Abstract

Tempeh is an Indonesian traditional fermented food produced using Rhizopus as a starter culture. In practice, however, the starter culture as well as fermentation processes would yield a polymicrobial fermentation, which generated a unique tempeh flavor and texture. This condition makes Indonesian tempeh as one of the most complex fermented food, while at the same time would make it difficult to scale up tempeh production with uniform quality and consistency. The aim of this study was to compare a number of tempeh microbial communities employing Amplified Ribosomal Intergenic Sequence Analysis (ARISA). Fresh tempeh samples were obtained from tempeh producers in Java and Moluccas. 16S rRNA gene libraries and DNA sequencing were employed to analyze further the nature of bacterial diversity in two selected tempeh samples. The results of our study showed that different tempeh producer possessed different Bacterial ARISA (BARISA) or fungi ARISA (FARISA) profiles.  However, BARISA profiles were found to be more discriminative than FARISA, and therefore BARISA would be more useful for tempeh genetic fingerprint or barcoding.

Abstract

Vanillin is one of the most important flavoring agents worldwide. Currently, consumers' awareness and concern for biovanillin production has been increasing. This study aimed to screen the potential of clove rhizospheric bacteria isolates producing vanillin through a biotransformation process of eugenol and to conduct the preliminary optimization of the biotransformation condition. Twenty-eight bacteria isolates were screened for their capability to transform eugenol into vanillin. BKL 15 isolate, which was identified as Lysinibacillus xylanilyticus, was selected as the highest vanillin producer among the isolates. The optimum molar yield of vanillin produced by the selected isolate was 4.99% (1.11 g/L) after 168 hours of biotransformation process in the medium consisting of TSB (30 g/L), eugenol (24 g/L), yeast extract (20 g/L), and concentration of casamino acid (20 g/L). Throughout the publications we have read, this is the first report of L. xylanolyticus that produces vanillin.

Abstract

Denitrification and nitrate-ammonification are the responsible processes for nitrate removal in the estuaries. Temperature, nitrate and organic carbon availability are key factors controlling a rate of the nitrate reduction processes. This mixed cultures chemostat study investigates the competition ability and their nitrate reduction end-products of the bacteria isolated from an estuary at different temperatures. This study will help us to understand the seasonal nitrate reduction processes in an estuary. The experiments showed that a nitrate-ammonifier was the predominant process in the steady-state chemostat at high temperature. While a facultative denitrifier-nitrate ammonifier was the predominant process at low temperature. However, the main end products of nitrate reduction at high temperature were up to 61% N2 indicating a denitrifier still had an important role in the end products of nitrate reduction in the estuary. The data also showed that a nitrite respiring bacterium reduced nitrite to N2, that responsible for approximately 6-9% of total N2 produced in the culture. This study confirmed that nitrate ammonifiers out-compete denitrifiers at high temperature, however, denitrifiers still had an important role in end products of nitrate reduction.

Abstract

Some of Gram-negative bacteria perform a phenomenon called quorum sensing (QS) to activate certain phenotypes such as pathogenicity. The bacterial cells performing QS produce N-acyl homoserine lactone (AHL) as signal molecules to communicate within a population. These molecules can be degraded by the enzyme, i.e. AHL lactonase. This study aimed to characterize the activity of AHL lactonase from Bacillus cereus INT1c and Bacillus sp. NTT3a in different pH and temperature levels. Both strains produce AHL-lactonase that could be found in intracellular and extracellular extracts. The dialysis process of extracellular AHL-lactonase of INT1c significantly increased the specific activity from 5.91 to 29.96, different from an extracellular enzyme of NTT3a that slightly increased from 4.08 to 5.39. Generally dialyzedAHL-lactonase of both B. cereus INT1c and Bacillus sp. NTT3a had activity in wide pH range with better activity in acidic pH and were not stable in high temperature with the highest activity at 30-40 oC.

Abstract

This study aimed to analyze the diversity of indigenous bacteria by comparing culture and non-culture methods and to analyze the physicochemical effects on bacterial diversity in polluted and natural mangrove sediments. The environmental parameter values of mangrove sediments for bacterial growth can change owing to differences in adaptation and tolerance to fluctuations in physicochemical conditions. The number of colonies in natural and polluted areas using the culture method was 6.2 × 104 CFU/g and 5.5 × 104 CFU/g, respectively. A total of 33 isolates were identified, with 17 and 16 isolates from the natural and polluted areas, respectively. The most common isolates found in both areas were Acinetobacter haemolyticus strain FBC636 and Exiguobacterium acetylicum strain IAE17. Using the nanopore sequencing method, the total number of colonies in the natural and polluted areas was 69,761 and 58,412 colonies, respectively. A total of 12,954 bacterial species were identified, with 6,837 species in the natural area and 6,117 in the polluted area. The most common isolate found was Sulfurovum aggregans. Physicochemical conditions influenced the differences in bacterial diversity between the natural and polluted areas in the mangrove areas of Ambon Bay.

Abstract

Tailing from mining activities affects soil fertility resulting in poor soil conditions that are challenging for plants to grow. Plants can interact with rhizosphere bacteria to enhance their growth in harsh environments. Rhizospheric bacteria possess numerous mechanisms that promote plant growth and induced resistance to various abiotic stress. This study aims to determine the diversity of rhizobacteria and their potential as plant growth-promoting rhizobacteria (PGPR) agents. Bacterial communities from rhizosphere soil samples from kaolin mining sites in Perawas, Tanjung Pandan district, Belitung Regency, Bangka Belitung Island, Indonesia were analyzed using Next Generation Sequencing based on the V3-V4 region of the 16S rRNA gene, while culturable bacteria were isolated from samples and screened for PGP activity. The results showed that the rhizosphere bacterial community was mostly dominated by Pseudomonadota, Acidobacteria, and Verrumicrobiota. There were 15 bacteria isolated from the sample and RKB-5 bacterial isolate had the potential to be PGP agent. The RKB-5 bacterial isolate was identified as Burkholderia cenocepacia based on its 16S rRNA sequence. The bacterial isolate produced IAA, utilized ACC, dissolved phosphate up to 209,5 mg/L, and formed a high potassium solubilizer index value of 5.00. Therefore, the B. cenocepacia RKB-5 has potential application as the PGPR to support plants growth by obtaining nutrients in ex-mining lands with poor soil conditions.

Abstract

Pond traditional system is the pond in still activity with a symple management system.  This activity indicated by low technology and relatively low production level.  Aquaculture activities in traditional pond not loss from nitrification and denitrification prosess, however this process is more low production rather than semiintensive and intensive system. This study aims to observe abundance of bacteria nitrification along with changes soil texture, and N-organic in the soil of traditional pond. Chemical and biological analyses were done using spectroscopy and Most Probable Number methods to determine the amount of nitrite and ammonium production of bacteria.  Based of the result, each stratum traditional ponds have relatively similar abundance in nitrite producing bacteria of 7.08-7.47 Log CFU/g.  Increasing abundance in ammonium producing bacteria was found in all stratum, range from 5.63 Log cfu/g to 8.12 Log cfu/g. From the first day of preparation, traditional ponds have a lot of nitrite and ammonium producing bacteria.Keywords: traditional, pond, nitrification, abundance of bacteri. ABSTRAKTambak sistem tradisional merupakan tambak yang dalam kegiatannya masih menggunakan sistem manajemen sederhana.  Hal ini ditandai dengan penerapan teknologi sederhana, dan tingkat produksi relatif rendah.  Kegiatan budidaya di tambak tradisional tidak akan terlepas dari proses nitrifikasi dan denitrifikasi, namun demikian proses ini relatif lebih rendah aktivitasnya daripada tambak sistem semiintensif dan intensif.  Tujuan dari penelitian ini adalah mempelajari kelimpahan bakteri penghasil senyawa nitrit, amonium seiring dengan perubahan tekstur tanah, dan N-organik pada tanah tambak tradisional. Media pertumbuhan bakteri dikondisikan bebas oksigen (oxygen free nitrogen/OFN method) , sedangkan kelimpahan bakteri dianalisis dengan rumus most porbable number (MPN). Berdasarkan hasil, setiap strata tanah tambak tradisional memiliki jumlah bakteri penghasil nitrit yang relatif sama, yaitu antara 7,08-7,47 Log cfu/g. Peningkatan kelimpahan bakteri penghasil amonium terjadi pada semua strata, yaitu berkisar antara 5,63 Log cfu/g sampai dengan 8,12 Log cfu/g.  Dari hari pertama persiapan, tambak tradisional telah memiliki kelimpahan bakteri penghasil nitrit dan amonium yang berlimpah.Kata kunci:  tradisional, tambak, nitritifikasi, kelimpahan bakteri.

Abstract

ABSTRACTOrganic material in the bottom of the pond is part of the land is a complex and dynamic system, which is sourced from the rest of the feed, plants, and or animals found in the soil that continuously change shape, because it is influenced by biology, physics, and chemistry. This study was aimed to see the profile of organic material consisting of C, N, and C/N ratio and phosphate in different depths of pond with different culture systems. Observation were conducted at Tambak Inti Rakyat, Karawang in traditional, semi-intensive and intensive culture systems. Observation at mangrove area was also observed as control. Sediment samples at the inlet and outlet at three different depths (0‒5 cm, 5‒10 cm, and 10‒15 cm) was taken every 30 days to measure the content of C, N, C/N ratio, and total phosphate. During the 120 day maintenance period could be known that in all pond systems were used (traditional, semi-intensive, and intensive) the concentration of C-organic and organic-N on average was located in the bottom layer which is a layer of 10‒15 cm. The lack of human intervention from ground pond system, the more diverse the type and amount of organic material contained therein.Keywords: organic materials, subgrade, depth, aquaculture systems, long maintenanceABSTRAKBahan organik di dasar tambak merupakan bagian dari tanah yang merupakan suatu sistem kompleks dan dinamis, yang bersumber dari sisa pakan, tanaman, dan atau binatang yang terdapat di dalam tanah yang terus menerus mengalami perubahan bentuk, karena dipengaruhi oleh faktor biologi, fisika, dan kimia. Penelitian ini bertujuan untuk melihat profil bahan organik yang terdiri dari C, N, dan C/N rasio serta fosfat pada kedalaman tambak yang berbeda dengan sistem budidaya yang berbeda pula. Pengamatan dilakukan di Tambak Inti Rakyat Karawang pada sistem budidaya tradisional, semi intensif, dan intensif. Pengamatan di daerah mangrove diamati pula sebagai kontrol. Sampel sedimen di inlet dan outlet pada tiga kedalaman yang berbeda (0‒5 cm, 5‒10 cm, dan 10‒15 cm) diambil setiap 30 hari sekali untuk diukur kandungan C, N, C/N rasio, dan total fosfatnya. Selama 120 hari masa pemeliharaan dapat diketahui bahwa pada semua sistem tambak yang digunakan (tradisional, semi intensif, dan intensif) nilai konsentrasi C-organik dan N-organik rata-rata terletak pada lapisan paling bawah yaitu lapisan 10‒15 cm. Minimnya campur tangan manusia dari tanah sistem tambak maka semakin beragam jenis dan jumlah dari bahan organik yang terkandung di dalamnya.Kata kunci: bahan organik, tanah dasar, kedalaman, sistem budidaya, lama pemeliharaan

Abstract

ABSTRACT This study aimed to determine optimum stocking density of mud crab Scylla serrata through the applied of different stocking density in every treatment in recirculation system. Experimental design used was complete randomized design (CRD) with three density treatments which were 5 (P1), 10 (P2), and 15 ind/container (P3). All treatments replicated three times. The crab with the average of body weight 150 g/ind cultured in a plastic box (40×30×30 cm). Crab was cultured within 60 days and were fed two times a day by at satiation method. The result showed that P2 treatment gave the best result of mangrove crabs production performance among all treatments with 73.33±5.77% survival rate, 0.68±0.01 g/ind/day absolute growth rate and food conversion ratio 10.11±0.01. Treatment P1 gave the good response of stress, it indicated by the lowest glucose of all tretamnets at the level of 31.91 mg/dL in the end of treatment periods. The water quality during study period was fluctuative as affected by different stocking density in the treatments. Keywords: mud crab, stocking density, production performance  ABSTRAK Penelitian ini bertujuan untuk menentukan padat tebar optimal kepiting bakau Scylla serrata melalui penerapan kepadatan tebar yang berbeda pada setiap perlakuan dalam sistem resirkulasi. Rancangan penelitian yang digunakan adalah rancangan acak lengkap dengan tiga perlakuan yaitu 5 (P1), 10 (P2), dan 15 ekor/wadah pemeliharaan (P3) dengan tiga ulangan. Kepiting bakau yang digunakan memiliki berat rata-rata 150 g/ekor. Wadah pemeliharaan yang digunakan selama pemeliharaan adalah kontainer plastik yang berukuran 40×30×30 cm. Pemeliharaan kepiting bakau dilaksanakan selama 60 hari dan diberikan pakan berupa ikan rucah dua kali sehari secara at satiation. Penelitian menunjukkan bahwa perlakuan P2 memberikan hasil kinerja produksi terbaik dibandingkan perlakuan lainnya dengan nilai kelangsungan hidup 73,33±5,77%, laju pertumbuhan mutlak 0,68±0,01 g/ekor/hari, dan rasio konversi pakan 10,11±0,01. Perlakuan P1 menunjukkan respons stres yang baik dengan memiliki nilai kadar glukosa paling rendah dari perlakuan lain yaitu 31,91 mg/dL pada akhir masa pemeliharaan. Kualitas air selama penelitian memiliki nilai yang fluktuatif di setiap perlakuan sebagai efek adanya perbedaan jumlah padat tebar setiap wadahnya Kata kunci: kepiting bakau, padat tebar, kinerja produksi
