Abstract

Penelitian ini bertujuan mendiagnosis kasus newcastle disease (ND) lapangan pada ayam bukan ras yang bersifat akut melalui hasil pemeriksaan di Laboratorium Patologi dan Laboratorium Biomedik Fakultas Kedokteran Hewan Universitas Udayana tahun 2008-2009. Sebanyak sepuluh ekor sampel ayam buras telah diperiksa. Gejala klinis yang teramati meliputi: anoreksia, lesu, bersin, batuk, dan diare putih kehijauan dengan diagnosis sementara sebagai penyakit ND yang bersifat akut. Sampel diambil dari organ yang mengalami perubahan patognomonis seperti pada proventrikulus, ventrikulus, seka tonsil, paru-paru dan otak. Perbanyakan virus menggunakan telur ayam bertunas (TAB) umur 10 hari melalui ruang alantois dan diinkubasikan pada inkubator telur suhu 37° C selama 3 hari. Koleksi cairan alantois dilakukan pada hari ke-3, selanjutnya diidentifikasi dengan uji serologi hemaglutinasi (Haemaglutination-Inhibition Test/HA/HI) dengan teknik mikrotiter baku dan dikonfirmasi dengan uji reverse transcriptase-polymerase chain reaction (RT-PCR) menggunakan primer FNDIFP (5’-CCCCGTTGGAGGCATAC-3’) dan FNDIBP (5’-TGTTGGCAGCATTTTGATTG-3’). Hasil penelitian menunjukkan bahwa semua sampel kasus ayam bukan ras yang diperiksa positif terinfeksi oleh virus penyakit ND akut. Kajian ini mengindikasikan bahwa penyakit ND di Bali masih bersifat endemis.

Abstract

Egg Drop Syndrome (EDS) is a poultry disease marked by a decrease of egg production up to 40% with a declined quality of eggs. In Indonesia, EDS is astrategically infectious disease that must be eradicated. Until now, EDS vaccines are available, vaccinations on egg-laying chicken have been conducted nevertheless the case of EBS are still found. This research collaborates with PT Sanbio Laboratories (Poultry vaccine producer in Bogor) aimed to prepare EDS seed vaccines from local isolates. Isolate samples are collected from ten egg-laying chicken farmswith signs of declined production and egg quality as well as unevenly shaped and thinning of egg shells. Virus isolation was conducted on embryonated duck eggs at the age of 11 days through the allantoic space. Embryonated duck eggs are then incubated for 3 days in the incubator at 370C and are observed daily. On the third day, the eggs are taken out of the incubator and then inserted in the cooler overnight. The allantoic fluid is harvested on the third day post inoculation; it is identified with haemaglutination test and Polymerase Chain Reaction (PCR). Six isolates are positively identified as EDS virus: two from the Medan isolates, there from the Rumpin isolates, and one from Surabaya isolate. One isolate is chosen; which was the EDS isolate of Medan, with the highest titer for passaging and characterization. The content of the virus is calculated with Reed and Muench formula and expressed in a unit of Egg Lethal Dose 50 (ELD50). Research results shows titer of Medan EDS virus isolate after the second passage was 1012 HA Unit, with virus content of 109,5 EID50, length base product of PCR that was successfully amplified as 500bp. Therefore the Medan isolate is a recommended candidate for EDS vaccine seeds.

Abstract

Peternakan ayam lokal tradisional, dibandingkan dengan peternakan unggas komersial, menghadapi banyak tantangan terkait penyakit, dengan salah satu ancaman utama adalah virus penyakit tetelo. Laporan kasus ini menggambarkan ayam lokal berumur 8 minggu yang terinfeksi virus Newcastle disease terdeteksi melalui pemeriksaan aspek epidemiologi, patologi, dan virologi. Angka kesakitan di peternakan ini adalah 65%, dengan angka kematian 20% dan angka kematian 30,7%. Hasil pemeriksaan anatomi dan histopatologi menunjukkan adanya perubahan patologis pada berbagai organ. Uji laboratorium menggunakan uji Haemmaglutination-Inhibition dan Haemmaglutination memastikan adanya virus penyakit tetelo. Berdasarkan gejala klinis, temuan patologi anatomi, analisis histopatologi, isolasi virus, dan identifikasi, disimpulkan bahwa ayam yang terkena penyakit tersebut terinfeksi penyakit Newcastle velogenik viscerotropik.

Abstract

Shigellosis is a bacterial infection caused by members of the genus Shigella and is rarely reported in chickens. In contrast, coccidiosis caused by Eimeria spp. is a common and devastating parasitic disease in broiler chicken. Both diseases can cause digestive disorders, decreased performance, and even death, and can potentially occur together under suboptimal rearing conditions. This case report aimed to identify the causative agent of a disease outbreak in broiler chickens on a farm in Kutuh Village through an epidemiological investigation, clinical examination, and a series of laboratory tests. The examination method began with interviews with farmers and field investigations (epidemiology), followed by clinical examination of sick chickens to collect samples and undergo supporting examinations in anatomical pathology, bacteriology, and parasitology laboratories. A 30-day-old broiler chicken showed clinical symptoms of weakness, severe lethargy, decreased appetite, dull and falling feathers, inability to stand, and brownish-yellow diarrhea. Anatomical pathology observations revealed cheilitis in several organs, a fibrin layer on the heart surface, and intestinal bleeding. Histopathological examination revealed the infiltration of polymorphonuclear inflammatory cells in the heart, liver, intestines, and cecum, and the presence of schizonts in the cecal epithelium of infected tissues. Bacteriological examination successfully identified Shigella spp., and parasitological examination revealed Eimeria spp. oocysts in fecal samples, with a total of 273,100 oocysts/gram. Based on these findings, it was concluded that the chickens in this case experienced co-infection with Shigella and Eimeria. Further research on Shigella infection in poultry is recommended, and farmers should improve biosecurity and provide appropriate treatment to their flocks.
