Abstract

ABSTRACTDeveloping of  drought tolerant hybrid rice varieties requires parental lines which have tolerance to drought. Polyethylene glycol (PEG) has been widely used as an osmotic solution for detecting drought tolerance at germination stage of rice. The objectives of this experiment were to evaluate variables of drought tolerance at germination stage  and to select drought tolerance of parental lines using osmotic solution PEG 6000 at concentration of 25%. The experiment was conducted at the greenhouse ICABIOGRAD Cimanggu Bogor, during April-May 2014. The design of the experiment was arranged in factorial randomized block design with three replications. The first factor (genotype) had 21 levels and the second factor (osmotic potential) had two levels (0 and 25% concentration levels of PEG 6000). Based on the principal component analysis, six primary indicators were proposed for drought tolerance of rice at germination stage, i.e., germination percentage, seed vigor, seminal root length, seedling length and dry weight of seminal root. Based on the clustering analysis, at 73.76% of similarity rate, the 21 tested genotypes were divided into 2 groups. The genotypes which showed similar responses with Salumpikit were IR 58025B, GMJ 14B, IR 80154B, GMJ 15B, R 3, PK 90, and PK 12 respectively. Abstract

Anther culture is one of tissue culture methods which can be applied to plant breeding programs in order to accelerate the process of obtaining pure lines. The successful development of rice varieties is highly dependent on genetic diversity and desirable traits. To obtain the genetic variability of doubled haploid lines through anther culture techniques, anther F1 or F2 were used as explants sources. The objectives of the study were to select and characterize doubled haploid lines of upland rice having the characters of new plant type, and to study the genetic variability and agronomic characters of tested doubled haploid lines. A total of 58 doubled haploid lines, and four parental lines i.e. Fatmawati, SGJT-28, SGJT-36, and Way Rarem were used in this study. The experiment used completely randomized design with three replications. Results showed that the characters of the doubled haploid lines vary considerably. Selection of the character i.e. number of productive tillers, number of fi lled grain per panicle, and percentage of empty grain was more effective to be selected because they were well correlated to weight of grain per hill, possessed high heritability values, and have wide genetic variability. Based on productive tillers number, number of fi lled grains per panicle, fertility, weight of 1,000 grains, and weight of grains per hill, the lines of P3-26, P3-27, P3-28, P4-45, P5-50, P6-103, P6-105, P3-120, P3-134, P3-135, P3-150, P3-158, P3-248, P3-249, P6-271, P6-272, P6-274, P6-276, and P6-295 were potential for further selection for new type of upland rice.

Abstract

Eighteen Indonesian new plant type (NPT) of rice varieties and lines were evaluated in a ratoon system. The results showed that ratooning ability was related to the vegetative growth prior to harvest, which gave high ability ratoon growth. New plant type varieties and several lines of rice had better vegetative growth and produced higher ratoon than other genotypes. Ratoons started to develop 2-7 days after harvest, with an average number of leaf 2-4 per tiller. Number of productive tillers of ratoon ranged from 6.0 to 30.0 per hill, with number of grain per panicle ranged from 38.0 to 228.2, and grain weight per hill of ratoon 10.4 to 31.2 g, dependent on genotypes. Maturity of ratoon was also dependent on genotypes. On average, maturity was 68 days after harvest of the main crop. Grouping of ratooning ability based on hierarchical cluster analysis resulted in three groups, namely nine genotypes had high potential, fi ve genotypes had medium potential, and four genotypes had low potential. This study indicated that ratoon is potential to increase rice planting index in Indonesia. The genotypes with high ratoon ability need to be evaluated for agronomic performance in the fi eld. Abstract

Anther culture is useful tool in rice breeding. The technique shortens the time to obtain full homozygous plant in just one generation. Information related to genetic variability among the regenerants is important for further analysis including selection activities. The objective of this study was to analyze agronomic character variability and yield potential among doubled-haploid lines (DH0) derived from anther culture. This study was carried out in Center for Agricultural Biotechnology and Genetic Resources Research and Development (BB Biogen) from October 2016 to February 2017. The experiment was arranged in randomized complete block design (RCBD) with three replications. Forty eight doubled haploid lines derived from anther culture and three check varieties were evaluated. The results showed high genetic variability as well as broad sense heritability (more than 90%) for all variables tested. Several lines showed superior agronomic characters compared to check (Ciherang, Inpari 18, Inpago 10). Anther culture could generate high genetic variability for further selection. Keywords: anther culture, broad sense heritability, haploid technology

Abstract

Observation and identification of rice germplasm with advanced traits are important activities in the process of obtaining a variety of rice because every genotype will show unique characters. Genotype derived from breeding program as anther culture need to be characterized to identify the doubled haploid lines that are potential to be developed in red rice breeding. This research was aimed to obtain agronomic characters of doubled haploid lines of red rice. Materials used were 59 genotypes, i.e., 57 doubled haploid (DH0) lines with red grain and 2 check varieties (Inpari 24 and Aek Sibundong) as control. The experiment used a randomized complete block design with three replications. The results exhibited there was agronomic variation among DH0 of red rice lines. Variance analysis showed that genotype affected most characters, except for grain dry weight per hill. Plant height, number of productive tiller, harvesting age, panicle length, number of grains per panicle, and number of filled grain per panicle can be used as a reference for effective selection to develop high productivity of red rice variety.

Abstract

Doubled haploid rice formation through anther culture becomes one of the alternatives in the plant breeding. The lines need to be characterized, clustered, and selected. This study aimed at determining heritability, characters, and cluster of doubled haploid lines obtained from anther culture. This experiment was conducted at IPB Rice Field Experiment Station, Darmaga, Bogor from October 2016 until January 2017. The experiment was managed according to randomized complete block design (RCBD) with 3 replications and genotype as the single factor treatment. Genotypes used were 60 genotypes consisting of 56 doubled haploid lines and 4 control varieties. The controls were Ciherang, Inpara 5, Inpari 29, and Inpari 34 Salin Agritan. The results showed that all characters have significant effect and high heritability (above 60%), so they could be used as a selection character. The tested genotypes could be grouped into three main groups. The first group consisted of 11 genotypes, the second group consisted of 44 genotypes, and the third group consisted of 5 genotypes. The main group determinant characters were productive tillers, percentage of filled grain number, plant height, panicle length, and weight of 1,000 grains. The number of filled grain and productive tillers can be used as selection characters together with yield potential.

Abstract

Rainfed rice tolerant to drought and high yielding would be an alternative to rainfed lowland areas prone to drought stress. Selection in the early phase of plant growth will accelerate the effort to obtain rainfed varieties. The objective of this experiment was to identify characters that significantly affect the selection of drought tolerance using PEG 6000 concentration of 25% in the germination phase. The experiment was conducted in greenhouse of ICABIOGRAD, Bogor, using a factorial randomized complete block design with three replications. The first factor was 32 rice genotypes while the second factor was 2 levels of PEG 6000, i.e., concentrations of 0 and 25%. The results showed that the correlation analysis and principal component analysis obtain the important characters namely the germination percentage, seminal root length, and the dry weight of seminal roots. Based on discriminant analysis, the index matching value of 96.77% was suitable to distinguish tolerant and sensitive genotypes using PEG 6000 concentration of 25% in the germination phase.

Abstract

Protocols for genetic transformation of rice have been widely developed, however the protocols are not universal and inapplicable for all types of rice plants directly. Transformation protocol on rice cv. Fatmawati needs to be developed to generate transgenic lines. The present research was carried out to optimize genetic transformation protocol in rice cv. Fatmawati mediated by Agrobacterium tumefaciens harboring pCambia1301 construct using immature embryo as an explant. The experiment was arranged in a completely randomized design. Factors influencing efficiency of transformation, i.e., sensitivity of callus to hygromycin antibiotic, acetosyringone concentration used in cultivation medium, hygromycin concentration for transformant selection were optimized. The results showed that genetic transformation of rice cv. Fatmawati mediated by A. tumefaciens using immature embryos have been successfully carried out with several parameters. Addition of 100 µM acetosyringone in co-cultivation medium and 30 mg L-1 hygromycin for transformant callus selection were optimal for genetic transformation of rice cv. Fatmawati mediated by A. tumefaciens. Transformation efficiency was found to be 7.84% based on the lines carrying the hpt gene. This result would be a valuable reference in genetic transformation of rice cv. Fatmawati using target genes. Keywords: immature embryo, Oryza sativa, pCambia1301, transformation efficiency

Abstract

Development of doubled haploid lines of upland rice through anther culture is one way to obtain superior upland rice varieties tolerant to drought. The objectives of this research were to determine the response of doubled haploid lines to drought stress at seedling stage and to select tolerant doubled haploid lines. The research was conducted at a screenhouse of Muara Research Station, Indonesia Center of Rice Research (ICRR) from November to December 2017. Fourteen doubled haploid lines and 4 check varieties, namely Inpago 10 and Limboto, Salumpikit (drought tolerant check) and IR 20 (drought sensitive check) were used in this study. The experiment was arranged in a randomized complete block design with three replications. The observations were conducted on leaf rolling, leaf drought, and recovery ability. Based on Friedman nonparametric analysis, HR-5-7-1-1 and HR-7-15-2-1 had moderate response with estimated median values for leaf rolling of 3.44 and 3.00, respectively. Three doubled haploid lines namely HR-2-27-2-7, HR-2-34-1-3 and HR-7-15-2-1 had moderate response to drought with estimated median value of leaf drought of 3.56. Nine lines of doubled haploid had tolerant response with estimated median value of recovery ability of 1.00-1.56. Based on weighted selection index, nine doubled haploid lines with positive and high selection index were identified; however, only two lines (HR-7-15-2-1 and B3-2) had selection index higher than Inpago 10 and Limboto. Keywords: drought tolerance, leaf rolling, recovery, weighted selection index
