Abstract

The objective of the present research is to study the activity of banana stem extract in ointment solutions in the acceleration of wound healing process on mice skins. Totally of 45 mice strain DDY 4-6 weeks old were devided in negativecontrol group (placebo ointment), positive control  group (Betadine® ointment) and Ambon banana stem extract ointment.All mice were aseptically wounded 1-1,5 cm in the anterior region of back skin using a sterile scalpel. The wound wassmeared with the ointment. The pathology anatomy observations was done inday 3, 5, 7, 14 and 21 post wounded. Parameters of the gross lesions (pathology anatomy) observations warecolour of the wound, blood coagulations, scab formations and size of the wound. Parameter for (microscopic lesions) histopathology were infiltrations of inflammatory cells (neutrophils, lymphocytes,macrofages), neo-capillarizations, re-epitelization percentage and the  thickness of fibroblast. All quantitative data were measure using ANOVA and continue with Duncan Test, moreover, the qualitative data were presented descriptively. The result shows that gross lesions observations, the extract ointment group was faster in scab formations and covers the wound without trace compared to the negative control group. The statistical test on the infiltrations of inflammatory cells parameter of the extract ointment group significantly different (P<0.05) compared to the negative control group. Histopatologycal observations shows that Ambon banana stem extract in ointment solutions can increase the infiltrations of inflammatory cells, neo-capillary formations, re-epitelizations percentage andacceleration of fibroblast formations. Base on the result the Ambon banana stems extract in ointment solutions can accelerate the wound healing process and it seems that this solution could be developed and uses for the medical purposes.

Abstract

Pegagan (Centella asiatica (L.)Urban) merupakan tanaman herba tahunan yangtumbuh menjalar dan berbunga sepanjang tahun.Pegagan dirujuk sebagaiantiinflamasi, antioksidan, antitumor, antibakteri atau untuk meningkatkan daya ingat(susunan syaraf pusat), eksim, luka bakar dan hepatitis.Tujuan penelitian ini adalahuntuk mengetahui antioksidan fraksi metanol dari ekstrak metanol daun pegagan.Melalui tahapan pemisahan, pemurnian dan menentukan aktivitas antioksidan denganmetode DPPH (1,1-difenil-2-pikril-hidrazil). Pada penelitian ini daun pegagandimaserasi menggunakan pelarut metanol kemudian dilakukan uji fitokimia danidentifikasi gula. Pemurnian dilakukan dengan metode kromatografi kolom denganelusi gradien menggunakan tiga eluen yaitu berturut-turut toluen : etil asetat (1:1), etilasetat, dan metanol. Identifikasi dilakukan menggunakan kromatografi lapis tipis(KLT) siliska gel dengan eluen etil asetat : metanol : air (8,1 : 1,25 : 0,65). Terhadapfraksi metanol dilakukan uji aktivitas antioksidan. Hasil uji fitokimia terhadap fraksimetanol diperoleh golongan senyawa terpenoid, flavonoid, dan alkaloid. Hasil KLTfraksi metanol menunjukkan nilai Rf pada 0,64. Aktivitas antioksidan fraksi metanoldiperoleh nilai IC50sebesar 481,64 ppm.

Abstract

 The purpose of this study was  to analyze the physicochemical stabilities  and in vivo antiaging  properties of the cream formulas of F1, F2, and F3 which contained the active compound of 0.1% 0.18, and 0.36%, respectively. The active compound was the combination of  ethyl acetate fraction of  Toona sinensis, Centella asiatica, and Acacia mangium  leaf (1:2:1). For physicochemical stability tests (color, odor, acidity, and antioxidant activity), the creams were stored for 21 days in the refrigerator, room temperature,   and extreme temperature (±40 ⁰C). For in vivo antiaging test,  the mice skin  were smeared with cream and UV irradiated for 15 minutes per day. After two and four weeks, the mice skins were  biopsied. The results showed that the physicochemical properties of creams stored in the refrigerator were  stable. The storage of creams at room and  extreme temperature decreased the physicochemical properties.  The application of cream formulas on mice skin after two to four weeks were able to increase the thickness of epidermis, but has not been able to increase the collagen skin of mice.Keywords: Acacia mangium, Centella asiatica, ethyl acetate fraction of leaf, physicochemical and antiaging properties of cream, Toona sinensis

Abstract

Edible bird’s nest (EBN) is often consumed as a health food due to its suggested health benefits, including anti-aging effects, however the mechanism is still unknown. This study investigated the effect of EBN extract using long term expansion bone marrow-derived mesenchymal stem cells (BMMSCs) as an aging model. Passage 5 (P5) and passage 8 (P8) BMMSCs were treated with EBN extract, and their proliferation, senescence-associated β-galactosidase (SA-β-Gal) activity, and expression of p16INK4a were analyzed. Treatment of BMMSCs with EBN extract decreased population doubling time (PDT) in P5 but not in P8 BMMSCs. In P5 BMMSCs, 200 ppm EBN extract increased BMMSCs proliferation, with PDT reduced by 27.6%. However, 200 ppm EBN extracts did not affect P8 BMMSCs proliferation, although it increased BMMSCs viability. Treatment of P5 and P8 BMMSCs with 200 ppm EBN extract decreased SA-β-Gal activity by 54.8% and 47.1% of the control, respectively (P<0.05). Levels of p16INK4a expression were 5.4-fold lower in P5 BMMSCs treated with 200 ppm EBN extract compared to control (P<0.05). Similarly, treatment of P8 BMMSCs with 200 ppm EBN extract reduced p16INK4a mRNA level by 7.9-fold compared to the control (P<0.05). In order to investigate the pathway of EBN extract inhibition, we further analyzed IL-6 and NF-κB1 expression. Treatment of P5 and P8 BMMSCs with 200 ppm EBN extract reduced IL-6 mRNA levels by 7.9-fold and 2.1-fold of control, respectively (P<0.05). We found that 200 ppm EBN extract reduced NF-κB1 mRNA level approximately 2.4-fold both in P5 and P8 BMMSCs (P<0.05). Thus, EBN extract reduces markers of senescence, indicated by decreased SA-β-Gal activity and p16INK4a mRNA level, and this correlated with reduced messenger RNA levels of the pro-inflammatory factor IL-6 and the transcription factor NF-κB1.

Abstract

Kucing jantan berumur 4 tahun bernama Rambo datang ke Klinik Star Vet Ciomas Bogor dengan keluhan menderita diare selama 6 bulan. Kucing memiliki kebiasaan diberikan minum air mentah, sering minum air toilet dan air got. Kucing di diagnosa menderita Giardiasis dengan menggunakan teknik fecal flotation yang ditunjukan dengan adanya kista Giardia sp. sebanyak >30 kista per lapang pandang. Kucing di terapi menggunakan antibiotika metronidazol dosis 20 mg/kg BB, imunomodulator (Echinacea extract) dosis 0.1 ml/kg BB dan Vitamin B12 dosis 0.025 mg/kg BB secara peroral selama 10 hari. Dilakukan pemantauan melalui pengamatan bentuk feses dan menghitung jumlah kista Giardia pada hari ke-0, 5, dan 10 pascaterapi. Pada hari ke-10 pascaterapi, feses sudah mulai berbentuk dan tidak ditemukan kista Giardia sp. sehingga kucing dinyatakan sembuh dari Giardiasis.

Abstract

Tingginya tingkat keterulangan kasus urolitiasis baik pada hewan maupun manusia, menuntut penelitian dalam mencari solusi menurukan angka keterulangan. Penelitian urolitiasis membutuhkan hewan model urolitiasis yang sesuai dengan kondisi sebenarnya. Salah satu metode yang digunakan adalah menciptakan kondisi hiperoksaluria. Penggunaan induksi etilen glikol menjadi metode yang paling umum digunakan. Induksi ini menghasilkan kristal kalsium oksalat monohidrat.

Abstract

Cyclophosphamide (CP) is a chemotherapeutic agent that belongs to the alkylating agent group that is widely used in the treatment of cancer. Cardiotoxicity is often a side effect of using CP in medical therapy. In this study, 24 Sprague Dawley rats were randomly divided into 4 groups. Group 1 (K1) was given injection with aqua pro injection intraperitoneally (IP) once a week for 21 days. Group 2 (K2) was given IP CP with a dose of 50 mg/kg BW, once a week for 21 days. Group 3 (K3) was given boswellic acid extract at a dose of 250 mg/kg BW orally, every day for 21 days. Group 4 (K4) was given boswellic acid nanoparticles at a dose of 250 mg/kg BW orally, every day for 21 days. During the treatment the body weight of the rats was weighed every day. At the end of the treatment, the rats were euthanized and blood samples were taken for blood biochemical evaluation, namely CPK, LDH, AST, and ALT. The results showed that the levels of CPK, LDH, AST, and ALT in K2 were significantly higher (p<0.05) than K1, K3 and K4. Statistically, the results of CPK, LDH, AST and ALT in K3 and K4 were not significantly different (p<0.05) compared to K1. The two groups (K3 and K4) were not significantly different (p<0.05) but on average the CPK, LDH, AST, and ALT results in K4 had lower scores than K3. This can indicate the protective effect of boswellic acid and boswellic acid nanoparticles on the heart against cyclophosphamide-induced cardiotoxicity.

Abstract

Penelitian ini bertujuan mengetahui aktivitas antibakteri, konsentrasi efektif, dan pengaruh peningkatan konsentrasi ekstrak etanol daun kari (Murraya koenigii) terhadap daya hambat pertumbuhan bakteri Staphylococcus aureus, Escherichia coli dan Pseudomonas sp. Ekstraksi dilakukan dengan cara maserasi menggunakan pelarut etanol 96%. Pengujian aktivitas antibakteri menggunakan metode difusi agar. Parameter yang diukur adalah besarnya diameter daya hambat yang terbentuk di sekitar kertas cakram. Hasil  uji aktivitas antibakteri dianalisis dengan metode one way anova dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa konsentrasi ekstrak 12,5; 25; dan 50% telah memberikan aktivitas daya hambat pertumbuhan bakteri uji. Konsentrasi efektif yang dapat menghambat bakteri Staphylococcus aureus dan Escherichia coli pada konsentrasi 50%, sedangkan bakteri Pseudomonas sp. pada konsentrasi 12,5; 25; dan 50%. Peningkatan konsentrasi ekstrak daun kari menunjukkan semakin luas diameter zona hambat pertumbuhan bakterinya. Penghambatan yang terjadi pada bakteri Staphylococcusaureus, Escherichia coli, dan Pseudomonas sp. tersebut, membuktikan bahwa daun kari mengandung senyawa aktif yang bersifat antibakteri, seperti flavonoid, fenol, alkaloid, dan saponin.

Abstract

Chemotherapy drug “cyclophosphamide (SP)” can trigger oxidative stress that causes hematotoxicity and myelotoxicity, therefore an agent that can suppress the negative effects of cyclophosphamide is needed. Boswellic acid is an herbal medicine that has anti-oxidant and anti-inflammatory effects. The aim of this study was to determine the effect of boswellic acid (BA) in hematology parameter rats induced by cyclophosphamid. Animal were allocated in to six groups, each group consists of 5 rats, the KS: healthy group, KSP: cyclophosphamide group; KSD: comparison group, and BA groups (K1;K2;K3). The treatment groups were given cyclophosphamide (SP) at dose of 150 mg/kg body weight (BW) i.p. The BA groups (K1,K2,K3) were given BA at dose of 250 mg/kg BW orally for 6 consecutive days and SP on day 1 (K1), day 4 (K2), day 6 (K3). The results showed that BA administration (K2 and K3) can help increase/maintain the amount of hematological parameters including hemoglobin, hematocrit, and erythrocytes. Administrating BA for 6 days consecutively before SP (K3) has shown best results on hematological parameters than the others including the comparison group. These results suggest that BA may have a potential herbal therapeutic agents that can mitigate cyclophosphamide-induced hematotoxicity manifestations.
