Abstract

This study aim at determining the activity of ethanol extract of Robusta green coffee beans as an antiproliferation agent in MCA-B1, MCM-B2, and Vero normal cell lines as a control. Ethanol extract of Robusta green coffee beans was obtained by maceration method, bioactive compounds in the extract were tested by phytochemical screening methods, testing levels of antioxidants were tested by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method, cytotoxicity effects (LC50) were tested by the method Brine Shrimp Lethallity Test (BSLT) and antiproliferation activity were carried out by counting the number of cells after being tested with several extract concentrations namely 100, 250, 400, 550, 700 and 850 ppm and doxorubicin as a positive control with a concentration of 100 ppm. The results showed that the ethanol extract of Robusta green coffee beans contained flavonoids and tannins, had very strong antioxidant levels with IC50 values of 40.9923 ppm, cytotoxicity effects obtained LC50 values of 430.64 ppm, and the highest antiproliferation activity was achieved at a concentration of 850 ppm at 69.58 % in MCM-B2 tumor cells, 60.46% in MCA-B1 tumor cells, and 14.2% in Vero normal cells. Based on the results, the ethanol extract of Robusta Lampung green coffee beans has antiproliferation activity on the test tumor cells and is relatively non-toxic to normal cells.

Abstract

Based on WHO’s data, about 80% peoples in the word use plant’s extract as herbal medicine. Avocadoleaves has used as herbal medicine which is works as diuretikum, reduce stone in renal, and cure sprue.The purpose of this research is to study activities of ethanol extract of avocado leaves to ethylene glycolinduction on rat’s kidney by histopatologically. This experiment was continuing Adha’s research (2009)with research design as follows: twenty male rats were divided into 4 groups, negative control group (KN),positive control group (KP), treated group I by ethanol extract of avocado leaves 100 mg/kg BW (E100), andtreated group 2 by 300 mg/kg BW (E300). Sampling kidney were fixed in BNF 10% for histopathologicalslide and stained by Hematoxylin-Eosin. The kidney histopathological changes were edema of glomerulus,and protein deposit in the lumen, hyalin droplet and necrotic tubules. The results showed that ethanolextract of avocado leaves can’t significantly to reduce glomerulus edema with percentage cases 31.9% and33.5% for E100 and E300 and tend not to reduce the occurrence of protein deposits in the lumen of thetubule. Ethanol extract of avocado leaves tend to reduce the incidence of tubular necrosis and it wassignificantly different from the KP group (p <0.05). It is caused by flavonoid in etanol extract of avocadoleaves works as diuretikum and antioxidant.

Abstract

The purpose of this study is to examine the activity of Ambon banana extract in gel formulation on thewound healing process of mice skin (Mus musculus albinus) based on gross and microscopic observations.The wound healing process were observe grossly everyday while the microscopic lesion was observed on the3rd, 5th, 7th, 14th and 21st days after skin incisition. The wounded skin were sampled after the mice wereeuthanized for further microscopic observation. The gross parameters were the existence of blood coagulation,scab formation, wound covering and wound size. The microscopic parameters observed the percentation ofwound reepitelization and the the tickness of wound connective tissues (fibroblast). Gross lesion and thefibroblast thickness were presented descriptively as a qualitative data. Grossly, it was observed that thescab formation was faster in mice treated with the extract of ambon banana than that of negative control.Based on the research the ambon banana extrack in gel can be used in the acceleratiof wound healing.

Abstract

Urine crystal is a crystal nucleus which tend to form urine stone. The case of urine stone seems to beincreased every year. Crystallization could induce acute tubular necrosis which impact on renal dysfunction.The signs of this condition are high level of urea, creatinine and decrease glomerulus filtration rate. Theobjective of this research was to evaluate the effects of infusum Persea americana Mill as an inhibitorcrystallization which induced by ethylene glycol on white male rats. 20 male rats were divided into 4groups; K1 as negative group received only distilled water ad libitum, K2 as positive group receiveddistilled water containing ethylene glycol, K3 (dose 5%) and K4 (dose 10%) as treatment groups receivedwater containing ethylene glycol and avocado leaves infusion. Phytochemsitry screening of infusion avocadoleaves consisted of flavonoid, saponin, tanine and quinone. Result of analysis showed that the level ofureum and creatinine on K2 was higher than K3 and K4 group. The increased level could be inhibited byinfusion avocado leaves. The measurement of glomerular filtration rate in treatment groups wassignificantly different (p<0.05). Descriptive histopathology observation showed that renal lesio in grouptreatment (K3 and K4) were declined. Large crystal calcium oxalate on K2 group was observed by usingpolarized microscope, whereas small crystal calcium oxalate were seen in the infusion of avocado leavesgroups. These result showed the ability of infusion of avocado leaves as an inhibitor on the growth ofcrystallization calcium oxalate

Abstract

This experiment was carried out to determine the mouse lethal dose (MLD50 of ethanolic extract ofthe turmeric (Curcuma longa) rhizomes following single oral administration and to evaluate thetoxicopathologic effects of the extract in certain organs ie stomach, liver and kidney. Forty five male micewere divided into nine groups. Four groups were treated orally with ethyl acetate fraction, four groups weretreated orally with hexane fraction of ethanolic turmeric extract, and one group was used as control. Theethyl acetate fraction groups were administered orally with the ethyl acetate fraction at doses of 7.5, 1.5,30 and 60 g/kg body weight. The hexane fraction groups were administered orally with the hexane fractionwith the dosage of 7,5, 15, 30 and 60 g/kg body weight. The control group received normal saline. MLD50 ofethyl acetate fraction was 27,98 gram/kg body weight by per oral administration. Oral MLD50 of hexanefraction was 19,50 gram/kg body weight. Histopathological features of the ethyl acetate and hexanefractions groups showed increased amount of parietal cells in stomach and parenchymal degeneration andnecrosis in their liver and kidney.

Abstract

The objectives of this research was to examine the efficacy of red betel extract ointment to eritrogram profiles in infected sheep myiasis. Fifteen female sheeps were divided into five treatments, which were sheep without wound, treated with asuntol 2%, without any treatment,with red betel leaf extract ointment 2% and 4 %. Three incision wounds were made on the sheep’s back, 50 larvae given to every wounds to four treatment, except the healthy sheep (K). Blood was taken on the 0, 3th and 7th to determine erythrocytes counts, hematocrit values (PCV) and hemoglobin levels (Hb). The result showed that oitment of red betel leaf extract do not change the erythrogram profiles. The administration of red betel leaf extract ointment 4% has better eritrogram profiles than 2% Oitment of red betel leaf extract can be used as myasis therapy.

Abstract

Nephrolithiasis is one of urologic disease due to crystallization urine in urinary system. Traditionally,avocado leaves has long been used for the treatment of nephrolithiasis because flavonoid in this plant canfacilitate the watering of crushing stone in the urinary tract. The aims of this study was to determine theeffect of aethyl acetate fraction of avocado leaves on ureum and creatinine level. Twenty white male rats(Sprague dawley, weight 200-300g) were divided into four groups. Normal control group (A) was givenaquadest ad libitum. Negative control group (B) and treatments group (C and D) were given ethyleneglycol 0.75% and ammonium chloride 2% to induce the formation of calcium oxalate crystal. Group C andD were given aethyl acetate fraction with different levels (100mg/kg bb and 300mg/kg bb for fourteen days).Blood samples were taken before and after treatment. The result showed that aethyl acetate fractioncould not decrease serum blood level of ureum and creatinine. Serum blood level ureum and creatinine ingroup D increased significantly compared to group A, B, C after fourteen days treatment. The used ofethylene glycol caused acute tubular necrosis. It therefore appears that ethyl acetate solvent was not ableto extract flavonoids to prevent renal dysfunction.

Abstract

Protein in albumen can be chelated and create strong bond with lead. The research was done toevaluate the speed of chelating process by local chicken egg, layers chicken egg, mojokerto duck egg, andpeking duck egg. The experiment was conducted by mixing lead acetat with albumen on 1:1 ratio. White eggconcentrations were divided into 25%, 50%, 75%, and 100%. Control group used 0% of albumen concentration(aquadestilate). Peking duck white egg had the fastest speed of protein chelation. Peking duck albumen by100% concentration had the fastest speed of chelation among the other egg used in experiment (p<0,05). Inconclusion, albumen can be used to overcome lead toxicity, with various speed of chelating process amongalbumen of various alvian.

Abstract

This study aims to determine the effect of the feed additive (zinc and herbs) in leucocyte profiles,performance, and observe the ability of broiler on E. coli challenge. A total of 160 chickens were dividedinto four groups: Group K-control negativewere given basal diet; Group two were given fed basal, turmeric1.5% + ZnO 180 ppm; Group two were given fed basal, garlic powder 2.5% + Zn0 180 ppm; and group K+ weregiven basal diet and treatment with antibiotics. At the age of three weeks all groups were challenged orallyby inoculation with E.coli at dose of 108CFU/mL. Observed parameters include performance (weight gain,consumption) and total leukocytes. Weight gain, consumption were observed at one to five weeks of age,whereas blood samplings for the examination of leukocytes were performed at week three (pre infection),week four and five (one week and two weeks after infection with E.coli). The results showed that thechicken body weight from age one to three weeks was increased sharply. One week after infection weightloss seemed to be decreased. The largest decrease was observed in the group given the combination ofgarlic-Zn, while providing a combination of curcumin-Zn shows that the weight tends to increase. Rationconsumption showed the same pattern as body weight. The highest consumption at the end of the studywas found in the group given the combination of curcumin - Zn, the lowest was in the garlic-Zn. Highleukocyte cells level at one week post infection, showing an animal in a state of infection and the leucocytedecreasing again at two weeks post infection. The conditions indicate the ability of chickens in eliminatinginfectious agents. These results explain the combination of curcumin-Zn showed the best results comparedwith other treatments. Providing supplementation of curcumin - Zn on chicken improved the ability toeliminate E.coli, leukocyte cell profile, and performance.

Abstract

Bisbul (Diospyros discolor Willd.) is a plant of the Ebenaceae family and red dragon (Hylocereus polyrhizus) is a plant of the Cactaceae family. Bisbul and red dragon have bioactive compounds are flavonoids that act as antioxidants. The purpose of this study was measure antioxidant activity from ethanol extract of bisbul pulp and ethanol extract of red dragon fruit stem by DPPH method (1,1-diphenyl-2-picrylhydrazin). Extraction by maseration method and using 96% ethanol as solution. Antioxidant activity assay using the immersion method of DPPH measured absorption at a wavelenght 517 nm and compared with vitamin C. The result showed that the ethanol extract of bisbul pulp (IC50 46.00 ìg/mL) more strong antioxidant activity compared the ethanol extract of red dragon fruit stem (IC50 1020.96 ìg/mL).
