Abstract

Oil palm (Elaeis guineensis Jacq.) is a plant that is native to Africa and South America. This plant can grow well in tropical areas, such as Indonesia. Indonesia is currently one of the largest producers of palm oil worldwide. As the area of palm oil plantations increases, the potential for waste from this plant continues to increase. The potential for palm oil leaf waste production in Indonesia is 658 kg dry matter/hectare/year. Palm oil leaf waste has been widely processed into organic fertilizers and animal feeds. This plant has the potential to be developed into a source of traditional medicinal ingredients. Based on the abundant sources of palm oil leaves and their pharmacological potential, palm oil leaves can be used as a source of raw materials for traditional medicine. This study aimed to compare the antioxidant activities of oil palm leaf extracts obtained by maceration and reflux to determine which extraction method produces the best antioxidant activity. Extraction was carried out using 70% ethanol solvent by maceration and reflux. The results of the phytochemical screening showed that oil palm leaves contain flavonoids, polyphenols, saponins, quinones, steroid-triterpenoids, and monoterpene-sesquiterpenes. Antioxidant activity using a UV-Visible spectrophotometer with DPPH free radicals showed that the extract obtained by maceration and reflux methods had IC50 values of 46.61 ± 1.76 ug/mL and 55.21 ± 2.91 ug/mL respectively. Therefore, extraction by maceration produces a better antioxidant activity.  Keywords: Palm leaves; Traditional medicine; Extraction; Active compound stability; Antioxidant

Abstract

Developing Orthosiphon aristatus (Blume) Miq. leaf extract of purple varieties into granule dosage forms is expected to facilitate consumption, storage, and can extend the shelf life of the product. This study aims to develop the formulation and evaluation of granule preparations of O. aristatus leaf extract of purple varieties, with a comparison of lactose and maltodextrin fillers to produce the best formula. The granule dosage forms were prepared using wet granulation method. Furthermore, the variation of granule preparation formula consists of F1 (extract with lactose), F2 (extract with maltodextrin), F3 (extract with a combination of lactose and maltodextrin 1:1), F4 (freeze dried extract with lactose), F5 (freeze dried extract with maltodextrin), and F6 (freeze dried extract with a combination of lactose and maltodextrin 1:1). The physical evaluation of granules comprised moisture content testing, flowability, compressibility index, particle size distribution, and granule dissolving time. Results from the physical evaluation of granules over 28 days of storage indicated that all six formulas met the criteria for good granules, with F3 emerging as the optimal formula due to its superior physical stability and visually appealing appearance upon dissolution. To conclude, this study demonstrates that O. aristatus leaf extract can be effectively formulated into instant granule preparations suitable for herbal beverage or alternative medicine. Moreover, by modifying the filler material, the physical stability of the granule preparation can be maintained.

Abstract

Oxidative stress is caused by an increase or decrease in the antioxidant defense activity. Orthosiphon aristatus (Blume) Miq and Stevia are two plants that contain antioxidant compounds and can bind free radical compounds. The combination of Orthosiphon aristatus (Blume) Miq and Stevia was expected to increase antioxidant activity. This study aimed to determine the antioxidant activity of a combination of the Orthosiphon aristatus (Blume) Miq white-purple variety and Stevia rebaudiana Bertoni extract. Antioxidant activity was tested using the FRAP method with a UV-visible spectrophotometer at a wavelength of 710 nm. The results of phytochemical screening showed that both plant extracts and dry material contained secondary metabolite compounds, including flavonoids, polyphenols, saponins, quinones, steroid-triterpenoids, and monoterpenes-sesquiterpenes. A qualitative test relating to antioxidants using TLC showed positive antioxidant activity, which was proven by the presence of green or blue light on the TLC plates. This was due to electron transfer in the compound, which reduced Fe3+ to Fe2+. On the other hand, quantitative measurements using the FRAP method showed the antioxidant capacity values consecutively, F1 combination extract (1:1) of 43.725 mg of QE/g extract, F2 combination extract (2:1) of 57.963 mg of QE/g extract, and combination extract of F3 (3:1) of 64.849 mg of QE/g extract. The extract combination of F3 showed the best formula, with a value of 64.849 mg QE/g extract.  Keywords: Orthosiphon aristatus (Blume) Miq, Stevia, antioxidants, FRAP
