Abstract

Ketela pohon (Manihot utilissima, Pohl) varietas mukibat berpotensi sebagai bahan baku bioetanol. Penelitian bertujuan untuk mengetahui efek interaksi ragi tape dan ragi roti terhadap kadar bioetanol ketela pohon mukibat dan menentukan interaksi terendah yang efektif menghasilkan bioetanol yang dapat terbakar. Sampel yang digunakan adalah ketela pohon mukibat umur 7 bulan dengan berat rata-rata 500 gram. Penelitian menggunakan Rancangan Acak Lengkap pola faktorial, yaitu konsentrasi ragi tape (0%, 0,3%, 0,6% dan 0,9%) dan konsentrasi ragi roti (0%, 0,3%, 0,6% dan 0,9%) dengan 3 kali ulangan. Parameter yang diukur meliputi jumlah sel khamir (data pendukung) dan kadar bioetanol (data utama). Data kadar bioetanol yang diperoleh dianalisis menggunakan anava dua arah dan dilanjutkan uji Duncan (DMRT) pada taraf kesalahan 5%. Hasil penelitian menunjukkan ada pengaruh interaksi ragi tape dan ragi roti terhadap kadar bioetanol ketela pohon mukibat p>0,05. Konsentrasi ragi tape dan ragi roti yang tinggi menghasilkan kadar bioetanol yang tinggi. Interaksi ragi tape 0,6% dan ragi roti 0,3% (T2R1) merupakan interaksi terendah yang efektif menghasilkan kadar bioetanol yang dapat terbakar (36%). Interaksi ragi tape 0,9% dan ragi roti 0,9% (T3R3) menghasilkan kadar bioetanol tertinggi dengan rata-rata mencapai 49,8%.Cassava (Manihot utilissima, Pohl) var Mukibat is potential for bioethanol feedstock. The study aimed to determine the effects of the interaction of bread yeast and tape yeast on the concentration of mukibat cassava bioethanol and to determine the lowest effective interaction can produce flammable bioethanol. The samples used were mukibat cassava aged 7 months with an average weight of 500 grams. Research design was completely randomized design factorial, tape yeast concentration (0%, 0.3%, 0.6% and 0.9%) and the concentration of yeast bread (0%, 0.3%, 0.6% and 0.9%) with 3 replications. Parameters measured include the number of yeast cells (supporting data) and ethanol concentration (primary data). Bioethanol content data obtained were analyzed using two-way ANAVA and Duncan continued test (DMRT) at the level of 5% error. The results showed that no effect of the interaction of breads yeast and tape yeast on levels of cassava bioethanol p> 0.05. The concentration of bread yeast and tape yeast yielded in high concentration of ethanol. The interaction of yeast tape 0.6% and 0.3% bread yeast (T2R1) gave the lowest effective interaction produced 36% ethanol. The interaction of tape yeast 0.9% and 0.9% bread yeast (T3R3) resulted in the highest ethanol concentration with an average reached 49.8%.

Abstract

Penelitian ini bertujuan untuk mengkaji efektivitas pemberian minyak sawit dan minyak lemuru dalam mempercepat pubertas tikus betina, pada parameter peningkatan berat badan, berat ovarium dan perkembangan folikel ovarium. Tikus betina usia 21 hari sebanyak 20 ekor dibagi menjadi 4 kelompok variasi pemberian minyak sawit dan minyak lemuru per oral, yaitu kelompok A (0%+0%), B (3%+3%), C (4%+4%), dan D (5%+5%). Pada akhir penelitian (hari ke-20), dilakukan pengambilan data peningkatan berat badan, berat ovarium, dan perkembangan folikel ovarium. Data dianalisis secara deskripsi dan statistik dengan anava satu arah. Jika terdapat perbedaan, dilanjutkan dengan uji Beda Nyata Terkecil. Hasil penelitian ini menunjukkan bahwa minyak sawit dan minyak lemuru dapat mempercepat pubertas tikus betina khususnya dari peningkatan berat badan, berat ovarium, dan perkembangan folikel ovarium. Dosis paling efektif untuk mempercepat pubertas adalah 4% minyak sawit + 4% minyak lemuru. This study was aimed to examine the effectiveness of palm and lemuru oil for the acceleration of female rat puberty, especially in increasing the body weight, the ovarian weight and the follicle development. Twenty female rats aged 21 days were divided into 4 based on the variation of orally feeding palm and lemuru oil, namely group A (0% +0%), B (3% +3%), C (4% +4%), and D (5% +5%). At the twentieth day, data of the increase of the body weight, the ovarian weight and the follicle development were collected and then analyzed descriptively and statistically using one-way ANOVA. For any difference, the Least Significance Difference Test would be performed. The result of this study showed that palm and lemuru oil can accelerate the puberty of female rats. The most effective dose was 4% palm oil and 4% lemuru oil.

Abstract

Tomato is one of the high antioxidant potential vegetables. Nowadays, there are many techniques of tomato processings instead of fresh consumption, i.e. boiled, steamed, juiced and sauteed. Every treatment of cooking will influence the chemical compound inside the fruits and the bodys nutrition intake. It is important to conduct the research on antioxidant compound especially lycopene, ?-carotene, vitamin C, ?-tocopherol, and its activity after processing. This research has been done using the experimental method. Tomatoes were cooked into six difference ways, and then it was extracted using the same procedure continued with antioxidant measurement. The research results showed that steaming had promoted the higher antioxidant numbers (lycopene. ?-tocopherol, ?-carotene and vitamin C) and higher TCA and antioxidant activities in the tomatoes than other processings. It was indicated that steaming was the best way to enhance amount, capacity and activities of antioxidants of the tomatoes.

Abstract

The study was aimed to determine the effectiveness of pedada fruit extract as a hepatoprotector in the experimental rat that fed by toxic dose of paracetamol. The total of 30 white rats (Wistar strain, two months age, and 150-200 g weight) were randomly divided into 5 groups. Group I (normal control) only given distilled water for 7 days). Group II (negative control) that given distilled water for 7 days and then treated by 270 mg/head single dose of paracetamol. Group III, IV, and V (treatment group) were given a pedada fruit extract at a dose of 28 mg/head/day, 56 mg/head/day, and 84 mg/head/day for 7 days and then treated by 270 mg/head single dose of paracetamol. On the 9th day of treatment, the blood samples were taken and were further measured for its SGOT and SGPT level using photometry enzymatic method. The result of LSD test on SGOT and SGPT data showed that III, IV, and V groups were not significantly different to the group I (p>0.05). However, it significantly different with the group II (p <0.05). Data of SGOT showed that group IV were significantly different (p <0.05) with the group V. Whereas, the data of SGPT among groups III, IV, and V were not significantly different (p>0.05). The result of linier regression test indicated that dose 28 mg/head was the most effective dose. It was concluded that pedada fruit extract was able to provide a hepatoprotective effects in rats that fed by toxic dose of paracetamol and most effective dose as a hepatoprotector was 28mg/head/day.Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak buah pedada sebagai hepatorotektor tikus putih yang diberi parasetamol dosis toksik.Sebanyak 30 ekor tikus putih (strain Wistar jantan berumur dua bulan dengan berat badan 150-200 g) dibagi secara acak dalam lima kelompok.Kelompok I (kontrol normal) diberi aquadest selama tujuh hari.Kelompok II (kontrol negatif),diberi aquadest selama tujuh hari dilanjutkan pemberian parasetamol 270 mg/ekor dosis tunggal.Kelompok III, IV, dan V (kelompok perlakuan) diberi ekstrak buah pedada pada dosis 28 mg/ekor/hari, 56 mg/ekor/hari, dan 84 mg/ekor/hari selama tujuh hari dilanjutkan pemberian parasetamol 270 mg/ekor dosis tunggal. Hari ke-9 darah diambil dan diukur kadar SGOT dan SGPT dengan metode fotometri enzimatik. Hasil uji LSD data SGOT dan SGPT menunjukkan bahwa kelompok III, IV, dan V tidak berbeda nyata terhadap kelompok I (p>0,05), namun berbeda nyata terhadap kelompok II (p<0,05). Data SGOT kelompok IV berbeda nyata (p<0,05) dengan kelompok V. Data SGPT tidak ada perbedaan nyata (p>0,05) antara kelompok III, IV, dan V. Hasil uji regresi linier, dosis 28 mg/ekor adalah dosis paling efektif.Disimpulkan bahwa ekstrak buah pedada mampu memberikan efek hepatoprotektor pada tikus yang diberi parasetamol dosis toksik dan dosis yang paling efektif sebagai hepatoprotektor adalah dosis 28 mg/ekor/hari.

Abstract

Ploceidae family’s birds in Indonesia consist of 41 species include 13 species as Java’s endemic. Some species of Ploceidae family were start to rarely oberved, because of new house developing and hunting as a pets, zoo collection and education kit. It’s need some efforts to conserve these rare species through improving the habitat, rehabilitation, nursery, controlling, law inforcement for hunters, and genetic conservation according to a knowledgement of genetic variation. The research aimed to knew the length fragmen of ND3 DNAmt Gen and genetic variation among species of Ploceidae family’s birds. 11 species were observe for morphology’s characteristic than blood sample were collected for DNA isolation with Dixit methode. ND3 gen were amplificated by DNA isolated with PCR used H11151 and L10755 primer. PCR’s gain were visualized with 2 % agarose gel. The length fragmen of ND3 DNAmt were 321bp for Bondol Jawa, 338 bp for Bondol Haji and Bondol Peking, 393 bp for Burung Gereja Erasia, 413 bp for Manyar Emas, 406 bp for Gelatik Jawa, 334 bp for Manyar Tempua and Manyar Jambul, 351 bp for Pipit Benggala, 333 bp for Bondol Hijau Binglis, 317 bp for Pipit Zebra. The conclusion of this research were : 1). the length variation of ND3 DNAmt Gen among 11 species range from 317 – 413 bp and 2). morphological variation and length variation of ND3 DNAmt Gen shows that there was genetic variation among species of Ploceidae family’s birds. Abstract

Meningkatnya kasus infeksi virus Avian Influenza (AI) subtipe H5N1 atau lebih dikenal dengan flu burung yang menyebabkan kematian pada manusia sangat dikhawatirkan dapat menular dari manusia ke manusia. Penelitian ini bertujuan untuk mendapatkan isolat virus Avian Influenza subtipe H5N1 pada unggas yang diperjualbelikan di pasar tradisional di Semarang. Sebanyak 55 sampel usap kloaka diambil dari unggas sehat dan belum divaksin di 6 pasar tradisional Kota Semarang. Inokulum ditumbuhkan pada telur ayam berembrio specific pathogen free (TAB-SPF) umur sembilan hari. Kemudian telur diinkubasikan selama 4 hari. Cairan alantois dipanen dan diuji kemampuannya mengaglutinasi sel darah merah. Cairan alantois yang menunjukkan aktivitas hemaglutinasi, selanjutnya diekstraksi RNA-nya dan diidentifikasi VAI subtipe H5N1 dengan metode Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) menggunakan primer spesifik H5 dan primer N1. Kemudian DNA hasil RT-PCR dianalisis dengan teknik elektroforesis. Hasil penelitian menunjukkan bahwa terdapat 4 isolat positif VAI subtipe H5N1 dengan sebaran 2 isolat dari sampel yang berasal dari pasar Mangkang, 1 isolat dari pasar Rejomulyo dan 1 isolat dari pasar Karimata. Berdasarkan hasil penelitian dan pembahasan dapat disimpulkan bahwa unggas yang diperjualbelikan di pasar tradisional di Kota Semarang ada yang terinfeksi VAI subtipe H5N1.The increasing cases of viral infection of Avian Influenza (AI) H5N1 subtype or more commonly known as bird flu that causes death in humans very feared to spread from human to human. The aim of this research was to obtain isolates of Avian Influenza virus (AIV) subtype H5N1 that marketable in traditional markets in Semarang. A total of 55 cloacal swab samples taken from healthy and unvaccinated fowl in the 6 traditional market in Semarang. Inoculum was grown in embryonated chicken eggs specific pathogen free (SPF TAB) nine days. Then the eggs were incubated for 4 days. Allantoic fluids were harvested and tested for their ability to agglutinate red blood cells. Allantoic fluid that showed hemagglutination activity, further their RNA was extracted and AIV subtype H5N1 identified with Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) method using specific primers primary H5 and N1. Then the results of RT - PCR were analyzed by electrophoresis technique. The results showed that there are 4 positive isolates with the distribution of the H5N1 subtype AIV 2 isolates samples derived from market Mangkang, 1 isolate from market Rejomulyo and 1 isolate from market Karimata. Based on the results of research and discussion, it can be concluded that the fowl that marketable in traditional markets in Semarang there were AIV infected with subtype H5N1.

Abstract

Phylogenetic or population genetic studies have been focused on finding genetic variations in mitochondrial DNA (mtDNA) because mutations in this region contained five times higher than other genetic materials. The aim of this research was to generate PCR-RFLP D-loop mtDNA profiles of some Indonesian domestic waterfowl. The samples were 35 local ducks, 5 muscovy duck, and 5 geese. These local ducks included on seven types of Javanese ducks in Central Java, i.e. Magelang duck, Peking duck, Pengging duck, Tegal Branjangan duck, Tegal Jarakan duck, Tegal Blorong duck, and Tegal Lemahan duck. PCR technique was used to amplify the D-loop genes, and RFLP analysis of the D-loop was performed with AluIand RsaI. The results show that muscovy duck has no restriction sites in D-loop gene. Moreover, goose has a restriction site of AluI in D-loop gene at 550 bp and 187 bp; and it has no restriction site of RsaI. The analysis of RsaI-RFLP in mtDNA D-loop region of ducks resulted in 1 restriction fragment pattern which can be applied in the identification of duck to differentiate it from goose and muscovy duck. PCR-RFLP analysis duck D-loop mtDNA using AluI and RsaI restriction enzyme resulted in 3 combinations of restriction fragment pattern shown in 3 haplotypes (A, B, and C). Genetic diversity of Indonesian domestic waterfowl population could be identified by using PCR-RFLP analysis on mtDNA D-loop region. So far, there are few results of research on Indonesian domestic waterfowl genetic characteristics based on PCR-RFLP mtDNA D-loop marker. Also, this research has a good contribution on genetic characterization of local duck. The characterization is an important aspect to maintain breed quality and manage the genetic resources of Indonesian germplasm and in facilitating the breeding program.

Abstract

This research was aimed to develop guided inquiry-based students’ worksheet in cell materials forstudents in senior high school. It has the characteristics of valid, readable, and executable. This researchwas a Research and Development (R&amp;D) type. The validity of worksheet was valued by validator andanalyzed using percentage description. The readability of worksheets can be obtained by small scaled tryout to 20 students taken by using purposive random sampling. The executability of worksheets wasobtained by bigger scale try out to 32 students. The result of the research showed that the guided inquirybasedstudents’ worksheets had characteristics of: 1) cell titled worksheets with its observation; 2) theactivities based on syntax model of guided inquiry learning; 3) the material is related to real example; 4)the illustration picture in worksheets was real and 5) the display of worksheet in full color. The averagepercentage of validity to the worksheet was 83.06%. The percentage score of the worksheet was 85.31%with executability of 93.18%. Based on the results, it can be concluded that this worksheet has severalcharacteristics and valid category to be used. It also had good readability and executability to thestudents.

Abstract

This aims of this research is to determine the application of NHT learning to student learning outcomes on the material of excretion system. The learning outcomes from this research is the result of learning in the affective and cognitive domain. Research with form of Pre-Experimental Design was held at SMPN 9 Semarang with class VIII D and VIII E as a research sample. Affective learning results show that 62,50% of students are in good categories and 29,69% are in very good category. The results of the N-gain test on students cognitive learning outcomes obtained from the posttest showed a gain of 0,61 with the medium improvement category. The final value analysis shows that 100% of students have complete their learning. Relatively, students give positive responses to the learning method that applied.&nbsp; Learning is accomplished with a value 90% on very high implementation criteria. The conclusion of this research is the method of learning using NHT method can improve student learning outcomes in the material excretion system

Abstract

Creative thinking is an ability of creating a new thought as a result from combination of previous knowledge. It is needed to be developed in the 21st century because it is importance in work world. This study aims to analyze the students’ creative thinking of SMA N 1 Pecangaan on environmental material. The research used observational method. Population is whole students of tenth grade of SMA N 1 Pecangaan. The samples used purposive samplings were X MIA 1, X MIA 2 and X MIA 4 with total number 116 students. The students’ creative thinking was tested by essay test and instrument non-test which is scoring rubric of writing article about environment problems. The result showed that students’ creative thinking of SMA N 1 Pecangaan in answering question is 14.7% very creative, 31% creative, 20.7% creative enough and 33.6% less creative. The students’ ability in writing article showed 0% very creative, 16.4% creative, 30% creative enough, 49.1% less creative and 4.3% not creative. The research concluded that the creative thinking ability of students SMA N 1 Pecangaan is low. The highest aspects of creative thinking that is reached by students are originality and fluency, while the lowest is elaboration.
