Abstract

Blood is one of the most important component of human body. In a case of crime, blood found at the site can be used as an evidence to track back the victim or the person who did such criminal. The main objective of this research were to investigate the time needed for the convertion of blood types and to identify bacterial species with capability to convert blood types. This research was conducted at Forensic Laboratory and Microbiology Laboratory, Faculty of Mathematics and Science, Udayana University. The methods used for detection of blood types and identification of Bacterias were elution method and vitex method, respectively. The results showed that blood type A on iron surface and ceramic media was changed into fake O type on day 25, while blood types of B and AB on ironic media were converted into fake type of O after 30 days of exposure. Two species bacteria (Rhizobium radiobacter and Serratia ficaria) with ability to convert blood type were identified in this research. Abstract

The main objective of this research was to isolate and identify microbes residing in the rhizosphere zone of strawberry (Fragaria x ananassa Dutch.), cultivated at Pancasari village, Bedugul Bali. This was an explorative and descriptive research conducted from April to July 2018. Soil samples were collected from the rhizosphere of strawberry plants and microbes residing in them were isolated by applying dilution and platting method, purified, and identified at the Food Microbology Laboratory, Faculty of Agricultural Technology, Udayana University. Important characteristics of the isolated microbes were compared to those specified in references, such as Bergey’s Manual of Determine Bacteriology 9th Edition (Holt et al., 1994) for bacterial isolates and Fungal Identification keys (Barnett and Hunter, 1972; dan Pitt and Hocking, 2009) for fungal isolates. Microbes successfully isolated and identified in this research included fungi and bacteria belong to Aspergillus flavus, Aspergillus niger, Fusarium oxysporum, and Colletotrichum sp., Pseudomonas sp., and Bacillus sp. Abstract

A research on in vitro inhibition of fungal antagonists, isolated from dragon fruit plantation in Sembung village, Bali, on Fusarium sp. (the disease causative agent of dragon fruit plant) was conducted with the main objective to investigate the effectiveness of these fungal antagonists to inhibit the in vitro growth of the pathogen. Dual assay method was applied in this experiment. The results showed that three potential fungal antagonists were successfully isolated in this research and they were identified as Trichoderma harzianum, Aspergillus niger, dan Paecilomyces lilacinus. All these fungal antagonists showed antagonistic activity against Fusarium sp. which was statistically significant (p<0.05) when compared to control. This indicated that all antagonist isolates were potential to be developed as biocontrol agent candidates.

Abstract

Hyperlipidimia merupakan suatu kelainan yang memicu terjadinya penyakit kardiovascular yang jumlah penderitanya cenderung mengalami peningkatan. Kelainan ini disebabkan oleh pola makan masyarakat yang cenderung mengkonsumsi makanan tinggi lemak, tetapi rendah kadar serat. Pemakaian obat paten, seperti simvastatin merupakan pilihan utama saat ini untuk mengatasi masalah tersebut. Namun, pemakaian obat ini dalam waktu lama tidak dianjurkan karena dapat menyebabkan efek samping merugikan bagi tubuh, sehingga perlu dicari bahan alam yang mempunyai efek sama dengan obat tersebut. Pada penelitian ini, potensi hypolipidemia daun salam diuji pada tikus jantan galur Wistar karena secara empirik daun tanaman ini sering dipakai sebagai obat tradisional. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak etanol daun salam (EEDS) dan rebusan daun salam (ARDS) terhadap menurunkan LDL-Kolesterol tikus galur Wistar. Penelitian ini menggunakan rancangan acak kelompok (RAK) dengan delapan kelompok perlakuan yaitu perlakuan dengan aquades (kontrol negatif), simvastatin 0,36 mg/kgBB (kontrol positif), EEDS 252mg/kgBB (EEDS 252), 504mg/kgBB (EEDS 504), 756mg/kgBB (EEDS 756), ARDS 25%/kgBB (ARDS 25), 50% mg/kgBB (ARDS 50), dan 75% mg/kgBB (ARDS 75). Perlakuan diberikan selama satu minggu pada tikus yang sebelumnya diberi pakan kaya lemak selama satu bulan. Hasil penelitian menunjukkan terjadinya penurunan kadar LDL-Kolesterol secara signifikan (p?0.05) pada semua kelompok perlakuan dengan kisaran potensi sebesar 8,00±0,82% sampai 56,16 ± 5,69% (< kontrol negatif dengan penurunan sebesar 6,25 ± 0,50%). Bahkan pada perlakuan 756mg/kgBB EEDS (EEDS 756) menunjukkan hasil yang lebih baik (p?0.05) daripada kontrol positif (simvastatin). Efek hypolipidemia dari ekstrak daun salam ini diduga disebabkan oleh golongan senyawa-senyawa flavonoid, alkaloid, dan tannin yang terkandung dalam ekstrak daun salam. Abstract

The main objectives of this research were to isolate and identify antagonists of Xanthomonas campestris from rhizosphere zone of broccoli plants. Soil samples were collected from broccoli farm located at Kembang Merta village, Tabanan, Bali. Isolation and identification of the antagonists were conducted at the Laboratory of Microbiology, Udayana University. Two fungal (Trichoderma harzianum and  Trichoderma viride) and two bacterial (Bacillus sp. and Pseudomonas sp.) antagonists potentially to be developed as biocontrol agents of Xanthomonas campestris were successfully identified in this research.

Abstract

Candidacies in female reproductive tract are mainly caused by Candida albicans. This infection often causes serious problems, particularly on their reproductive tract (genital part). Until recently, control of this infection has relied on the use of antibiotics. However due to numerous bad side effects of antibiotics, lactic acid bacteria have been proposed as an alternative method to control the growth of Candida albicans. Therefore, this research was aimed to isolate, screen, and characterize lactic acid bacterial isolates (LAB) antagonistic against Candida albicans (the causative agent of candidacies infection in reproductive tract of human). LABs were isolated from various fermented foods, such as tape ketan and kimchi. Isolation of LABs was conducted by applying dilution and spread plate method on MRS agar medium supplemented with BCP indicator to distinguish LABs from non acid-producing bacteria. Colonies with indication to produce acid were screened for antagonistic activity against C. albicans on MRS agar and followed by characterization of those isolates (Gram stain, catalase production test, oxydase production, gas production test, resistance test to low pH conditions and to high level of NaDC (sodium deoxicolic), and test for ability to convert colic acid (CA) into deoxicolic acid (DCA)). The results showed that 46 LAB isolates were successfully isolated from samples of tape ketan and kimchi. Among those, 7 isolates showed antagonistic activity against C. albicans in in vitro tests. All these 7 candidates were also found to be resistance to low pH conditions (up to pH 2) and to high level of NaDC (up to 0.6 mM). Four most potential isolates were further testes for ability to convert colic acid into deoxycolic acid and none showed positive result, indicating that they all showed initial potential and safe for future human probiotic development (especially to be used to treat patients infected by C. albicans).

Abstract

Pneumolysin is a protein that plays an important role in cell death and bacterial colonization of Streptococcus pneumoniae. Ceftriaxone, clarithromycin, dan quercetin has been proven (in vivo and in vitro) to prevent the action of pneumolysin. The purpose of this study was to identify interactions between ceftriaxone, clarithromycin, and quercetin in inhibiting the pneumolysin activity through in silico. The 3D structure of pneumolysin was downloaded via RCSB PDB (PDB ID:PQQA), while 3D structure of ceftriaxone (ID:5479530), clarithromycin (ID:84029), and quercetin (ID:5280343) were downloaded via PubChem. Protein preparations were done with Discovery Studio, while ligands were prepared with OpenBabel. The docking process used HEX 8.0.0 and visualization analysis with Discovery Studio. Pneumolysin-ceftriaxone interaction produces 4 hydrogen bonds, 5 hydrophobic bonds, 1 electrostatic bond, and 2 unfavorable residue bonds with a bond energy value -329.98kJ/mol. Pneumolysin-clarithromycin interaction produces 3 hydrogen bonds, 1 electrostatic bond, and 3 unfavorable residue bonds with a bond energy value -311.12kJ/mol. Pneumolysin-quercetin interaction produces 2 hydrogen bonds, 4 hydrophobic bonds, and 2 electrostatic bonds with a bond energy value -246.52kJ/mol. Abstract

The main objective of this research was to investigate the potential of Lactic Acid Bacterial isolates and their supernatants, isolated from yoghurt and kefir, to control the growth of A. flavus FNCC6109 in chicken feed concentrate. The experiments were conducted both In vitro and directly applied to chicken feed concentrate. The results showed that two Lactic Acid Bacterial were successfully isolated, identified and both showed initial potential to be developed as biocontrol of A. flavus FNCC6109. Only one best isolate (isolate I) was used in the further investigation. Percentage of growth reduction of A. flavus FNCC6109 in the chicken feed concentrate treated with 30%(v/v) of Lactobacillus sp. supernatant was 72% lower than control (chicken feed concentrate without addition of Lactobacillus sp. Abstract

Cananga odorata and Cymbopogon citratus are two plant species producing inhibitory compounds in their essential oils. In our current research, the potential of their essential oils to inhibit the growth of Staphylococcus aureus and Candida albicans were investigated. MIC (Minimum Inhibitory Concentration) values of mixture of those essential oils with a ratio of 1:1 on those microbes were also assayed. Bioassays were conducted on Nutrient Agar (NA) or Potato Dextrose Agar (PDA) medium added with various ratios of the two plant species essential oils. Medium added with sterile distilled water served as control. The data obtained were analyzed quantitatively with SPSS software for Windows version 22. When significant different at p<0.05 was indicated, the analysis was continued with Duncan's multiple range test. The result showed that all treatment of essential oils applied effectively eliminated the growth of S.aureus. C.albicans appeared to be relatively resistant to 100% of C.odorata essential oil. Elimination of C.albicans was observed when the essential oil of C.odorata was combined with that of C.citratus. When compared to control, all ratios of essential oils treatments were statistically significant (p<0.05). The MIC values of essential oil mixture a ratio of 1:1 on S.aureus and C.albicans were 0,1% and 2,6% respectively. Abstract

Carbapenem Resistant Enterobacteriaceae (CRE) is a group of Enterobacteriaceae resistant to Carbapenem antibiotics (Imipenem, Ertapenem, Meropenem, Doripenem). One species of Enterobacteriaceae frequently showing resistant characteristic to Carbapenem is Klebsiella pneumoniae. Early detection methods on the CRE existence (phenotypically or genetically) are urgently needed. The methods should be able to detect carbapenemases or genes encoding these types of enzymes. In the recent years, phenotypic characteristics can be done by applying methods, such as Modified Hodge Test (MHT), Carba Nordmann-Poirel (Carba NP), or Modified Carbapenem Inactivation Method (MCIM). To detect the presence of genes encoding production of carbapenemases, Polymerase Chain Reaction (PCR) is the most reliable method available in the recent years.
