Abstract

Polyethylene (PE) plastic is one of the most widely used for multiple purposes and leading the environmental problems. Lipolytic bacteria are promising agents to reduce the plastic waste. The dry weight of PE plastic was reduced by 33 % in the Winogradsky's column after 45 days of incubation. However, the lipolytic bacteria responsible for those reduction was unknown. This study aimed to characterize and identify the potential lipolytic bacterial isolates forming biofilm on polyethylene PE plastics. Samples of lipolytic bacterial isolates were screened on tributyrin selective media based on the formed clear zone. Moreover, the 16S rRNA genes of the two most potential lipolytic bacterial isolates were amplified. Then, the amplicons of the 16S rRNA gene were sequenced. This study found two potential lipolytic bacteria isolates, AB A-2 and AB M-3, which had the characteristics of round colonies, wavy edges, convex surfaces, and milky white color. The two isolates are gram-positive and have the shape of Coccobacillus cells. The molecular identification showed that AB A-2 isolate was Bacillus sp., while AB M-3 isolate was Bacillus amyloliquefaciens. This finding contributes to novel bacterial isolates that potentially overcoming the plastic waste problem.

Abstract

The need for potential bacteria as plant-promoting agents is still in high demand. Previous reports suggested five potential rhizobacteria isolated from the frangipani plant (Pulmeria accuminata) and producing indole acetic acid (IAA). However, evidence of its potential as a plant growth promoter has not been obtained. Therefore, this study aims to test the effect of rhizosphere bacterial inoculation on chili plants (Capsicum annuum L.). The method was a quantitative experimental method, with 15 repetitions for each treatment. The data were analyzed using a 95% student t-test confidence test or a significant level of 5% (a = 0.05). This inoculation begins with the germination of chili seeds until the radicula is observed. The germinated chili seedlings were soaked in bacterial suspension for an hour at room temperature. The results showed that inoculation using P38 isolate incubated for 6 hours significantly increased plant height and number of roots. The P31 isolate that was incubated for 6 hours inhibited the height of plants and the number of leaves of chili plants. Thus, these results provide important information about the effect of inoculation of newly isolated rhizosphere bacteria.

Abstract

Frangipani trees (Plumeria acuminata) are commonly found in cemeteries. Cemeteries are used as a location for interment so that the corpse actively decomposes to provide soil minerals that encourage the formation of microbes, including bacteria. Bacteria around the roots of frangipani trees are thought to have potential as plant growth-promoting rhizobacteria (PGPR). Based on reference searches, there has been no research on frangipani tree rhizosphere bacteria from burial grounds. The aim of this study was to investigate the phosphate-dissolving and blast disease-causing Pyricularia oryzae fungus-inhibiting properties of frangipani tree rhizosphere bacteria from Pracimaloyo cemetery, Surakarta. A total of 39 isolates of frangipani tree rhizosphere bacteria were tested for their ability to dissolve phosphates using PKA media (Pikovskaya), while the antagonism test against the fungal pathogen Pyricularia oryzae used the dual assay method. The results of the study showed that 15% of the rhizosphere bacteria of the frangipani tree were able to dissolve phosphate, and 33% of the isolates were able to inhibit the growth of the fungus Pyricularia oryzae with an inhibitory power of more than 40%. Based on the results, the rhizosphere bacterial isolates from Pracimaloyo TPU show the characteristics of a possible PGPR.

Abstract

A public cemetery (TPU) is land provided by the government to bury bodies. The culture of funerals in Indonesia is varied but generally involves being buried in the soil with wrapped clothes, covered wooden boards, and tombs. The material contains cellulose that will be broken down by cellulolytic microorganisms, including bacteria. The exploration of the cellulolytic bacteria of TPU in Indonesia has not been carried out, so this study aims to investigate and identify cellulose bacteria isolates from TPU. This research is a non-experimental study for the isolation of bacteria in the soil of TPU Bonoloyo, Surakarta. A selection of cellulite bacteria is grown in CMC media. Isolates that show cellulolytic activity are identified by observing colonial morphology and Gram coloring. Selection results of cellulolytic bacteria obtained isolates with cellulolytic index (IS) including a high category of 1 isolate (B14), a medium category of 3 isolates (B16, B27, and B45), and 9 low category isolates. The study concluded that 28.8 % showed cellulolytic activity with irregular morphological forms of white dyeing on flat surfaces with flat edges and dominated by gram-negative coccyx-shaped.

Abstract

In The Pracimaloyo Public Cemetery, the body’s decomposition occurs continuously, producing soil nutrients and minerals that affect the growth of bacteria. Bacterial population data from the cemetery area is still very limited, whereas cemeteries hold the risk of contamination with pathogenic bacteria. This research aims to find out how the population and diversity of bacteria in the Pracimaloyo Public Cemetery. Soil samples were taken from the Pracimaloyo cemetery at 2 locations (blocks 8 and 18), each at 20 and 50 cm depth. Soil samples are inoculated in Nutrient Agar media using the spread plate method. After 48 hours, colony counting, colony morphology, and gram staining observations were carried out. The rate of soil bacterial populations in blocks 8 and 18 at a depth of 20 cm was 4.23 × 107 CFU/g and 9.79 × 107 CFU/g, while at depths of 50 cm, it was 1.94 × 107 CFU/g and 1.92 × 107 CFU/g. The morphology of the bacterial colonies is dominated by circular shape, entire margin, flat elevation, and white color. 20 isolates are gram-negative and 16 isolates are gram-positive, the cell form is dominated by the bacillus.

Abstract

Public cemeteries are considered to be a potential source of groundwater pathogen contamination. Many settlements in Indonesia are located near cemeteries and rely on groundwater for consumption. However, there is no information available on whether the simple method of boiling can improve the microbiological quality of groundwater around cemeteries. Therefore, this experimental study aimed to determine the effectiveness of boiling in improving the microbiological quality of groundwater around Bonoloyo Cemetery. The study had two treatments: groundwater without boiling (control) and groundwater that had been boiled. Samples were collected from two points (S1 and S2) inside the cemetery and three points (S3, S4, and S5) outside the cemetery. The microbiological quality of groundwater was analyzed using the MPN method for each treatment. Samples that produced gas in positive tubes were inoculated on eosin methylene blue (EMB) media. The results of the study showed that S1, S2, S3, and S5 had total coliform levels that exceeded the normal threshold for water. Fecal coliform was found in S3 and S5. However, after boiling, there was an average decrease in total coliforms in samples S1, S3, S4, and S5. There was also no growth of E. coli in samples S3 and S5. The decrease in MPN values ​​in the samples indicates that boiling can improve the microbiological quality of groundwater at cemetery Bonoloyo Surakarta.

Abstract

Daun kelor (Moringa oleifera) dikenal kaya nutrisi, sementara kulit lemon (Citrus limon) memiliki senyawa antioksidan. Penelitian ini bertujuan untuk menghasilkan formulasi teh celup inovatif yang menggabungkan kedua bahan tersebut dengan pemanis alami stevia (Stevia rebaudiana) serta menganalisis karakteristik produk yang dihasilkan. Formulasi ini terinspirasi dari penelitian sebelumnya mengenai pembuatan teh celup daun kelor dengan stevia dan potensi antioksidan minuman serbuk lemon dengan variasi stevia. Metode penelitian meliputi eksperimentasi dengan lima perlakuan berbagai rasio daun kelor, kulit lemon, dan stevia, diikuti dengan pengujian karakteristik fisikokimia meliputi kadar air, abu, dan antioksidan. Hasil penelitian memberikan informasi mengenai formulasi teh celup kelor-lemon-stevia dengan karakteristik yang dapat diterima konsumen yaitu kadar air 9,41-10,26%, kadar abu 6,74-10,28%, dan pH 7,073-7,183. Simpulannya, penelitian ini berpotensi menghasilkan produk teh celup herbal yang inovatif dan bermanfaat bagi kesehatan.

Abstract

Indonesia faces dual environmental challenges from plastic pollution and agricultural waste accumulation. To address these issues, innovative solutions are needed to transform waste into sustainable materials. This study valorizes rice straw (Oryza sativa L.) cellulose and used cooking oil (UCO) glycerol to develop biodegradable plastics through an accessible method. Cellulose was extracted from rice straw via alkaline (10% NaOH) and bleaching (20% H₂O₂) treatments, while glycerol was recovered from UCO through saponification. Bioplastic films were synthesized by solution casting with varying glycerol volumes (6, 9, 12 mL) and cellulose particle sizes (fine, semi-fine, coarse). Results demonstrated exceptional biodegradability, with ≥ 80% weight loss within 14 days in EM4-supplemented soil burial tests outperforming commercial cassava-based bioplastics (25% degradation). Higher glycerol volumes (12 mL) combined with coarse cellulose maximized water solubility (69.39%) and absorption (14.29%), while lower glycerol (6 mL) with fine cellulose minimized water uptake (5.88%). All formulations exceeded thickness standards (0.438–0.554 mm vs. JIS ≤ 0.25 mm), enhancing rigidity but reducing flexibility. Organoleptic analysis confirmed glycerol content inversely affected transparency and texture smoothness.

Abstract

Original "SuleMu" soy milk currently has limited shelf life without refrigeration. Therefore, it is crucial to assist partners in enhancing product competitiveness and diversification. The primary objectives of this community engagement activity were to improve SuleMu's marketability through the addition of new flavors and the provision of nutritional value analysis, as well as to enhance product quality by developing derivative products such as soyghurt and frozen yogurt. The activity stages included: (1) mentoring for the diversification of SuleMu derivative products, (2) nutritional value analysis of derivative products, (3) designing product labels compliant with standards, and (4) conducting a market survey on consumer preferences for SuleMu derivative products. Results indicate that SuleMu is now available in new flavors: melon, chocolate, and strawberry. The nutritional content per 100 grams is 3.4–3.5 grams of soluble protein, 3.3–3.7 grams of fat, 5–5.7 grams of reducing sugars, 2.6–3.6 grams of crude fiber, and approximately 39–40 mg of calcium. SuleMu production increased from 5 kg of soybeans to 6 kg per day. Furthermore, consumers showed high interest in the "Soyoghurt" frozen yogurt derivative product, with 42.4% being very interested and positive evaluations regarding taste (36.1%) and affordable price (29.2%). Overall, this community engagement activity successfully increased SuleMu's production by 17%, and soyghurt demonstrates significant potential for development as a highly competitive new innovation.
