Abstract

Abstract

Panus lecomtei (Polyporales) is reported for the first time from Indonesia at the IPB University Campus Forest (IPBUCF). This species has similar macroscopic features to P. strigellus and P. neostrigosus. This study aimed to confirm the taxonomic position of P. lecomtei found at IPBUCF based on morphological and molecular evidence. The specimens were described by their macroscopic and microscopic characteristics. A phylogenetic approach was used with the internal transcribed spacer 1/2 sequence data. The combination of morphological and molecular analyses confirmed the taxonomic position of our specimens as P. lecomtei. The recorded species was distinguished by gregarious basidiomata, an infundibuliform resembling a funnel, violet during the immature stage to orangish brown in the mature pileus and ellipsoid basidiospore stages. Our specimens were distinguished from P. strigellus by having smaller basidiospores. It was difficult to separate P. lecomtei BO24428 from P. neostrigosus based on morphological characters. However, the BLAST results showed that specimen BO24428 had 99.56% similarity with P. lecomtei. In addition, our phylogenetic tree nested P. lecomtei BO24428 in the P. lecomtei clade. Morphological illustrations and a phylogenetic tree of P. lecomtei are provided. We suggest combining the morphological and molecular evidence in future studies of P. lecomtei from Indonesia

Abstract

The exploration of ectomycorrhizal fungi diversity around the campus could provide insights into the presence and distribution of these symbiotic fungi in the area. This study aimed to observe, identify, and provide information regarding ectomycorrhizal fungi diversity in the IPB University Campus Forest. The sampling sites represent various tree species including forests, parks, and green spaces. The exploration was done using opportunistic sampling method. The obtained basidiomata were examined for macromorphological and micromorphological examination to identify the ectomycorrhizal fungi. The identification results based on the morphological characteristics confirmed our specimens as Inocybe cf. squarrosolutea (Corner & E. Horak) Garrido and Suillus bovinus (L.) Roussel. The Inocybe was characterized by having small to medium-sized, pileus squamules, dry surface, light brown to yellowish cap, lamellae crowded, cylindrical or attenuated stipe towards the apex, clavate basidia, and hemispheric knobs basidiospores. The Suillus was distinguished by having a cap that is convex in shape with a slightly brownish yellow color and the surface texture of the cap is smooth, lamella in the form of pores that are quite large and angular, brownish yellow in color, yellow tubular stipe, clavate basidia, and oval spores. These two macrofungi are new records in the sampling area and I. cf. squarrosolutea could be the new information for Indonesia. Our finding provides valuable information for ecological studies and contributes to the additional data on Indonesian ectomycorrhizal fungi diversity.

Abstract

Mold are multicelular -microscopic fungi known as heterotrophic-cosmopolitan organism which exist almost at any place in the environment. Textiles, particularly those composed of natural organic such as jeans are easily colonized by mold. Till time, there is no report ever found regarding fungal of moldy jeans in Indonesia. This study aimed to isolate fungi from moldy jeans, identified, and described it. The result revealed that the isolate studied was Chaetomium globosum, by using several macroscopic and microscopic characters. However, since Chaetomium are known as species complex fungi, further observation is needed using molecular approach. Therefore, author consider that the isolate should be treated as Ch. globosum sensu lato for time being. Interestingly, Ch. globosum was frequently reported as endophytic fungi, which pose the potency to be used as biocontrol and bioinducer in agriculture. Further research is needed to test the isolate potency as plant pathogenic biocontrol and plant growth inducer.

Abstract

Clarkeinda trachodes is an interesting agaric and has been acknowledged as one of the poisonous mushrooms worldwide. This species has mentioned to be found in tropical regions of Asia, including Indonesia. However, no clear record has been found regarding the primary information and herbarium collection of this species in Indonesia. During our regular mushroom foraging at IPB University campus forest Indonesia, the basidiomata of Clarkeinda was encountered. The current study aimed to confirm the taxonomical position of our specimens based on morphological and molecular evidence. Fresh basidomata was observed on the macro- and micromorphological features. The molecular analysis and phylogenetic tree construction were done based on ITS 1/2 sequence. The combination of morphological and molecular evidence confirmed our specimen as C. trachodes. Morphologically, Clarkeinda trachodes BO24637 can be determined by a large basidiocarp of Agaricales, prominent pellicle on disc pileus, notable ring, reddish brown context and stipe when injured, and truncate apex of basidiospores. The BLAST result showed that our specimen has a high similarity to C. trachodes as the top hits. In addition, the phylogenetic tree constructed by ITS sequence committed specimen BO24637 as C. trachodes with 100% BS value. The current study resolves a clear and accessible document of C. trachodes in Indonesia. In addition, we provided the morphological and molecular information as well as herbarium collection of C. trachodes in Indonesia, which can be used for future study of this species.
