Abstract

Oxidative stress is caused by an increase or decrease in the antioxidant defense activity. Orthosiphon aristatus (Blume) Miq and Stevia are two plants that contain antioxidant compounds and can bind free radical compounds. The combination of Orthosiphon aristatus (Blume) Miq and Stevia was expected to increase antioxidant activity. This study aimed to determine the antioxidant activity of a combination of the Orthosiphon aristatus (Blume) Miq white-purple variety and Stevia rebaudiana Bertoni extract. Antioxidant activity was tested using the FRAP method with a UV-visible spectrophotometer at a wavelength of 710 nm. The results of phytochemical screening showed that both plant extracts and dry material contained secondary metabolite compounds, including flavonoids, polyphenols, saponins, quinones, steroid-triterpenoids, and monoterpenes-sesquiterpenes. A qualitative test relating to antioxidants using TLC showed positive antioxidant activity, which was proven by the presence of green or blue light on the TLC plates. This was due to electron transfer in the compound, which reduced Fe3+ to Fe2+. On the other hand, quantitative measurements using the FRAP method showed the antioxidant capacity values consecutively, F1 combination extract (1:1) of 43.725 mg of QE/g extract, F2 combination extract (2:1) of 57.963 mg of QE/g extract, and combination extract of F3 (3:1) of 64.849 mg of QE/g extract. The extract combination of F3 showed the best formula, with a value of 64.849 mg QE/g extract.  Keywords: Orthosiphon aristatus (Blume) Miq, Stevia, antioxidants, FRAP

Abstract

Community service activities carried out by students of Universitas Jenderal Achmad Yani were carried out on August 29, 2022, taking place in a medicinal plant garden, Faculty of Pharmacy, Universitas Jenderal Achmad Yani which was attended by UMKM Spice Nusantara Bandung participants. The socialization carried out was the presentation and delivery of post-harvest processing materials (wet sorting, washing, drying, and dry sorting) and the standardization of traditional raw materials. Conventional medicine is a form of treatment efforts in the community to support health which is currently being recommended by the government. Communities can use the toga as an economical and efficient measure to maintain body immunity because the community can obtain and use the toga quickly and simply to maintain family health. The postharvest process greatly determines the existence of active compounds that are efficacious for treatment. Incorrect postharvest management can change, reduce, or destroy the active substance of a plant into a substance that has no therapeutic effect and can even endanger health. The results of the participant's level of understanding were carried out post-test with the results of what is the purpose of post-harvest processing (100%), the sequence of post-harvest processing (80%), what is the general drying temperature (100%), a medicinal plant has good quality and efficacy then we need to know (80%), and what is the function of the washing process (90%) with the conclusion that socialization activities can significantly increase the knowledge of participants from MSMEs regarding planting, post-harvest processing, and the properties of medicinal plants significantly.

Abstract

Moringa oleifera Lam. is recognized for its versatile nutritional and medicinal benefits, largely attributed to its rich polyphenolic content, which includes bioactive compounds with significant antioxidant, anticancer, antimicrobial, and antidiabetic properties. This study addresses the gap in knowledge regarding the enhancement of phenolic compounds in Moringa leaves through innovative biotechnological methods. The primary objective is to analyze the extraction and quantification of polyphenols from Moringa leaves subjected to boiling and fermentation processes to produce kombucha. Using the Folin-Ciocalteu method, quantified the total polyphenol content, revealing that boiled Moringa leaves contained 67.66 ± 2.79 mg/g of polyphenols, while the resulting kombucha tea exhibited a higher concentration at 70.66 ± 2.79 mg/g. These findings demonstrate the potential of Moringa leaves as not only a dietary supplement but also as a functional ingredient in health-promoting beverages, such as kombucha. The implications extend to enhancing the nutritional value of plant-based diets and establishing Moringa as a sustainable antioxidant source. This research underscores the importance of advanced biotechnological approaches in optimizing extraction and cultivation methods, thus potentially increasing phenolic yields. By providing crucial insights into the functional applications of Moringa leaves, this study contributes to the broader fields of plant biotechnology and food science, encouraging further exploration into their health benefits and utility in promoting sustainable nutrition.

Abstract

The present study investigates the antioxidant properties of in-vitro cultured leaves of cat's whiskers (Orthosiphon stamineus) compared to the well-known antioxidant quercetin. Antioxidants are crucial for neutralizing free radicals and play a significant role in protecting cells from oxidative stress, a contributing factor to various diseases and aging processes. The research aims to quantify and compare the radical scavenging activity of extracts from the in vitro cultured leaves and quercetin, using the DPPH method, a widely recognized assay in antioxidant research. Extraction involved macerating in-vitro cultured cat's whiskers leaves with 96% ethanol, which facilitates the dissolution of active phytochemicals. Antioxidant activity was subsequently assessed through the DPPH assay, where colorimetric change indicates the extract’s scavenging ability towards free radicals. The findings revealed that the in vitro cultured leaves extract exhibited a potent antioxidant activity, with an IC50 value of 0.74 μg/mL, significantly lower than quercetin’s IC50 value of 7.51 μg/mL. These results highlight the potential of in-vitro cultured cat's whiskers as a natural antioxidant source, suggesting possible applications in the food and pharmaceutical industries, where combating oxidative stress is essential for health maintenance and disease prevention. Further research is necessary to elucidate the specific biochemical pathways and mechanisms underpinning the antioxidant capacity of the in vitro cultured leaves extracts.

Abstract

Temulawak (Curcuma xanthorrhiza Roxb) a plant indigenous to Indonesia, is increasingly recognized for its medicinal properties, formulating a cornerstone of traditional medicine with annual usage marking a significant growth rate of approximately 5.4%. As awareness of its therapeutic potential expands, too does the demand for this valuable resource, necessitating enhanced efforts in its propagation and the extraction of its bioactive compounds. This study investigates the efficacy of different growth regulators in the induction of callus tissue from C. xanthorrhiza through plant tissue culture, a key method for regenerating valuable plant material. Specifically, the research focuses on the comparative effects of natural growth regulators, particularly water extracts from Phaseolus aureus, and synthetic growth regulators such as 2,4-Dichlorophenoxyacetic Acid (2,4-D) and Indole-3-butyric Acid (IBA) in varied concentrations. The experimental design included varying concentrations of P. aureus water extract at 10%, and 15%, alongside a fixed concentration of the synthetic agents. Findings indicated that synthetic growth regulators, particularly the combination of 2,4-D and IBA at a concentration of 0.4 ppm each, yielded superior callus proliferation compared to the natural extract treatments. However, P. aureus aqueous extract showed positive effects also on callus emergence and on increasing explant size when used at 10% concentration in Murashige-Skoog medium. Consequently, the study underscores the potential of integrating both natural and synthetic growth regulators to enhance callus induction and ultimately facilitate the sustainable use of C. xanthorrhiza as a medicinal resource.
