Abstract

Electrogenic microbes have been exploited in the microbial fuel cell (MFC) system for harvesting bioelectricity. Electrogens are widely distributed in various environments, but the exploration of this microbial group from ecosystems in Indonesia is still limited. This present study aimed to isolate the electrogenic bacteria from sediments of two waterlogged ecosystems (dam and paddy field) using serial dilution into extinction method prior to streak plate method on the solid thioglycollate media supplemented with Fe3+. Electrochemical evaluation was conducted in glucose-fed dual-chamber MFC by using Arduino UNO-based data logger for the accurate monitoring of electricity production in the term of open circuit voltage (OCV). A total of 54 electrogens were successfully isolated from these two ecosystems, ranging from weakness to strongest electrogens (OCV >800 mV) and ranging from microaerophilic, aerotolerant and facultative anaerobes, to obligate anaerobes. This result also suggested that sediment of waterlogged ecosystems rich in electron donor and solid acceptor electron compounds could potentially host electrogenic microbes. The exploration of electrogens from many other waterlogged ecosystems in Indonesia, both natural and anthropic ecosystems, could be conducted to collect genetic resources of novel electrogenic bacteria for the development of MFC technology in Indonesia.

Abstract

AHPND causes up to 80% of shrimp mortality, severely impacting Indonesia's aquaculture. Rising antibiotic resistance necessitates sustainable alternatives. This study investigates the probiotic potential of Pseudomonas alcaligenes SG03, a bacterium with unique genomic traits, to address antibiotic resistance and promote animal health. Genome analysis revealed a 6.17 Mb genome with 4,446 protein clusters, including genes for carbohydrate metabolism (26.63%), amino acid metabolism (19.93%), and energy metabolism (10.88%). Key probiotic-related genes, such as acid resistance (rpoS, actP), salinity tolerance (gshA, cysK), antibiotic resistance (vanB, gyrA), and metal resistance (copZ, zwf), were identified. In vitro, assays evaluated probiotic properties under conditions mimicking the shrimp gastrointestinal tract. Osmo-tolerance tests showed optimal growth at 10% glucose, with a significant drop in viability at higher concentrations. Autoaggregation increased progressively, reaching 0.78 at 18 hours and 0.13 at 48 hours. Optimal growth occurred at 30°C, with a 2.5-fold increase in optical density compared to 25°C. NaCl tolerance peaked at 2% (20.33×1012 CFU at 48 hours), while bile salt tolerance was highest at 0.1% (27.00×1012 CFU at 48 hours). pH tolerance was optimal at pH 5 (20.00×1012 CFU at 48 hours). Antioxidant (53.00%) and anti-inflammatory (60.33%) activities peaked after 48 hours. Phenotypic antibiotic resistance was observed against Tetracycline, Amoxicillin, Cefixime, Streptomycin, and Chloramphenicol, with proteolytic activity but no lipolytic or hemolytic properties. These findings highlight P. alcaligenes SG03’s potential as a probiotic in aquaculture to support shrimp health and reduce antibiotic dependency. Future studies should focus on in vivo validation and formulation development.

Abstract

Bees are one of the pollinators that a honey producing agent as well. Honey derives from nectar that stored and procees in honey stomach. The quality of honey is determined by physical, chemical and microbes in the honey stomach as well. This study was aimed to analyze the diversity of bacterial communities (bacterial metagenome) derived from bee’s honey stomach of A. mellifera, A. cerana, and T. laeviceps using Denaturing Gradient Gel Electrophoresis (DGGE) method. Previous researches of bacterial communities are more focused on the entire digestive tract, which is not specifically in honey stomach of the bees. The diversity of bacterial communities showed differences among three bee’s species. The number one of A. cerana has the highest number of Operational Taxonomic Units (OTUs), i.e., 15 OTUs compared to the other sample. While the T. laeviceps number two show the highest percentage of OTU dominant at 59.4%. The bacterial community in bees honey stomach of A. mellifera, A. cerana, T. laeviceps the similarity of bacteria from phylum Firmicutes, Proteobacteria, Acidobacteria and Fusobacteria. Actinobacteria and Bacteroidetes phyla are found only in A. cerana honey stomach.

Abstract

Household organic waste generated daily holds potential for conversion into bioenergy through biodrying technology. Biodrying is a biological drying process that relies on microbial activity to reduce the moisture content in organic waste, producing solid fuel. This study aimed to formulate a microbial consortium for biodrying applications and to compare the effectiveness of solid (talc-based) and liquid (molasses-based) carriers on biodrying performance and inoculant stability. The consortium comprised Lysinibacillus halotolerans, Bacillus soronensis, Lactobacillus rhamnosus, Pichia manshurica, and phosphate-solubilizing bacteria. Isolate rejuvenation was performed using selective culturing followed by characterization. Microbial formulations were developed in both solid and liquid forms and applied to organic waste. Key parameters observed included moisture content, temperature, pH, and inoculant viability over a three-month storage period. The results showed that both talc and molasses carriers were more effective than commercial controls, as indicated by higher temperatures (44.1°C and 43.66°C), lower moisture content (44.5% and 45.25%), and favorable pH values (5.8 and 6.35), which supported mesophilic and thermophilic microbial activity during the biodrying process. Peak microbial activity occurred on day four, predominantly driven by Bacillus species during the thermophilic phase. Inoculant stability was also higher in the talc-based formulation, potentially due to its magnesium and silicon content, which contributed to maintaining microbial viability during storage.

Abstract

A study has been conducted to develop indigenousBacillusthuringiensisbioinsecticide. Preliminary study has been conducted to explore B. thuringiensisfrom sample of soil, leaf, and infected larvae from selected tea area as well as another area such as cocoa, and acasia. The result showed that based on the colony morphology, it was found 10 isolateswith the characteristics of B. thuringiensis colony.  Assessed of the ability to formed crystal using phase contrast microscope and staining revealed that 4 isolates as crystal protein forming isolates. The four isolates used as active coumpound of bioinsecticide. The best formula based on the viability of bacteria was the one withwhite clay as a carrier. While the best liquid formula based on the similar characteristic was using maltosa as osmoprotectant. Theassessment ofthetoxisity B. thuringiensistowards Hyposidra talacalarvae showed that B. thuringiensiscould control H. talacalarvaeup to 37.5%. However, the toxicity need longer periode compared to that commercial B. thuringiensisbioinsecticide. [Keywords:Bt insecticide, cypermethrine, integrated pest management, Tea looper].AbstrakPenelitian telah dilakukan untuk mengembangkan bioinsektisida berbahan aktif B. thuringiensisdari isolat asli Indonesia. Eksplorasi B. thuringiensisdari contoh berupa tanah, daun, dan ulat dari kebun teh telah dilakukan demikian pula dari habitat lain seperti kebun kakao dan akasia.  Berdasarkan morfologi koloni diperoleh 10 isolat yang menunjukkan ciri-ciri koloni B. thuringiensis. Selanjutnyaisolat yang diperoleh diuji kemampuan pembentukan kristal protein dengan pewarnaan dan pengamatan mikroskop phase kontras dan menghasilkan 4 isolat yang mampu membentuk kristal protein. Selanjutnya keempat isolat yang diperoleh digunakan sebagai bahan aktif dalam formulasi bioinsektisida. Formula terbaik berdasarkan kriteria viabilitas bakteri adalah formula yang menggunakan bahan pembawa berupa white clay. Formula terbaik untuk bioinsektida cair berdasarkan kriteria viabilitas B. thuringiensisdan kejernihan bioinsektisida adalah menggunakan maltosesebagai osmoprotektan.Pada pengujian toksisitas isolat B. thuringiensisterhadap larvaulat jengkal(Hyposidra talaca)menunjukkan bahwa B. thuringiensishasil percobaan dapat mengen-dalikan larva ulat jengkalhingga 37,5%. Namun demikian toksisitasnya memerlukan waktu yang lebih lama dibandingkan dengan bioinsektisida berbahan aktif B. thuringiensiskomersial. [Kata kuci: Bioinsektisida Bt, pengendalian hama terpadu, sipermetrin, ulat jengkal teh].

Abstract

Kaur Regency is a high-potential vannamei shrimp (Litopenaeus vannamei) aquaculture area in Bengkulu Province that frequently experiences high mortality rates due to vibriosis caused by pathogenic Vibrio spp. The high shrimp mortality rates resulting in decreased production or crop failure due to attacks or infections of vibriosis disease. Data on these Vibrio spp. in traditional Kaur ponds is required for illness prevention. To identify pathogenic Vibrio spp., this study included purposeful sampling and testing, as well as data analysis. Seven Gram-negative isolates (OALF 1, 2, 3, and 5 from shrimp organs; WALF 7, 8, and 10 from pond water) produced green/yellow colonies on TCBSA medium. Biochemical tests (Gram KOH 3%, catalase, oxidase, fermentation (O/F), TSIA, MIO, lysine decarboxylase, gelatin, Simon citrate, methyl red, urease, glucose, maltose, lactose, mannitol, and sorbitol) revealed that OALF isolates from vannamei shrimp organs have morphological and physiological similarities to the pathogenic species Vibrio parahaemolyticus, whereas WALF isolates from pond water are similar to the pathogenic species V. pluvialis. The study identified Vibrio spp. as the cause of vibriosis in L. vannamei in traditional Kaur ponds, emphasizing the need for improved water and feed management, as well as proper pond environmental practices, potentially including probiotics derived from healthy local shrimp digestive bacteria, to reduce losses in vaname aquaculture in Kaur Regency, Bengkulu. Abstract

Growth of mangosteen essentially depends on its root system.  Therefore, it needs technology to obtain stringer mangosteen root system.  The use of Agrobacterium rhizogenes bacterium is an alternative.  The objectives of this experiment were : 1) to find the effective strain of A. rhizogenes bacterium for inoculation of mangosteen seedling root, 2) to find the best inoculation method for inducing mangosteen seedling root.  The materials used in this experiment were ; mangosteen fruit and A. rhizogenes collection from Puslit Biotechnology LIPI Cibinong-Bogor.  The experiment was arranged in completely randomized design with two factorial treatments.  The first factor : 11 strains A. rhizogenes (ATCC-15834, ATCC-8196, R-1000, 07-20001, A4, A4-J, 509, 510, 511, MAFF 01-1724, and control), the second factor : 2 inoculation methods (cutting and dipping).  The results showed that A. rhizogenes  of ATCC-15834, 509, 07-20001, A4, and R-1000 increased stem diameter, plant height, leaf number, lateral and tertiary root number, better than ATCC-8196, MAFF 01-1724, 510, 511, A4-J, and control.  Cutting root method of inoculation resulted in higher live plant percentage compared to dipping root method.   Key words :  Agrobacterium rhizogenes, Garcinia mangostana, inoculation

Abstract

Methane is a greenhouse gas and has potency to deplete ozone layer.  Rice fields are a significantly sources of atmospheric methane.  Chemical fertilizers application in rice fields can increase methane emission. Methanotrophic bacteria are unique in their ability to utilize methane as a sole carbon and energy source.  This research was successfully isolating and characterizing methanotrophic bacteria from rice fields in Bogor and Sukabumi, West Java, Indonesia.  Determination of methane oxidation activity using Gas Chromatography showed that all isolates performed methane oxidation activity.  The highest methane oxidation activity was performed by BGM 9 isolate. And DNA amplification of BGM 9 genome was performed single band of mmoX in the size of 500 bp and three bands of pmoA  in the size of 1000, 750 and 500 bp respectively

Abstract

Ketersediaan shelter (tempat perlindungan) pada kepadatan spesifik diharapkan dapat meningkatkan produksi kepiting bakau (Scylla serrata) dalam sistem resirkulasi. Shelter sebagai salah satu faktor abiotik berperan penting dalam mengurangi kematian yang disebabkan oleh kanibalisme kepiting dan menekan tingkat stres biota. Sistem Resirkulasi Akuakultur (RAS) dengan bak kotak kultur berkapasitas 60 L yang didukung oleh shelter diperkirakan menghasilkan respons fisiologis dan pertumbuhan kepiting bakau yang paling baik. Penelitian ini bertujuan untuk mengevaluasi dampak penambahan shelter pada lingkungan dengan sistem resirkulasi terhadap respons fisiologis dan produksi kepiting bakau S. serrata dengan kepadatan 10 kepiting per satu bak kultur yang berisi 60 L air laut. Penelitian ini dilakukan dengan tiga perlakuan penambahan shelter, yaitu 2 shelter (S2), 4 shelter (S4), 6 shelter (S6), dan kontrol tanpa shelter (C). S6 adalah perlakuan terbaik dengan tingkat kelangsungan hidup 73,33±5,8%, laju pertumbuhan spesifik 0,886±0,014%, pertumbuhan lebar karapas 0,024±0,004 cm/hari, dan rasio konversi pakan terendah dibandingkan dengan perlakuan lain. Perlakuan S6 secara signifikan mempengaruhi jumlah hemosit total kepiting pada awal budidaya (P <0,05). Penambahan 6 shelter dapat mengoptimalkan pertumbuhan kepiting lumpur dengan kepadatan 10 kepiting dalam satu bak kotak kultur.
