Abstract

Terap (Artocarpus elasticus Reinw. ex Blume) is an underutilized fruit tree native to Indonesia, valued both for its edible fruit and its diverse applications in traditional medicine. Terap seeds are recalcitrant, so cryopreservation is the most effective method for their long-term preservation. This study aimed to determine the optimal loading and vitrification times for cryopreservation to maintain seed viability. A factorial, completely randomized design was employed with two factors: loading time (0 and 20 min) and vitrification time (0, 30, and 60 min). Seed viability after loading and vitrification treatment with or without freezing in liquid nitrogen was evaluated using tetrazolium chloride (TTZ) and germination tests. Results indicated that the loading and vitrification treatments did not yet improve the viability of terap seeds after cryopreservation, as determined by the TTZ and germination tests. The TTZ test showed that the viability of all treated seeds decreased, regardless of whether they were frozen in liquid nitrogen. Germination tests revealed that all treated seeds without freezing had reduced germination, while none of the seeds treated with freezing germinated. Structural analysis indicated that freezing alters cell degradation, likely due to the formation of ice crystals and mechanical stress. To improve the success of terap seed cryopreservation, future efforts should focus on optimizing exposure times, reducing PVS2 toxicity, and enhancing cryoprotectant penetration by removing the seed coat or using the embryo axis as an explant.

Abstract

Foxtail millet (Setaria italica L. Beauv.) is an emerging carbohydrate-producing crop. It is considered a climate-resilient crop due to its tolerance to abiotic stresses. Breeding for abiotic-tolerant crops requires powerful tools such as molecular markers. The single-nucleotide amplified polymorphism (SNAP) marker, developed from a single-nucleotide DNA variation at a specific location in the plant genome, is a simple yet powerful PCR-based marker widely used in phenotype-related selection. A SiDREB2-based SNAP marker was previously developed based on a base variation at the 558th base pair (an A/G transition) in the SiDREB2 gene and used to estimate the drought tolerance in foxtail millet. However, the specificity of the marker depends on technical aspects, including the type of DNA polymerase used, primer concentration, and the number of PCR cycles. Here, we reported that non-specific and false positive amplicons can be eliminated by utilizing DNA polymerase with no 3’ to 5’ proofreading activity and reducing the final primer concentration to 1.25 μM. PCR cycle number 25 yielded the optimum specificity, while increasing the cycle to 30 resulted in false positive results. Altogether, our results showed that technical optimization is necessary for improving the specificity of the SNAP marker.

Abstract

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo) is a significant disease attacking rice crops worldwide. This disease attacks at various stages of plant growth and causes significant yield loss. Breeding new varieties resistant to BLB is important to support sustainable agriculture in the future. This study aimed to identify new superior green super rice (GSR) lines resistant to BLB disease. The experiment evaluated the resistance of lowland rice lines obtained from anther culture using a factorial randomized complete block design. The 1st factor was genotype, consisting of 20 lines, 2 checks of commercial varieties (Inpari 42 Agritan GSR and Inpari 18), a resistant check (Conde), and a susceptible check (Taichung Native 1). The 2nd factor was BLB pathotypes (i.e., III, IV, and VIII). Quantitative data on disease severity and severity index were analyzed using analysis of variance and t-Dunnett test at 5% level. The results showed that the interaction between genotype and pathotype affected the disease severity and severity index in both growth phases. The tested lines exhibited varying resistance, from susceptible to resistant, to BLB. Four lines (SN 11, 13, 57, and 58) showed moderate to resistant criteria for BLB disease of 3 pathotypes in both growth phases. The selected lines can be used as a source of parents for breeders and candidates for new superior varieties with BLB resistance properties to support the reduction of synthetic chemical bactericide inputs and control BLB disease. However, further field evaluations are necessary to assess their performance.

Abstract

The improvement of eggplant yield through breeding is crucial. Doubled haploid technology has accelerated the development of varieties. This study utilized doubled haploid anther culture-derived lines (confirmed through flow cytometry and morphology) developed from embryos from a previous study to evaluate the morpho-agronomic performance and yield of doubled haploid eggplant lines. The experiment used a randomized complete block design (RCBD), three replications, and one factor, namely genotype (35 doubled haploid lines, three F1 varieties). Observation was made on plant height, dichotomous height, stem diameter, days to flowering and harvesting, fruit length, fruit diameter, weight per fruit, number of fruits, and fruit yield per plant. The data were analyzed using ANOVA, t-Dunnett, Tukey-Kramer at a 5% level, Pearson correlation coefficient, and selection index. The results showed significant variability in the population of doubled haploid lines. Fruit yield was positively correlated with plant height, dichotomous height, stem diameter, days to flowering, fruit length, and weight per fruit, but negatively correlated with the number of fruits per plant. These variables can be used as selection criteria because of their high heritability and genotypic coefficients of variation. The selection index revealed that the high-yielding doubled haploid lines with desirable morpho-agronomic traits were RS-P2, RS-P6, RS-P9, RS-P14, RS-P18, RS-H19, RS-H20, RS-H23, RS-H27, RS-H3, RS-M31, RS-M32, RS-M33, RS-M34, and RS-M37. The selected lines with high yield and good quality fruit, similar to the commercial hybrid parent, were Hitavi’s derived lines. All selected lines serve as the genetic basis for production improvements and long-term breeding programs for sustainable and productive eggplants that benefit farmers.

Abstract

Rice production is facing a decline due to climate change and land conversion. The development of high-yielding and multi-tolerant abiotic stress rice varieties (such as drought, salinity, and flooding) is needed for adaptation to a changing climate. This research aimed to identify potential doubled-haploid (DH) rice lines with good agronomic performance and high yield through index selection. The study was conducted in the rainy season of 2024, and used a randomized complete block design with a single genotype factor and three replications. The genetic material evaluated consisted of 56 DH rice lines and 5 check varieties, i.e., Inpari 18, Inpari 30, Inpari 34, Inpari 35, and Ciherang. Observation was conducted on plant height (PH), number of vegetative tillers (NVT), number of productive tillers (NPT), days to 50% flowering (DF), days to harvesting (DTH), number of filled grains (NFG), total grains per panicle (TG), panicle length (PL), and productivity. The results showed high heritability in all observed variables, indicating that the variables could be used as selection criteria. The weighted index selection involving PH, NPT, DTH, and productivity resulted in 30 DH rice lines with good agronomic traits such as medium PH, medium NPT, medium maturity, and productivity above the check varieties. Those selected DH lines can be evaluated further in more diverse environments to study the effects of genotype, environment, and G×E interactions.
