Abstract

 AbstrakSARS-CoV-2 merupakan virus yang menyebabkan Coronavirus Disease 2019 (COVID-19) di seluruh dunia dan sampai saat ini kasus terbaru masih terus dilaporkan. Diagnostic test merupakan hal yang krusial untuk dikembangkan. Prinsip diagnostic test COVID-19 berbasis antigen, yaitu mendeteksi virus SARS-CoV-2 melalui respon antibodi dari penderita. Penelitian ini bertujuan untuk memproduksi antibodi poliklonal menggunakan protein rekombinan RBD-Spike untuk mendeteksi virus SARS-CoV-2 berbasis antibodi. Penelitian dimulai dengan penentuan domain RBD-Spike menggunakan pensejajaran asam amino, dan konstruksi DNA untuk RBD-Spike pada vektor ekspresi pET28a menggunakan sintetik nukleotida. Produksi protein rekombinan RBD-Spike diekspresikan pada sel bakteri Escherichia coli. Purifikasi dilakukan untuk memperoleh protein RBD-Spike dan selanjutnya digunakan sebagai antigen untuk induksi antibodi poliklonal pada kelinci. Hasil penelitian menunjukkan bahwa ekspresi protein rekombinan RBD-Spike SARS-CoV-2 memerlukan induksi IPTG 0,1 mM dan terekspresi dalam bentuk inclusion bodies dengan ukuran 39 kDa. Purifikasi protein RBD-Spike dilakukan menggunaan resin afinitas NiNTA, elektroelusi, dan dialisis. Total protein RBD-Spike yang diperoleh sebanyak 4 mL dengan konsentrasi 10 mg/mL. Analisa Ouchterlony menunjukkan bahwa antibodi poliklonal terdeteksi pada minggu kedua setelah injeksi booster dan analisa spesifitas antibodi terhadap antigen menunjukkan bahwa antibodi poliklonal dapat mendeteksi protein RBD-Spike pada konsentrasi 0,1 µg. Selanjutnya diharapkan antibodi poliklonal dapat digunakan untuk deteksi keberadaan virus SARS-CoV-2 dan dapat dikembangkan untuk kit deteksi berbasis antibodi.AbstractSARS-CoV-2 is the virus that causes Coronavirus Disease 2019 (COVID-19) worldwide and the latest cases are still being reported until now. The diagnostic test is a crucial to be developed. The principle of the antigen-based COVID-19 diagnostic test is to detect the SARS-CoV-2 virus through antibody response from the patients. This study was conducted to produce polyclonal antibodies using recombinant protein RBD-Spike. The research was carried out by determining the RBD-Spike domain using amino acid alignment and constructing the DNA of RBD-Spike to the expression vector of pET28a using nucleotide synthesis. Production of RBD-Spike recombinant protein was expressed in Escherichia coli. Purification was carried out to obtain RBD-Spike protein and used to induce polyclonal antibody ina rabbit. The results showed that the expression of RBD-Spike recombinant protein required induction of IPTG 0.1 mM and was expressed in inclusion bodies with molecular size of 39 kDa. The purification of RBD-Spike protein was carried out using resin affinity, electroelution, and dialysis. The total protein of RBD-Spike obtained was 4 mL with a concentration of 10 mg/mL. Ouchterlony analysis revealed that polyclonal antibody was detected in the second week after booster injection and analysis of antibody specificity showed that polyclonal antibodies detected RBD-Spike protein at the concentration of 0.1µg of RBD-Spike protein. Moreover, it is expected that our polyclonal antibody detect the SARS-CoV-2 virus and can be developed for antibody-based detection kits.

Abstract

Critical thinking skills are needed in the 21st century, so efforts are being made to improve them, one of which is by developing research-based teaching materials, and research into the use of Extremely Low Frequency Electromagnetic waves to increase food security is currently being carried out very intensively. This research aims to describe the development of research-based teaching materials to improve critical thinking skills in the use of Extremely Low Frequency Electromagnetic waves to increase food security. This development research uses the Borg and Gall method, which consists of 10 steps. The subjects of this research were 90 students at the University of Jember. Undergraduate Physics Education students in classes A, B, and C in environmental physics courses and on material on the use of Extremely Low Frequency electromagnetic waves to increase food security. This research uses quantitative analysis. The results of this research state that research-based teaching materials are effective in improving students' critical thinking skills in the material on the use of Extremely Low Frequency Electromagnetic waves to increase food security with a score of 82.33 based on critical thinking indicators.

Abstract

Perkembangan ilmu pengetahuan dan teknologi semakin pesat sehingga bioteknologi sebagai bidang ilmu pengetahuan harus dikuasai oleh bangsa Indonesia. Namun perkembangan bioteknologi saat ini yang semakin pesat membuat beberapa penerapan melanggar etika dan menimbulkan banyak perdebatan di berbagai bidang sehingga perlu adanya norma yang mengatur penerapannya. Upaya peningkatan mutu pembelajaran bioteknologi dapat ditempuh dengan menggunakan bahan ajar yang bermutu, salah satunya adalah buku siswa. Penelitian ini merupakan penelitian pengembangan dan menggunakan model ADDIE yang terdiri dari lima tahap (analisis, desain, pengembangan, implementasi, dan evaluasi). Nilai validitas buku siswa yang dikembangkan sebesar 83,43% dengan kategori sangat valid. Nilai rata-rata tes keterbacaan dan tingkat kesukaran buku siswa sebesar 91,35% dengan kategori baik. Nilai praktikum buku siswa 80,67% dengan kategori praktis. Buku siswa efektif dalam mempengaruhi siswa untuk mencapai kategori baik dalam ranah kognitif, psikomotor, dan afektif. Hasil ini menunjukkan bahwa buku siswa yang dikembangkan dapat digunakan dalam pembelajaran bioteknologi di kelas XII SMA dan MA di Jember. Abstract

This study aimed to determine the potential antagonism performed by the endophytic fungi of lemongrass against Escherichia coli. Five species of endophytic fungi were tested using agar well method. These fungi consist of Paecilomyces lilacinus, Penicillium sp., Trichoderma viride, Fusarium oxysporum, and Fusarium sp., each was taken from roots, stem, and leaves, respectively. Chloramphenicol and aquadest were used as control. The results showed that there is significant inhibition of E. coli compared to the negative control. Paecilomyces lilacinus showing the most prominent inhibition with 74.67%, followed by unknown species of Fusarium 47.33%, Penicillium sp 42.67%, Fusarium oxysporum 21.67%, and Trichoderma viride 17.43%. It can be concluded that all five isolates of endophytic fungi from lemongrass (Cymbophogon nardus (L.) Rendle) have the ability to inhibit the growth of E. coli, and thus have antagonism potential against E. coli.

Abstract

The Apocynaceae family, including Tapak Dara (Catharanthus roseus), is one of the most popular medicinal plants due to its secondary metabolites. Secondary metabolites are naturally produced in small amounts, but have very high economic value. Therefore, exploration of plant species within the same family is an interesting issue to be conducted. The purpose of this study is to discover members of the Apocynaceae family that have close relatives with Catharanthus roseus so that expected to have the similar active compounds. The method used was phylogenetic analysis using DNA sequences of maturase K (matK) and riboluse 1,5-biphosphate carboxylase/oxygenase large subunit (rbcL) regions. A total of 22 plants of the Apocynaceae family were used, and the sequences of both genes from these plants were retrieved in the NCBI database. Phylogenetic tree was constructed using MEGA 11 and two Rubiaceae species as outgroups. The study results showed that 10 plant groups were formed based on the suprageneric in both trees generated. It is consistently obtained that Rauvolfia serpentina is in one group with Catharanthus roseus in the Vinceae tribes of Rauvolfioideae subfamily. Morphologically, this group of plants has similar flower and leaf characters. It can be conclude that phylogenetic analysis with DNA sequences supports the grouping of morphological data, and indicates that R. serpentine has a biochemical profile similar to Tapak Dara.

Abstract

This study explores the effectiveness and challenges of implementing the Contextual Teaching and Learning (CTL) model in multicultural classrooms, particularly focusing on its impact on interpersonal skills development. The primary aim is to investigate how the CTL model enhances student engagement by connecting academic content to real-world contexts and to identify the barriers that impede its effective application. This research utilized a qualitative approach with a case study method to examine the application of the CTL (Contextual Teaching and Learning) model in building inclusive learning communities and fostering interpersonal skills in multicultural classrooms. The data collection techniques used in this study included interviews, observations, and document analysis. The findings indicate that while the CTL model significantly enhances student engagement and fosters the development of interpersonal skills, its implementation is fraught with challenges. Key obstacles include insufficient teacher training, misalignment with existing curriculum standards, limited resources, and institutional barriers such as rigid schedules and lack of administrative support. Despite these challenges, the research highlights the importance of sustained professional development and a supportive learning community to overcome these barriers. By addressing these issues, educators can better leverage the CTL model to create inclusive and effective learning environments. Overall, this study underscores the potential of the CTL model to transform teaching practices and improve student outcomes in multicultural settings. It calls for a concerted effort to provide the necessary support and resources for educators to implement the CTL model successfully, thereby enhancing educational experiences and fostering inclusive learning communities.

Abstract

This research aims to develop teaching materials based on local wisdom to improve the interpersonal skills of prospective elementary school teachers. The background of this research is based on the low interpersonal skills of prospective students of Elementary School Teachers at the Faculty of Teacher Training (FKIP) Cenderawasih University (Uncen) Papua. The strategy used to improve students' interpersonal skills is to integrate Khombouw into the teaching materials of the Basic Concepts of Science Elementary School. Khombouw is a local wisdom made of bark from red khombow trees (Ficus nodosa teijsm & Binn) and white khombow (Ficus variegata Blume), which grow in Sentani, Papua. This research is a research development (R&D) of the ADDIE (Analyze, Design, Development, Implementation) design model. The results of student self-assessment on the implementation of teaching materials based on Khombouw Local Wisdom Model Guided Inquiry on interpersonal skills were 69.5% in the "good" criterion, 28.2% in the "very good" criterion, and 5.1% in the "quite good" criterion. Lecturers' observations of interpersonal skills showed results that were not much different, namely 74.3% on the good criteria, 25.6% on the very good criteria, and 7.6% on the good criteria. The value of N < g> obtained was 0.27 and was included in the low criteria. A thorough evaluation and in-depth analysis of the factors that affect the low N-Gain value is required.

Abstract

Decision-making skills are an activity to make the best decision from several alternative solutions offered with certain considerations. This study aims to examine the effect of Socio-Scientific Issue (SSI)-based science learning on the decision-making skilss of class VIII Junior High School students. The research method used is a quasy experiment with a non-equivalent control group design. The population in this study was class VIII SMP Negeri 7 Jember in the academic year 2021/2022. The selection of research samples using purposive sampling technique obtained class VIII B as the control class and class VIII E as the experimental class. The results of the analysis used the Independent Sample T-test with a significance value og 0.041 < 0.05. In addition, based on the N-Gain test in the control class the average N-Gain is 0.4233 with moderate criteria in the less effective category, while the average N-Gain in the experimental class is 0.5568 with moderate criteria being quite effective. Based on the results of the study, it can be concluded that there is a significant effect on Socio-Scientific Issue (SSI)-based science learning on the decision-making skills of class VIII Junior High School students. Analysis with the N-Gain test shows that SSI-based science learning is included in the category of being quite effective in learning with a precentation of 56 percent. Abstract

Pathogenic Escherichia coli infect fruit through irrigation contaminated water. Bacteriophage ϕPT1b has ability to infect E. coli and act as food security. Bacteriophage formulations as emulsion preparations increase the efficiency of application and the stability of long-term storage, one of it is W/O/W emulsions. The objective of this study was to examine the effect of cremophor RH 40, span 80 and their interaction on viscosity, droplet size and inhibition of bacteriophage ϕPT1b W/O/W emulsions against pathogenic E. coli. Factorial design method was used to get the optimum formula. The results showed that the cremophor RH 40, span 80, and their interactions increased the viscosity response, decreased the emulsion droplet size, and increased the inhibition against E. coli. FB is the optimum formula for ϕPT1b W/O/W emulsions which has 1% cremophor RH 40 and 4.5% span 80. The formula produced a viscosity of 29 dPas, an average emulsion droplet diameter size of 10.483, and an inhibition against E. coli of 8.65 m. The desirability index value of 0.905 indicates that FB meets 90.5% of the specified criteria.
