Abstract

This research aims at finding out characterization of a hydrolyzed fatty acid from moringa seeds oil (Moringa oleifera). Samples analyzed are dried and wet seeds which extracted through n-hexane to obtain oil. The extracted oil is analyzed for acid value and saponification value. Then, the hydrolysis of extracted oil with KOH solution and H2SO4 catalyst becomes free fatty acids, fatty acids analysis is then converted to be methyl ester using CH3OH solvent and H2SO4 as catalyst. This research uses gas chromatography mass spectrometry analysis, fatty acids of dried moringa seeds were lauric acid, paltoleic acid, palmitic acid, oleic acid, stearic acid, and arachidic acid. Meanwhile, fatty acids of wet moringa seeds were paltoleic acid, palmitic acid, oleic acid, stearic acid, eicosanoid acid, arachidic acid, behenic acid, and lignoceric acid. The highest component of methyl ester on both moringa seeds are methyl oleate with each presentation is 38,08% and 38,84%Keywords: Moringa seeds, Fatty acid, GC-MSPenelitian ini bertujuan untuk mengetahui karakterisasi asam lemak hasil hidrolisis biji kelor (Moringa oleifera) dengan metode kromatografi gas-spektroskopi massa. Sampel yang digunakan dalam penelitian ini adalah biji kelor kering dan basah. Sampel biji kelor diekstraksi menggunakan n-heksan untuk memperoleh minyak. Ditentukan bilangan asam dan bilangan penyabunan. Menghidrolisis minyak hasil ekstraksi dengan larutan KOH dan katalis H2SO4 menjadi asam lemak bebas, analisis asam lemak kemudian dikonversi menjadi metil ester dengan menggunakan pelarut CH3OH dan H2SO4 sebagai katalis. Penelitian ini menggunakan Kromatografi Gas-Spektroskopi Massa untuk mengidentifikasi asam lemak dalam sampel. Dari analisa Kromatografi Gas-Spektroskopi Massa yang telah dilakukan, asam lemak minyak biji kelor kering yang dihasilkan adalah asam laurat, asam palmitoleat, asam palmitat, asam oleat, asam stearat, dan asam arakidat. Sedangkan untuk asam lemak minyak biji kelor basah yang dihasilkan adalah asam palmitoleat, asam palmitat, asam oleat, asam stearat, asam eikosenat, asam arakidat, asam behenat, dan asam lignoserat. Dimana komponen terbesar metil ester pada biji kelor kering dan biji kelor basah adalah metil oleat dengan persentasi masing-masing 38,08% dan 38,84%.Kata Kunci: Biji Kelor, Asam Lemak, KG-SM

Abstract

Eceng gondok adalah salah satu tumbuhan bahan serat alam yang memiliki kandungan selulosa yang cukup tinggi. Kadar selulosa yang tinggi dapat digunakan dalam pembuatan Karboksimetil Selulosa Sodium (Na-CMC). Tujuan dari penelitian ini adalah: untuk mengetahui karakteristik Karboksimetil Selulosa Sodium (Na-CMC) dari tanaman eceng gondok. dan mendapatkan variasi media reaksi terbaik dalam proses sintesis. Penelitian ini terdiri dari tiga tahap yaitu tahap pertama ekstraksi selulosa yang terdiri dari (dewaxing, dehemisellulose dan bleaching), tahap kedua yaitu tahap sintesis yang terdiri dari tahap pencampuran menggunakan media reaksi etanol-isobutanol (20:80, 50 : 50, 80:20). Tahap alkalisasi menggunakan NaOH 10% b / v, tahap karboksimetilasi menggunakan ClCH2COONa. Tahap ketiga adalah karakterisasi CMC yang terdiri dari uji organoleptik, sifat fisikokimia (pH, susut pengeringan sampel, viskositas dan derajat substitusi), uji kadar NaCl, uji kemurnian, analisis FT-IR. Berdasarkan hasil penelitian didapatkan bahwa karakteristik CMC mendekati karakteristik standar Na-CMC dengan campuran media reaksi etanol-isobutanol 20:80 (v / v) dengan nilai DS 0,8560, kemurnian 93,7463% level, pH 6,5 dan viskositas. 302 cP.

Abstract

Laju sedimentasi dari beberapa sungai yang bermuara di danau Limboto semakin tinggi sehingga menyebabkan pendangkalan. Pertumbuhan eceng gondok yang sangat cepat menutupi permukaan danau menyebabkan perubahan ekosistem danau dan kerugian finansial bagi nelayan lokal. Program Kuliah Kerja Nyata Pembelajaran Pemberdayaan Masyarakat (KKN PPM) di kelurahan Kayubulan bertujuan untuk mengedukasi masyarakat melalui implementasi teknologi pengolahan eceng gondok dan sedimen danau Limboto. Kegiatan pemberdayaan masyarakat melalui 5 (lima) tahapan yakni tahap persiapan, tahap sosialisasi, tahap pelatihan, tahap pendampingan dan tahap evaluasi. Metode yang digunakan berupa  FGD (forum group discussion), perencanaan partisipatif dan metode praktek. Hasil kegiatan ini memberi pemahaman pada masyarakat tentang pentingnya menjaga danau. Pemahaman masyarakat kelurahan Kayubulan tentang pengolahan eceng gondok menjadi biogas, briket, pupuk dan sedimen menjadi batako, batu bata dan pot tanaman meningkat secara keseluruhan hingga 48%. Kegiatan ini berdampak positif pada peningkatan pengetahuan dan ketrampilan kelompok Alata sebagai mitra sasaran.

Abstract

Ikan nike adalah salah satu jenis ikan endemik di provinsi Gorontalo. Ikan nike ini memiliki keunikan tersendiri yaitu siklus pemunculannya terjadi dalam jumlah besar pada lokasi tertentu terdapat di muara Sungai Bone, keberadaan nike yang musiman perlu dimanfaatkan secara baik-baik. Tujuan penelitian ini menganalisis aktivitas antioksidan dari hidrolisat protein ikan nike (Awaous melanocephalus) secara enzimatis menggunakan enzim bromelin variasi konsnetrasi 4%, 5% dan 6% (b/v) serta variasi waktu hidrolisis selama 2, 4 dan 6 jam. Konsentrasi optimum enzim bromelin dalam pembuatan hidrolisat protein yaitu 5% (b/v) dengan waktu hidrolisis optimum selama 6 jam. Parameter yang diamati adalah TVB-N (Total Volatil Basa Nitrogen), uji kualitatif protein (biuret, ninhidrin, xanthoprotein dan Pb-sulfida), Kadar protein menggunakan BSA (Bovine Serum Albumin), dan aktivitas antioksidan menggunakan metode DPPH (1,1-difenil-2-pikrilhidrazil). Hasil menunjukkan bahwa hidrolisat protein ikan nike memiliki nilai TVB-N kurang dari 10 mgN/100g. Hidrolisat protein ikan nike positif mengandung protein pada uji kualitatif. Kadar protein yang dihasilkan sebesar 6,25%. Nilai aktivitas antioksidan tertinggi pada konsentrasi enzim 5% dengan waktu hidrolisis 6 jam memiliki nilai IC50 sebesar 193 ppm.

Abstract

Abstract. Infectious and parasitic diseases is one of various disease that the most common. Based on data from the World Health Organization (WHO) in 2011, infectious and parasitic diseases become the third largest cause of death in the world. One of the plants that have potential as an antimalarial is suruhan (Peperomia pellucida). The purpose of this study is to test antimalarial activity against Plasmodium falciparum and analysis photochemistry consituen from extract suruhan. Antimalarial activity test using Desjardin method. Extraction was done by using maceration wuth methanol as a solvent and fractionation using partition methods. Results of phytochemical screening revealed the presence of alkaloids, flavonoid, steroid, saponins and triterpenoid test results antimalarial activity fraction of n-hexane, ethyl acetate and water show IC50 values are 12.80, 2.90 and 10.74 mg/ mL respectively.Keyword: Peperomia pellucida, suruhan, antimalarial

Abstract

This study aims to determine the protein content and activity of nike fish (Awaous melanocephalus)  extract as an antioxidant. Nike fish (amphidromous goby larva) is an endemic fish of Gorontalo which is consumed as food and contains minerals, fats, carbohydrates, amino acids, and high protein. Nike fish extract uses a buffer with several components such as water, Tris HCl 0.1 M pH 8.3, NaCl 2 M, CaCl 0.01 M, 2-Mercaptoethanol 1%, and Triton X-100 0.5%. Based on qualitative analysis, Nike fish protein extract was positive through biuret, ninhydrin, xantoproteate, and Pb sulfide tests. Judging from the quantitative analysis, nike fish extract using the biuret method using UV VIS and TVB-N spectrophotometers obtained 47.60% and 0.098%, respectively. The antioxidant activity of nike fish extract using the DPPH method (1,1-diphenyl-2-pycryhydrazyl) was 20.29 mg AEAC/g (Ascorbic acid Equivalent Antioxidant Capacity). IC50 (Inhibitory Concentration) is 520 ppm at low levels but has potential as an antioxidant.

Abstract

Daun ketapang (Terminalia catappa L.) merupakan tumbuhan pesisir pantai yang memiliki kandungan senyawa metabolit sekunder yaitu flavonoid, alkaloid, steroid, saponin dan tanin yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder daun Terminalia catappa L. dan menguji aktivitas antioksidannya. Metode penelitian eksperimen laboratorium kuantitatif dan kualitatif meliputi ekstraksi, isolasi dan identifikasi senyawa metabolit sekunder menggunakan spektrofotometer FTIR, penetapan total kadar flavonoid dengan spektrofotometer UV-Vis, dan uji aktivitas antioksidan menggunakan metode DPPH. Hasil uji fitokimia ekstrak metanol mengandung senyawa flavonoid, steroid, saponin, tanin dan isolat menunjukkan hasil positif senyawa flavonoid. Hasil identifikasi menunjukkan keberadaan gugus fungsi flavonoid dengan adanya pita serapan seperti O-H, C-H aromatik, C-H alifatik, C=O aromatik, C=C aromatik dan C-O alkohol. Hasil pengukuran total kadar flavonoid sebesar 114,65 mgQE/g. Nilai aktivitas antioksidan ekstrak metanol sebesar 460,37 mg AEAC (Ascorbic Acid Equivalent Antioxidant Capacity) /g dan isolat diperoleh sebesar 19,62 mg AEAC/g.

Abstract

UMKM Puncak Sejahtera are micro, small and medium enterprises that produce virgin coconut oil (VCO) and palm sugar which are faced with production and marketing problems. The problems faced from the production aspect are that the product is still limited, does not have technological skills innovation, good product packaging and storage, does not have a machine with a large capacity and produces good quality. The problem from another aspect is not having extensive business management and marketing capabilities so that financially the results obtained are insufficient for daily needs. Even though the potential of coconut and palm sugar can be developed various products with a fairly high economic value. The community partnership program (PKM) aims to help and develop VCO and palm sugar businesses in UMKM Puncak Sejahtera. VCO manufacturing innovation training has been carried out with several methods and has been tested in the laboratory, assistance in processing technology for coconut and palm sugar-based product innovations, VCO training and palm sugar business management through packaging technology, design labeling techniques, storage time and marketing. Business license assistance for business development and online marketing is also carried out in this program. The methods used are the RRA (Rapid Rural Appresial) and PRA (Participatory Rural Appresial) approaches, including: 1) priority of problems to be addressed, 2) training and assistance based on problems in stages, 3) product improvements are carried out based on the results of laboratory tests. The result of this activity is an increase in understanding, skills of members of the UMKM Puncak Sejahtera group in Bukit Aren village in processing VCO and palm sugar with good quality and quantity of production. VCO products and cooking oil are certified for nutritional value in the laboratory. Product marketing is expanded online to make it easier to sell products.

Abstract

COVID-19 is a disease that caused a prolonged pandemic in many countries caused by the SARS-CoV-2 virus. This study aims to identify the antiviral potential of secondary metabolites in Gorontalo traditional medicinal plants, which are believed to have the ability to inhibit the main protease protein of this virus. The methods used in this research were molecular docking and molecular dynamic. The main protease proteins for SARS-CoV-2 used based on the homology modeling results were 3V3M and 7TE0. The results of the active compounds in the paxlovid drug were also compared to obtain accurate data comparisons. The validation of the docking method on the 3V3M protein using the natural ligand 0EN revealed an RMSD of 0.75 Å. The RMSD value for validating the 7TE0 protein and natural ligand 4WI was 1.65 Å. The best molecular docking results were obtained using physalin F with a binding affinity of −10.3 kcal/mol for the 3V3M protein and physalin J with a binding affinity of −8.9 kcal/mol for the 7TE0 protein. The outcomes of the molecular dynamic method on the best complexes were determined by examining the value of changes in system energy, changes in system temperature, changes in system pressure, RMSD, RMSF, and bond-free energy (ΔG) of the complex. The standard 0EN ligand had a ΔG of −26.53 kcal/mol, while the standard 4WI ligand had a ΔG of −47.16 kcal/mol. The ΔG of the 3V3M-physalin F and 3V3M-physalin J complexes were respectively −28.22 kcal/mol and −26.62 kcal/mol. The ΔG of the 7TE0-Vitexin 2”-O-gallate and 7TE0-physalin J complexes were found to be −28.08 kcal/mol and −26.62 kcal/mol, respectively. The ΔG produced in paxlovid with complexes 3V3M and 7TE0 was −19.38 kcal/mol and −25.44 kcal/mol, respectively. Physalin F, physalin J, and Vitexin 2”-O-gallate have great potential to become SARS-CoV-2 inhibitor agents. However, in terms of structural stability and binding-active residues, these three compounds do not outperform the active substance in paxlovid.

Abstract

AbstrakMakanan tradisional sudah mulai kurang dikenal dan bahkan ditinggalkan oleh generasi muda termasuk di Gorontalo. Mengantisipasinya, di Gorontalo telah dilaksanakan kebijakan pelestarian dan pengembangannya melalui mata pelajaran muatan lokal (mulok) ilmu gizi berbasis makanan tradisional Gorontalo (MTG) pada pendidikan dasar (SD,SMP) dan menengah (SMA/SMK). Kebaruan dalam penelitian ini karena meneliti tentang pembelajaran Ilmu Gizi terhadap sikap konsumsi makanan traditional Gorontalo. Tujuan penelitian ini adalah  menganalisis pengaruh pembelajaran Ilmu Gizi terhadap sikap konsumsi MTG pada siswa yang menerima mata pelajaran mulok dan tidak mulok. Metode penelitian dilakukan secara purposive ditentukan contoh SMP di kabupaten/kota yang punya kesamaan letak geografi dan tingkat akreditasi. Provinsi Gorontalo punya 6 daerah kabupaten/kota sehingga contoh sekolah berjumlah 24 SMP (12 sekolah mulok dan 12 tidak mulok) dan setiap sekolah diwakili oleh 13 contoh siswa. Penentuan contoh siswa ini secara stratified random sampling karena populasi terdiri dari siswa yang mendapat mulok dan tidak mulok.Total contoh siswa dalam penelitian ini ada 305. Hasil penelitian bahwa sikap konsumsi MTG meliputi suka dengan alasan karena penampilan, tekstur, aroma khas, cita rasa, menyehatkan dan mudah diperoleh pada siswa mulok berbeda nyata dengan tidak mulok (p0,05). Kesimpulan bahwa pembelajaran mulok ilmu gizi berbasis MTG berpengaruh pada perubahan sikap contoh siswa. Anticipating this, in Gorontalo a policy of preservation and development has been implemented through the subject of local content (mulok) of Gorontalo traditional food-based nutrition science (MTG) in primary (SD, SMP) and secondary (SMA / SMK) education. The novelty in this study is because it examines the learning of Nutrition Science towards the attitude of traditional Gorontalo food consumption. The purpose of this study was to analyze the effect of Nutrition Science learning on the attitude of MTG consumption in students who received mulok and non-mulok subjects. The research method was carried out purposively, determining the example of junior high schools in districts/cities that have similar geographical locations and accreditation levels. Gorontalo Province has 6 districts/cities so that the example schools are 24 junior high schools (12 mulok schools and 12 non-mulok) and each school is represented by 13 examples of students. The determination of this student example is stratified random sampling because the population consists of students who get mulok and not mulok. The total number of examples of students in the study was 305. The results of the study that the attitude of MTG consumption includes liking with the reason because the appearance, texture, distinctive aroma, taste, healthy and easy to obtain in mulok students are significantly different from not mulok (p0.05). The conclusion that mtg-based nutrition science mulok learning has an effect on changing the attitudes of student examples.Keywords: Nutrition science learning; Gorontalo traditional food; Consumption attitude.
