Abstract

The purpose of this study was to determine the potency and optimum pH of AAS thermophilic bacteria isolates in producing bioethanol. This research was carried out from September 2021 to December 2021, at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Padang State University. This research is an experimental study consisting of 6 treatments and 3 replications with variations in incubation pH of 6.5, 7, 7.5, 8, 8.5, and 9. The data obtained were analyzed by means of variance (ANOVA) with Duncan's further test. New Multiple Range Test (DNMRT). The results showed that the isolates of thermophilic bacteria SSA had the potential to produce bioethanol with isolates SSA 8 being the producer of bioethanol with the highest concentration of 1.001%, and variations in incubation pH affected the concentration of bioethanol produced with the optimum pH of thermophilic bacteria in producing bioethanol, namely pH 8 which produced the highest concentration of bioethanol is 1.001%.Tujuan dari penelitian ini adalah untuk mengetahui potensi serta pH optimum isolat bakteri termofilik SSA dalam menghasilkan bioetanol. Penelitian ini dilaksanakan pada bulan September 2021 hingga bulan Desember 2021, di Laboratorium Mikrobiologi  Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri Padang. Penelitian ini merupakan penelitian eksperimen yang terdiri dari 6 perlakuan dan 3 ulangan dengan variasi pH inkubasi 6,5, 7, 7,5, 8, 8,5, dan 9. Data yang diperoleh dianalisis dengan sidik ragam (ANOVA) dengan uji lanjut Duncan’s New Multiple Range Test (DNMRT). Hasil penelitian menunjukkan bahwa isolat bakteri termofilik SSA memiliki potensi dalam menghasilkan bioetanol dengan isolat SSA 8 merupakan penghasil bioetanol dengan kadar tertinggi yaitu 1.001%, dan  variasi pH inkubasi berpengaruh terhadap konsentrasi bioetanol yang dihasilkan dengan pH optimum bakteri termofilik dalam menghasilkan bioetanol yaitu pH 8 yang menghasilkan konsentrasi bioetanol tertinggi yaitu 1.001%.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi dan memetakan kebutuhan peserta didik terhadap booklet pada materi virus. Metode yang digunakan adalah metode deskriptif dengan pendekatan kuantitatif dan kualitatif. Sampel pada penelitian ini yaitu satu orang guru biologi yang mengajar di kelas X dan 27 peserta didik dari kelas X IPA1 yang dipilih berdasarkan rata-rata nilai ulangan yang terendah pada materi virus. Hasil penelitian ini yaitu guru belum pernah menggunakan booklet pada materi virus, peserta didik kesulitan dalam memahami materi virus, dan bahan ajar yang digunakan belum terintegrasi nilai-nilai islam. Kesimpulan dari penelitian ini adalah booklet terintegrasi nilai-nilai islam pada materi virus dibutuhkan sebagai suplemen atau tambahan bahan ajar yang sudah ada sebelumnya, untuk mendukung dan memaksimalkan proses pembelajaran.

Abstract

LKPD (Student Activity Sheet) in student-centered learning serves as a guide for learning activities. Based on the results of observations and interviews with teachers at SMA AL-Istiqamah it is known that the LKPD in this school has not been adapted to the learning model used, therefore it needs to be developed. The purpose of this study was to develop problem solving based worksheets on viruses for class X SMA/MA. This type of research is development research using the 4-D model. The subjects of this study were 39 students of class X MIPA 1 SMA AL-Istiqamah. The product that is produced is a problem solving based LKPD on virus material. The validators consisted of two lecturers from the Department of Biology FMIPA UNP and one Al-Istiqamah High School Biology teacher. The research data is in the form of validation data and practicality of LKPD. Data analysis is in the form of descriptive statistics. The results showed that problem solving-based worksheets on virus material fulfilled the valid category with a validity value of 89.26% and practical with 88.45% practicality. It could be concluded that problem solving-based Student Activity Sheets were produced on virus material for class X SMA. Valid and practical /MA.

Abstract

This study aims to determine the effect of the two stay two stray learning model based on a scientific approach to the competence of class XI students at SMAN 3 Pariaman. This type of research is a quasi-experimental research with a randomized control-group posttest only design. The research population was all class XI MIPA students at SMAN 3 Pariaman in the 2022/2023 academic year. Sampling used a purposive sampling technique, so that class XI MIPA 1 (experimental class) and class XI MIPA 3 (control class) were selected. Data analysis techniques using normality test, homogeneity test and hypothesis testing. Testing the hypothesis in the domain of knowledge uses the t test while testing the hypothesis in the domain of attitudes and skills using the Mann Whitney test. The results of the study found that the two stay two stray learning model based on a scientific approach had an effect on the competence in the realm of knowledge, attitudes and skills of class XI students at SMAN 3 Pariaman

Abstract

Mudiak Sapan hot spring, South Solok Regency, West Sumatra is one of the hot springs where there are various species of thermophilic bacteria that produce xylanase enzymes. The xylanase enzyme is a group of enzymes that has the ability to hydrolyze xylan (hemicellulose) or polymers into xylose and xylo-oligosaccharides. The purpose of this study was to test the compatibility of bacterial isolates to become a xylanase-producing bacterial consortium. This research is a descriptive research that informs the compatibility of thermophilic bacterial isolates from the Mudiak Sapan hot springs. The method used in this study was disk diffusion, based on the research, it was found that 5 combinations of compatible xylanase enzyme-producing isolates were MS18 x MSS11; MS18 x MSS15; MSS18 x MS16 ; MSS11 x MS16 ; MSS15 x MS16 and 1 combination of incompatible isolates namely MSS11 x MSS15 which had an inhibition zone of 0.645 cm.

Abstract

Endophytic bacteria have been extensively studied as a source of new antimicrobial compounds. Research into new antimicrobials is important as an alternative to the treatment of resistant microbes. One potential source of antimicrobial compounds is the Taxus (Taxus sumatrana). This study aimed to investigate the antimicrobial activity produced by the endophytic bacteria of the Taxus. Endophytic bacteria were isolated from the roots of Taxsus plants growing in Mt. Singalang, X Koto District, West Sumatra. Bacteria were isolated by inoculating pieces of Taxus root tissue into a PDA medium. Endophytic bacteria were identified macroscopically and microscopically. Antimicrobial activity was tested with the test microbe using the spot test diffusion method. As a result of the research, 23 isolates of endophytic Taxus bacteria were obtained, of which 21 isolates were gram-positive and 2 isolates were gram-negative bacteria. The results of the antimicrobial activity test yielded only 2 isolates of endophytic bacteria with antibacterial activity.

Abstract

Advances in the field of biotechnology have led to the use of enzymes in industry increasingly. The high public awareness of environmental pollution makes enzyme technology as an alternative to replace various chemical processes. Enzymes can be used in various industrial sectors such as textiles, food, detergents, paper and cosmetics, as well as biofuels. One of the enzymes that is often used is the enzyme xylanase. Thermophilic microorganisms that can produce thermostable enzymes are thermophilic bacteria. One of the important factors affecting the success of the manufacturing process of the production of the xylanase enzyme is the concentration of the starter. The production of the thermophilic bacterial xylanase enzyme SSA 4 produced the highest specific enzyme activity at a concentration of 4% which was 0.137 U/mg. This shows that low starter concentrations can overwhelm microbial growth well. The increase in the concentration of the starter causes the enzyme activity to decrease, this happens.

Abstract

Biofuel is an alternative energy to replace fossil fuels. The most popular form of biofuel today is bioethanol. Biofuels are considered as a suitable alternative to fossil fuels because they are more environmentally friendly. Production of bioethanol utilizing thermophilic microorganisms is more profitable because thermophilic microbes generally have the characteristics of being able to use a variety of substrates, low contamination, and resistance to high temperatures. The use of microbial consortia tends to give better results than the use of monoculture isolates to optimize bioethanol yields. The aim of this study was to determine the compatibility and ability of the best thermophilic bacterial triculture consortium from the Mudiak Sapan hot springs in producing biofuel. This research is a descriptive research, to test isolates of a consortium of thermophilic bacteria that produce bioethanol. The results of this study showed that consortium isolates tended to give better results than monoculture isolates. The best thermophilic bacterial triculture consortium from the Mudiak Sapan hot springs in producing biofuels namely MS 12, 17, 18 produced a bioethanol content of 0.863%.

Abstract

One of the factors for improving the quality of education in schools is teaching materials. Based on the results of observations at SMAN 1 Sitiung that students have not used e-modules but use electronic teaching materials in the form of pdf files downloaded from the internet and only in the form of writing and a few pictures. Based on the results of the questionnaire, it is known that students have difficulty understanding the concept of the respiratory system material. This causes low student learning outcomes. Based on the results of the creative thinking questionnaire, it was found that students' creative thinking ability is still low with an average score of 33.8, so it is included in the less creative category. So it takes teaching materials that can help in visualizing the subject matter. This type of research is Plomp model development research which consists of three stages, namely the initial investigation stage, the development or prototyping stage, and the assessment stage. The research subjects were class XI students at SMAN 1 Sitiung for the 2022-2023 academic year. Validation results by the validator on nuanced e-modules Science Technology Engineering And Mathematics (STEM) shows an average grade 88.05% (very valid). Practical results by Biology teachers showed an average score of 87.5% (very practical) and 88.28% students (very practical). The test data for the effectiveness of creative thinking was obtained from the average essay questions 81.22% (very creative).

Abstract

The xylanase enzyme has high commercial value in the industrial sector, including the food industry, animal feed, bleaching of pulp/pulp, lignocellulosic bioconversion as fuel, and in the food industry, namely the cheese, bread and meat industries, while in the non-food industry this can be used in detergents. Xylanase enzymes can be produced from microorganisms, one of which is thermophilic bacteria. Thermophilic bacteria are known to be able to produce thermostable enzymes and proteins that are heat resistant. Xylanase production in compatible bacterial consortia was higher than monoculture. The purpose of this study was to determine the bicultur consortium that has the potential to produce xylanase enzymes. This research is an experimental research. Enzyme activity testing used the Miller method with DNS (Dinitrosalycilic acid) reagent using a completely randomized design (CRD) with 3 repetitions. Data on the results of xylanase activity were analyzed by ANOVA test and DMRT follow-up test with a 5% significance level.The results of the thermophilic bacterial isolates in the consortium that have the potential to produce xylanase enzymes are MS18 & MSS15,  MSS15, MS16 and MS18 & MS16.
