Abstract

The development of antibiotic resistance has become an important issue in medicine. Many types of bacteria are pathogenic to humans were reported to have become resistant to one or more types of antibiotics. It is pushing for a new search for alternative sources of antibiotic-producing compounds. One source of potential alternatives to be developed as a source of new antibiotic compound s are symbiotic bacterium with gastropods. The purpose of this study was to determine the characteristics of bacterial symbionts in gastropods ( TOV 12.16 bacterial isolates), the bioactivity of crude bacterial extracts against bacterial MDR and find a group of compounds from TOV 12.16 crude bacterial extracts which most potentially. The main materials used in this study were samples of bacterial isolates TOV 12.16 in isolation from the gastropods (Oliva vidua) from Aquatic Ternate (North Maluku). While the bacterium MDR test are Escherichia coli and Staphylococcus aureus obtained from the Laboratory of Microbiology Faculty of Medicine UNDIP. Biochemical tests used to determine the characteristics of bacterial isolates TOV 12.16. Meanwhile, to identify the bioactive compounds using phytochemicals screening and GC/MS. Bacterial symbionts Oliva vidua Gastropoda is Vibrio ordalii. Ethyl acetate crude extract of bacterial symbionts Oliva vidua Gastropoda species showed antibacterial activity of the most active against MDR bacteria test. Phytochemical screening test results showed the ethyl acetate crude extract showed two groups of compounds which contain flavonoids and triterpenoid compound that has both the antibacterial activity. Being on the analysis of the GC/MS_are_known_isobutyric_acid_and_2-metilbutanoic_acid.

Abstract

Teripang merupakan hewan jenis Echinodermata. Beberapa species teripang, misalnya H. scabra, H. fuscogilva dan T. ananas memiliki nilai jual yang tinggi. Meskipun tidak semua spesies memiliki nilai jual yang tinggi, teripang berpotensi untuk dikembangkan dalam bidang kesehatan, terutama dalam kemampuannya untuk meredam radikal bebas dan mencegah berbagai penyakit degeneratif. Tujuan dari penelitian ini yaitu untuk mengidentifikasi, menentukan aktivitas antioksidan, kandungan total fenol, serta kadar karotenoid pada ekstrak beberapa teripang yang berasal dari Perairan Karimunjawa Jepara. Hasil identifikasi dari ke empat spesies adalah Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, dan Holothuria atra. Ekstraksi dilakukan dengan teknik maserasi dinding tubuh teripang menggunakan pelarut metanol. Uji aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (1,1-difenil-pikrilhidrazil) sebagai agen radikal bebas. Penentuan total fenolik dilakukan menggunakan metode folin-ciocalteu, dan penentuan kadar karotenoid dilakukan dengan menggunakan spektrofotometer. Hasil menunjukkan bahwa aktivitas antioksidan Bohadschia vitiensis, Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, dan Holothuria atra tergolong sangat lemah dengan nilai IC50 secara berturut-turut adalah sebesar 454,28 ± 14,4; 713,51 ± 9,1; 801,57 ± 8,4 dan 1535,47 ppm. Kandungan total fenol sebesar 21,08 ± 0,49; 14,325 ± 0,21; 14,033 ± 0,33 dan 10,67 ± 0,12 mg GAE/g sampel, dan kadar karotenoidnya sebesar 23,28 ± 0,07; 25,78 ± 0,56; 11,85 ± 0,04; dan 41,44 ± 0,008 µmol/g sampel.Kata kunci: Teripang, Antioksidan, DPPH, Fenolik, Karotenoid Sea cucumbers are sea benthos from phylum Echinoderms phylum. Some sea cucumbers such as H. scabra, H. fuscogilva, T. Ananas have a high economical price. Eventhough, some sea cucumbers have the potency of being a healthy food. It it scientifically proven that some sea cucumbers have the ability to reduce free radicals and prevent various degenerative diseases caused by the excessive free radicals. The aims of this study were to identified, determine antioxidant activity, total phenol content, and carotenoid levels in sea cucumber extracts originally from Karimunjawa waters. Some of the sea cucumbers obtained are Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, and Holothuria atra. The extraction was done by macerating the body walls with methanol. The antioxidant activity test were tested using the DPPH method as free radicals, the total phenolic content tested using the folin-ciocalteu method, and determined the carotenoid levels spectrophotometrically. Based on the results, antioxidant activity of  B. vitiensis, S. cf. quadrifasciatus, P. graeffei, and H. atra were  classified as very weak with IC50 454.28 ± 14.4; 713.51 ± 9.1; 801.57 ± 8.4 dan 1535.47 ppm. Total phenolic content were 21.08 ± 0.49; 14.325 ± 0.21; 14.033 ± 0.33 dan 10.67 ± 0.12 mg GAE / g samples, while carotenoid levels were 23.28 ± 0.07; 25.78 ± 0.56; 11.85 ± 0.04; dan 41.44 ± 0.008 µmol / g sample.Keyword : Sea cucumber, Antioxidant, DPPH, Phenolic, Carotenoid

Abstract

Along with the increasing resistance of bacteria in the health world, it is necessary discover a new drug. Sources of new antibacterial bioactive compounds can be obtained from seaweeds. The purposes of the research were to determine the antibacterial activity of crude extract seaweed againts the skin disease bacteria, Pseudomonas aeruginosa, Staphylococcus epidermidis and Micrococcus luteus, and to identify classes of compounds contained in crude extract of seaweed. This research was done from January to Mei 2012. Seaweeds sampling (Caulerpa sp, Eucheuma sp, Gracilaria sp dan Sargassum sp) was conducted in territorial water of Jepara. Extraction process and analysis of bioactive compound of crude extracts was conducted at Laboratory, Marine Science Department, Diponegoro University, Semarang. Test antibacterial activity was conducted at Medicine Laboratory, Ungaran. The method used in this research was experimental laboratories. Extraction process was conducted with solid-liquid extraction method. Extract were tested for antibacterial activity by agar diffusion method according to Kirby-baurer. Based on the antibacterial activity’s assay, 12 extracts which have antibacterial activity against test bacteria P. aeruginosa, S. epidermidis and M. luteus were obtained the 12 seaweed extracts the highest consentration of inhibition zone was 200 µg/disk in ethyl acetate extract of the Sargassum sp was the most active extracts against bacteria P. aeruginosa and M. luteus, while the methanol extract of Sargassum sp was the most active extracts against the bacteria S. epidermidis. Steroids was the most dominant compounds on all of 12 seaweed extracts.

Abstract

ABSTRAK : Bagian-bagian pohon dari R. mucronata seperti serasah daun, kulit kayu, maupun limbah propagul diketahui memiliki kandungan pewarna yang ramah lingkungan. Potensi pewarna alami dari tumbuhan ini dapat menjadi alternatif bahan produksi bagi industri batik di Indonesia. Pengambilan sampel dilakukan di Teluk Awur, Jepara, Jawa Tengah pada bulan Oktober 2015 kemudian dilakukan ekstraksi pewarna, pewarnaan, dan berbagai analisis. Tujuan penelitan adalah untuk mengetahui kualitas pewarna alami dari bagian-bagian pohon R. mucronata sebagai pewarna alami pada kain katun. Sampel R. mucronata yang diambil yaitu kulit kayu, limbah propagul, dan serasah daun. Ekstraksi pewarna dilakukan menggunakan air panas pada suhu 30°C, 50°C, dan 70°C. Pengikatan warna setelah pencelupan menggunakan kain katun mori primissima dengan mordan tawas. Hasil penelitian menunjukkan bahwa warna cokelat dihasilkan oleh ekstrak R. mucronata. Analisis FTIR dan UV Vis mengindikasikan adanya senyawa tanin terkondensasi. Pengujian Total Phenol Content (TPC) dan Total Flavonoid Content (TFC) menunjukkan hasil positif sebesar 2,4950 mg GAE/g untuk TPC dan 0,6516 mg QE/g untuk TFC. Hasil pindaian dengan Scanning Electron Microscope menunjukkan granula-granula yang pecah pada spesimen. Kualitas pewarnaan pada kain menunjukkan kisaran hasil antara 3 (cukup) hingga 4 (baik) dan telah memenuhi standar SNI. Pewarna dari serasah daun pada ekstraksi 70°C menunjukkan hasil terbaik di antara yang lain. ABSTRACT : The tree parts of R. mucronata such as the leaves litter, bark, and propagule are known for containing dyes that are environmentally friendly. This natural dye potential could be used for the production resource alternative for the batik industry in Indonesia. Sampling was conducted in Teluk Awur, Jepara, Central Java on October 2015 and followed by dye extraction, dying process, and also several analysis. The purpose of this research was to discover the quality of natural dye from R. mucronata on cotton fabric. Samples that were taken from R. mucronata were bark, leaves litter, and propagule litter. Dye extraction used hot water method with variants of temperature: 30°C, 50°C, dan 70°C. The color locking after dyeing used cotton fabric with mori primissima type and alum as the mordant. The obtained result from extract R. mucronata showed brown color. Condensed tannin was indicated from FTIR and UV Vis analysis. Total Phenol Content and Total Flavonoid Content assays showed positive result as follows: 2,4950 mg GAE/g for TPC and 0,6516 mg QE/g for TFC. Scanning result using Scanning Electron Microscope showed that the granules break on the specimen. Color fastness quality showed the range of result from colored fabric from 3 (enough) until 4 (well) and already fulfilled SNI standard. The dye extracted from leaves litter in 70°C showed the best result among the others.

Abstract

It is known that the bacterial symbionts are symbiotic with gastropods Conus miles has the potential to be able to produce antibacterial compounds against bacterial MDR (Multi Drug Resistant). Information found previously from bacterial symbionts and isolation based on screening results, the size of the zone of inhibition and inhibitory properties against several types of bacteria tests showed that isolates derived from Conus miles Gastropoda symbionts characterized by kinship rate of 98% in bacteria Pseudoalteromonas sp. The research objective was to determine the characteristics of the bacterial symbiont bioactive compounds associated with the type of mollusk Conus miles. The study was conducted three phases namely the isolation and purification of bacteria; characterization of isolates identified through test results on the reaction of physiological and biochemical tests, as well as the identification of secondary metabolites by GC-MS method QP2010S Shimadzu. The results showed that the bacterial symbiont isolates had a number of compounds with a percentage of the highest peak to the lowest compound the compound, Acetic acid (CAS) ethylic acid (35.22%); propanoic acid, 2-methyl-(CAS) Isobutyric acid (11.78%); Iso -valeric acid (9:38%), butanoic acid, 2-methyl-(CAS) 2-methylbutanoic acid (4.74%) and 1,2-Benzenedicarboxylic acid, dioctyl ester (CAS) Dioctyl phthala (4:36%). The results concluded that the bacterial symbiont isolates obtained in this study provide information on the results of potentially new antibiotics.

Abstract

Gastropod is one of the marine organisms that can be used as a source of natural bioactive compounds, one of which is Telescopium telescopium. T. telescopium until now it has not been yet exploited optimally, some people still use it as feed ingredients that contain high protein nutrition, however it must be supported by scientific information about the efficacy and side effects. This research aims at determining the secondary metabolite by crude extract T. telescopium, and the lethal toxicity value (LC50-24 hours). The research consisted of two steps, namely the phytochemical and toxicity test. The method used exploratif and experimental laboratory with a complete randomized design, which consited of five concentration treatments namely 25.12, 63.1, 158.5, 398.15, and 1000 ppm with three replications. Each treatment used 10 Artemia salina larvae, and observations were made during 24 hours of Artemia salina larvae mortality. LC50 was determined by probit analysis using the EPA Probit Analysis Program Version 1.5 Finney. The results of the phytochemical test showed that crude extract T. telescopium contain alkaloid compounds, steroids, flavonoids. The results BSLT (Brine Shrimp Lethality Test) of crude extract T. telescopium LC50-24 hours showed that the crude extract clorofom (229.562 ppm), crude extract ethyl acetate (244.906 ppm), and crude extract methanol (197.242 ppm). LC50-24 hours < 1000 ppm showed that the crude extract Telescopium telescopium potent antitumor compounds.

Abstract

Sargassum sp. is a known genus of brown seaweed whose dominant component is alginic acid or alginate. Various studies found that alginate having proinflammatory activity, so it may have potential to improve the wound healing process. This study aimed to obtain sodium alginate extract and to know the potency of the sodium alginate extract in incision wound treatment in mice abdomen. Lotion application was performed once a day in a span of five consecutive days. Observation on the wound was carried out to obtain data on blood coagulation, tissue reparation, and fibroblast development rate on the wound using histology. The results showed that the average yield from the Sargassum sp. extraction process was 31.3%. Quality control of the prepared lotion included organoleptic test; light brown coloration, lotion specific odor, semi- solid texture. The acidity of the lotion was measured as 6.61 with 6.731 cp of viscosity. The lotion adhesiveness test showed 0.21 seconds and the lotion was shown to be oil-in-water. Application of the lotion on incision wounds made in the abdomen of rats showed blood coagulation on the first day and onset of wound reparation process on the second day. Wound observation by the third day showed that the inflicted area had undergone a near-complete reparation. The study also showed that on the fifth day of reparation Sargassum sp. extract lotion treatment group showed fibroblast formation, as opposed to the non-extract lotion treatment group. Previous study use alginates as wound dressing material, but in this study alginate used as active ingredient in wound healing treatment. Therefore, this study concludes that sodium alginate in Sargassum sp. seaweed has a potential application in the field of medicine.

Abstract

Ekosistem lamun memiliki fungsi ekologi dan ekonomi yang tinggi. Peran ekosistem lamun dalam penyimpanan karbon akan tetapi masih belum menjadi sorotan. Tujuan dari penelitian ini yaitu untuk mengetahui nilai biomassa dan estimasi simpanan karbon pada ekosistem lamun di Perairan Batulawang, Pulau Kemujan serta Pulau Sintok, Taman Nasional Karimunjawa. Penelitian ini dilaksanakan pada 7 – 14 Noevmber 2019 di Perairan Batulawang dan Pulau Sintok, Taman Nasional Karimunjawa. Metode penelitian di lapangan menggunakan metode SeagrassWatch, sementara nilai biomassa dan nilai estimasi simpanan karbon dihitung menggunakan metode Metode Loss of Ignition (LOI) di laboratorium. Data yang diperoleh berupa pengukuran berat kering untuk menghitung biomassa dan analisa kandungan karbon pada lamun dan sedimen. Hasil penelitian didapatkan empat jenis lamun di Perairan Batulawang yaitu Enhalus acoroides, Thalassia hemprichii, Cymodocea serrulata, dan Thalassodendron ciliatum sedangkan di Pulau Sintok terdapat tiga jenis lamun yang ditemukan yaitu Thalassia hemprichii, Cymodocea rotundata, dan Halophila ovalis. Nilai total biomassa lamun terbesar pada Perairan Batulawang yaitu Enhalus acoroides dengan nilai 849,75 gbk/m2 dan nilai total biomassa lamun terkecil Thalassodendron ciliatum dengan nilai 29 gbk/m2. Nilai total biomassa lamun terbesar pada Pulau Sintok yaitu Cymodocea rotundata dengan nilai 177,75 gbk/m2dan nilai total biomassa lamun terkecil Halophila ovalis dengan nilai 4,75 gbk/m2. Hasil pengukuran karbon lamun pada Perairan Batulawang yaitu 12,97 – 359,87 gC/m2­ dan 258,20 – 541,51 gC/m2 pada sedimennya. Hasil pengukuran karbon pada lamun di Pulau Sintok yaitu 2,35 – 85,80 gC/m2 dan 204,92 – 765,92 gC/m2 pada sedimen. Kandungan karbon paling besar terdapat pada bagian bawah substrat (below ground). Kandungan karbon pada bagian bawah substrat tidak terganggu oleh faktor lingkungan (gelombang, arus, dan ulah manusia) sehingga terakumulasi baik. Seagrass ecosystems have high ecological and economic functions. The role of seagrass ecosystems in carbon storage, however, has not yet been highlighted. The purpose of this study was to determine the value of biomass and estimated carbon storage in seagrass ecosystems in Batulawang waters, Kemujan Island and Sintok Island, Karimunjawa National Park. This research was conducted on 7 − 14 November 2019 in Batulawang waters and Sintok Island, Karimunjawa National Park. The research method in the field uses the SeagrassWatch method, while the biomass value and the estimated value of carbon storage are calculated using the Loss of Ignition (LOI) method in the laboratory. The data obtained were measurements of dry weight to calculate biomass and analysis of carbon content in seagrass and sediments. The result shows that there are four species of seagrass in Batulawang Waters, they are Enhalus acoroides, Thalassia hemprichii, Cymodocea serrulata, and Thalassodendron cliatum meanwhile in Sintok Island there are three species, they are, Thalassia hemprichii, Cymodocea rotundata, and Halophila ovalis. The measurement of carbon is done by using Loss on Ignition Method. The highest total seagrass biomass in Batulawang waters is Enhalus acoroides with a value of 849.75 gbk/m2 and the lowest total seagrass biomass is Thalassodendron ciliatum with a value of 29 gbk/m2. The highest total seagrass biomass on Sintok Island is Cymodocea rotundata with a value of 177.75 gbk/m2 and the lowest total seagrass biomass is Halophila ovalis with a value of 4.75 gbk/m2. The results of measurements of seagrass carbon in Batulawang waters are 12,97 – 359,87 gC/m2­ and 258,20 – 541,51 gC/m2 on the sediments. The result of seagrass carbon measurement in Sintok Island is 2,35 – 85,80 gC/m2 and 204,92 – 765,92 gC/m2 on the sediments. The largest carbon content is at the bottom of the substrate (below ground). The carbon content at the bottom of the substrate is not disturbed by environmental factors (waves, currents, and human activities) so that it accumulates well.

Abstract

Rajungan merupakan komoditas perikanan yang banyak diminati baik dalam maupun luar negeri. Permintaan rajungan meningkat setiap tahunnya. Desa Tunggulsari merupakan desa penghasil rajungan di Rembang. Desa ini terletak di Kecamatan Kaliori dengan 78,35% penduduknya nelayan. Penangkapan rajungan terjadi setiap hari tanpa adanya restocking. Kondisi ini dapat menyebabkan overfishing dan berdampak pada kelestarian rajungan. Penelitian ini bertujuan untuk mengkaji pertumbuhan rajungan (Portunus pelagicus) hasil tangkapan nelayan di Desa Tunggulsari, Rembang meliputi distribusi lebar karapas dan pola pertumbuhan. Metode yang digunakan pada penelitian ini adalah deskriptif. Penelitian ini berlangsung selama 30 hari dengan pengambilan 100 sampel rajungan per hari. Hasil penelitian ini diketahui ukuran rajungan terdistribusi antara 49–178 mm dengan pola pertumbuhan allometrik negatif dengan nilai b=2,8575. Berdasarkan penelitian terdapat 64 % rajungan yang layak tangkap dan 36 % rajungan yang tak layak tangkap meliputi rajungan muda berukuran <10 cm. Berdasarkan data, nelayan belum menerapkan PERMEN KP No. 12 Tahun 2020 tentang penangkapan rajungan. Hal ini akan berdampak pada kondisi rajungan di wilayah Tunggulsari.  Blue swimming crab fishery commodity that has high demand both at home and abroad. Tunggulsari is one of the villages in Rembang where people catch blueswimmingcrab. It’s located in Kaliori Sub-district with 78.35% population of fishermen. Blue swimming crab is catched every day without any restocking. This condition cause overfishing and impact on the sustainability of blue swimming crab. This research aims to study the growth of blueswimming crab (Portunus pelagicus) in Tunggulsari, Rembang include distribution of width and growth pattern. The method used in this research is descriptive. This research lasted for 30 days by taking 100 sample/day. The results is the width is distributed between 49–178 mm and has a negative allometric growth pattern with value of b=2.8575. Based on the research, there are 64% blueswimming crab that worth to catch and 36% blueswimming crab that doesn’t worth to catch include young crab which size <10 cm. Based on the data, fishermen have not implemented Government Regulation No. 12, 2020 regarding catching crabs. This impact on the condition of the crabs in the Tunggulsari area.‎ Based on the research, there are 64% of crabs that are suitable for catching and 36% of crabs that are not suitable for catching include young crabs measuring <10 cm Based on the data, fishermen have not implemented Government Regulation No. 12, 2020 concerning the Crab Regulation. This will have an impact on the chopping conditions in the Tunggulsari area.‎

Abstract

Teripang Stichopus hermanii merupakan biota laut yang mempunyai senyawa dengan bioaktivitas sebagai antibakteri terhadap patogen. Stichopus hermanii mampu menghambat bakteri gram positif dan negatif. Produk teripang yang sudah ada banyak memperlihatkan manfaatnya salah satunya untuk gigi. Produk komersil yang terlihat di masyarakat yakni pasta gigi, namun belum diketehaui  secara mendetail mengenai seberapa besar peran pasta gigi tersebut dalam mengatasi permasalahn gigi.  Tujuan dari penelitian ini untuk mengetahui senyawa yang aktif didalam ekstrak teripang Stichopus hermanii. Metode ekstraksi yakni dengan padat ke cair, pencarian senyawa aktif menggunakan skrining fitokimia dengan pereaksi yang berbeda setiap pengujiannya, dan uji aktivitas antibakteri menggunakan metode difusi agar dengan konsentrasi yang berbeda-beda. Uji aktivitas antibakteri pada ekstrak S. hermanii menggunakan bakteri patogen Streptococcus mutans. Hasil senyawa aktif yang didapat dari skrinining fitokimia meliputi flavonoid, alkaloid, triterpenoid dan saponin. Uji aktivitas antibakteri menunjukkan bahwa ekstrak metanol S. hermanii mampu menghambat bakteri S.mutans. Zona hambat tertinggi pada S. mutans  5,86 mm ± 4,92 dengan konsentrasi 80 µg/disk pada waktu 24 jam. Disimpulkan bahwa senyawa aktif pada ekstrak metanol S. hermanii mempunyai bioaktivitas antibakteri pada bakteri S. mutans.  The sea cucumber Stichopus hermanii is a marine biota that has compounds with bioactivity as an antibacterial against pathogens. Stichopus hermanii can inhibit gram-positive and gram-negative bacteria. Sea cucumber products that already exist have shown many benefits, one of which is for teeth. The commercial product seen in the community is toothpaste, but it is not yet known in detail how big the role of toothpaste is in overcoming dental problems. The purpose of this study was to determine the active compounds in sea cucumber extract Stichopus hermanii. The extraction method is solid to liquid, the search for active compounds uses phytochemical screening with different reagents for each test, and the antibacterial activity test uses the agar diffusion method with different concentrations. Antibacterial activity test on S. hermanii extract using the pathogenic bacterium Streptococcus mutans. The results of the active compounds obtained from phytochemical screening include flavonoids, alkaloids, triterpenoids, and saponins. The antibacterial activity test showed that the methanol extract of S. hermanii was able to inhibit S. mutans bacteria. The highest zone of inhibition in S. mutans was 5.86 mm ± 4.92 with a concentration of 80 g/disk at 24 hours. It was concluded that the active compound in the methanolic extract of S. hermanii had antibacterial bioactivity on S. mutans bacteria.
