Abstract

Minyak wijen (MW) yang banyak mengandung polyunsaturated fatty acid (PUFA) berfungsi menurunkan kadar lipid serum melalui induksi β oksidasi di mitokondria. Proses ini menghasilkan produk sampingan berupa radikal bebas. Vitamin E (α-tocopherol) diketahui menghambat aktifitas radikal bebas. Penelitian ini bertujuan untuk membuktikan efek sinergisme MW dan α-tocopherol dalam menghambat steatosis yang diakibatkan keadaan hiperkolesterol. Hasil penelitian menunjukan bahwa diperoleh perbedaan bermakna (p<0,05) terutama dosis MW 1,2 ml pada semua parameter antara kelompok hiperkolesterol, MW dan MW+α-tocopherol. Kadar kolesterol kelompok hiperkolesterol (140,925±10,5) mg/dl; kelompok MW (93,845±4,37) mg/dl dan kelompok MW+α-tocopherol (92,90±8,5) mg/dl. Kadar trigliserida kelompok hiperkolesterol  (154,78±11,09) mg/dl; kelompok MW (184,64±3,87) mg/dl dan kelompok MW+α-tochopherol (66,89 ± 6,03) mg/dl. Pada kelompok MW kadar MDA (0,257±0,013) nmol/mg protein, kelompok hiperkolesterol (0,847±0,036) nmol/mg protein dan kelompok kombinasi MW dengan α-tocopherol (0,092±0,006) nmol/mg protein. Kadar SOD pada kelompok kombinasi MW dengan α-tocopherol (253,82±16,63) U/mg protein, kelompok MW (208,7±11,27) U/mg protein, kelompok hiperkolesterol (139,7±2,82) U/mg protein. Dari gambaran histologis steatosis lebih banyak didapatkan pada kelompok hiperkolesterol yang disertai dengan keradangan, sedangkan pada kelompok kombinasi minyak wijen dengan α-tochopherol memiliki gambaran histologis terendah mengalami steatosis. Abstract

Advanced Glycation Endproducts (AGEs) or so-called glycotoxin can be triggered by heated food in which the Maillard reaction occurs. One type of glycotoxins is CML. Accumulated N-Carboxymethyl-Lysine (CML) can cause inflammation of organs, e.g. liver. Rosella flowers contain anthocyanin compound that has anti-glycation and antioxidant effects. This study aimed to determine the effect of ethanol extract of Rosella on CML serum level, IL-6 level, and NF-ƙB activation in the liver of mice fed with baked food. This study used post-test design using 25 Wistar rats aged 3-4 months old that were divided into 5 groups, namely negative control group, positive control group, treatment groups given Rosella extract dose of 200mg.kgBW-1, 300mg.kgBW-1, and 400mg.kgBW-1. Baked feed was given for 12 weeks, and Rosella ethanol extract was administered in the 9th to 12th week. The examination on CML serum and IL-6 of the liver was using ELISA method. Immunofluorescent staining was used to determine NF-ƙB activation in the liver using a confocal microscope. CML serum is proven to increase significantly (p = 0.000). The effective dose of Rosella flower extract to prevent CML Serum increase is 200 mg.kgBW-1, whereas a dose of 400 mg.kgBW-1 can decrease IL-6 level and NF-ƙB activation. Ethanol extract of Rosella flower decrease the levels of N-carboxymethyl-lysine serum, IL-6, and NF-ƙB activation in the liver of rats given baked-food diet. Abstract

Coronary heart disease (CHD) is a non-transmitted disease which is particular concern at the global, national, and local level. CHD has become a concern because it has caused a lot of deaths. Risk factors linked with CHD consist of permanent risk factors and variable risk factors. The purpose of the research is to analyze the correlation between variable risk factors and CHD in the Minahasa ethnic society in Manado City. This research was an observational case-control study conducted by Prof. dr. R. D. Kandou Hospital, Manado, from August to October 2016. In total, there were 220 patients included. The sampling used a simple random sampling method and the data obtained were analyzed by chi-squared test. According to the results, hypertension, smoking, and behavioral type were connected to cases of CHD. The correlation degree showed that the respondents suffering hypertension were 5.70 times more likely to suffer CHD, the smoking respondents were 2.25 times more likely to experience, and behavior type A respondents were 2.96 times more likely to suffer CHD. Hypertension, smoking, and behavioral type are linked with CHD, so there should be some promotion and preventive actions from the health governmental institution to society, especially aimed at adults, about the quality of life enhancement by healthy behavior and avoiding CHD risk factors. Abstract

The food processing by heating can increase the formation of AGEs (Advanced Glycation End Products). AGEs are compounds that formed from a non-enzymatic continuous glycation reaction between proteins and sugar residues. The CML (N-carboxymethyl-lysine) is used as a marker for AGEs cause most commonly found in vivo. The bond of AGEs and RAGE (receptor for AGEs) induce various signaling pathways that trigger inflammation and increase oxidative stress. The AGE and RAGE interaction activate the transcription factor of NF- κβ. NF- κβactivates gene transcription to release proinflammatory cytokines such as TNF-α. Anthocyanins are compounds that can prevent the formation of AGEs and muffle the adverse effects of AGEs. Rosella contains anthocyanin such as: delphinidin-3-O-glucoside, delphinidin-3-O-sambubioside, and cyanidin-3-O-sambubioside. This study is to determine whether the daily intake of Rosella extract can reduce the levels of n-carboxymethyl-lysine in serum, expression of NF-κβ and TNF α in the rats fed with heated food. This study applied experimental post test control using Rattus norvegicus Wistar strain. The samples were divided into 5 groups: KN (negative control/fed without heating), KP (positive control/fed heated food but not treated by Rosella extract), R1 (fed heated food and treated by 200 mg.kg-1BW Rosella extract), R2 (fed heated food and treated by 300 mg.kg-1BW Rosella extract), and R3 (fed heated food and treated by 400 mg.kg-1BW Rosella extract). The n-carboxymethyl-lysine levels were measured by using the ELISA, the expression of NF-κβ is analyzed by using immunofluorescence, and expression of TNF - α is observed by immunohistochemistry. There was significantly decreased the levels of n-carboxymethyl-lysine in all groups which were treated by Rosella extracts (R1,R2,R3); p= 0.000, α = 0.05 (p< α). Decreased activation of NF-κβ in all groups which were treated by Rosella extract is significant (p = 0.000), and decreased expression of TNF α in all groups which were treated by Rosella extract is also significant (p = 0.000). Rosella extract can reduce the levels of n-carboxymethyl-lysine, expression of NF-κβ, and TNF α. Keywords: AGEs, anthocyanin, Rosella flower extract, n-carboxymethyl-lysine, NF-κβ, TNF α.

Abstract

Heated food is a source of formation of various types of Advanced Glycation Endproducts (AGEs). One of which is N-carboxymethyl-lysine (CML) that is often used as a sign of the formation of AGEs in the diet. The accumulation of AGEs in the brain can trigger neurodegenerative conditions. It also accelerates the accumulation of Amyloid β and proinflammatory cytokines which exacerbate neurodegeneration. Hibiscus sabdariffa L or rosella known to have antiglycation, antioxidant, and anti-inflammatory effects. This study aims to prove the effect of food heating to the increasing levels of CML serum and evaluate the effects of daily intake of Rosella extract IL-6 levels and Amyloid β in the Wistar rats' brain given a diet high of AGEs. This experimental study with post test only control group design was conducted using 25 white male rats Wistar strain (Rattus novergicus) which were divided into 5 groups namely positive control, negative control, the treatment group with rosella dose 200mg.kgBW-1, 300mg.kgBW-1, and 400mg.kgBW-1. The high diets of AGEs are given for 12 weeks, and it was added to the rosella extract with various doses on the 9th to 12th weeks. The examination of CML serum level, interleukin 6 brain level, and Amyloid β of the brain was carried by ELISA. This study found differences between CML serum level of rats which fed a standard diet and the one that was given a heated diet (p= 0.0001). It also found that rosella flower extract can prevent the increasing levels of interleukin 6 and Amyloid β in the brain tissue of rats which were given a heated diet, and both effective doses were on 200mg.kgBW-1.
