Abstract

Specific changes in morphology, anatomy, biochemistry and molecular occur due to changes in light intensity. Changes in growth is closely related to the mechanism of plant resistance when low light conditions. In addition, the environmental stress conditions of supply nutrients plants need in order to remain able to grow optimally. This experiment was conducted in experimental garden Sawah Baru, Post-harvest Laboratory and Micro-engineering Laboratory Bogor Agricultural University, beginning in December 2014 until July 2015. This experiment was aims to determine the effect of shade and fertilization on the growth and change of morpho-anatomy of leaves jack bean. The experiment was arranged in a split plot design based on randomized complete block design. Shade as the main plot that consists of three levels: without shade, shade 25% and 50% and combination doses of fertilizer as a subplot which consists of three levels ie: 50 kg urea ha-1 + 100 kg SP-36 ha-1 + 75 kg KCl ha-1, 75 kg urea ha-1 + 125 kg SP-36 ha-1 + 100 kg KCl ha-1, 100 kg urea ha-1 + 150 kg SP-36 ha-1 + 125 kg KCl ha-1.The results showed increased percentage of shade caused etiolation, reduction number of branches, change the leave size, depletion of palisade tissue which decreases the thickness of the leaves, and decreased of pod/plant.Whilefertilizationfactoronlyinfluence onthe thickleaves andthickpalisade.

Abstract

  Characterization is important in breeding program and developing superior varieties of jatropha.  Jatropha characterization can be analyzed based on its morphological and agronomical characteristics.  In this study, a molecular marker, Random Amplified Polymorphic DNA (RAPD), was applied for cluster analysis.  This study was aimed at analyzing character and similarity of 30 jatropha germplasms of SBRC-IPB collection. Results showed that 30 jatropha accessions have high character variability.  Dendogram of RAPD marker showed that at 33% similarity level, the 30 jatropha accessions could be classified into three main groups.  It was also shown that Papua and China accessions had similarity levels higher than 80%.  Based on production characteristic identification, the potential accessions to be developed further included Palembang, Pontianak, Pidi, Palembang I, Pagar Alam, Medan, Curup, Lampung I, Lampung II, Komering, and Indralaya.   Key words:  Jatropha curcas L., characterization, cluster analysis, morphological marker, RAPD marker

Abstract

This research was aimed to develop selection criteria based on correlation and path coefficient analysis.  The experiment was carried out at CETROFS (Center for Tropical Fruit Studies) field station of Bogor Agricultural University, Tajur.  The result showed that plant height, pseudostem diameter, number of leaves, weeks to harvest,  finger number per plant, and mean of  finger weight were positively correlated with bunch weight. Pseudostem diameter,  number of leaves, finger number per plant have strong direct effect on bunch weight,  with path coefficient 0.321, 0.264, 0.297, and 0.722, subsequently.  However, the plant height showed strong indirect effect on bunch weight  through  pseudostem diameter. These characters  are useful as a selection criteria for developing new variety of banana  with higher bunch weight   Key words: path coefficient, selection criteria, banana

Abstract

Banana accessions introduced from INIBAP Transit Center (ITC) have been studied their diversity revealed on phenotypic and RAPD markers.  Both of them showed different clustering pattern and not fit to genomic composition. In order to know fitness level of clustering pattern both of them, comparison analysis have done to their similarity coefficient matrix, and followed by partial correlation analysis among phenotypic traits, and genomic composition.  The comparison analysis resulting very poor correlation (r = 0.491). Partially correlation analysis among traits, DNA profile, and Musa genomic composition at 95-99% confidence revealed only OPA-18 line 2 related to accessions possessed pure "A" genome, and they were together associated to green color of petiole margins.  OPD-10 line 3, significantly, associated to accessions with genome dominated "B", but there was no correlation between both of them to any characters. Abstract

Laboratory and field experiments were carried out to analyze genetic diversity of 22 pepper genotypes (Capsicum spp.) and their resistance to Phytophthora capsici Leonian. Resistance screening was performed in plastic flats 72 cells. Inoculation was done on 28-day old pepper plant soon after watering by pipeting 5 ml of inoculum (105 zoospore/ml) at the base of each plant. P. capsici  isolate used in this experiment was TG01, identified as race 3 based on AVRDC differential pepper lines (PI 188478, PBC 602, PBC 137 and Early Calwonder).  The evaluation of pepper genotypes characteristic was conducted in the field.  Principle Component Analysis, Clustering Analysis and Biplot Analysis were used to analyze genetic diversity based on 37 characters. Result of resistance evaluation showed that two genotypes (C4 and C13) were identified as resistant, 7 genotypes (C2, C3, C5, C8, C10, C15, and C20) as slightly resistant, 6 genotypes (C7, C9, C17, C19, C21 and C27) as slightly susceptible, and 7 genotypes (C1, C11, C18, C28, C48, C64, and C65) as susceptible.  Based on genetic diversity analyzed, all genotypes could be divided into 4 clusters.  Cluster I consisted of 18 genotypes i.e. C1, C2, C3, C4, C5, C7, C8, C9, C10, C13, C14, C15, C17, C18, C19, C28, C64, and C65. Cluster II consisted of only 1 genotype i.e. C48.  Cluster III consisted of 2 genotypes (C20 and C21) which were characterized by the colour of corolla, corolla spot, and filament. Cluster IV consisted of 1 genotype (C27) which was characterized by fruit cross-sectional corrugation.   Key words :  Capsicum, resistance, Phytophthora capsici, genetic diversity, clustering.

Abstract

The objective of this research was to study the effect of seed coating using Benomil and curcuma powder on seedborne diseases of  antrachnose and seed viability of Capsicum annuum L.. This research was conducted in Laboratory of Plant Mycology and Laboratory of Seed Science and Technology IPB, from June to July 2006. This research consisted of three experiments, i.e.: (1) the effectiveness of seed coating in decreasing infection level of antrachnose fungus in the seed of chilli, (2) the effect of seed coating on seed viability, (3) the effect of seed coating in reducing infection level of antracnose at hypocotyl of chilli.  The Experiment 1 and 3 used Randomized Complete Design and the experiment  2 used Randomized Complete  Block Design. The result of this research showed that seed coating with Benomil and curcuma powder significantly decreased the infection  level of antrachnose fungus on seed and hypocotyl (Benomil 2.5 g/l and curcuma powder 1 g/l). However, seed coating with curcuma 1 g/l had the lowest  vigour index, seedling growth rate and seed germination (3%, 2.26%, 23%). Seedcoating with Benomil 2.5 g/l and curcuma powder did not significantly affect the maximum growth potential, seed germination, and vigour index.  However, Benomil significantly decreased  the seedling growth rate.   Key wods : chilli, seed coating, antrachnose, curcuma, benomil, viability

Abstract

The objective of this research was to study and cluster of Jatropha germplasm belonging to University of Sriwijaya. This research was conducted from September 2007 until July 2008. The research used 14 accessions of Jatropha taken from some regions in Indonesia, namely: Komering, Palembang, Yogyakarta, Indralaya, ATP2, Pontianak, Lahat, Pagaralam, Curup, Lampung, Medan Aceh Besar, Pidi and Gorontalo. Accessions of Jatropha curcas L. were planted at Agro Techno Park (ATP) Bakung village, Indralaya Utara district Ogan Ilir, South Sumatera using Randomized Complete Block Design. RAPD analysis using 20 primers was done at RGCI (Research Group on Crop Improvement), Bogor Agricultural University. Dendrogram based on RAPD analysis produced five groups that were: the first group was Komering, Lahat, Pidi, Indralaya, Aceh Besar, Pontianak and Curup. The second group was Palembang and ATP2. The third group was Pagaralam, Gorontalo, and Medan. Lampung was included in to fourth group. The fifth group was Yogyakarta. Dendrogram from morphological marker had also five groups. First group was: Komering, Indralaya, Pontianak. Lahat, and Pagaralam. Second group was: Palembang, Lampung, Pidi, Medan, and ATP2. Third group was: Curup. Fourth group was: Yogyakarta and Gorontalo. Fifth group was: Aceh Besar. The difference of member from each groups between dendrogram using RAPD and morphological markers indicated that the bands resulted from RAPD did not have relation with characters observed. A complete genetic map, including genetic markers for all regions of all the chromosomes in a genome, serves .as framework,for determining the location of genes responsible for variat!on in plant growth and development. Strategies for mapping both discrete and contino us phenotypes are well - established as are efficIent experlmental approaches which minimize the time and cost of genetic mapping. GenetIc map represents a powerful tool In studying the basIs of variation in plant growth and development, and in testing hypothesis. about possible candidate genes. Fifty seven individuals of recombinant inbred lines (RlLs) of faba bean (Viciaf aba L.) descended from the cross between 34Morocco x Kristal125 have been analyzed for RAPD markers. Abstract

The objective of this experiment was to determine the effect of triacontanol on cucumber production and viability. A comletely randomized design was used, consisting two factors i.e. concentration and time of application. Concentration consists of 5 levels: 0, 0.01, 0.1, 1.0, and 10.0 mg/l while time of application consists of 3 leveles: 2 weeks after planting (W AP), 4 W AP, and 2 & 4 W AP. Concentration, time of application and interaction of the two factors were not significant on vegetative and reproductive variables but were significant on seed weight index, and seed variability. Concentration of O.O1 mg/l and application of 4 WAP gave the best effect on seed viability. (p.11)   Key words: Triacontanol, Seed, Cucumber
