Resistance screening were conducted on 28-days-old pepper plants grown in 72-cell fl ats by inoculating 5 mL of inoculum (contain 105 zoospore mL-1) to the base of each plant. Phytophthora capsici isolate used in this experiment was TG01, which then identifi ed as race-3. The evaluation of pepper genotypes characteristics were conducted in fi eld. Eight characters demonstrated a wide genetic variability; those characters were resistance to Phytophthora blight, infection period, dicotomous height, canopy width, fruit weight, fruit length, fruit diameter and production. These characters along with fruit wall thickness, chlorophyll-a, chlorophyll-b, total chlorophyll had high heritability values. Using path analysis infection period, dicotomous height and fruit diameter are recommended to be simultaneously used as selection criteria for developing Phytophthora blight resistant pepper varieties. To get more information about its ripening off, the tree characteristics a respiration pattern was determined. The data were collected on fi ve trees for each ten selected accessions (n=50) for fruit ripening uniformity on its trees study and fi fteen bunches for each seven selected accessions (n=105) for fruits bunches ripening uniformity study at two jatropha pilot projects at Sabah state of Malaysia at the end of March 2009 when the trees were exactly one year old. This study confi rmed that heterogeneous ripening occured in all jatropha accessions and within individual jatropha bunches. Respiration tests confi rmed that jatropha is a climacteric fruit. The results showed an upsurge in CO2 production at the end of ripening and at the beginning of senescence. This study has revealed indications of the possibility to harvest jatropha fruit in bunches rather than harvesting individual ripe fruit, which could potentially improve harvest effi ciency by reducing harvesting time and drudgery. Abstract

Agronomic performance and heterosis effect of progeny are important criteria for selection of parental hybrid. Theagronomic performance evaluation of Jatropha progeny which was derived from crossing of some Jatropha accessions hasbeen carried out for those goals. The study was conducted at Agro Techno Park, Indralaya South Sumatra. The crossingcombinations were Pidie x Medan, Curup x Lampung, Pidie x Lampung, Lampung x Pidie, Curup x Medan, Indralaya xPontianak, Medan x Palembang, Curup x Pidie, and Jogya x Komering. Oil production, nitrate reductase enzyme activity,leaf chlorophyll content and relative plant growth rate were evaluated. The result showed that the relative plant growth ratecould be used as early detection parameter for yield potential. Progenies of Pontianak x Indralaya, Palembang x Indralayaand Pidie x Lampung were selected as promising hybrids based on high productivity and heterosis effect.Keywords: Jatropha curcas, heterosis, relative plant growth rate, nitrate reductase activity

Abstract

The objective of this research was to improve seed vigor, plant growth, and productivity of black soybean through seed invigoration treatment. The experiment was arranged in split-plot design. As the main plot, there were two black soybean varieties (Detam-1 and Detam-2) and as the sub-plot there were seed invigoration treatments applied (untreated control, matriconditioning, biofertilizer, matriconditioning plus biofertilizer). Matriconditioning was conducted using ratio of seeds to carrier (rice hull charcoal) to water of 9:6:7 (w/w/v) for 12 h in ambient room. For biofertilizer treatment, biofertilizer (consists of nitrogen-fixing bacteria, phosphate-solubilizing bacteria, fungicide-producing bacteria, and growth-regulator-producing endophytic bacteria) was applied just before planted by mixing seeds with 6.25 g biofertilizer (kg seeds)-1 and small amount of water. Matriconditioning plus biofertilizer was conducted by integrating the biofertilizer in matriconditioning. Results of the experiment showed that matriconditioning or matriconditioning plus biofertilizer improved germination percentage and rate of germination. Biofertilizer or matriconditioning plus biofertilizer improved vegetative growth. Although there was no significant effect of invigoration treatment on productivity, biofertilizer treatment produced seeds 15% higher than control, and matriconditioning treatment produced seeds 13% higher than control. Viability and vigor of harvested seeds were not affected by invigoration treatments, however, Detam-2 had higher viability and vigor than Detam-1 based on germination percentage, index vigor, and rate of germination. Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh perlakuan benih untuk meningkatkan mutu dan produksi benihserta mengendalikan penyakit bulai pada jagung manis. Galur jagung manis 06 digunakan pada percobaan di rumah plastikyang disusun berdasarkan rancangan acak lengkap satu faktor, yaitu 13 perlakuan benih. Percobaan di lapangan disusunberdasarkan rancangan petak terbagi dengan dua faktor. Faktor pertama merupakan galur jagung yang terdiri atas duagalur yaitu galur 06 dan 07, dan faktor kedua merupakan perlakuan benih yang terdiri atas 7 perlakuan. Matriconditioningmenggunakan rasio benih : arang sekam : pelarut = 3:0.5:1 (g) dalam botol tertutup dan ditempatkan pada ruangan bersuhu20±2 C selama 24 jam. Fungisida sintetik yang digunakan mengandung metalaksil dan dimethomorf. Agen hayati yangdigunakan adalah Bacillus megaterium dan Brevibacillus laterosporus. Hasil percobaan di rumah plastik dan lapanganmenunjukkan bahwa perlakuan fungisida sintetik paling efektif menekan penyakit bulai. Percobaaan di lapangan menunjukkanbahwa galur 07 lebih tahan terhadap penyakit bulai dibanding galur 06. Perlakuan matriconditioning + fungisida sintetik+ B. laterosporus pada galur 07 menghasilkan bobot tongkol per tanaman paling tinggi. Perlakuan matriconditioning +fungisida sintetik + B. laterosporus pada galur 06 mampu meningkatkan mutu fisiologis benih hasil panen.Kata kunci: Bacillus megaterium, Brevibacillus laterosporus, matriconditioning, Peronosclerospora maydis

Abstract

ABSTRACTThe recent scarcity of fuel has stimulated many efforts to remove dependence on petroleum oil by finding alternative sources of new and renewable energy. Jatropha curcas L. has been chosen as a source of biofuel that has an important role as a petroleum oil substitute, especially for producing biodiesel. Possible efforts for the development of Jatropha in Indonesia include exploration of Jatropha plants from various regions of Indonesia, introducing the plants from abroad and plant breeding. The purpose of this study was to study the genetic parameters of morpho-agronomic traits and selection of potential genotype for genetic improvement of Jatropha curcas. The experiment was conducted at the Biotech Center, BPPT from November 2011 to July 2012. The experiment was performed using a complete randomized block design with sixteen genotypes of J. curcas. Five plants were used for each genotype, with three replications. Results of this study showed that genetic diversity of the genotypes based on morpho-agronomic characters was relatively narrow, with genetic diversity coefficient ranged from 2.73% to 9.02%. The broad-sense heritability was high for all characters, ranged from 32.26% to 85.89%. None of morpho-agronomic characters observed in the 16 genotypes was directly correlated to productivity and seed oil content. Number of fruit and total seed number per bunch were the best characters for selection criteria to obtain high dry seed weight. Based on the bi-plot analysis on seed oil content and dry weight characters among the observed genotypes, LMP and CRP x PDI could be recommended as potential F1 parents to be used in constructing F2 population.Keywords: biplot analysis, broad-sense heritability, genetic diversity coefficient

Abstract

Increased productivity of maize can be done with the use of high quality seeds from improved varieties such as hybrid seed. The objectives of this study were Increasing productivity of maize female parent is important in order to reduce the price of hybrid seed. The objectives of this study were to determine the nitrogen fixing bacteria compatible with phosphate solubilizing bacteria (PSB), and using those bacteria to increase physiological seed quality and seedling growth of maize female parent. The research consisted of laboratory and field experiments. Laboratory experiment for the isolation and identification of rhizobacteria resulted in 25 Azotobacter and 29 Actinomycetes non-pathogenic isolates capable of fixing nitrogen and PSB selected for compatibility tests were AB3, B28, P12, P14, P24, and P31. The compatibility test showed 25 pairs of BPF with Azotobacter and 16 pairs of BPF with Actinomycetes were mutually compatible. The BPF pair with Azotobacter or Actinomycetes P24-AzL7, P24-AzL9, B28-AcCKB4, P24-AcCKB9, P24-AcCKB20, and P24-AcCKW5 were able to increase the vigor index of hybrid maize female parent seed. Field experiment was arranged in a split plot design with three replications. The main plot was dosage of N-P fertilizer (0%, 25%, 50%, 75%, and 100% of recommendation dosage), and the subplot was 12 rhizobacteria treatments selected from 25 compatible pairs of BPF with Azotobacter and 16 pairs of BPF with Actinomycetes and 1 control. The application of compatible pairs of bacteria had a significant effect on plant height, the number of leaves at 3 and 4 weeks after planting and plant dry weight. However, the best treatment i.e. B28-AcCKB4 was not significantly different with the nutrient broth treatment (as control).

Abstract

Nutmeg is an Indonesian native plant, produces essential oill as an export commodity. However, it has low seed uniformity that mengurangi keseragaman tanaman di lapang. The main objective of this research was to obtain morphological characteritics as parameter for nutmeg mother plants selection. This field research was carried out from August-December 2014 at Toisapu, Wakal and Lula nutmeg seed source plantations in Ambon, and Seed Storage and Testing Laboratory, IPB. Characterization was undertaken on ten mother plants from each  Wakal and Lula seed source plantations and eight mother plants from Toisapu seed source plantation. Quantitative and qualitative characters of vegetative, fruit, mace and seed from every mother plants were analyzed with  Gower’s similarity test. Mother plants that produced high uniformity seed was the 7th (0.80), the 8th (0.93) and the 10th (0.80) mother plants in Wakal; the 2nd (0.80) mother plant in Toisapu; the  2nd (0.82), the 5th (0.80), the 6th (0.82), the 8th (0.84), the 9th (0.80) and the 10th (0.93)  mother plants in Lula. Those selected mother plants produced morphological uniform seed morphologically. Lula seed plantation had the highest uniformity level among mother plants (0.77-0.87) compared to Toisapu (0.70-0.87) and Wakal (0.69-0.84) mother plants. The morphological characteristic can be use to select the source seed mother plants which able to produce high morphological uniformity seeds.Keywords: Gower’s similarity, seed production, uniformity

Abstract

A field research was conducted to study morpho-ecotype and proximate aspects of Banda nutmeg (Myristica fragrans Houtt.) in three ecotypes: Banda Islands, Ambon Island, and Ceram Island in Maluku. The objectives of the study were to characterize morpho-ecotype of the Banda nutmeg, to investigate the trees productivity across three ecotypes, and to identify the proximate characteristics of the fruit. Ten productive nutmeg sample trees aged ranging 25 to 50 years were chosen for observations. In morphological observation, 21 items were described based on IBPGR procedure, and then subjected to UPGMA cluster analysis. The proximates of nutmeg flesh including edible portion (EP), water content, protein content, fat content, and pectin content were analyzed by AOAC procedure. Morphological and proximate data were analyzed using SAS. Results show that Moluccas ecotypes characterized by hill and mountain has tectonic mountain physiography and karst soils with a slope of 16 to 40%. Ambon and Banda ecotypes are mainly composed of volcanic soils, whilst Luhu has sediment. The climate of Moluccas ecotype is dominated by IIIC type, except Banda Island which has IIB. The morphological traits of the nutmeg are stabile across three ecotypes (similarity index, SI 90%). Productions of fruit, nutmeg, and mace show no difference between the three sites. They are 137.73, 19.27, and 3.07 kg per tree, respectively. All proximate parameters analyzed are not statistically different, except EP. EP of Ambon ecotype is statistically different from that of Ceram but it is similar to Banda ecotype.
