial invasion. Previous studies indicated that probiotics and prebiotics may act as an immunomodulator agents. Nevertheless, research on the immunomodulatory effect of local materials has never been performed.Objective: To study the effects of Lactobacillus plantarum Mut7 and sweet potato fiber on the activity and Nitric Oxide (NO) production of peritoneal macrophages of Sprague Dawley rats.Method: Ninety six Sprague Dawley rats aged 8 weeks were divided into two groups; A (not infected with Salmonella typhimurium) and B (infected with S. typhimurium). Each group was divided into 4 subgroups and assigned to standard AIN-93M diet (KON), 109 CFU/ml of L. plantarum Mut7 (PRO), modified AIN-93M diet with sweet potato fiber (PRE), and both component (SIN). After 3 weeks of treatment, 6 rats of each subgroup were sacrificed and the peritoneal macrophages were isolated and analysed for its activity and NO production. The rest of the rats continued the treatments for another 2 weeks. At the end of the experiment, they were sacrificed and the peritoneal macrophage were isolated and analysed for its activity and NO production.Results: Oral administration of  L. plantarum Mut7, sweet potato fiber, or both improve phagocytic activity of peritoneal macrophage which was indicated by an increase in the percentage of macrophages that phagocyte latex particles (p<0.05) and an increase in the number of latex particles engulfed by macrophages either after 3 or 5 weeks of treatment (p<0.05). Oral administration of L. plantarum Mut7, sweet potato fiber, or both were unable to increase the nitric oxide production after 3 weeks of treatment (p>0.05), but after 5 weeks of treatment the production of NO was significantly increased (p<0.05).Conclusion: L. plantarum Mut7, sweet potato fiber, or both increase the non specific immune response as they could improve the activity and NO production of peritoneal macrophages.

Abstract

Background: Self-medication by using ceplikan leaves (Ruellia tuberosa L.) has been practiced in society for many times in the after-boiled form. It had been proved that liquid extract of ceplikan leaves (Ruellia tuberosa L.) was able to reduce blood glucose level on diabetic rat. It is also well known among diabetes mellitus patient that it has following effect on increasing triglyceride and reducing HDL-cholesterol level.Objective: This research was aimed to give scientific evidence on the society habit on self-medication by using ceplikan leaves (Ruellia tuberosa L.) by examining its oral administration on serum lipid profile pre-clinically.Method: Two months old Rattus novergicus within average weight of 150-200 g were divided into experimental and control groups. The control group consisted of 6 normal rats; while the experimental group consisted of 24 alloxan induced-diabetic rats. The experimental groups were divided into four groups i.e. non-treatment group and oral administrations of Ruellia tuberosa L. liquid extract with different concentrations that were 1.6 mg, 3.2 mg, and 6.4 mg, respectively. The feeds were given ad libitum. The oral administrations of Ruellia tuberosa L. liquid extract were given everyday for 30 days. In the end of the treatment, all rats were fastened and the blood was taken from the eye and being observed for its serum lipid profile. The data were analyzed statistically by using ANOVA and Tukey’s HSD mean difference tests.Results: The results showed that oral administration of 3.2 mg Ruellia tuberosa L. liquid extract was significantly reduced triglyceride and LDL-cholesterol level (p < 0,05) and increased HDL-cholesterol level (p < 0,05).Conclusion: The 3.2 mg of Ruellia tuberosa L. liquid extract was able to reduce triglyceride and LDL-cholesterol level and increase HDL-cholesterol level. Moreover, ceplikan leaves (Ruellia tuberose L.) could be taken as traditional drugs in a non-formal way with control on dietary intake and regular physical exercise.
