Abstract

Sea urchins are organisms that have antibacterial, antitumor, and anticancer abilities. The gonads can be used as a food source because they store 28 kinds of amino acids, vitamin B complex. This research aims to determine the type of bacteria from the gonads of sea urchins (Diadema setosum) which have inhibitory power against the growth of Pseudomonas fluorescens and Staphylococcus aureus bacteria. This research used an exploratory method which was analyzed descriptively, using stages in which sea urchin samples were taken and antibacterial tests against Pseudomonas fluorescens and Staphylococcus aureus from sea urchin gonads using the spread method. The media used for the process of isolating bacteria from the gonads of the sea urchin Diadema setosum is Nutrient Agar (NA) media. Isolate colonies that grew after incubation for 24 hours were separated based on differences in colony morphology. Morphological characterization of bacterial cells begins with gram staining. The diameter of the inhibition zone in the test bacteria shows different shapes. The research obtained 8 bacterial isolates, namely isolates B1-B8 which were gram-negative. The eight bacterial isolates have the potential as antibacterials against the test bacteria P. fluorescens and S. aureus. Isolates B1, B2, B3, B7 are similar to the genus Proteus, while isolates B4, B5, B6, and B8 are similar to the genus Citrobacter. The diameter of the largest inhibition zone in the P.fluorescens test bacteria, namely isolate B1, was 9.03 mm. Meanwhile, the smallest diameter for isolate B7 was 6.97 mm. In the S.aureus test bacteria, the largest diameter in isolate B1 was 11.09 mm. Meanwhile, the smallest diameter for isolate B4 was 8.57 mm.

Abstract

Pakoba fruit is one of the endemic flora of North Sulawesi which is rich in nutrients and has the potential to be fermented. The fermentation process occurs due to the bacteria in the fruit, including lactic acid bacteria (LAB). One of the interesting products from LAB fermentation is that it can produce secondary metabolites which have the potential to produce exopolysaccharides, namely high molecular weight and biodegradable polymers. This research aims to determine whether LAB can be obtained from fermentation of Pakoba marination and what types of LAB produce exopolysaccharides based on their phenotypic characteristics. This research uses a qualitative descriptive method to characterize the type of LAB and observe the exopolysaccharides produced by the bacteria. This research succeeded in isolating and identifying 10 LAB isolates based on their morphological and biochemical characters. Four isolates named MP(1)5.2, MP(1)7.1, MP(3)6.3, MP(3)7.2 can produce exopolysaccharides with a soft character but not mucus, where bacterial exopolysaccharides are believed to increase probiotics in the digestive tract. Genus level recognition (generic assignment) based on profile matching revealed that these four bacteria were from the genus Lactobacillus.

Abstract

Tonsealama is a village that has the potential to develop an environmentally friendly organic-based agribusiness farmer group business that has maximum agribusiness products and high economic value. The livelihood of the villagers is dominated by farmers and farm laborers and only a small part is engaged in services. The development of agribusiness entrepreneurship of raw chilli bara F1 variety for women's farmer groups in Keroit village is deemed necessary to be developed, in order to utilize idle land and organic household waste products. This is also a business opportunity to increase household economic income and the welfare of women's farmer groups and their members. Based on the above, the Community Partnership Program for Women Farmers Group in Tonsealama Village was implemented. This program includes the use of agricultural technology products in the form of ULiBiCo (Unima Liquid Biofertilizer Combination) and Effective Microorganism (EM4) applications that help the decomposition process of chicken manure waste into quality organic fertilizer, and combined with the use of silver black plastic mulch technology (MPHP) and bioinsecticide-based pest control, it turns out to be able to provide good yields in the cultivation of cayenne pepper plants of the bara F-1 variety. The morphological character of the first harvest obtained is the quality of the cayenne pepper fruit, which has a sharp fruit, with a length of ± 3 cm, a diameter of ± 0.4 cm, shiny red skin, spicy taste, and can be harvested at the age of 78 - 100 days after planting (HST), the average yield of this cayenne pepper is 0.85 - 1.0 kg per plant, with a weight per fruit ranging from 0.9 - 1.8 grams and has a potential yield of 8.9 - 10 tons / ha. ABSTRAK Tonsealama merupakan desa yang memiliki potensi untuk dikembangkannya sebuah usaha kelompok tani agribisnis berbasis organik ramah lingkungan yang memiliki produk hasil agribisnis yang maksimal dan bernilai ekonomi tinggi. Mata pencaharian penduduk desa masih didominasi oleh para petani. Pengembangan wirausaha agribisnis cabai rawit varietas bara F1 pada kelompok tani wanita di desa Tonsealama dirasa perlu untuk dikembangkan, guna memanfaatkan lahan tidur dan produk limbah organik rumah tangga. Hal ini juga merupakan sebuah peluang usaha untuk meningkatkan pendapatan ekonomi rumah tangga dan kesejahteraan kelompok tani wanita beserta anggotanya. Berdasarkan hal tersebut di atas, maka dilaksanakanlah Program Kemitraan Masyarakat Kelompok Tani Wanita di Desa Tonsealama. Program ini meliputi pemanfaatan produk teknologi pertanian berupa aplikasi ULiBiCo (Unima Liquid Biofertilizer Combination) dan Effective Microorganism (EM4) yang membantu proses dekomposisi limbah kotoran ayam menjadi pupuk organik berkualitas, dan dikombinasikan dengan pemanfaatan teknologi mulsa plastik hitam perak (MPHP) serta pengendalian hama berbasis bioinsektisida, ternyata mampu memberikan hasil panen yang baik dalam budidaya tanaman cabai rawit varietas bara F-1. Karakter morfologi hasil panen pertama yang diperoleh yaitu kualitas buah cabai rawit memiliki buah lancip, dengan panjang ± 3 cm, diameter ± 0,4 cm, kulit buah berwarna merah mengkilap, berasa pedas,  dan dapat dipanen pada umur 78 - 100 hari setelah tanam (HST), rata-rata hasil cabai rawit ini yaitu 0,85 - 1,0 kg per tanaman, dengan bobot per buah berkisar 0,9 – 1,8 gram dan memiliki potensi hasil 8,9 – 10 ton/ha.

Abstract

Lactic Acid Bacteria (LAB) are a group of Gram-positive, cocci or rod-shaped microorganisms characterized by their catalase-negative phenotype. These bacteria are known to produce various antibacterial compounds, including organic acids, hydrogen peroxide, carbon dioxide, and bacteriocins, which exhibit bacteriostatic or bactericidal properties against pathogenic bacteria. The aim of this study is to isolate LAB that produce antibacterial compounds during the fermentation of Pakoba fruit (Syzygium luzonense) into sweets, with a focus on inhibiting the growth of Escherichia coli and Staphylococcus aureus. This research employs a quantitative descriptive method, involving the isolation of LAB from candied Pakoba fruit samples and subsequent testing of their antibacterial activity against the aforementioned pathogens. The barrier zone formed around the wells is then measured. The data from the study showed that BAL isolation from Pakoba fruit candied was obtained from as many as 10 isolates, and antibacterial activity tests showed that 10 BAL isolates produced antibacterial compounds with inhibitory diameters of 7.6 – 15.3 mm. The isolates with the greatest antibacterial activity are  LAB isolate MP(1)5.2  for E. coli and LAB isolate MP(3)6.3 isolates for S. aureus. Based on the results of identification using the profile matching method, it was shown that the selected LAB isolates, namely MP(1)5.2 and MP(3)6.3, were classified in the genus Lactobacillus with the characteristics of stem cell shape, single cell arrangement, catalase-negative, non-motile, did not form spots, and did not produce gas from glucose.

Abstract

Tridax procumbens L. and Aloe vera L. are plants commonly used in the treatment of burns. This study was conducted to determine the effect of giving a combination gel of T. procumbens and A. vera leaf extract on second-degree burn healing in mice. This study used 24 male mice (Mus musculus) which were divided into four treatment groups, namely positive control (KP, Bioplacenton®); negative control (KN, gel base); leaf extract of T. procumbens 0.5% and A. vera 0.5% (P1); and leaf extract of T. procumbens 1% and A. vera 0.5% (P2). The process of inducing burns on the backs of mice requires the use of a 1x1 cm iron, which is subjected to heat in a fire for three minutes. The iron is placed on the mice's back for five seconds, which leads to the development of a second-degree burn. The gel was applied twice a day for 21 days. The highest level of burn wound healing was recorded with the Bioplacenton® treatment, which achieved a full recovery of 100% on day 15. Following this, the combined gel treatment of 1% T. procumbens leaf extract and 0.5% A. vera also reached 100% healing by day 18. While it may not be as potent as Bioplacenton®, the gel formulation containing 1% T. procumbens and 0.5% A. vera leaf extract presents a viable alternative for treating second-degree burns.

Abstract

Turtles They are among the world's protected animals and one of the rare, almost extinct species. The preservation of their habitat and knowledge of the traits of turtle-friendly habitats are two ways that the rescue and protection of these turtles must go on. For rescue operations, turtle sustainability, and sustainable coastal management, research is being conducted on the features of the turtle nesting habitats on Popareng Beach. This research aims to determine the characteristics of turtle nesting habitats and identify the parameters that hinder turtles from nesting at Popareng Beach, Tatapaan District, South Minahasa Regency. The method used in this research is the descriptive-exploratory method. With characteristic parameters consisting of beach width measurement, beach slope, sand temperature, sand humidity, sand texture, beach vegetation, and the presence of predators. From the research results, it shows that station 1 and station 2 have results that are suitable for turtle nesting habitats, with values obtained at station 1 being 88.33% and station 2 being 92%. Meanwhile, at station 3, with an evaluation score of 72%, it is considered fairly suitable and the station has tolerable limitations.

Abstract

Red beans are a horticultural crop belonging to the annual legume group that grows upright and has a fairly high nutritional content. To obtain maximum growth and yield from red beans, several factors must be considered, including balanced fertilizer application. The factor of providing organic fertilizer from chicken manure with the right dosage is the target of this study. This study aims to determine the effect of red bean growth on the dosage of chicken manure fertilizer. This study was conducted from December 2024 to February 2025 and was carried out on the plantation land of the Department of Science Education, Faculty of Mathematics, Natural Sciences and Earth Sciences, Manado State University. The method used is a quantitative method with a type of experiment in the form of a Randomized Block Design (RAK) with five treatment combinations with each repeated three times. A0 = 0 (control), A1 = 1.00 kg, A2 = 1.50 kg, A3 = 2.00 kg, A4 = 2.50 kg, so there are 15 experimental bed units, and the data obtained are then analyzed using analysis of variance (ANOVA). The results of this study indicate that good plant height growth, number of leaves and stem diameter are found in treatment A1 with a dose of 1.00 kg of chicken manure fertilizer and A2 with a dose of 1.50 kg of chicken manure fertilizer because there is the most maximum growth.
