Abstract

Adiantum lunulatum Burm. f. has been widely used in aesthetic needs and medical fields. Detail information about the life cycle of A. lunulatum is still unknown, particularly about its gametophyte generation. Present work aimed to study and compare the growth and development of the gametophytes of A. lunulatum from dry and humid areas. Research method consists of two stages: (1) Spores were collected from two locations, Pasuruan and Bogor and (2) Spores were planted and observed the growth and development of gametophyte. The result showed that natural habitat of the plant in various dry and humid areas, affecting the rate of growth and development rates of A. lunulatum gametophyte. The gametophyte from the dry area showed faster rate of growth and development than that of humid areas. The spores collected from dry area need 9 weeks for germination, growth, and development while the spores of humid area took 22 weeks.

Abstract

The morphology of sporophyte, the type of reproduction, and cytology of Pteris had been reported, while the gametophyte morphology of Pteris in Java island has not been studied yet. The objective of this study was to describe the gametophyte morphology and development of P. biaurita, P. ensiformis,  P.exelsa, P.longipinnula, P. tripartita, and   P. vittata in Java island. Spores were obtained from fertile leaves of Pteris plants originated from several locations in Java island. The number of spores per sporangium were counted from fresh fertile leaves with mature sporangia. As much as 0.002 g spores was sown in transparent box with sterile medium contain of vermiculite, spaghnum moss, and perlite with ratio 2:2:1. The gametophyte development of each species was observed under a microscope every 7 days. The spores of P. ensiformis were germinated faster, 10 days after sowing, while the spores of P.longipinnula were germinated slower, 18 days after sowing.  The pattern of spore germination is Vittaria-type. The development of gametophyte is Ceratopteris-type in common, but in a few cases is the Adiantum-type. The gametophyte development of observed Pteris spesies are varied in six characters including the number of filament cell, germinated time, formation time of notch and gametangia, margin shape, and development type.

Abstract

Spathoglottis plicata is a ground orchid that displays large variation in flower features. In spite of large variation on morphological characters, its anatomical and cytological characters are not well known. This paper described the leaf anatomical and chromosomal features of S. plicata. Leaf anatomy was observed from both transverse and paradermal sections of leaves. Chromosome numbers were observed from root tip cells. Epidermis of all S. plicata variants comprised of two cell shapes, the rectangle cells located above veins, while the polygonal one located in between veins. Stomatal type was brachitetracyclic, which were only found on lower epidermis. Leaf transversal section comprised of one layer of upper and lower epidermis, spongy mesophyll, and vascular tissue. The number of leaf hair varied depended on the original habitat and altitute. All variants of S. plicata had same chromosome number (2n=40).

Abstract

Abstract- A few people know Indigofera, but as a admirer of batik and tenun, these plant gives hope for the preservation of batik. Indigofera has a variety of benefits , but more commonly known as blue dye in batik and tenun. The problems of restore the glory of Indigofera from Indonesia who had experienced the triumph can be divided into three principal. There are the increasing scarcity of Indigofera plant, the lack of willingness of the community batik for the cultivation, the process of indigo dyeing longer and complicated, and the results of staining with the natural color is not uniform. The problem by the business presence of more efficient synthetic dyes,  the price of synthetic dyes is cheaper. While research on Indigofera dye is still limited and stagnate, this is a problem should be a concern Botanist .  Keywords: Indigofera, batik, tenun, natural dye

Abstract

Abstrak Selaginella yang banyak dijumpai melimpah di Indonesia sangat  berpotensi sebagai sumber antioksidan alami. Kadar bahan bioaktif dalam tumbuhan  sangat tergantung pada lingkungan tumbuhnya, namun penelitian aspek budidaya dari Selaginella di Indonesia yang berkaitan dengan bahan bioaktif belum banyak dilakukan.  Penelitian awal tentang aspek budidaya Selaginella telah dilakukan khususnya untuk mengetahui naungan yang tepat bagi pertumbuhannya serta pengaruhnya terhadap kandungan bahan bioaktif. Penelitian ini menggunakan rancangan petak terpisah dengan naungan sebagai petak utama dan jenis Selaginella sebagai anak petak. S. plana, S. willdenovii dan S. mayeri diberi perlakuan naungan dengan paranet 40%, 65%, 80%, dan tanpa naungan. Uji kandungan tanin, saponin, dan flavonoid secara kualitatif diketahui dengan melakukan ekstraksi dengan alkohol. Pertambahan cabang total, pertambahan bobot basah total, bobot basah, dan bobot kering biomassa dipengaruhi secara nyata oleh naungan dan jenis Selaginella. Masing-masing jenis Selaginella memerlukan intensitas cahaya yang berbeda untuk mendukung pertumbuhannya. Semua jenis yang diuji juga mengandung kadar tanin, saponin, dan flavonoid berbeda secara kualitatif. Kata kunci:  Selaginella, naungan, tanin, saponin, flavonoid Abstract Selaginella found abandonly in Indonesia is very potensial as the source of natural antioxidant. The amount of bioactive material of Selaginella depends on the environment, but research on the amount of bioactive material correlated to the cultivated aspect has not been done in Indonesia. The preeliminary research of the cultivation of Selaginella was  investigated especially to know the right shading for the best growth of Selaginella. The right shading may also affect bioactive content of Selaginella. This research used split plot design with level of shading as main plot and species as sub plot. S. plana, S. willdenovii, and S. mayeri were grown under paranet with 40%, 65%, 80% shading, and non shading treatments. Bioactive compound was extracted using alcohol 70% and analyzed for tannin, saponin, and flavonoid contents. Results indicated that level of shading and species affected total branch increase, total fresh weight increase, fresh and dry weight of biomass.  Each species required different light intensity to support its growth. All species in the experiment contained different qualitative level of tannin, saponin, and flavonoid. Keywords: Selaginella, shade, tanin, saponin, flavonoid

Abstract

Transferability of 25 rye and 20 wheat simple sequence repeat (SSR) markers were examined across Secale spp. to elucidate their genetic relationships.  One hundred percent of rye and 75% of wheat SSR markers could be amplified in weedy and wild forms of Secale. However, only 84% of rye SSR markers were amplified in S. sylvestre indicating that this species had the furthest relation to the cultivated rye. The PCR products resulting from rye SSR tended to have similar size as the original size, while wheat SSRs tended to have a shorter size than that of the original size. Polymorphism in the allelic number and size was found in 68% of the rye and 40% of the wheat SSR markers.  Several alleles were unique for the annual and perennial rye taxa, which were counted for 22 and 15%, respectively.  Some SSR markers also produced specific alleles that could be used to identify certain taxa.  The genetic relationships within the genus Secale based on SSR markers were discussed.

Abstract

The revision of Ampelocissus was performed by observing the anatomical character of Ampelocissus leaves. The leaves were collected from 33 collection numbers of Herbarium Bogoriense (BO) and four collection numbers from Sumatra exploration. The purpose of this study is to update the information of diversity and to support species concept delimitation of Ampelocissus based on the anatomical study, especially on the sterile plant. Anatomical characters were observed on nine species and one variety of Sumatran Ampelocissus. Ampelocissus species is varied by the anatomical characters, i.e.: shape of the anticlinal wall of abaxial and adaxial epidermal cell, number of palisade layers, upper epidermal cell thickness, leaf thickness, presence of papillae on stomata neighboring cell, type, and shape of calcium oxalate crystals, also stomata position. The cluster analysis of Ampelocissus in Sumatra based on 16 anatomical characters showed the coefficient similarity in the range of 0.48 - 0.81. The research showed that the leaf anatomical characters can be used as additional characters to distinguish the species of Ampelocissus. Keywords: calcium oxalate crystals, diversity, papillae, similarity coefficient, species concept   ABSTRAK Ciri Anatomi Ampelocissus (Vitaceae) di Sumatera Revisi Ampelocissus dilakukan dengan mengamati ciri anatomi daun dari marga Ampelocissus. Sebanyak 33 nomor koleksi Herbarium Bogoriense (BO) dan empat nomor koleksi hasil eksplorasi di Sumatera digunakan. Penelitian ini bertujuan untuk memperbarui informasi mengenai keanekaragaman dan mendukung konsep jenis Ampelocissus berdasarkan ciri anatomi, terutama pada tumbuhan steril. Studi anatomi dilakukan pada sembilan jenis dan satu varietas Ampelocissus di Sumatera. Jenis-jenis Ampelocissus di Sumatera bervariasi pada ciri anatomi yaitu bentuk dinding antilkinal sel epidermis pada bagian abaksial dan adaksial, jumlah lapisan jaringan tiang, ketebalan sel epidermis atas, ketebalan daun, kehadiran papila, jenis dan bentuk kristal kalsium oksalat, serta posisi stomata terhadap sel epidermis pada bagian abaksial daun. Analisis pengelompokan Ampelocissus di Sumatera menggunakan 16 ciri antomi dan menghasilkan fenogram dengan koefisien kemiripan 0,48 - 0,81. Ciri anatomi daun dapat digunakan sebagai ciri tambahan yang memiliki nilai taksonomi dalam membedakan jenis-jenis Ampelocissus di Sumatera.

Abstract

Peoples of Traditional Cikondang Village, located in the village of Lamajang, Pangalengan, Bandung City West Java have a very close relation to the environment along with its natural resources. This relation is expressed in the form of procedures and rules on the management and utilization of landscape units as well as the diversity of plants as a form of adaptation to the natural environment. This research was conducted through ethnographic method for photographing people’s lives and ethnobotany to know the biological resources that are important to Cikondang’s people’s daily lives. Descriptive and analytical methods are used through interviews with 87 respondents and 4 key informants. Results of research on landscape units that were identified by the community, indicates that in terms of the character, function, and its management there are 9 units of landscape that is both natural landscape and which has been modified by Cikondang community activities. Each unit of the landscape is characterized by vegetation cover according to the type of value in order for daily life. Meanwhile, cultural factors, economic, and political could be a trigger to the landscape transformation of one type of landscape into other landscapes.

Abstract

Selaginella, a genus of Pteridophyte, is known as an herb that contains of several bioactive compounds. The growth and bioactive compounds of plant were affected by plant environment, such as media types. The objective of the research was to obtain an appropriate plant media for growing S. plana dan S. willdenovii, and its effect on the content of bioactive compounds. The experiment consisted of two factors, plant growth media and the species of Selaginella. The first factor consisted of eight plant growth media, i.e.: soil, rice husk, rice husk : soil = 1:1 (v/v), burned rice husk, soil : burned rice huks = 1:1 (v/v), burned rice husk : rice husk = 1:1 (v/v), burned rice husk : rice husk = 3:1 (v/v), and burned rice husk : rice husk = 1:3 (v/v). The second factor was two spesies of Selaginella, i.e.: S. plana and S. willdenovii. Ethanol extract of Selaginella was qualitatively analyzed for flavonoid, tannin, and saponin content. The result showed that both fresh and dry weights of Selaginella were influenced by the type of plant media, Selaginella species, and their interactions. S. plana and S. willdenovii grew best on medium containing soil: burned rice = 1:1 (v/v). The highest flavonoid content of S. plana and S. willdenovii was also obtained from the plant grown on the same media. Comparing to the bioactive compound content of natural plant, saponin content of S. plana and S. willdenovii was increased by the treatment of plant media. The treatment also increased tannin content of S. plana, but not of S. willdenovii.

Abstract

A molecular analysis was conducted to determine whether Eugenia boerlagei Merr. (Myrtaceae) belongs to genus Eugenia or Syzygium based on sequences of cpDNA fragments namely atpB-rbcL intergenic spacer. The study used seven specimens of Syzygium sect. Jambosa, three of Syzygium sect. Syzygium, two of Eugenia s.s. and one of Eugenia boerlagei Merr. with Baeckea ovalifolia and B. tuberculata as the outgroup. The results show that Eugenia boerlagei is appropriately placed under the genus Syzygium.
