Abstract

Abstract- The purpose of this research was to determine the antibacterial activity of garlic extract and black garlic against E.coli sensitive and resistant to antibiotics. This research is an experiment with 2 factors : 1) the type of extract (garlic and garlic Black), 2) test bacterial strain (E. coli sensitive and resistant to antibiotics). Solvent extraction using 20% ethanol, 1: 1 and then centrifuged. Antibacterial activity test by well diffusion method. Parameter study is inhibition zone formed around the wells. The results showed that extracts of garlic has antibacterial activity against E.coli sensitive (14.22 mm) and antibiotic-resistant E.coli (22.2 mm), while the black garlic extract showed no antibacterial activity. Keywords: garlic, black garlic, E.coli

Abstract

ABSTRAK Antibiotik merupakan bagian penting dalam terapi infeksi bakteri  karena itu penemuan sumber antibiotik baru yang potensial sangat diperlukan untuk mengatasi masalah akibat  infeksi bakteri. Streptomyces adalah bakteri yang mampu memproduksi agen antimikroba dan banyak ditemukan di tanah pada rizosfer tanaman tingkat tinggi. Tujuan penelitian ini adalah mengetahui potensi Streptomyces dari rizosfer Familia Poaceae yang dapat menghasilkan antibiotik untuk menghambat bakteri S. aureus dan  E. coli. Isolasi Streptomyces dari rizosfer rumput kembangan (Digitaria microbachne (Presl.) Henr), rumput gajah (Pennisetum purpureum Schumach) dan alang–alang (Imperata cylindrica L) yang tumbuh dari tanah yang kering, sedikit berpasir dan tidak tergenang air. Tahap awal isolasi Streptomyces adalah pre treatment sampel tanah. Isolasi menggunakan media raffinosa-histidine agar. Isolat dipurifikasi pada media benneth agar kemudian dipindah ke media oatmeal agar untuk menumbuhkan spora. Identifikasi isolat Streptomyces dilakukan dengan pengecatan Gram. Uji potensi isolat Streptomyces dilakukan dengan metode agar block menggunakan bakteri uji S. aureus dan  E. coli ATCC 25922. Hasil isolasi diperoleh 57 isolat Streptomyces dengan warna miselium vegetatif, spora aerial dan pigmen difus yang berbeda. Dari 57 isolat diperoleh 10 isolat (17,50%) berpotensi ”sangat kuat” (diameter lebih dari 20 mm), 6 isolat (10,52%) berpotensi ”kuat” (diameter 10-20 mm) terhadap S. aureus, sedangkan terhadap E. coli ATCC 25922 diperoleh 8 isolat (14,04%) berpotensi antibiotik “sangat kuat” (20-30 mm), 11 isolat (17,54%) berpotensi ”kuat” (10-19,5 mm), dan 1 isolat (1,75%) berpotensi ”sedang” (7 mm).   Kesimpulan yang diperoleh adalah Streptomyces dapat dijadikan sebagai sumber penghasil antibiotik baru yang sangat potensial di Indonesia.   Kata Kunci:  Antibiotik, Streptomyces,  Familia Poaceae, Rizosfer

Abstract

The high cost of instant media such as nutrient agar encourage researchers to find an alternative media from raw materials that are easily available and cheap. This study aims to determine of using a source of carbohydrates to used as an alternative media. The source of carbohydrates that can be used is Canna edulis, Dioscorea esculenta, and Maranta arundinaceae and the results were compared with nutrient agar. The sample used are 300 gr of tubers with addition 10 gr of sugar and 15 gr of agar in 1000 ml of destilled water. This study used a Completely Randomized Design (CRD) two factors. The first factor is a type of bacteria: Escherichia coli and Staphylococcus aureus and the second factor is the types of alternative media: Canna edulis, Dioscorea esculenta, and Maranta arundinaceae. The bacteria were inoculated with a spread plate method and incubated at 37 ° C for 24 hours. the parameters are a number of population and the size of the bacterial colonies.The result showed that in Escherichia coli, the highest population is in the Canna edulis (7.56x107CFU/ml), Dioscorea esculenta (6,86x107CFU/ml), nutrient agar media (6.21x107CFU/ml), and the last is Maranta arundinaceae (5.49x107CFU/ml) while in the the Staphylococcus aureus, the highest population is in the Canna edulis (8.17x107CFU/ml), Maranta arundinaceae (5.13x107CFU/ml), Dioscorea esculenta (4.08x107CFU/ml), and the last is nutrient agar media (4.02x107CFU/ml). Based on the size of the bacteria, the best result is in the  Dioscorea esculenta, because the bacterial colonies are large and looks more clear. The results showed that the media of Canna edulis, Dioscorea esculenta, and Maranta arundinaceae can be used as an alternatives media for bacterial growth and the most optimal media is  Dioscorea esculenta.

Abstract

Fungi needs utrient, a source of energy and certain environmental conditions in order to growth. PDA was a common medium used to grow fungi in laboratories. The high of instant media PDA encourage researchers to find out alernatif media of the raw materials was easily available and more cheap. This researche determine the fungal growht in alternative media on carbohydrate source of the Canna edulis, Dioscorea aculeata and Maranta arundinacea. This study was an experimental studi using Completely Randomize Design (CDR), two factor. The first factor is a type of fungi: Candida albicans (J1), Aspergillus niger (J2). The second factor was the types of alternative media: the media of Canna edulis (M1),Discorea esculenta (M2) and Maranta arundinaceae L (M3). The collected data is analized by using qualitative descriptive. In this research Candida albicans was tested by spread method at 28oC for 2 days, Aspergillus niger by agar block method was tested at 30oc for 7 days. The result show that growth of Candida albicans best on medium Canna edulis with a population of 2,87×108CFU/ml  and the size of fungal colony was large white, while the growth of Aspergillus niger best on medium Canna edulis with a diameter about 39,7 mm and heavy sporulation. Both of fungi test showed that almost comparable growth of PDA. Media Canna edulis, Dioscorea aculeata and Maranta arundinacea can be used as  alternatives media for fungal growth. Keyword: fungal growth media, fungal, the source of carbohydrates, PDA.

Abstract

The purpose of this study was to determine the response of nodal explants and leaves of plants binahong on MS medium with the addition of 2,4-D with BAP variation. Research was designed using two factors: factor 1: types of explants (E1 = node, E2 = leaf),  factor 2: the concentration of plant growth regulator BAP (B0 = 2,4-D 2 mg / L (without BAP); B1 = 2, 4-D 2 mg / L and BAP 1 mg / L; B2 = 2,4-D 2 mg / L and BAP 2 mg / L). Explants nodes and leaves of plants Binahong sterilized using Bayclin 45 "sterile distilled water and then rinsed 3 times further incubated in a culture room. Parameters include callus formation time (Days After Planting / HST), size, and color callus. Additional parameters: the presence or absence of roots and shoots. Observations made during the 35 days and the result that the treatment of E1 (explants nodes) more quickly induced to form callus and callus size larger than E2 (leaf explants). B0 treatment induced to form roots (E1 and E2), while B1 forming buds at E1. The conclusion is the response of nodal explants Binahong better for the formation of callus on MS medium with the addition of 2 ppm 2,4-D and variations in the concentration of BAP (0, 1, 2 ppm) compared explants of leaves and the most excellent in the treatment E1B1 (explants nodes on MS + 2 ppm 2,4-D and 1 ppm BAP).Keywords: Binahong, Callus, 2,4-D, BAP
