Abstract

Enzim protease merupakan salah satu enzim komersial yang memiliki berbagai manfaat dan aplikasi pada bidang industri. Enzim protease dapat dihasilkan oleh mikroorganisme dan dapat dilakukan optimasi untuk menghasilkan aktivitas enzim yang optimum. Tujuan penelitian adalah untuk mengetahui jenis sumber karbon dan sumber nitrogen terbaik dalam menghasilkan enzim protease. Penelitian ini dilakukan dengan metode eksperimen menggunakan beberapa variasi jenis sumber karbon dan nitrogen untuk melihat aktivitas enzim protease melalui ekstrak kasar enzim (crude enzyme extract) yang dihasilkan. Hasil didapatkan dari  penelitian ini adalah sumber karbon yang terbaik berupa maltosa, sedangkan sumber nitrogen terbaik merupakan kalium nitrat (KNO3) konsentrasi 1% (v/v), dengan aktivitas protease sebesar 1,136 U/mL

Abstract

Enzim protease memiliki kemampuan untuk memecah protein menjadi oligopeptida dan asam amino. Enzim yang dihasilkan oleh bakteri endofitik dapat diproduksi dalam skala besar tanpa perlu merusak atau menebang tanaman inangnya. Penelitian ini bertujuan untuk mengetahui suhu, pH, dan waktu inkubasi optimum terhadap isolat bakteri endofitik EUA-136 dalam meningkatkan aktivitas enzim protease. Penelitian ini merupakan penelitian eksperimen dengan menggunakan Response Surface Methodology (RSM) tipe rancangan Central Composite Design (CCD) pada software Design Expert 13. Hasil dari penelitian ini didapatkan aktivitas protease dari isolat bakteri endofitik EUA-136 optimum pada suhu 30 ºC, pH 8 dalam waktu inkubasi 15 menit. Keywords: optimasi, protease, bakteri endofitik, response surface methodology

Abstract

Protease merupakan enzim yang paling banyak dimanfaatkan diberbagai industri seperti industri kesehatan, farmasi, deterjen, makanan, dan tekstil. Protease termasuk golongan enzim hidrolase yang mampu memecah protein menjadi oligopeptida pendek atau asam amino. Enzim mudah terdenaturasi pada suhu tinggi. Sedangkan dalam industri, protease lebih banyak diaplikasikan pada suhu tinggi. Sehingga diperlukan enzim yang tahan terhadap suhu tinggi (termostabil). Enzim termostabil yang dihasilkan oleh bakteri termofilik sangat dibutuhkan dalam bidang industri karena kemampuannya yang tahan terhadap suhu tinggi. Tujuan dari penelitian ini untuk mengetahui sumber karbon dan sumber nitrogen optimum terhadap isolat bakteri termofilik TUA-504 produksi protease. Metode penelitian dilakukan secara eksperimen dengan menggunakan variasi sumber karbon dan sumber nitrogen konsentrasi 1% untuk mendapatkan kondisi optimum dari aktivitas protease. Hasil dari penelitian ini didapatkan kondisi optimum isolat bakteri termofilik TUA-504 dalam memproduksi protease yaitu pada sumber karbon glukosa dan sumber nitrogen NaNO3

Abstract

Protease is a vital enzyme used in industries such as detergents, pharmaceuticals, and animal feed, with a growing demand in the enzyme market. Endophytic microorganisms can produce stable proteases with a rapid synthesis process. This study optimized conditions of temperature, pH, salinity, agitation, and nutrient sources for protease production by EUA-136 and EUA-139 bacterial isolates. The research used Response Surface Methodology (RSM) with a Central Composite Design (CCD) in Design Expert Software 13.1 to identify optimal conditions and the bacterial isolates. The optimum conditions for the EUA-136 bacterial isolate to produce protease were 3% inoculum at 30 ºC, pH 7, 28.5 ppt salinity, and 150 rpm agitation. For the EUA-139 bacterial isolate, the optimum conditions were a carbon source of 1% (v/v) maltose, a nitrogen source of 1% (v/v) KNO3, casein as the inducer, and an inoculum concentration of 7.5% (v/v). Molecular identification of isolates EUA-136 and EUA-139 revealed similarities to Bacillus cereus strain 3TC-3 and Bacillus paramycoides 3665, respectively.

Abstract

Coconut pulp has potential as animal feed but is limited in poultry diets due to its high fiber content and low protein levels. Fermentation using indigenous microorganisms offers a practical approach to improving its nutritional value. This study aimed to evaluate the nutritional quality of coconut meal using indigenous microorganisms (IMOs). The initial phase of the study involved producing IMOs, followed by measuring pH, total colony, and cellulase and mannanase enzyme activities. IMOs were applied to ferment coconut meal using a completely randomized design with four treatments (0, 7, 14, and 21 days) and four replications. Observed parameters included cellulase and mannanase enzyme activities, pH, crude fiber, crude protein, crude fat, and nitrogen-free extract. The characteristics of IMOs were analyzed descriptively, while enzyme activities and the nutritional content of coconut meal were analyzed using ANOVA and Duncan’s Multiple Range Test. The results showed that IMOs had a pH of 3.3, were dominated by lactic acid bacteria with a total colony of 5.4 × 10¹² CFU/mL, had cellulase activity of 2.1 U/mL, and had mannanase activity of 7.73 U/mL. Cellulase activity at 7, 14, and 21 days was significantly higher (P<0.01), while mannanase activity was highest at 21 days (P<0.01), with the lowest pH observed at 14 and 21 days. The lowest crude fiber and crude fat content, as well as the highest crude protein content (P<0.01), were observed at 14 days of fermentation. Fourteen days of fermentation were identified as the optimal duration for enhancing the nutritional quality of coconut pulp.

Abstract

ABSTRAK. Probiotik konsorsium merupakan kombinasi dari berbagai mikroorganisme yang saling bekerja sehingga menghasilkan manfaat yang lebih baik dibandingkan probiotik tunggal. Tujuan penelitian adalah untuk mendapatkan dosis pemberian probiotik konsorsium yang terbaik dilihat dari uji organoleptik daging broiler meliputi tekstur, rasa, warna dan aroma. Penelitian menggunakan ayam broiler yang dipelihara selama 35 hari dengan pemberian level probiotik konsorsium berbeda dalam air minum. Pakan yang digunakan adalah BR 1 Wonokoyo. P0 = pemberian level 0 ml probiotik konsorsium, P1 = pemberian level 0,5 ml probiotik konsorsium, P2 = pemberian level 1 ml probiotik konsorsium dan P3 = pemberian level 1,5 ml probiotik konsorsium dengan 5 kali pengulangan. Parameter yang diamati adalah tekstur, rasa, warna dan aroma daging. Hasil penelitian ini menunjukkan pemberian probiotik konsorsium pada ayam broiler memberikan pengaruh berbeda sangat nyata (P<0,01) terhadap tekstur daging, rasa daging, warna daging dan aroma daging. Hasil penelitian ini disimpulkan pemberian level 1 ml probiotik konsorsium pada ayam broiler diperoleh tekstur daging 4,40; rasa daging 4,31; warna daging 3,82 dan aroma daging 4,14.Kata kunci: Probiotik, konsorsium, tekstur, rasa, warnaThe Effect of Probiotic Consortium Dosage on the Organoleptic Quality of Broiler MeatABSTRACT. Consortium probiotics are a combination of various microorganisms that work together to produce better benefits than single probiotics. This research aimed to obtain the best dose of probiotic consortium based on organoleptic tests of broiler meat, including texture, taste, color, and aroma. This study used broiler chickens that were reared for 35 days by administering different levels of probiotic consortium in drinking water. The feed used is BR 1 Wonokoyo. P0 = Giving probiotic consortium level 0 ml, P1 = Giving probiotic consortium level 0.5 ml, P2 = Giving probiotic consortium level 1 ml, and P3 = Giving probiotic consortium level 1.5 ml with 5 times covered. The parameters observed were texture, taste, color, and aroma of meat. The results of this study showed that giving a probiotic consortium to broiler chickens had a very significantly different effect (P<0.01) on meat texture, meat taste, meat color, and meat aroma. From the results of this study, it was concluded that by administering 1 ml of the probiotic consortium to broiler chickens, the meat texture was 4.40, the meat taste was 4.31, the meat color was 3.82, and the meat aroma was 4.14.

Abstract

This study aimed to isolate, screen, and partially characterize protease-producing thermophilic bacteria from the Padang Dama hot spring, Solok, Indonesia. The main issue addressed in this study is the limited information regarding thermophilic bacteria capable of producing protease enzymes from this geothermal environment, despite their potential use in industrial processes requiring stable enzymes. Isolation was carried out using Nutrient Agar followed by incubation at 50 °C for 24 hours. Screening of protease activity was performed using Skim Milk Agar based on clear zone formation, and the proteolytic index (PI) was calculated to evaluate enzyme activity. Selected isolates were further characterized through macroscopic, microscopic, and biochemical analyses. The results showed that three isolates exhibited proteolytic activity, with isolate TUA-109 showing the highest proteolytic index, indicating superior enzymatic potential compared to other isolates. Partial characterization revealed that isolate TUA-109 is a Gram-negative, rod-shaped bacterium with positive catalase activity and motility, which are important traits supporting its adaptability and potential for enzyme production under extreme conditions. These findings highlight the potential of thermophilic bacteria from the Padang Dama hot spring as sources of protease enzymes, with isolate TUA-109 identified as a promising candidate for further investigation, particularly for industrial enzyme production. Future studies are recommended to optimize enzyme production and evaluate its application in industrial processes.

Abstract

The research was conducted to study the biodegradation capability on palm oil sludge substrate by Phanerochaete  chrysosporium   with increasing urea  as  nitrogen  source. The  experiment was  designed using Completely Randomized Design (CRD) with 5 treatments and 4 replications, namely : U-0 = 0,0 %  Urea,    U-1=      0,5  %  Urea,    U-2  =  1,0  %  Urea,    U-3  =  1,5  %  Urea  and  U-4  =  2,0  %  Urea. The observed variables were the amount of spore, C/N ratio, pH, crude fiber, crude protein, lignin, cellulose and enzyme activities. The result of this study showed that added urea were significantly (P<0.01) crude fiber, crude protein, lignin, cellulose and enzyme activities. The use of urea on the first level amounting to  1.5  %  in palm  oil  sludge  can  help  Phanerochaete  chrysosporium   to  reduce  crude  fiber to (30,71%),  lignin (29.89  %),  cellulose (36.42  %) and it  can  increase  the  content  of crude  protein    to    (34.50  %), reduction sugar and enzyme activities.

Abstract

West Sumatra Province has relatively extensive agriculture with feed ingredients such as rice straw, Rumput Gajah and legume groups that can be used for beef cattle needs. The total area of Padang City is 694.96 km², and more than 60% of this area, about ± 434.63 km² is a hilly area covered by protected forests, while the rest is an effective urban area. While the topography of the city varies, 49.48% of the land area of Padang City is in a slope of more than 40% and 23.57% is in the slope of the slope. The eastern region consists of several sub-districts, starting from north to south, respectively from Koto tangah, Pauh, Kuranji, and Lubuk sub-districts. The Pauh region, such as the village of Lambung Bukik, belongs to the Pertides forum program (universities for villages) which collaborates with the 2019 Chancellor's MOU with the Ministry of Education PDTT) to prepare and implement programs to help build villages. Some of their income is obtained from raising livestock and farming. In general, both agricultural and livestock businesses are still carried out traditionally, so it is not surprising that the results obtained are relatively low. Cows are only ground or tethered at night, while in the afternoon they are released to look for food on the grass or in the sleeping area around the village. Only a small amount of effort has been made to raise livestock intensively by supplying and providing sufficient and regular food. The low level of livestock production is caused by the lack of knowledge of farmers in the proper way of raising livestock, also because of the lack of food both forage and the high price of concentrate. With the increase in the livestock population, it certainly requires more and sufficient forage throughout the year. However, the provision of forage has experienced serious obstacles. One of them is the existence of a dry season that causes a decrease in forage production. Therefore, the business of developing beef cattle will be more profitable if you can find alternative substitutes for conventional forage with the use of silos as feed fermentation sites. The use of agricultural products (such as rice straw) and other food crops as animal feed is expected to address the above problems. This is possible because cattle breeding is generally integrated with other farming businesses, especially food crops (rice fields) so that the results of agricultural follow-up will be available throughout the year. Therefore, there is a need for a kind of touch of feed technology with the use of agro-industry by products such as straw which are proven to be available at all times. The implementation of the application of rice straw ammoniation technology in the field with the direct administration of ammonia which has been removed and chicken manure is given. Participation and motivation of farmer groups in participating in a series of service activities is very high. Because all this time there has never been any guidance related to the technical and management aspects in the maintenance of beef cattle. Farmers already know how to manage fattening beef cattle such as giving concentrates such as coconut cake, soybean meal, fish meal, tofu pulp and bran and giving premix minerals.

Abstract

Abstract. Makmur M, Zain M, Marlida Y, Khasrad, Jayanegara A. 2019. Fatty acids composition and biohydrogenation reduction agents of tropical forages. Biodiversitas 20: 1917-1922. The study was conducted to determine the composition of fatty acids, measured rumen biohydrogenation reduction agents (total phenols and total tannins) content and selected promising plants in various species of tropical forages. Ten species of tropical forages, namely, Panicum maximum, Cynodon plectostachyus, Pennisetum purpurephoides, Pennisetum purpureum, Brachiaria decumbens, Glyricidia sepium, Calliandra calothyrsus, Stylosanthes guaianensis, Leucaena leucocephala and Indigofera zollingeriana were used in this study. The fatty acids composition (% of total identified fatty acids) which were dominant in grasses were C18: 3n-3 (29%), C16: 0 (28%) and C18: 2n-6 (23%). Whereas in legumes, the significantly higher composition of fatty acids was C18: 3n-3 (42%) followed by C16: 0 (17%) and C18: 2n-6 (17%). The average poly-unsaturated fatty acids (PUFA) composition in grasses was relatively lower (44.6%) than legumes (59%). Likewise the content of total phenols and total tannins (g/100g DM) of grasses (0.91 and 0.41) and legumes (1.72 and 0.70). The selection of the forage plant species was based on the criteria of PUFA composition and biohydrogenation reduction agents using TOPSIS method. The results obtained show that B. decumbens (grass) and I. zollingeriana (legume) had the highest preference value of 0.74 and 0.87, respectively. In conclusion, B. decumbens and I. zollingeriana are forage species that have potential to provide healthier ruminant products.
