Abstract

Lais (Krypyoterus apogon) is highly economic important catfish in South Sumatera. Investigating fish diversity of catfish is of importance for species conservation. Many species or subspecies extincted due to environment decreasing caused by habitat loss, pollution and over fishing. Cytochrome C Oxidase subunit I (COI) is one of mitochondrial DNA markers used for species barcoding in freshwater, brackishwater and marine fish. This research aim to explore the use of COI gene for species barcoding in investigating the diversity of COI nucleotide, constructing phylogenetic tree of lais, and knowing physical and chemistry characteristic of habitat at Musi and Kelekar River, South Sumatera. The methods used in the research consisted of DNA  extraction, PCR (Polymerase Chain Reaction) amplification and sequencing mtDNA COI gene of Kryptopterus apogon obtained from Musi and Kelekar River (South Sumatera). A 615 bp (sample code CM1) and 693 bp sequences (sample code CK1 and CK2) of parsial coding were sequenced from this fish. Nucleotide BLAST analyzes showed COI gen can distinguish lais at Musi and Kelekar River from others fish at GenBank database with high identity (92.29% and 92.21%) to Phalacronotus bleekeri (Thailand) and Phalacronotus apogon (Australia).Phylogenetic analyzes indicated that obviously, lais was at the same cluster with others genus in Siluridae family, but different cluster from Hemibagrus nemurus (Bagridae), Chanidae and Oreochomis niloticus (Cichlidae). Water quality criteria (water tranparancy, temperature, ammonia, pH and alkalinity) at Musi and Kelekar River were still in tolerance for maintainning survival rate of lais. Further study using more species in Siluridae family and various habitat are needed to investigate the diversity of DNA of Siluridae at South Sumatera water resources.Keywords: Catfish, DNA barcode, COI gene, Musi River, Kelekar River

Abstract

ABSTRACT The success in egg hatching is determined by internal and external factors. One of external factors is the acid level. The use of pH in egg hatching is to stimulate chorionase enzyme that can make chorion become soft. The purpose of this research is to determine the best pH value for hatching of snakehead eggs. This research had been conducted in Laboratorium Dasar Perikanan, Departement of Aquaculture, Agriculture Faculty, Sriwijaya University on January until February 2016. The research method used a completely randomized design with five treatments and three replications. The treatment were P1 (pH 5±0.2), P2 (pH 6±0.2), P3 (pH 7±0.2), P4 (pH 8±0.2) dan P5 (pH 9±0.2). The results showed that different values of water pH in snakehead hatching gave significant effect on hatching percentage, incubation time and survival rate of larvae but did not indicate  significant effect on percentage of abnormal larvae. The highest hatching percentage was in treatment P5 (90.75%), the fastest incubation time was in treatment P4 (20.00 hours), the highest survival rate of larvae was in treatment P2 (85.31%) and the highest percentage of abnormal larvae was in treatment P1 (1.67%). During the research, water quality were in tolerance range for hatching and snakehead larvae rearing where DO (5.27-6.01 mg/l), ammonia (0.00-0.29 mg/l), and alkalinity (26-106 mg/l).   Key words : Snakehead, Hatching, pH different

Abstract

ABSTRACT In fish cultivation, Aeromonas hydrophila could couse desease epidemic with mortality rate of 80-100% for 1-2 weeks. The purpose of research was to know the effect of addition of guava leaf in feed to prevalence, survival rate and growth of pangasidae which infected by A. hydrophila. The research was conducted on August- September 2015 in Laboratorium Budidaya Perairan and UPT. Klinik Kesehatan, Sriwijaya University. The research used Completed Randomized Design (CRD) with  six treatments and three replications, there were fish fed commercial feed and infected by A. hydrophila, fish fed by commercial feed with addition of guava leaf powder 1.5%, 2%, 2.5%, 3%, 3.5% and infected. The result of the research showed the addition of guava leaf to feed has a significant effect to prevalence and survival rate of pangasidae, but has n’t have significant effect to growth of pangasidae. The treatment on P1 has the best result with prevalance 42.22%, survival rate 75%, absolute weight 6.34 g and absolute length 2.41 cm. Keywords : Aeromonas hydrophila, Bacteria infected, Guava leaf, Pangasidae

Abstract

ABSTRACT The purpose of this study was to determine the effect of different incubation temperatures to the rate of the kissing gouramy (Helostoma temminckiiC.V) yolk absorption, and to know the right time to start feeding in the larval stage. This study was held on January 09th to 13th, 2013 using Completely Randomized Design (CRD) with five treatments namely P1 (26±0.50C), P2 (28±0.50C), P3 (30±0.50C), P4 (32±0.50C) and P5 (34±0.50C).Each treatment was repeated three times. The results showed that the temperature gave significant effect (P>0,05) to rate of yolk absorption, periode of yolk absorption and percentage of larval survival.The highest rate of yolk absorption was P5 (42.24 x 10-5 mm3/hour), but not significantly different fromP4 (41.63 x 10-5 mm3hour-1), and P3 (41.83 x 10-5 mm3hour-1). The fastest periode of yolk absorption was P5 (72 hours) and the longest was P1 (78.67 hours). While the highest larval survival rate was found on P3 (89.87%), on the other hand the lowest survival rate was found on P5 (62.35%). The natural feeding of kissing gouramy fish should be started at 74th hours (3 days, 2 hours) after hatching and larval rearing should use temperature of 300 ± 0.50 C.   Keywords: Helostoma temminckii C.V., temperature, yolk absorption

Abstract

ABSTRACT The problem of development of climbing perch culture was low in growth rate. The effort to increase growth rate of climbing perch is hormonal treatment, including the hormone thyroxine. The aim of this research was to determine the effective consentration and duration of immertion of thyroxine hormone to increase yolk sack absorbtion and growth rate. This research held in August to September 2014 in Laboratorium Dasar Perikanan, Aquculture Study Departement, Faculty of Agriculture, Sriwijaya University, Indralaya. This research using factorial completely rondomezed design with two factors, the first was thyroxine consentration with immersion media (K1=0.01 mg.L-1, K2=0.05 mg.L-1, K3=0.1 mg.L-1) and the second was the long immersion (W1=24 hours, W2=48 hours, W3=72 hours) with three replication. The parameters that observed were yolk sack absorption, growth, survival rate and water quality. The result showed that the highest yolk sack absorbtion was K2 (0.05 mg.L-1) and K3 (0.1 mg.L-1). The highest growth was K3W2 (0.1 mg.L-1 of consentration and immersion for 48 hours) with the weight was 316.1 mg and length was 26.56 mm. The best survival rate was 88.67% (K1W1). The value of water quality during larva rearing were temperature 26-28 oC, pH 5.6-6.5, dissolved oxygen 4.43-5.93 mg.L-1 and ammonia 0.00-0.03 mg.L-1   Keywords : Climbing perch larvae, thyroxine hormone, duration of immertion, growth

Abstract

ABSTRACT The use of extract root ginseng in the process of Betta fish masculinization is expected to increase the testosterone. The purpose of this research is to determine the effect ginseng root extract to the percentage of the male betta fish by masculinization. This study has been conducted at the Basic Aquaculture Laboratory, Faculty of Agriculture, Sriwijaya University. This study use a completely randomized design with 4 treatments and 3 replications. The treatments in this research was betta fish larvae aged 7 days that submersed on ginseng root extract media with different concentration for 8 hours. The treatment were the addition of ginseng root extract as much as 1 mg/L (P1), 2 mg/L (P2), 3 mg/L (P3), 4 mg/L (P4) and without ginseng root extract addition as control (P0). Parameters observed consist of percentage of betta fish male, survival rate and quality of water. The result of this research shows that the addition of extract panax ginseng with difference concentration has significant effect to male betta fish percentage, but there is no a significant effect on survival rate. Treatment P3 with concentration 3 mg/L is the best result, it produced 95.05% of male betta fish after immersion for 8 hours. Keywords : Betta fish, Ginseng root extract, Immersion, Masculinization

Abstract

ABSTRACT The aims of this research was to study Gonad Maturity Level (GML), Index Gonado Somatik (IGS), absolute fekundity and egg diameter of Channa striata injected with the Human Chorionic Gonadotropin hormone. The research was conducted in 26th of December 2013 until 26th of January 2014 at the Fish Hatchery Unit (FHU) Batanghari Sembilan, North Indralaya District, Ogan Ilir, South Sumatra. Channa striata which as used had weights 85-105 gram per fish. The experiment was arranged in a Completely Randomized Design (CRD), (P0) No injection of HCG, (P1) 200 IU per kg fish, (P2) 250 IU per kg fish, (P3) 300 IU per kg fish. Parameters observed Gonad Maturity Level (GML), Index Gonado Somatik (IGS), absolute fekundity and egg diameter. From the four treatment experiment, P3 treatment produced the highest GML V whose the percentage was 20%, while P0 treatment did not reach GML IV and V, most of the P0 treatment (80%) only reached GML II. Based on further test of Duncan Signipicant Differences with 95% credibility level, the IGS data of Channa striata was significantly different and influential among treatments, the smallest IGS was produced from P0 treatment that is 0.43% while the largest IGS was produced from P3 treatment that is 5.91%. The smallest absolute fecundity was on the P0 treatment which was 582 eggs while the largest was on P3 treatment that is 5,775 eggs. The smallest egg diameter was on the P0 treatment which was 0.35 mm while the largest was on P3 treatment that is 0.93 mm. Hormone HCG injections of 300 IU per kg weight of the fish proved to be finalizing a Channa striata gonad.   Keywords : Channa striata, HCG hormone, GML, IGS, egg diameter

Abstract

ABSTRACT The aims of this research was to know the effect of fed with addition of probiotic to bacteria population, and water quality of media maintenance, histology of snakehead (Channa striata) fry. This research used randomized completely design (RCD) with six treatments and three repeat with treatments code (D). treatment tested were D0 (control without probiotic), D1 (2.5 ml.kg-1 feed), D2 (5 ml.kg-1 feed), D3 (7.5 ml.kg-1 feed), D4 (10 ml.kg-1 feed), D5 (12.5 ml.kg-1 feed). The research was conducted November to December, 2012 at the Laboratorium Dasar perikanan, Agriculture Faculty Sriwijaya University, Indralaya. Population bacteria, water quality and histology were analyzed descriptively. The result research was conducted that addition probiotic with a cocentration of 10 ml.kg-1 feed in snakehead medium give good influence to survival rate (63.33 %). Histology of intestine were enteritis, loss of epithelial tunica serosa, and histology of lever were congestion, necrosis, hemorrhagic.   Key word : Snakehead, probiotic, population bacteria

Abstract

The aims of this study was to know the effect of different temperatures on the percentage of climbing perch eggs hatching. This study has been done from October 2012 to November 2012 at Fish Breeding Unit “Batanghari Sembilan” Indralaya. The implementation of this study include selection of the brood, rearing of brood stock, injection ovaprim, spawning, hatching eggs, larval rearing. The results  indicate that the most rapid hatching time obtained in P4 treatment (34° C) with a incubation time 964 minutes. Optimal incubation temperature to produce the maximum hatching percentage was 98.66% at a temperature of 34 oC. Water quality of medium were 31-340C for temperature, pH  6,7-7,6 and disolved oxygen 3,3-3,8 mg.L-1. Keywords : temperature, hatching percentage hatching time, water quality.

Abstract

The thyroxine has been known as a hormone that can increase fish growth especially in the larvae stage. The provision of hormone thyroxine on larvae fish can be conducted through dipping. Determination of dipping time are very important to increase growth. The purposes of this research are determining the optimum dipping time for the growth and survival rate of snakehead larvae. This research was done at Laboratorium Dasar Perikanan, Aquaculture, Faculty of Agriculture, University of Sriwijaya, Indralaya. The method used Competely Randomized Design (RAL) with five treatments and three replicates, with dipping time are 24, 27, 30, 33 and 36 hours. The parameters observed are the absolute length growth, absolute weight growth, survival rate of larvae and water quality (temperature, pH and disolved oxygen). The results showed that the duration of immersion in thyroxine hormone solution in snakehead larvae showed relatively faster growth on treatment (P5) with 36 hours of dipping time. While the regression test result showed the optimal growth of snakehead larvae at the time immersion for 27,92 – 29,49 hours. Thyroxine hormone increased the growth of snakehead larvae but has no statistically significant effect. The length of time of thyroxine hormone 24-36 hours with a dosage of 0.1 mg l-1 did not affect the survival of snakehead larvae. Water quality of maintenance media is still tolerance for supporting the growth and survival of snakehead larvae.
