Abstract

Penelitian ini bertujuan untuk mengungkapkan hirarki dan diferensiasi  genetik tanaman sagu berdasarkan penanda molekuer RAPD. Keragaman genetik tanaman diungkapkan dengan menggunakan 10 jenis primer RAPD yang diamplifikasi dengan menggunakan mesin PCR. Hasil penelitian menunjukkan bahwa ditemukan sebanyak 86 lokus dari 10 primer RAPD yang digunakan pada 100 individu sampel tanaman sagu. Hirarki genetik tanaman sagu menunjukkan bahwa pada level antar individu dan populasi berbeda sangat nyata, sedang pada level pulau berbeda nyata. Diferensiasi genetik sampel tanaman sagu menunjukkan bahwa populasi tanaman sagu mengalami diferensiasi berdasarkan nilai probabilitas Fst dan exact test (chisquare). Populasi dari Pontianak berbeda nyata untuk semua populasi begitu pula populasi dari Selat Panjang berbeda nyata dengan semua populasi kecuali populasi dari Bogor. Hasil kalkulasi diferensiasi genetik menunjukkan bahwa sampel telah mengalami diferensiasi pada level individu, populasi. Berdasarkan observasi, tiap-tiap populasi sagu di tiap-tiap pulau perlu dipertimbangkan sebagai sumber plasma nutfah untuk program kegiatan koleksi plasma nutfah sagu.

Abstract

The essential oil of Banda nutmeg was extracted using hydro-distillation method to characterize their essential oil and volatile components.  Banda, Ambon, and Luhu nutmegs were chosen as sample ecotypes. Nutmeg oils were subjected to physico-chemical and GC-MC analyses.  Results indicated that nutmeg from the three ecotypes produced uncolored oils with the contents in mature seed were 11.69, 11.92, and 9.99%, respectively. Meanwhile those of immature seeds contained 13.32, 11.99, and 11.03% respectively.  Furthermore, the physico-chemical of the oil are specific gravity 0.897 to 0.909 g/ml; refraction index, 1.489 to 1.491; and optical rotation, +11.40 to +16,30.GC-MS analysis suggested that essential oils Maluku nutmegs composed of 28 to 31 components and also showed that nutmeg from Banda comprised 52.8% monoterpene hydrocarbon (MH), 21.11% oxygenated monoterpene (OM), and 18,04% aromatic compound (AC); Ambon’s 45.12% MH, 24.51% OM, and 16.97% AC; and Luhu’s 56.06% MH, 27.34% OM, and 13.62% AC.  Further analysis indicated that there were four important volatile oils fractions in nutmegs i.e. myristicin, elemicin, safrole, and eugenol. Maluku nutmeg contain 5.57 to 13.76% myristicin and 0.97 to 2.46% safrole. In conclusion, nutmeg oil shows a high stability in all physico-chemical properties.  Nutmeg from Banda ecotype has the highest content in myristicin.

Abstract

Pemerintah Kabupaten Sukoharjo membentuk program corporate farming di Desa Dalangan, Kecamatan Tawangsari, Kabupaten Sukoharjo sejak tahun 2017 sebagai contoh pengembangan pertanian modern di pedesaan. Namun demikian, program corporate farming tersebut belum berjalan terus menerus karena terkendala modal dan memiliki ketergantungan yang tinggi terhadap keputusan para stakeholder. Tujuan penelitian ini adalah: (1) mengidentifikasi persepsi petani terhadap program corporate farming; (2) mengidentifikasi dan menganalisis tingkat kepentingan dan pengaruh stakeholder corporate farming di Desa Dalangan, Kecamatan Tawangsari, Kabupaten Sukoharjo; (3) menganalisis dampak penerapan corporate farming terhadap pendapatan petani anggota dan bukan anggota corporate farming; dan (4) menganalisis tingkat pengaruh faktor-faktor karakteristik petani terhadap keputusan mengikuti corporate farming. Metode analisis data yang digunakan yaitu analisis deskriptif, analisis stakeholder, analisis pendapatan R/C ratio dan B/C ratio, dan analisis regresi binomial logistik. Hasil penelitian menunjukkan bahwa sebagian besar responden petani setuju bahwa pelaksanaan corporate farming di Desa Dalangan dapat meningkatkan kinerja usahatani, namun corporate farming belum mempermudah akses petani terhadap permodalan gapoktan. Berdasarkan analisis stakeholder, Dinas Pertanian Kabupaten Sukoharjo dan Gapoktan Tani Mandiri memiliki tingkat kepentingan dan pengaruh tertinggi dalam pelaksanaan corporate farming. Berdasarkan analisis pendapatan, nilai R/C ratio dan B/C ratio atas biaya tunai pada petani anggota dan bukan anggota corporate farming lebih dari 1, sehingga usahatani pada kelompok menguntungkan dan layak diusahakan. Faktor-faktor yang memengaruhi keputusan petani mengikuti corporate farming yaitu jumlah tanggungan keluarga dan pendapatan usahatani. Abstract

Penyakit layu fusarium yang disebabkan oleh Fusarium oxysporum merupakan salah satu penyakit penting pada bawang merah (Allium ascalonicum). Sebagian besar petani menggunakan umbi sebagai benih dan diketahui bahwa beberapa patogen dapat terbawa oleh benih seperti Fusarium oxysporum. Oleh karena itu diperlukan pengujian kesehatan benih untuk mencegah penyebaran penyakit tersebut. Salah satu metode sederhana dan efektif untuk digunakan di laboratorium adalah blotter test. Namun beberapa strain F. oxysporum terbukti tidak bersifat patogenik, serta tidak dapat dibedakan secara morfologi dengan strain yang bersifat patogenik. Penelitian ini bertujuan menentukan parameter uji dan jumlah sampel minimal dalam deteksi Fusarium oxysporum pada umbi bawang merah dengan metode blotter test. Hasil pengujian menunjukkan bahwa parameter nekrosis pada basal plate umbi bawang merah mempunyai koefisien korelasi (r) sebesar 0,77 terhadap tingkat infeksi pada growing on test (GOT) dan lebih besar dari tingkat infeksi fusarium (0,34) pada blotter test. Dari 195 isolat Fusarium spp. yang diuji, diketahui bahwa sebagian besar isolat bersifat nonpatogenik. Penentuan jumlah minimal umbi dengan plot kurva rerata jumlah nekrosis pada basal plate dan standar deviasi, menunjukkan jumlah sampel umbi minimal untuk blotter test adalah 150 umbi. Perhitungan jumlah sampel dengan formal probability statement yang menunjukkan jumlah umbi minimal untuk blotter test adalah 138 umbi.

Abstract

The Effect of Giberelin Acid (GA3) Application to Seed Yield of Hybrid Rice. Seed yield of rice hybrid on three line system has weakness in panicle exertion and outcrossing that cause low yield.  GA3 has been proved to increase the seed set through increasing panicle exertion and improved outcrossing. The purpose of this study was to determine the effect of GA3 application to increase seed yield of hybrid rice (F1). The study was conducted in Singamerta Experimental Farm, Banten Assesment Institute for Agricultural Technology from May to October 2013. The experiment was arranged in a split plot designed with four replications. The main plot were parental lines (cytoplasmic male sterility/CMS and restorer/R) of hybrid i.e. HIPA 8 (A1 and BP51-1), HIPA 6 (A2 and B8094), HIPA Jatim 3 (A6 and PK88) and HIPA 14 SBU (A7 and BH33d-Mr-57-1-2-2). Sub plot were the  frequency of GA3 treatments i.e. control (W0), two (W1), and three (W3) times application of GA3. GA3 dosage was 200 ppm, the best dose from the previous studies. The results showed that the GA3 applications increased plant height, stigma exertion, panicle exertion, duration of floret opening and panicle length. Two times application (W1) of  GA3  gave the best results as indicated by the highest seed yield (1429 kg/ha) that was better than three times application of GA3 that gave seed yield 1215 kg/ha and control with yield of 703 kg/ha.Keywords : GA3, hybrid rice, seed yieldABSTRAKHasil benih padi hibrida sistem tiga galur memiliki kelemahan, yaitu rendahnya eksersi malai dan tingkat penyerbukan silang,sehingga hasilnya rendah. Percobaan terdahulu menunjukkan bahwa aplikasi GA3 dapat meningkatkan hasil melalui peningkatan eksersi malai dan penyerbukan silang. Penelitian ini bertujuan untuk mengetahui pengaruh frekuensi aplikasi GA3 terhadap peningkatan hasil benih padi hibrida (F1). Penelitian dilakukan di Kebun Percobaan Singamerta, Balai Pengkajian Teknologi Pertanian (BPTP) Banten pada Mei sampai Oktober 2013. Penelitian menggunakan rancangan petak terbagi dengan empat ulangan. Petak utama yaitu tetua padi hibrida (galur mandul jantan/GMJ dan restorer) yaitu HIPA 8 (A1 dan  BP51-1), HIPA 6 (A2 dan B8094), HIPA Jatim 3 (A6 dan PK88) dan HIPA 14 SBU (A7 dan BH33d-Mr-57-1-2-2). Anak petak ialah tiga taraf frekuensi penyemprotan GA3 terdiri atas : kontrol (W0), dua kali aplikasi (W1) dan tiga kali aplikasi (W2). Dosis GA3 yang digunakan adalah 200 ppm yang merupakan konsentrasi terbaik dari hasil penelitian terdahulu. Hasil penelitian menunjukkan bahwa aplikasi GA3 mampu meningkatkan tinggi tanaman, eksersi malai, eksersi stigma dan durasi bunga membuka dibandingkan kontrol. Hasil yang terbaik dicapai pada perlakuan GA3 dengan dua kali aplikasi (W1). Hasil benih  pada perlakuan W1 mencapai 1429 kg/ha, lebih tinggi dibandingkan dengan aplikasi tiga kali GA3 (1215 kg/ha) dan kontrol (703 kg/ha).Kata kunci : GA3, padi hibrida, hasil benih

Abstract

Disease incidence of Stewart’s wilt on the seed and response of several maize varieties to Pantoea stewartii subp. stewartii. Stewart’s wilt disease of maize is caused by Pantoea stewartii subsp. stewartii. This bacterium is seed-borne pathogens, when attacked maize caused yield lost 40-100%. The objective of this research was to detemine the incidence level of stewart’s wilt disease, growth of some varieties of maize and their response to stewart’s wilt pathogens Pantoea stewartii subsp. stewartii. The research was conducted in the Laboratory of Bacteriology and Greenhouse Cikabayan IPB from November 2011 to March 2012. In experiment I, nineteen samples of maize were used for symptom test in the maize seedling stage, using Randomized Block Design with three replications. Experiment II used a Randomized Block Design with 2 factors: maize varieties (8 hybrids varieties, 3 open pollinated varieties, and 7 sweet corn varieties) and bacteria isolates (BGR 2, BGR 4, BGR 28, BGR7 and PSM 27), with three replications. The results showed in experiment I, the incidence of stewart’s wilt disease ranged 2.00 – 15.33%, germination and vigor index of maize seed were 68.00 – 95.33% and 55.33 – 90.67% respectively. While in experiment II, hybrid and open pollinated of maize varieties were resistant to moderately susceptible while all sweet corn varieties were susceptible to infection of Pantoea stewartii subsp. stewartii.

Abstract

ABSTRACT Sugarcane (Saccharum officinarum L.) is one of importance crop grown in marginal in Indonesia. Phosphorus (P) is critical to the growth and development of plant in the marginal land. P is stored in plant  as phytic acid (myo-inositolhexakisphosphate).  Phytic acid is hydrolyzed by the activity  of phytases to yield inositol and free phosphate. Genetic transformation of sugarcane with phytases gene holds promise to provide enough P during period of rapid cell division and growth of plant.  Plant transformation mediated by Agrobacterium tumefaciens, has become the most used method for the introduction foreign genes into plant cells and the sub sequens regeneration of transgenic plant. The selection and regeneration of embryogenic callus of transformed plant was done on MS medium containing kanamycin. The main objective of this study were: (1) To find the best kanamycin  concentration for selectable marker; (2) Insert phytase gen into  varieties of sugarcane (cv. PA 175) through Agrobacterium tumefaciens GV 2260 (pBinPI-IIEC); and (3)  To analyze of intregated transgene into genom of sugarcane using PCR  method.  Result of the experiment showed: (1) kanamycin selectable marker lethal doses for transformed sugarcane calli: 100 mg l-1;   (2)  Efficiency transformation of putative transgenic line  was cv. PA 175= 24 %; (3)  The first culture of transformed calli become 24 (Triton cv.), 18 (PSJT 94-41 cv.), and 30 (PA 175 cv.) putative plants; the second sub cullture of putative eksplant regenerate become new plant: 380 (PA 175 cv.) plants.  (4) Analyzed of integrated phytase gene was proven by appearance of 900 bp of PCR band (5) transgenic plants (cv. PA 175)  with highest activities respectively: 45 %; with medium phytase activity: 27 %, and low phytase activity: 27 % from total of sample.  Non transgenic plants, most of sample show low phytase activity respectively:  100 % , noneh show medium - hight activy of phytase.  Key words: Sugarcane, transformation, phytase,  Agrobacterium tumefaciens

Abstract

Benih kedelai memiliki daya simpan yang rendah karena penurunan kualitas benih selama penyimpanan berlangsung lebih cepat dibandingkan benih tanaman lain. Kedelai setelah dipanen akan mengalami penurunan benih baik secara kualitatif maupun karena beberapa faktor eksternal. Pengadaan benih sering dilakukan beberapa saat sebelum musim tanam sehingga benih harus disimpan dengan baik agar memiliki daya tumbuh yang tinggi saat ditanam kembali. Pengemasan benih merupakan tindakan penyediaan lingkungan mikro yang optimal agar benih tidak dipengaruhi oleh faktor lingkungan selama penyimpanan. Penyimpanan benih kedelai dilakukan di Gudang Pusat Perbenihan, Leuwikopo IPB mulai April 2018 hingga Juli 2018. Penelitian ini mempelajari pengaruh jenis kemasan dalam mempertahankan umur simpan benih selama penyimpanan. Jenis kemasan tidak berpengaruh terhadap perkecambahan benih dan kadar air, tetapi berpengaruh terhadap potensi tumbuh maksimal dan bobot kering kecambah normal, menggunakan benih yang dikemas menggunakan jerigen plastik dan plastik kedap udara memiliki potensi tumbuh maksimal yang lebih tinggi dibandingkan kertas semen. dan kantong plastik selama umur simpan 4 bulan. Benih yang disimpan selama 1 tahun dalam 4 kemasan mengalami penurunan viabilitas yang drastis, meskipun disimpan pada suhu 17-19° C dan RH 53-55%.

Abstract

Bambara groundnut is a legume plant that has several advantages, including droughttolerance, has ability to grow on infertile land and high nutrient content. The production ofbambara groundnut is still low, so it is necessary to obtain high yielding bambara groundnut line.The experiment was conducted to evaluate the genetical variation and agronomic characters of 30bambara groundnut lines. This experiment was carried out in experimental field of SEAMEOBIOTROP Tajur Bogor (± 280 m above sea level), on Februari-June 2018. Plant materials used inthis research were 30 bambara groundnut lines derived from pure line selection of Sukabumilandrace and unselected Sukabumi landrace uses as control. The experiment was arranged in arandomized complete block design with three replications. Results showed that there werevariations among several agronomic characters of bambara groundnut lines. Several lines showedsuperior characters. R59.30, A90.8, A41.4, A56.10, A43.5, A103.5, A41.4, A55.8, R59.31 andA28.10 lines were lines with the higest dry pods weight compared to the other lines and controlline. These lines had potential as new superior varieties of bambara groundnut.Keywords: landrace, new superior variety, pure line selection

Abstract

Demostration rice plot of IPB 3S and IPB 9G varieties were at Mekarharja village, Purwaharja district, Banjar city. The demostration plot area of one hectare is spread over three hamlets including Randegan, Cibentang, and Pasirleutik hamlets. Stasiun Lapang Agro Kreatif (SLAK) program aims to develop IPB 3S and IPB 9G varieties of rice in an effort to increase farmer resources and rice production. Application of IPB 3S dan IPB 9G rice cultivation technology through training and field assistance to farmers. Farmers are given training and assistance to utilize cow dung and rice straw, seed treatment and tillage technology, and balanced fertilization technology. There are farmers’ knowledge increases about the use of cow dung and rice straw to increase rice production, and balanced seed treatment and fertilization. Farmers can apply cultivation technology easily because the materials used are easy to obtain and simple technology implementation procedures. The application of rice cultivation technology that has been carried out by most farmers in the village of Mekarharja. Farmers’ knowledge increases about the management of agricultural and livestock waste which can be used as organic fertilizer. The submitted science and technology was accepted and applied well by the farmers.
